Using a combination of gas-exchange and chlorophyll fluorescence measurements, low apparent CO2/O2 specificity factors (1300 mol mol?1) were estimated for the leaves of two deciduous tree species (Fagus sylvatica and Castanea sativa). These low values contrasted with those estimated for two herbaceous species and were ascribed to a drop in the CO2 mole fraction between the intercellular airspace (Ci) and the catalytic site of Rubisco (Cc) due to internal resistances to CO2 transfer. Cc. was calculated assuming a specificity of Rubisco value of 2560 mol mol?1. The drop between Ci and Cc was used to calculate the internal conductance for CO2 (gi). A good correlation between mean values of net CO2 assimilation rate (A) and gi was observed within a set of data obtained using 13 woody plant species, including our own data. We report that the relative limitation of A, which can be ascribed to internal resistances to CO2 transfer, was 24–30%. High internal resistances to CO2 transfer may explain the low apparent maximal rates of carboxylation and electron transport of some woody plant species calculated from A/Ci curves.  相似文献

In Europe and North America the western flower thrips,Frankliniella occidentalis, is an important pest in various greenhouse crops, such as sweet pepper and cucumber. Two species of predatory mite are commercially applied for biological control of this pest:Amblyseius cucumeris andA. barkeri. Thrips control is generally successful from March onwards. During winter, however, thrips control by these predatory mites is less effective. An important reason for this is that the commercially applied strains of both mite species enter reproductive diapause under short-day photoperiods, whereas the western flower thrips does not enter diapause. In this paper we report on selection experiments for non-diapause in strains of both mite species, aimed at obtaining predators that do not enter diapause under light- and temperature conditions prevailing in winter. Additional experiments were done to estimate the potential of the selected lines as control agents ofF. occidentalis. Selection for non-diapause proved highly successful in both predatory mite species. In a New Zealand strain ofA. cucumeris diapause incidence decreased from 41% to 0% in about ten generations; in a Dutch strain ofA. barkeri diapause incidence decreased from 67% to 0% in about six generations. Furthermore, selection for non-diapause had no influence on predator performance, measured as predation rate and oviposition rate on a diet of first instar thirps larvae. Rates of predation and oviposition were the same for selected and unselected lines in both species; rates of predation and oviposition were higher forA. cucumeris than forA. barkeri. After 18 months under non-diapause conditions, no less than 92% of a sample of the selected non-diapause line ofA. cucumeris did not enter diapause when tested under diapause-inducing conditions. This indicates that ‘non-diapause’ is a stable trait in these predatory mites. Finally, a small-scale greenhouse experiment in a sweet pepper crop showed that the selected non-diapause line ofA. cucumeris established successfully under diapause-inducing short-day conditions.  相似文献

甜味蛋白(thaumatin)是世界上已知最甜的物质,具有很大的应用前景.Thaumatin的基因核苷酸和蛋白质氨基酸序列都已测定.晶体分析表明它具有高稳定的四级结构.在味蕾小孔中发现了介导thaumatin发生作用的物质.Thaumatin自身的功能仍不清楚.在多种生物中发现了thaumatin类似蛋白质,具有不同的生物活性.用基因工程手段实现了thaumatin在多种原核和真核生物中的表达,但迄今仍未得到理想的基因工程产品.  相似文献

Callus formation from stem internodes of the apple rootstocks M.9, M.25, M.26, M.27 and the cherry rootstock Colt, and from pith of Nicotiana tabacum cv. Wisconsin 38 was initiated on 4 -naphthaleneacetic acid (NAA)-based media (2.0–10.0 mg1-1). Transfer of callus to corresponding media lacking NAA allowed regeneration of shoots from callus of M.25, M.27, Colt and tobacco but not of M.9 and M.26. With M.25 phloroglucinol (PG) depressed regeneration from 30 to 10% and no regeneration was observed in cultures grown in the presence of casein hydrolysate (CH) and glutathione (GSH).Organogenesis was also obtained from leaf discs of M.27 employing 6-benzyl-aminopurine (BAP) at 5.0mg 1-1 and 2,4-dichlorophenoxy acetic acid (2,4-D) at 0.1 mg1-1. The regenerated shoots have been multiplied and rooted.Organogenesis also occurred in M.26 from small (1–2mm), green, compact embryoid-like structures derived from stem and leaf surfaces of excised axillary shoots. These structures differentiated into shoots at a low frequency (< 1%) on media containing BAP (1.0mg1-1) and indole-3-butyric acid (IBA) (0.1 mg1-1) and could also be micropropagated by subsequent axillary shoot proliferation.  相似文献

为评价新入侵害虫草地贪夜蛾对我国不同品种玉米的为害潜能,本文采用组建实验种群生命表的方法研究了草地贪夜蛾对3种甜质型玉米(穗甜1号、正甜68和华金甜1号)以及3种糯质型玉米(京科糯2000、广黑甜糯和广糯1号)的生存适合度,分析了草地贪夜蛾生物学指标与玉米叶片主要营养物质及粗纤维含量间的相关性。结果表明: 取食甜质型玉米的草地贪夜蛾的幼虫存活率、蛹重、雌成虫产卵量均显著高于取食糯质型玉米;取食甜质型玉米草地贪夜蛾的内禀增长率介于0.1566~0.1843,净生殖力介于187.97~353.35,也均高于取食糯质型玉米(内禀增长率介于0.0998~0.1465,净生殖力介于25.89~95.34),表明取食甜质型玉米的草地贪夜蛾具有更高的种群增长能力。甜质型玉米叶片中主要营养物质维生素C、淀粉、可溶性糖、蛋白质、脂肪、总氨基酸及粗纤维的含量普遍高于糯质型玉米,草地贪夜蛾的净生殖力与维生素C、淀粉、可溶性糖、蛋白质及粗纤维的含量均存在显著正相关关系。草地贪夜蛾在甜质型玉米上具有更高的生存适合度。  相似文献

