利用平板分离法从药用植物益母草(Leonurus heterophyllus Sweet)中分离到15株内生真菌,来源于根、茎、叶分别有2,6,7株。经形态学观察和染色等方法,初步鉴定了益母草内生真菌有3个属,曲霉属(Aspergillus sp.)2株、头孢霉属(Cephalosporiumsp.)3株和囊孢霉属(Capsule sp.)10株。益母草内生真菌生物多样性较为单一,但不同部位内生菌的数量、种类及分布存在明显差异。  相似文献

为探索内生真菌与广藿香互作间对宿主活性成分形成机制的影响,该研究以成分差异较大的牌香和湛香为对象,采用传统形态学方法对所获菌株归类,通过真菌通用引物ITS1/ITS4扩增菌株rDNA-ITS序列,鉴定其分类地位并研究其多样性。结果表明:(1)用PDA和LBA培养基对苗期、分枝期和成株期广藿香茎叶组织块进行内生真菌分离,共获得3 070株菌株,其中牌香(PX)分离出1 624株,鉴定出1 319株,分属于36属;湛香(ZX)分离出1 446株,鉴定出994株,分属于33属。牌香分离出7种特有内生真菌,分别为香柱菌(Epichloe typhina)、盘长孢状刺盘孢菌(Colletotrichum gloeosporioides)、座腔孢菌(Botryosphaeria sp.)、丝核菌(Rhizoctonia sp.)及截盘多毛孢菌(Truncatella sp.),并首次分离到疫霉菌(Phytophthora sp.)和指疫霉菌(Sclerophthora sp.),这2种菌属于卵菌门内生菌。湛香分离出拟青霉菌(Paecilomyces sp.)和尾孢菌(Cercospora sp.)...  相似文献

采用显色反应、薄层层析色谱和紫外吸收光谱法,对甘蔗叶内16株内生真菌的代谢产物进行黄酮类化合物检测。结果共筛选到3株能够产黄酮类化合物的内生真菌(GZ-1、GZ-4和GZ-5)。依据真菌的形态特征及ITS序列分析,鉴定结果表明菌株GZ-1、GZ-5为棘孢曲霉(Aspergillus aculeatus),菌株GZ-4为黑曲霉(Aspergillus niger)。  相似文献

从云南程海湖分离到473株淡水真菌,用滤纸片法测定其抗菌活性,活性菌株经显微形态观察进行分类鉴定。结果显示有290株菌对至少1种病原菌具有抗菌活性,占总菌株数的61.31%;高抗菌活性菌株59株(抑菌圈直径≥15 mm),占总菌株的12.47%。将具有抗菌活性的菌株鉴定为46个属,优势属为青霉属(Penicillium)、曲霉属(Aspergillus)和枝孢属(Cladosporium)。  相似文献

为研究夹竹桃(Nerium oleander)内生真菌的多样性并评价其次生代谢产物的活性,该研究对广西夹竹桃(Nerium oleander)的内生真菌进行分离纯化,采用形态学和ITS序列分析结合的方法进行鉴定,以5种指示菌(其中有3种弧菌)对内生真菌提取物进行抑菌活性筛选。结果表明:(1)从广西夹竹桃中共得到19株内生真菌,这19株内生真菌都属于子囊菌门,涵盖5个目7个属,包括炭疽菌属(Colletotrichum)、球座菌属(Guignardia)、叶点霉属(Phyllosticta)、新壳梭孢属(Neofusicoccum)、曲霉属(Aspergillus)、隔孢壳科新属(Nothophoma)和间座壳属(Diaporthe),优势属为炭疽菌属(分离率为36.85%)和球座菌属(分离率为21.05%),其中炭疽菌属主要分布于茎,球座菌属全部来源于叶。(2)jing-117(Neofusicoccum sp.)和ye-130(Guignardia sp.)对坎氏弧菌有较为特异的抑菌效果,ye-136(Aspergillus sp.)能同时抑制蜡样芽孢杆菌和坎氏弧菌,ye-135(A...  相似文献

