胡萝卜软腐欧文氏菌甜菜亚种(Erwinia carotovora subsp. betavasculorum) EcbCSL101菌株具有很强胞外酶分泌活性, 接种非寄主植物烟草引起过敏反应。Southern blotting结果表明EcbCSL101菌株中含有hrpN 基因。PCR扩增含EcbCSL101完整开放阅读框的DNA片段并克隆到表达载体pET28a(+)中。核苷酸序列分析表明, EcbCSL101菌株的hrpN 基因的ORF为1113 bp, 编码36.65 kD HarpinEcbCSL101蛋白(GenBank, DQ355519),与其它几种软腐欧文氏菌Harpin蛋白有较高的同源性。将含有hrpNEcbCSL101基因的重组质粒转化到大肠杆菌JM109(DE3)中进行表达,纯化后的HarpinEcbCSL101能诱导烟草发生过敏反应。  相似文献

用PCR方法从胡萝卜软腐欧文氏菌的基因组DNA中扩增出信号分子合成酶expI基因,将其克隆到大肠杆菌表达载体pET-28α(+)上,转化大肠杆菌BL21(DE3),获得高效表达expI基因的重组大肠杆菌BL21(pET28α-expI).重组菌经IPTG诱导表达,SDS-PAGE检测表达蛋白相对分子质量约为24.8kD,与预期分子量相符.经薄层层析和高效液相色谱分析发现该重组菌产生的信号分子种类为N-3-羰基己酰高丝氨酸内酯和N-己酰高丝氨酸内酯与胡萝卜软腐欧文氏菌产生的一致.  相似文献

魔芋软腐病菌分子鉴定与遗传多样性   总被引:11,自引:0,他引:11  
通过对分离的魔芋软腐病菌株和其它参试菌株的致病性测定、选择性培养基培养性状观察和16S-23S rDNA转录间隔区PCR(ITS-PCR)分析,将测试的33株软腐病菌株主要分为3个组群。第1组群为胡萝卜软腐欧文氏杆菌胡萝卜软腐亚种(Erwinia carotovorasubsp.carotovora,E.c.c.);第2组群为菊欧文氏杆菌(Erwinia chrysanthemi,E.ch.);还有一组未能确定的菌株。利用细菌基因组重复序列通用引物BOX和J3进行Rep-PCR特异性扩增,引起软腐病的菌株E.c.c.和E.ch.(ITS-PCR鉴定)种内的Rep-PCR指纹存在明显的遗传分化,经聚类分析,在0.1水平上把E.c.c.13株区分为5个类群。  相似文献

用两种方法分别洗除菌体胞外多糖(EPS)和脂多糖(LPS),经在大白菜幼苗上测定表明,菌体洗除LPS后对寄主根表的吸附能力明显减弱,吸附菌量随无LPS的菌体在混合接种体中比例的增高而降低,在无LPS的菌体接种体中加入LPS提取物,可使吸附能力恢复到正常水平;平行测定的EPS不发生上述影响。用EPS和LPS对根表进行预处理,均可明显抑制完整菌体(不洗,含EPS和LPS)和无EPS的菌体对根表的吸附,但只有LPS预处理使无LPS的菌体吸附量提高2—3倍。用大白菜对该菌的专化性细菌凝集素Agin—SD60所作的菌体凝集试验表明,菌体洗除LPS后不再被 Agin—SD60所凝集。根据以上结果认为,菌体脂多糖在大白菜软腐欧文氏菌与寄主的接触识别中可作为细菌识别子起作用。  相似文献

采用人工分段合成梨火疫病细菌(Erwinia amylovora)Harpin基因A、B、C、D片段,然后连接构建全长harpin基因,克隆到pET-23(+)表达载体中,在E.coli BL21中表达Harpin蛋白.通过His@Bind Resin Ni++ 层析柱纯化,获得Harpin蛋白.经SDS-PAGE和Western blot分析,该蛋白分子量为45 kDa.采用叶片穿刺法,5μL 30μg/mL Harpin能诱发烟草叶片的过敏反应.采用叶脉注射法,5μL 30μg/mL Harpin能诱发辣椒叶片的过敏反应.将烟草和辣椒用30μg/mL Harpin喷雾处理5d后,接种烟草花叶病毒(Tobacco mosaic virus, TMV),观察植株病症,并采用ELISA检测TMV含量.结果表明经Harpin处理的烟草和辣椒对TMV侵染有一定的抑制作用.  相似文献

【目的】为了研究鞭毛钩基因flgK在胡萝卜软腐果胶杆菌胡萝卜亚种(Pectobacterium carotovorum subsp.carotovorum,P.c.c)的功能。【方法】本研究采用两亲同源交换法构建了基因缺失突变体ΔflgKpcc并构建了互补菌株ΔflgKpcc-KH,测定突变体及其互补菌株的菌体形态、运动性、致病因子、致病性等表型。【结果】与野生菌株PccS1相比,ΔflgKpcc鞭毛缺失,菌体易沉降,在0.3%半固体培养基上运动能力明显降低,生长速率无明显变化,但是纤维素酶和蛋白酶的活性、生物膜形成能力明显下降,对感病寄主的致病力显著减弱。基因互补可以使上述突变表型恢复。【结论】实验表明,鞭毛基因flgK突变导致了菌体的运动性降低、病原菌毒性相关的酶活力下降,从而导致致病力下降。  相似文献

【目的】对贵州省半夏块茎腐烂病病原菌进行分离和鉴定,为开发有效的防治技术体系提供依据。【方法】采用组织分离法进行病原菌分离,根据柯氏法则进行致病性测定,结合形态学、生理生化特性和分子生物学方法进行鉴定。【结果】从贵州3个半夏产地患病样品中分离出12株病原细菌和5株病原真菌。经形态学、生理生化特性和分子生物学方法鉴定,12株病原细菌为胡萝卜软腐果胶杆菌胡萝卜软腐亚种;ZJ、Z3病原真菌为尖孢镰刀菌,D3、D5、H1为茄病镰刀菌。【结论】贵州省半夏块茎腐烂病的病原菌有病原细菌和病原真菌。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Microbial resistance to antibiotics is an unresolved global concern, which needs urgent and coordinated action. One of the guidelines of the Centers for Disease Control and Preventions (CDC) to combat antibiotic resistance is the development of new antibiotics to treat drug-resistant bacteria. In our effort to find new antibiotics, we report the synthesis and antimicrobial studies of 30 new pyrazole derivatives. These novel molecules have been synthesized by using readily available starting materials and benign reaction conditions. Some of these molecules have shown activity with MIC values as low as 0.78?µg/mL against four bacterial strains; Staphylococcus aureus, methicillin-resistant S. aureus, Bacillus subtilis, and Acinetobacter baumannii. Furthermore, active molecules are non-toxic to mammalian cell line.

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Cyclin-dependent kinases (CDKs) and Polo-like kinases (PLKs) play key role in the regulation of the cell cycle. The aim of our study was originally the further development of our recently discovered polo-like kinase 1 (PLK1) inhibitors. A series of new 2,4-disubstituted pyrimidine derivatives were synthesized around the original hit, but their PLK1 inhibitory activity was very poor. However the novel compounds showed nanomolar CDK9 inhibitory activity and very good antiproliferative effect on multiple myeloma cell lines (RPMI-8226).  相似文献
