在大肠杆菌中克隆肺炎支原体P1蛋白羧基端基因片段,为P1蛋白基因片段的扩增、表达及探讨羧基端基因片段功能打基础.采用PCR扩增方法获取P1结构基因.扩增产物用SalI和EcoRI酶切消化,回收1kb大小的DNA片段并与pUC19DNA连接,转入大肠杆菌JM109菌株.用X-gal平板及质粒图谱分析方法筛选重组克隆株,再用限制性核酸内切酶酶切图谱分析鉴定.经PCR扩增MPDNA获得1条5.0kbDNA片段.重组质粒限制性内切酶指纹图谱显示出2条带,1条为pUC19载体DNA带,另1条是1kb的插入片段.实验获得肺炎支原体P1蛋白结构基因及含P1蛋白羧基端DNA片段的重组克隆株.  相似文献

利用PCR方法从输血传播性病毒 (transfusiontransmittedvirus,TTV)阳性标本中获得不同长度且重叠覆盖TTV基因组的DNA片段。将PCR扩增片段克隆到pT Adv载体中 ,筛选获得阳性克隆。DNA序列测定结果表明所克隆的片段为TTV基因组序列。利用DNA片段中特有的限制性内切酶位点将TTV的DNA片段首尾相连 ,得到近全长的基因组克隆 ,命名为TTV0 2 1。对TTV0 2 1的核酸序列进行分析 ,TTV0 2 1长 3472nt,存在 2个阅读框架ORF1和ORF2 ,分别编码 785和 1 46个氨基酸。将TTV0 2 1与其它已知的TTV基因组全序列进行了同源性比较 ,并进行进化分析。结果表明 ,TTV0 2 1序列与TTV分离株CHN2、BDH1的遗传距离较近 ,而与其它分离株相对较远。  相似文献

HindtTI和艮o Rl双重酶解p1)G3|质粒DNA制备了29个硷基对DNA片段。该DNA片段含有l‘个硷基对A拟操纵基因(pseud~perator),其一端为HindLTI接头,另一端为EcoRI接头。29个玲基对片段与pBR325质粒连接、转化、筛选获得克隆pQHl00(Ap‘Tc‘。M’)。FQHl00DNA分别以Hznd[1l,EcoRl单独酶解或双重酶解,经凝胶电泳分析,其结果表明,pQH1~)0DNA含有两个A拟操纵基因,这两个DNA片段在HtnI川位点上,以头对头的方式相连。  相似文献

通过直接施用除草剂、荧光诱导和正反杂交试验等鉴定出4种抗阿特拉津的龙葵生物型。从抗性最强的B_(13)株制备叶绿体DNA,构建限制性内切酶Bam HI片段的基因文库,并从中筛选出1个5kb Bam HI片段的克隆pSB 135,通过与探针的分子杂交,证明在pSB 135克隆的Bam HI片段中含有编码叶绿体32k蛋白质的阿特拉津抗性基因。  相似文献

目的:克隆汉滩病毒84FLi株L片段的cDNA。方法:提取病毒总RNA,反转录为cDNA模板,用高保真的Toq酶扩增出含有L片段部分克隆的片段84L1(1-1875)、84L2(1725—3352)、84L3(2921-4961)和84L4(4727—6533),将这4个片段分别与T载体pGEM-T-Easy连接,转化感受态大肠杆菌后获得4个克隆pGEM—L1、pGEM—L2、pGEM—L3和pGEM—L4。利用酶切连接方法获得pGEM—L12和pGEM-L34,2段产物覆盖全长的L片段,并且于2921—3352nt区域内相互重叠;再利用酶切连接的方法获得了含有L片段全序列的cDNA克隆。结果:获得6个含有L片段部分片段的克隆,进而酶切组装为含有L片段全序列的cDNA克隆。结论:应用分段克隆和逐次拼接法成功构建了84FLi株L片段全序列的cDNA克隆。  相似文献

鸡肠道微生物菌群经PCR-DGGE分析,回收PCR-DGGE分析胶上的一条DNA片段,回收的DNA片段再重复进行2次PCR-DGGE分析,以及分别用PCR反复循环扩增和PCR高保真酶扩增后再进行DGGE分析等方法研究PCR-DGGE分析中多条带产生原因。结果显示PCR-DGGE分析中多条带产生原因可能是作PCR扩增模板的DNA混杂有少量其他DNA片段,多条带现象不易被消除。DGGE分析胶上的DNA片段测序时,将该DNA片段回收、PCR扩增后克隆,提取多个阳性克隆菌的质粒DNA片段,分别与其原目的DNA片段进行DGGE分析,在DGGE分析胶上选取与原目的DNA片段处于同一电泳位置的质粒DNA测序,提高测序的准确性。  相似文献

通过构建人源毒素源性大肠杆菌野生株E519/66A大质粒的基因文库,成功地筛选出能表达定居因子抗原CS6菌毛的阳性克隆,初步确定了克隆DNA片段的限制性内切酶图谱。CS6抗原的编码和调控基因集中在一大小为4.6kb的DNA区段中,该片段能产生两种分子量大小不同、但抗原反应交叉的菌毛蛋白。本研究获得的CS6抗原阳性的重组菌株可用于人源ETEC多价疫苗的研制,克隆的基因片段亦可作为研究CS6菌毛蛋白基因表达及调控的基础。  相似文献

用家鼠型流行性出血热病毒R22株RNA,经polyA接尾,以Oligo-dT做引物,合成cDNA。用pUC18为载体转染E.coli Mc1061,建立cDNA克隆。再经菌落杂交,选择病毒特异性的5个阳性克隆制成缺口翻译探针,与病毒RNA3个片段进行反杂交,确定RNA片段的特异性。结果表明,3个克隆为中(M)片段的cDNA,另两个分别为大(L)和小(S)片段cDNA。核苷酸序列分析证明,克隆的DNA中含病毒特异的核苷酸序列。  相似文献

为了对1株中国棉铃虫核型多角体缺失病毒HZ-9进行基因组测序,采用了一种新的方法,通过超声波振断HaBacHZ9细菌人工染色体质粒(bacterial artificial chromosome plasmid,Bacmid)基因组DNA,用Taq酶在DNA片段两端加腺噤呤A,胶回收后得到预期的1—2kb的DNA片段,然后与pGEM-Teasy载体连接,构建了中国棉铃虫缺失病毒HaBacHZ9的亚克隆文库。结果随机挑选10个克隆子酶切分析,显示9个克隆子有1500bp左右的插入片断,并对HaBaeHZ9进行了全基因组测序。结论成功构建了HaBaeHZ9的DNA测序文库,为HZ-9功能基因组学研究奠定了基础,这是一种简单快速的构建DNA病毒测序文库的方法。  相似文献

以穿梭质粒pCN60为载体,大肠杆菌C600为受体构建了扣囊拟内孢霉(Endomycopsis fibuligera)G45 Sau 3A基因文库。从基因文库中提取重组质粒DNA并转化酿酒酵母(Saccharomyces cerevisiae)BJ1991,选出四个具有o-淀粉酶活性的转化子,琼脂糖凝胶电泳结果证实插入的DNA片段为9.0kb。对插入DNA片段亚克隆,确定。-淀粉酶基因位于PstI-Sall 3.9kb片段上,启动子位于PstI-EcoRI 的1.3kb片段上。用亚克隆PGK11.9kb片段置换。-淀粉酶启动子区,其转化子的e-淀粉酶活性有明显提高。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
