Addition of glucose to Saccharomyces cerevisiae inactivates the galactose transporter Gal2p and fructose-1,6-bisphosphatase (FBPase) by a mechanism called glucose- or catabolite-induced inactivation, which ultimately results in a degradation of both proteins. It is well established, however, that glucose induces internalization of Gal2p into the endocytotic pathway and its subsequent proteolysis in the vacuole, whereas FBPase is targeted to the 26 S proteasome for proteolysis under similar inactivation conditions. Here we report that two distinct proteolytic systems responsible for specific degradation of two conditionally short-lived protein targets, Gal2p and FBPase, utilize most (if not all) protein components of the same glucose sensing (signaling) pathway. Indeed, initiation of Gal2p and FBPase proteolysis appears to require rapid transport of those substrates of the Hxt transporters that are at least partially metabolized by hexokinase Hxk2p. Also, maltose transported via the maltose-specific transporter(s) generates an appropriate signal that culminates in the degradation of both proteins. In addition, Grr1p and Reg1p were found to play a role in transduction of the glucose signal for glucose-induced proteolysis of Gal2p and FBPase. Thus, one signaling pathway initiates two different proteolytic mechanisms of catabolite degradation, proteasomal proteolysis and endocytosis followed by lysosomal proteolysis.  相似文献

Comparative mineral and hormonal analyses were made on tissue culture derived truncated leaf syndrome and wild type oil palm seedlings. Mineral analysis confirmed that Boron, Zinc and chlorophyll levels were significantly lower in truncated leaf syndrome leaves than those of wild type. Hormonal analysis also revealed various cytokinin derivatives such as trans-zeatin, trans-zeatin riboside, trans-zeatin O-glucoside and trans-zeatin riboside 5??mono phosphate were significantly higher in truncated leaf syndrome leaves compared to wild type leaves. Brassinolide level was also significantly higher in truncated leaf syndrome leaves than those of the wild type. These observations suggest that the truncated leaf syndrome abnormality could be associated to high cytokinin and brassinosteroid production which affects the uptake of Boron and Zinc.  相似文献

AimTo evaluate the outcome of prostate cancer patients with initial PSA value >40 ng/ml.BackgroundThe outcome of prostate cancer patients with very high initial PSA value is not known and patients are frequently treated with palliative intent. We analyzed the outcome of radical combined hormonal treatment and radiotherapy in prostate cancer patients with initial PSA value >40 ng/ml.MethodsBetween January 2003 and December 2007 we treated, with curative intent, 56 patients with non-metastatic prostate cancer and initial PSA value >40 ng/ml. The treatment consisted of two months of neoadjuvant hormonal treatment (LHRH analog), radical radiotherapy (68–78 Gy, conformal technique) and an optional two-year adjuvant hormonal treatment.ResultsThe median time of follow up was 61 months. 5-Year overall survival was 90%. 5-Year biochemical disease free survival was 62%. T stage, Gleason score, PSA value, and radiotherapy dose did not significantly influence the outcome. Late genitourinal and gastrointestinal toxicity was acceptable.ConclusionRadical treatment in combination with hormonal treatment and radiotherapy can be recommended for this subgroup of prostate cancer patients with good performance status and life expectancy.  相似文献

Extremely miniaturized longipedes insects (body length c. 0.3 mm) embedded in two pieces of Cretaceous amber from Myanmar are described and interpreted. Using inverted fluorescence and light microscopy for detailed analysis of microstructures, the inclusions were identified as primary larvae of the beetle family Ripiphoridae, subfamily Ripidiinae. While the structure of thoracic and abdominal segments including appendages corresponds well with the groundplan known in recent members of Ripidiinae, a curved prosternal ridge with prominent spines (each c. 5 μm), the reduced condition of stemmata and antennae and the lack of sharp mandibles are unique features within the entire family, apparently apomorphies of the longipedes larvae. A sinuate prosternal edge with a dense row of spines (prosternoctenidium) might be homologous with ‘head ctenidia’ in some previously described miniaturized conicocephalate larvae, but further investigation is needed. The morphological differences between the head of longipedes larvae and extant Ripidiinae are interpreted as adaptations to different groups of hosts and life strategies. Palaeoethology of the longipedes larvae is briefly discussed. In addition, the systematic placement of conicocephalate larvae from Canadian, Myanmar and Russian Cretaceous ambers, already interpreted by various authors as primary instars within Coleopterida (assigned to either Strepsiptera or to the coleopteran Tenebrionoidea: Ripiphoridae), is discussed.  相似文献

Intrahepatic biliary lesions were observed in two of 12 lambs, seven of 12 yearlings and 10 of 25 adult impala (Aepyceros melampus) surveyed in the Kruger National Park, Republic of South Africa. Lesions were associated with the nematode Cooperioides hepaticae, a trichostrongyloid parasite that inhabits the bile ducts of impala, and ranged from a mild chronic-eosinophilic cholangitis to foci of florid hyperplastic cholangitis with duct ectasia. The latter almost always contained viable worms and, after the worms died, the lesions appeared as foreign-body granulomas. Infection was acquired early in life; severe lesions were seen most frequently in yearlings. Adults were less severely infected, which suggested an acquired immunity. Although the incidence of infection was high, cooperiiasis did not appear to be a serious herd-health problem at the time of this study.  相似文献

Insect and veteran trees are important parts of ecosystems and are usually included in ecological studies of forest management. The loss of veteran trees in woodlands and open landscapes would lead to the loss of saproxylic organisms—an important part of biodiversity. Hence, the persistence of many specialized insects depends on the presence of veteran trees scattered in woodlands (e.g. ancient wood pastures, game parks or protected areas), cities, towns and villages (e.g. avenues, parks or chateau parks) or open landscapes (e.g. fishpond dams, solitary trees or fruit orchards). Veteran tree conditions could be fairly well described by three components—diameter, age and microhabitats present. The problem is that diameter belongs to the most studied characteristics, while age and microhabitats, which can be quite complicated to measure, are much less studied. This paper illustrates that, due to this unbalanced use of indicators of veteran-tree conditions, we are still missing some important information on saproxylic species ecology—and sometimes only large trees might be studied, rather than real veterans. Although we already know that veteran trees are essential habitat for a range of saproxylic organisms, there are still gaps in our knowledge of the specific conditions that veteran trees provide. It is vital that these are quantified and understood so that this information can be used to conserve veteran trees and their associated species.  相似文献

Mountain ecosystems are considered to be sensitive to global change and human disturbance. Our retrospective analysis of archival aerial imagery showed dynamics of arctic-alpine tundra vegetation in the Krkono?e Mountains, Czech Republic; image classification revealed a change in land cover classes in 44% of the study area over the past eighty years. This particular ecosystem holds many rare features, such as high numbers of endemic, glacial relict and rare species as well as relict geomorphological components such as sorted patterned ground. Our study revealed an accelerating expansion of the native and planted shrub, Pinus mugo, from 30.6% of the study area in 1936 to 48.6% in 2018, mostly at the expense of grasslands that decreased from 59.3% in 1936 to 44.2% in 2018. Shrub expansion represents a threat to relict periglacial landforms, covering 8% of the sorted patterned ground in 1936 and 26.5% in 2018. Shrub encroachment was shown to be due to artificial planting of the pine in the past as well as the cessation of former farming (mowing and cattle grazing) and, most probably, also by global change. Both dwarf pine stands and tundra grasslands hold high conservation value (Natura 2000 habitats); a balance between different nature protection interests must, therefore, be ensured. Detailed spatio-temporally, explicit outputs of the remote sensing analysis can serve as a baseline for nature conservation in order to prepare corresponding management plans.
