The goal of this study is to understand the evolution relationship of the members of the B-hordein gene family in hull-less barley by analysis of their structure and to explore their utility in grain quality improvement. Six copies of the B-hordein gene (Hn1-Hn3, Hn7-Hn9) were cloned from six hull-less barley cultivars collected from Qinghai-Tibet Plateau and molecularly characterized. Comparison of their predicted polypeptide sequences with the published data suggested that they all share the same basic protein structures. In addition, we found that the C-terminal end sequences of all B-hordeins shared a similar feature. In the six clones and the other three published genes (Hn4, Hn5, and Hn6) from hull-less barley, Hn2 and Hn7 contained the identical C-terminal end sequence DIMPVDFWH. Hn3, Hn4, Hn5, Hn8 and Hn9 also shared the common sequence DIMPPDFWH, which was similar to that of a B-hordein reported previously. Both Hn1 and Hn6 exhibited differences in their C-terminal end sequences, and they clustered into different subgroups. The B-hordeins with identical C-terminal end sequences were clustered into the same subgroup, so we believe that B-hordein gene subfamilies possibly can be classified on the basis of the conserved C-terminal end sequences of predicted polypeptide. Phylogenetic analysis also indicated that there is a relatively weak identity between our predicted B-hordeins and those reported from H. chilense and H. brevisubulatum. All of our nine predicted B-hordeins were clustered together and other B-hordeins formed another cluster. The possible use of these genes in relation to barley quality is discussed. 对产自湖北武当山的柴黄姜(Dioscorea nipponica subsp.rosthornii Prain et Burkill)的花部特征和访花昆虫种类及访花行为、访花频率和携粉量进行了研究,并对柴黄姜的传粉方式、结实和种子萌发状况进行了检测。结果表明,柴黄姜具有虫媒花的典型花部特征,共观测到访花昆虫4目34种,其中传粉昆虫6种,隧蜂属(Halictus sp.)和地蜂属(Andrena sp.)的2种昆虫是柴黄姜的主要传粉昆虫。访花昆虫对雄株的访花高峰时段为11:00至14:00,对雌花没有明显的访花高峰时段。在自然条件下,柴黄姜的座果率为18.86%,结实率为7.12%,种子萌发率为64.76%。在武当山,柴黄姜种群扩展仍以有性繁殖为主,无性繁殖为重要补充。  相似文献

丙酮酸磷酸双激酶(pyruvate phosphate dikinase, PPDK; EC 2.7.9.1)能够可逆催化磷酸烯醇式丙酮酸(phosphoenolpyruvate, PEP)、单磷酸腺苷(adenosine monophosphate, AMP)和焦磷酸盐(pyrophosphate, PPi)生成三磷酸腺苷(adenosine triphosphate, ATP)、无机磷酸盐(orthophosphate, Pi)和丙酮酸(pyruvate).以热玫瑰小双孢菌基因组DNA为模板,PCR扩增得到了编码PPDK的基因,将此基因片段插入表达载体pET24a (+),在大肠杆菌中表达C端融合His-Tag的重组PPDK.与我们先前表达的N端融合His-Tag的PPDK相比,酶的活性提高了20倍,提示该酶的N端对活性十分重要.重组PPDK单体分子量为98 kD.经过镍亲和层析和超滤后,重组PPDK基本达到电泳纯.重组PPDK与荧光素酶偶联能够形成1个ATP-AMP循环反应,在该循环反应中,荧光素酶催化ATP生成的AMP和PPi能够被PPDK重新转化成ATP,产生一个持续稳定的信号.  相似文献