从黄山地区红豆杉中分离得到107株内生真菌,利用薄层层析(TLC)方法对107株内生真菌的发酵代谢物进行了初筛,首次筛出8株可产紫杉醇或其类似物的菌株。再通过高效液相色谱(HPLC)对其作了进一步分析,发现有1株内生真菌菌株发酵代谢物的吸收峰与紫杉醇标准品吸收峰保留时间一致。同时结合以中国仓鼠卵巢CHO细胞株作为肿瘤细胞模型,用Resazurin法检测生长抑制率,对经筛选出的1株内生真菌次生代谢产物进行体外抗肿瘤活性试验,该菌株的代谢物对CHO细胞的抑制率高达71.28%。通过对该菌株的显微形态观察,从菌丝、孢子的形态和产孢子的特征等初步判定HQ-24是曲霉(Aspergillus sp.)。  相似文献

从三尖杉,南方红豆杉及香榧中分离出172株内生真菌,对其进行抗菌活性检测,结果表明共90株内生真菌对一种或多种植物病原真菌,如红色面孢霉(Neurospora sp.),木霉(Trichoderma sp.),镰刀菌(Fusarium sp.)等有抑制作用,来自三尖杉、南方红豆杉和香榧的抗菌活性菌株比例分别为40%,54.2%及57.1%。其中平板抑菌圈直径大于15mm的高抗菌株有35株。按Ainsworth等鉴定系统和方法进行鉴定,具有抗菌活性的内生真菌主要分布于心青霉属、镰孢菌属等18个属中。  相似文献

从植物黄花夹竹桃 (Thevetiaperuviana)的根、茎、叶、果实中分离出内生真菌 10 1株。以鼠伤寒沙门氏菌 (Salmonellatyphimurium)、肺炎链球菌 (Streptococcuspneumoniae)、乙型溶血链球菌 (Streptococcushemolyticus)、金黄色葡萄球菌 (Staphyllococcusaureus) 4种人类病原菌为指示菌 ,对其进行抑菌活性筛选 ,结果有 17株内生真菌对 1株或 1株以上人类病原菌有抑菌活性 ,其中平板抑菌圈直径大于 2 0mm的菌株有9株。具有抗菌活性的内生真菌分别来自木霉属 (Trichoderma)、曲霉属 (Aspergillus)等 9个属。  相似文献

水稻秸秆还田时间对土壤真菌群落结构的影响   总被引:7,自引:0,他引:7  
为揭示水稻秸秆还田对土壤真菌群落结构的长期影响,采用荧光定量PCR和PCR-DGGE技术分析了秸秆还田90,180,270 d和360 d的土壤真菌基因丰度和群落结构组成演变趋势,并利用冗余分析(RDA)研究土壤真菌群落结构变化与环境因子的关系。结果表明:随着秸秆还田时间的增加,土壤真菌群体数量和多样性指数(H、R和E)显著增加,在360 d时达到最高。对DGGE图谱的特征条带进行胶回收、测序,系统进化分析表明,土壤真菌主要种群包括:接合菌(Zygomycete sp.)、盐腐霉菌(Pythium salinum)、肉盘菌(Uncultured Sarcosomataceae)、牛粪盘菌(Ascobolus stercorarius)、大链壶菌(Lagenidium giganteum)、青霉菌(Penicillium sp.)、曲霉属真菌(Aspergillus sp.)和疏绵状丝孢菌(Thermomyces lanuginosus)、灰绿曲霉菌(Aspergillus glaucus)、禾谷多粘菌(Polymyxa graminis)和枝顶孢霉菌(Acremonium sp.),其中青霉菌(Penicillium sp.)、曲霉属真菌(Aspergillus sp.)和枝顶孢霉菌(Acremonium sp.)具有纤维素降解能力,而枝顶孢霉菌(Acremonium sp.)在90 d时成为新的优势菌群。RDA分析表明,90 d和180 d秸秆还田与对照土壤的真菌群落结构较为类似,270 d和360 d的秸秆还田与对照土壤的真菌群落结构发生了明显变化。土壤有机碳、pH和速效磷是引起土壤真菌群落结构及多样性变异的主要因素。  相似文献

