沙田柚系列遗传关系的RAPD标记研究   总被引:9,自引:0,他引:9  
采用 RAPD标记技术分析了沙田柚系列 1 2个样品的遗传关系。利用经筛选具多态性的 1 4个 1 0碱基随机引物对 1 2个样品进行 DNA随机扩增 ,获得清晰可重复的位点 99个 ,其中多态性位点 5 8个 ,占 5 8.89% ;在部分样品中检出了特异性 RAPD标记 1 9个 ,占 1 9.1 9% ;通过样品间遗传相似性系数比较与 UPGMA聚类分析 ,并根据历史事实可知 ,广西沙田柚、梅州金柚为沙田柚的无性繁殖系 ,软枝系沙田柚、沙田柚早熟单株为沙田柚的芽变后代 ,梅花早柚、菊花心沙田柚、冬瓜圈沙田柚、段氏柚、垫江沙田柚、古老钱沙田柚为沙田柚的实生变异品系。  相似文献

利用22个经筛选在样品间具多态性的10碱基随机引物,对柚 [Citrus max ima (Burm.) Merr.] 的主要品种及柚的一个野生近缘种云南红河橙 (C. hongh eensisY. M. et al.)、一个栽培近缘种葡萄柚 (C. × paradisi Macfad) 共79个样品的总DNA进行PCR扩增.在样品间共产生164条RAPD标记带,其中多态性带134条,多态性百分率为81.71%;以NTSYS-pc (Version 1.80)软件计算样品间的Jaccard遗传相似系数,并进行UPGMA聚类分析.结果表明野生近缘种云南红河橙及栽培近缘种葡萄柚与柚三个种种内聚合良好,种间分异明显,为柚分类上的地位奠定了遗传学基础.特异性RAPD分子标记的研究为柚类品种的鉴定提供了重要指标.  相似文献

四川农业大学小麦研究所侯永翠、郑有良、魏育明等研究人员对黑麦遗传多样性课题作了研究。他们采用随机扩增多态性DNA(RAPD)标记 ,对黑麦属 (SecaleL) 7个品种共 1 2份材料进行了遗传多样性检测 ,发现被检测材料间RAPD标记多态性较高 ,在 4 0个随机引物中 ,有 2 5个引物约占整个的 6 2 .5 %的扩增产物具有多态性。这 2 5个中共扩增出 1 6 7条带 ,其中 89条带约占 5 3.2 %具有多态性 ,每个引物可扩增出 1~ 1 0条多态性带 ,平均为 3~ 6条。RAPD标记遗传距离GD变异范围为 0 .1 382~ 0 .4 5 1 2 ,平均为 0 .2 71 2。通过聚类分析表…  相似文献

利用RAPD与ISSR分子标记检测手段,分析了哈茨木霉T2-16肽类代谢产物处理豇豆土著根瘤菌,对其遗传性状的影响,同时,比较了RAPD和ISSR两种不同分子标记在检测根瘤菌种间的遗传相似性以及遗传变异性的分辨力.实验中,从100条引物中筛选到具有多态性的ISSR引物5条,从80条引物中筛选到具有多态性的RAPD引物6条,用5条ISSR引物扩增出54条带,多态性条带比率为75.93 %;6条RAPD引物扩增出61条带,多态性条带比率为68.85 %.两种分子标记均能揭示出处理前后根瘤菌间的遗传差异,但ISSR标记比RAPD标记可检测到更大的遗传变异.根据两种标记的结果,对供试的根瘤菌进行聚类分析,结果表明,土著根瘤菌经木霉肽类代谢产物处理后,与出发菌株相比,表现出一定程度的遗传分化和遗传差异性.  相似文献

分子标记鉴定常山胡柚优良基因型的初步研究   总被引:1,自引:0,他引:1  
本研究利用RAPD和ISSR分子标记对常山胡柚的优良基因型进行鉴定,并探讨常山胡柚的起源。从100个RAPD引物中筛选出12个多态性引物用于正式扩增,共得到117条DNA带,其中多态性DNA带64条,占扩增片段的54.7%;从105个ISSR引物中筛选出11个多态性引物用于正式扩增,共得到94条DNA带,其中多态性DNA带58条,占扩增片段的61.7%。RAPD和ISSR分析揭示了常山胡柚及其近缘种的一些特异性条带。ISSR共产生了15条特异条带,RAPD共产生12特异性条带。实验数据用AMOVA软件计算遗传距离,用NTSYS-pc软件构建UPGMA聚类树状图。结果显示,所有的基因型及不同种之间均能够彼此区分,分析得到的指纹图谱对常山胡柚种和基因型的鉴定具有潜在的应用价值,可用于优良基因型的鉴定。聚类分析结果显示常山胡柚和甜柚聚为一枝,确定了甜柚是杂交亲本之一,但是常山胡柚和柚的遗传距离较远,说明常山胡柚可能是甜橙、柚和柑桔属其他种的多重自然杂交的结果。  相似文献