The effects of CO2 concentration (C a) on growth, photosynthesis, and the activity of enzymes associated with the translocation and assimilation of CO2 were studied in sugar beet (Beta vulgaris L. subsp. saccharifera, cv. Ramonskaya) plants. The plants were grown in controlled-climate chamber to the stage of 3–4 leaves and then used in experiments. Experimental plants were exposed in boxes to doubled C a (700 µl/l, 2C plants), whereas control plants were kept in a chamber with ambient atmosphere (350 µl/l, 1C plants). As compared with 1C plants, in 3 and 8 days, the leaf area of 2C plants increased by 14 and 26%, respectively. The rate of their photosynthesis (P n) measured in 3, 6, and 8 days increased by 85, 47, and 52%, respectively, whereas in normal air, the values of P n in 2C plants were by 12, 19, and 15% lower than in 1C plants. After 8-day growth, the content of soluble carbohydrates in the leaves of 2C plants attained 7.2%, being by 80% greater than in 1C plants; the content of starch did not exceed 3%. The total content of chlorophylls a and b in the leaves of 2C plants was by 14% greater than in 1C plants, but their ratio was essentially the same. The level of protein in 2C plants was by 13.4% lower than in 1C plants. The activity and content of Rubisco in 1C and 2C plants were similar. As compared with 1C plants, in 2C plants the activity of soluble carbonic anhydrase (sCA) was lower by 34% in 3 days and by 18% in 8 days; the activity of carbonic anhydrase of membrane preparations (mCA), was lower by 24 and 77%, respectively. Catalase activity in 2C plants became by 8% lower than in 1C plants only after 8 days. Changes in the activity of peroxidase, a component of the NADPH oxidase signaling pathway, in potato cells were studied. This activity increased sharply during ring rot pathogenesis. Two mechanisms of peroxidase activation were distinguished. One of them was the enzyme de novo synthesis; it was characteristic of the potato cultivar susceptible to the pathogen. Another mechanism characteristic of the resistant cultivar included not only the enzyme synthesis but also the activation of preexisting enzyme molecules. Bacterial infection and exopolysaccharides secreted by the pathogen induced changes in the pattern of intra- and extracellular peroxidases of the susceptible cultivar. No changes were noted in the peroxidase patterns of the resistant cultivar. A sharp activation of the extracellular peroxidase of R f 15 occurred in the infected or exopolysaccharide-treated cells of the resistant cultivar.  相似文献

The culture liquid of Luteococcus japonicus subsp. casei was found to be able to reactivate cells of this bacterium inactivated by UV irradiation or heat shock. The antistress activity of the culture liquid was due to the presence of an extracellular exometabolite of a protein nature with a molecular mass of more than 10 kDa. When the bacterium was grown in a nutrient broth or glucose-containing mineral medium, the antistress protein was secreted by cells in the logarithmic growth phase. The reactivating effect of the antistress protein was inversely proportional to the survival rate of stressed cells.  相似文献

嗜水气单胞菌为该病的条件致病菌。在适宜的条件下 ,该菌可通过体表伤口进入鳖体引起疾病。此病以体表腐皮与赤斑 ,有的中华鳖背部有明显的乳白色小点和出血性败血症为主要病理学表现 ,以各器官功能衰竭为致病原因。解剖后 ,肝脏病变呈污泥色 ,腐烂液化 ,肾呈青紫色、肿大 ,肠内无食物 ,呈灰白色 ,部分肠壁有出血点 ,脾脏明显肿大。经分离鉴定 ,生理生化特性分析 ,结果为典型的嗜水气单胞菌嗜水亚种 (Aeromonashydrophilasubsp .hydrophila)。  相似文献

1. Trichome‐producing (hairy) and trichomeless (glabrous) plants of Arabidopsis halleri subsp. gemmifera were investigated to test whether plant resistance to herbivory depends on the plants' phenotypes and/or the phenotypes of neighbouring plants (associational effects). 2. A common garden experiment was conducted in which the relative frequency of hairy and glabrous plants was manipulated. Two species of leaf‐chewing insects (larvae of a white butterfly and a cabbage sawfly) were found less often on hairy plants than on glabrous plants. By contrast, the numbers of aphids and flea beetles did not differ significantly between hairy and glabrous plants. For none of these insects did abundance depend on the frequency of the two plant morphs. 3. A field survey was conducted in two natural populations of A. halleri. In the first population, a species of white butterfly was the dominant herbivore, and hairy plants incurred less leaf damage than glabrous plants across 2 years. By contrast, in the other population, where flea beetles were dominant, there were no consistent differences in leaf damage between the two types of plants. In neither of the two populations was any evidence found of associational effects. 4. This study did not provide any conclusive evidence of associational effects of anti‐herbivore resistance, but it was discovered that trichomes can confer resistance to certain herbivores. Given the results of previous work by the authors on associational effects against a flightless leaf beetle, such associational effects of the trichome dimorphism of A. halleri were herbivore‐specific.  相似文献