【目的】从11份南海海洋沉积物中分离耐盐真菌,并对其物种多样性及产酶活性进行研究。【方法】利用平板涂布法分离耐盐真菌,基于形态学和ITS序列的系统进化研究耐盐真菌多样性;利用6种筛选培养基对耐盐真菌进行产酶活性筛选。【结果】分离得到1689株耐盐真菌,共41个形态种。形态学和ITS序列分析表明,这些真菌归于15个属,其中曲霉属(Aspergillus)和青霉属(Penicillium)为优势菌群。对已测序的41株耐盐真菌的产酶活性研究表明,8株产纤维素酶,9株产淀粉酶,5株产复合酶,16株产蛋白酶,3株产脂肪酶,未发现产壳聚糖酶的菌株,其中Acrodontium sp.8m和Aspergillus sp.86b产复合酶的活性相对较高,而Penicillium sp.41m产蛋白酶的活性相对较高。【结论】南海局部海洋沉积物中耐盐真菌丰富,多数菌株具有产酶活性。  相似文献

A new species ofPhaeosphaeria is described from mangroves in South Africa. InPhaeosphaeria capensis ascospores are 29–36×7–8 μm, cylindric-fusiform or somewhat clavate, widest at the middle, narrow at the base and rounded at the apex, (6-)7-septate, light brown or brown, minutely verruculose and surrounded by a spreading, mucilaginous sheath. Chaetosphaeria tortuosa is described as the newly discovered teleomorph of Menispora tortuosa, based on specimens from Canada and the Czech Republic, and single spore isolations from both morphs. The fungus produces superficial, more or less globose, papillate, dark brown to black smooth perithecia (200–)220–250 × (220–)230–260 μm. The asci are unitunicate, 8-spored, cylindrical-fusiform, (110–)120–133(–145) × 12–14 with a distinct apical, nonamyloid annulus 1–1.5 μm high, 3.5–4 μm wide. The ascospores are fusiform, 19–24 × 5–6 μm, hyaline, 3-septate, smooth, and 2-seriate in the ascus. The morphology of the teleomorph and anamorph are similar to that of C. ovoidea (anamorph: M. glauca), differing in dimensions of asci and ascospores, and in the disposition and morphology of the phialides of the anamorphs. The generic concept and phylogeny of Menispora is briefly discussed, and a key to the 11 species currently accepted in the genus is provided.  相似文献

During a survey of bryophilous fungi from boreal and montane habitats in central Alberta, a hitherto undescribed species of Cladophialophora was recovered from Polytrichum juniperinum, Aulacomnium palustre, and Sphagnum fuscum. On potato dextrose agar (PDA) colonies grew slowly, attaining a diameter of 25 mm after 30 d, were dark grey, velvety, radially sulcate, and convolute and cracked at the centre. Micronematous conidiophores gave rise to branched chains of small (1–2 × 8–22 μm), cylindrical to fusiform conidia with truncate, swollen scars at each end. Phylogenies built on the ITS and ribosomal SSU regions indicate the isolates form a monophyletic clade within the family Herpotrichiellaceae (Chaetothyriales) that is composed of two geographically based groups, each with 99 % within-group sequence similarity and 97–98 % between-group sequence similarity. A teleomorph has not been found but would likely be similar to species of Capronia. In vitro inoculation of the isolates onto axenically grown P. juniperinum produced no discernible host symptoms, and host penetration could not be detected using light microscopy. The production of polyphenol oxidases by the fungus and the role of other Cladophialophora species as latent endophytes and saprobes suggest that a potential role for the fungus is the degradation of the polyphenol-rich cell walls of mosses. A dichotomous key to species of the genus Cladophialophora is provided.  相似文献

Mesophilic fungi isolated from organic fertilizer compost samples accounted for 70.94% of the total fungal count, while thermophilic and thermotolerant fungi constituted 29.05% of that count. Eight mesophilic fungal species, namelyAspergillus niger, Monilia sitophila, Paecilomyces divaricata, Penicillium chrysogenum, P. fellutanum, Scopulariopsis brevicaulis, S. brumptii andZygorhynchus japonicus; two thermophilic fungiHumicola grisea andOidiodendron flavum and three thermotolerant speciesAspergillus fumigatus, Thermomyces lanuginosus andZygorhynchus vuilleminii were isolated during the study. Most of the tested fungi showed a proteolytic activity and liquified gelatin in the test tube method and in cup plates. The thermophilic fungusO. flavum was the most potent proteolytic fungus. The comparative fibrinolytic assay revealed the following sequence in the ability of the tested fungi to hydrolyse fibrin:O. fiavum>S. brevicaulis>H. grisea>A. fumigatus>T. lanuginosus.  相似文献