The efficiency of some inducer resistance chemicals (IRCs) like bion, chitosan, humic acid and salicylic acid as well as the fungicides like Folu-Gold, Galben Copper, Previcure-N and Redomil Gold Mancozeb on management of sweet melon downy mildew, caused by Pseudoperonospora cubensis was evaluated in vitro and in vivo. Also, the efficiency of the alternation between the sprayed two fungicides and IRCs on management of the disease and the produced fruit yield and its total soluble solids (TSS) under field conditions were assessed. The inhibitory effect of the IRCs and the tested fungicides on sporangial germination of P. cubensis resulted in a significant reduction in the germinated sporangia. In addition, IRCs were less effective than the fungicides. Disease management revealed the same trend of the in vitro experiment when they sprayed fungicides on sweet melon plants artificially inoculated with the sporangia of the causal fungus under greenhouse conditions. Furthermore, under field conditions, spraying sweet melon plants with the two tested fungicides was the most efficient in decreasing the disease and increasing fruit yield and its TSS, to somewhat, followed by the alternation between them and the tested IRCs. In addition, IRCs treatments showed the lowest efficiency in this regard.  相似文献

The sweet protein brazzein, a member of the Csβα fold family, contains four disulfide bonds that lend a high degree of thermal and pH stability to its structure. Nevertheless, a variable temperature study has revealed that the protein undergoes a local, reversible conformational change between 37 and 3°C with a midpoint about 27°C that changes the orientations and side‐chain hydrogen bond partners of Tyr8 and Tyr11. To test the functional significance of this effect, we used NMR saturation transfer to investigate the interaction between brazzein and the amino terminal domain of the sweet receptor subunit T1R2; the results showed a stronger interaction at 7°C than at 37°C. Thus the low temperature conformation, which alters the orientations of two loops known to be critical for the sweetness of brazzein, may represent the bound state of brazzein in the complex with the human sweet receptor. Seed potato crops are currently sprayed weekly with mineral oil to prevent transmission of the Potato virus Y (PVY; Potyviridae: Potyvirus), one of the most prevalent and important non‐persistent viruses affecting potato production. In spite of its wide usage as inhibitor of virus transmission, the mode of action for mineral oil is poorly known. The objective of this study was to quantify the effect of dosage and time from application of mineral oil on the inhibition of PVY acquisition. The bird cherry‐oat aphid, Rhopalosiphum padi (L.) (Hemiptera: Aphididae), known as vector of PVY, was used in all the experiments. The results indicated that mineral oil efficiently decreased PVY acquisition by 75 and 70% 1 day after application of 5 and 10 l ha?1, respectively. The inhibition effect decreased with time from application; mineral oil inhibits acquisition for less than 4 days at 5 l ha?1 and between 8 and 12 days at 10 l ha?1. As mineral oil was detected in the body of fewer aphids when they fed on plants 1 day after oil application, a change in the aphid probing behaviour on mineral oil‐treated plants was deduced. These results support the hypothesis that mineral oil physically inhibits the binding of the virus at the tip of the stylets.  相似文献

Drosophila suzukii, Matsumura, is a relatively new pest in the United States attacking a variety of fruit crops. Studies were conducted to develop a standardized, economical trap for monitoring. Laboratory bioassays found that flies were attracted to dark colours ranging from red to black. Similarly, fly catches in 237‐ml plastic ‘spice’ jars with ten 0.48‐cm holes and baited with apple cider vinegar were significantly higher in jars with red or black than white caps. The use of an alternating set of three, horizontal red, black and red bands (‘Zorro’ trap) significantly increased fly catches compared with the use of all‐red or all‐black strips. This increase was associated with a significantly higher proportion of flies first landing on the side near the openings of the trap instead of on the cap with the ‘Zorro’ trap compared with the other traps. Laboratory data were used to develop a predictive model to define total fly capture as a function of trap colour/colour pattern, cumulative area of entry holes and the length of the trapping portion of the trap. Total fly catches by the ‘Zorro’ trap were compared with other red and clear plastic traps in five field trials conducted in several cultivated and uncultivated sites. Comparisons included a commercial red‐capped 200‐ml trap with two 0.63‐cm holes, an all‐red spice jar with ten 0.48‐cm holes and clear and red 473‐ml and clear 946‐ml plastic cups with six or ten 0.48‐ or 0.63‐cm holes. The model was successfully validated, suggesting that performance of cup traps can be predicted based on a few characteristics. The current ‘Zorro’ trap did not catch most of the flies among trap designs, but showed some advantages, including the durability and potential to recycle the plastic, small size, commercial availability and its greater selectivity for D. suzukii than the other traps tested.  相似文献