【背景】松树的内生真菌会影响红脂大小蠹及其伴生真菌的生长及扩散,从而影响红脂大小蠹的入侵。【目的】掌握赤峰地区红脂大小蠹寄主树种内生真菌的物种多样性,筛选对其伴生菌有拮抗作用的菌株,为红脂大小蠹的生物防治提供资源。【方法】采用组织分离法、形态学鉴定和internal transcribed spacer (ITS)序列分析相结合的方法对红脂大小蠹寄主树种油松、樟子松和潜在寄主落叶松进行内生真菌多样性研究,并用两点对峙法进行拮抗菌株筛选。【结果】松树内生真菌鉴定为2门6纲10目18科19属,其中落叶松韧皮部分离到的内生真菌数量最多,为39株,隶属于9属12种,真菌检出率为43.33%;油松次之,为30株,隶属于7属8种,真菌检出率为33.33%;樟子松最少,为29株,隶属于10属13种,真菌检出率为32.22%。3个树种的内生真菌相似性较低,无共有的菌种,青霉属(Penicillium)和篮状菌属(Talaromyces)是唯二的共有菌属,且青霉属在3个树种中均为优势菌属。平板对峙结果表明90%以上的树木内生真菌均能够与伴生真菌形成稳定的对峙,抑制率在50%-86%之间,且Phialocephala sp.和Pochonia bulbillosa对伴生菌的抑制率能高达93.7%。【结论】松树韧皮部的内生真菌具有较高的生防潜力,Phialocephala sp.和P. bulbillosa对红脂大小蠹伴生菌有较好的抑制效果,可作为红脂大小蠹的生防资源。  相似文献

Trimmatostroma abietis from the needles of conifers and from stone, mostly in Germany, is described. On the natural substrate it is characterized by stromatic conidiomata. Conidia are mostly two-celled, are arranged in long chains, and originate by intercalary dilatation of fertile hyphae. The wide morphological plasticity of the fungus and its physiological profile are described. Its occurrence as a saprophyte and possibly opportunistic pathogen on plants and as an invader of environments with low water activity, particularly stone surfaces, are discussed. Clohiesia lignicola sp. nov. (freshwater ascomycetes) is introduced based on a specimen collected on submerged wood in the Tung Chung River, Hong Kong. Ascomata are clypeate, asci are cylindric-clavate with a relatively massive apical apparatus and ascospores are fusoid-ellipsoidal.Clohiesia lignicola differs fromC. corticola in having wider asci and wider fusoid-ellipsoidal ascospores, and larger ascomata.Clohiesia lignicola is described and illustrated with light micrographs and is also compared with species in the genus,Annulatascus.  相似文献

A new defoliating disease was observed on 20-year-old trees of Lecythis pisonis in Viçosa, Minas Gerais, Brazil. The disease is characterized by forming one or more large zonate epiphyllous leaf spots with a light brown to white center, followed by two to three zones gradually varying from brown to red-brown. On the corresponding abaxial leaf surface, the spots are lighter with a profuse greenish gray coloration caused by the fungal colonies. In PDA culture, the fungus is characterized by a slow growth of the colonies with brown-violet pigmentation at the reverse. Inoculation tests carried out on healthy plants of L. pisonis confirmed the pathogenicity of this fungus. Periconiella lecythidis sp. nov., the causal agent of the zonate leaf spot disease of L. pisonis is described, illustrated, discussed and compared with the other species of the genus Periconiella. The intricate generic affinity within a complex of morphologically similar hyphomycete genera consisting of Metulocladosporiella, Parapericoniella, Penidiella and Periconiella is discussed in detail.  相似文献

During a survey of plant-inhabiting fungi in a botanical garden in Japan, some noteworthy fungi were collected from leaf spots of some herbal and arboreal plants. Among them, five new species are described, namely: Phyllosticta. ardisiicola on Ardisia crenata, Phy. aspidistricola on Aspidistra elatior, Phy. kerriae on Kerria japonica, Phy. fallopiae on Fallopia japonica, and Pseudocercospora davidiicola on Davidia involucrata. Passalora pyrrosiae, a new combination for Pseudocercospora pyrrosiae on Pyrrosia lingua, is proposed based on its morphological characteristics designating the neotype specimen.  相似文献