以随机扩增多态DNA技术(RAPD)分析了奥利亚罗非鱼和尼罗罗非鱼两个养殖群体的群体内及群体间遗传关系,并探讨了该技术在种群鉴定中的应用。RAPD引物筛选结果表明,所测试的20个随机引物中(Table 1),除一个引物未扩增出任何片段外,其余19个引物均扩增出1~11个大小不等的片段,长度大部分在500—3000bp之间,共扩增出220个片段,平均每个引物产生55个片段。两群体间共有片段70条,大部分引物的扩增产物具有种间多态性,种群间相似系数为0.727。以筛选的引物对两种群不同个体(Fig.1,Table 2)及种群混合样品(Fig.2,Table 3)进行RAPD分析。结果表明,不同引物在扩增图谱上表现很大差异:奥利亚罗非鱼不同个体间表现为一致的扩增图谱,种内相似系数达1000,显示了其种群内遗传变异的缺乏;尼罗罗非鱼种内相似系数为0.827,个体间存在不同程度的多态性;两个种群间的相似系数分别为0.767和0.742,表明种间有较高的同源性,遗传距离为0.235,略低于国外的报道.此外,两个养殖群体间的扩增图谱比较也暗示了遗传渐渗现象的存在。实验表明,RAPD标记可以作为一种可靠的遗传标记,用于不同鱼类种群的鉴定,RAPD分析方法是一种快速,简便且行之有鼓的鉴定鱼类种群的方法。  相似文献

甜樱桃品种及其砧木的RAPD分析   总被引:4,自引:1,他引:3  
利用RAPD技术,从130个随机引物中筛选出46个引物,对欧洲甜樱桃、欧洲酸樱桃、马哈利樱桃和野生中国樱桃4个类型樱桃种,以及欧洲甜樱桃与中国樱桃的种间杂交种共15个品种的基因组遗传变异进行分析。结果表明,46个随机引物均得到了稳定可重复的RAPD图谱,扩增出的DNA条带大小在100~2625bp之间,多态性位点数517个,多态性位点百分率为98.85%,每个随机引物扩增出的多态性DNA条带数在4~23条。品种间Nei遗传距离在0.166~0.479之间,平均遗传距离0.329;甜樱桃新品种‘秦樱1号’与‘秦岭玛瑙’、‘CDR-1’等10个樱桃砧木之间的遗传距离在0.248~0.376,并且根据遗传距离可以相互区分,所分析的15个樱桃品种均扩增出了特有的DNA条带,每个樱桃特有标记带在2~17个之间,共扩增出149个特有标记,据此可以进行樱桃品种及砧木的RAPD鉴定。研究认为利用RAPD技术可以在分子水平上对甜樱桃品种及其砧木进行快速鉴定。  相似文献

中国啤酒大麦品种RAPD标记的遗传多样性分析   总被引:3,自引:0,他引:3  
采用RAPD技术对中国38个啤酒大麦品种的遗传资源进行了聚类分析。结果表明:从筛选出的28个有多态性的随机引物中,共扩增出153条谱带,其中91条谱带具有多态性,占59.4%。每个引物可扩增出1~8条多态性谱带,平均3.3条。聚类分析表明,在遗传距离GD值0.27水平上38个啤酒大麦品种可聚成两大类,下分5个亚类。品种间遗传距离GD变幅为0.00952~0.37846。RAPD标记揭示出这38个啤酒大麦品种遗传变异较小,遗传基础比较狭窄。  相似文献

采用RAPD分子标记技术,从100个随机引物中筛选出多态性强、重复性好且稳定性高的引物18个,对38份野生鸢尾属材料进行扩增,共扩增出409条带,其中多态性带405条,多态性比率为99.0%,表明野生鸢尾属植物种间有丰富的遗传多态性;根据DNA谱带计算物种间遗传距离,聚类分析结果将鸢尾属38份材料划分为6组,其结果与传统生物学特性划分的6个亚属的分类结果基本一致;物种特有RAPD标记分析表明,利用18个引物可以较好地将鸢尾属38种植物区分开,其中9个材料得到了单一标记的扩增带,表明运用RAPD分子标记对研究鸢尾属植物特异性基因及标记的筛选等有一定的理论和实际应用价值。  相似文献

Six parent and their 12 gamma ray-induced somatic flower colour mutants of garden rose were characterized to discriminate the mutants from their respective parents and understanding the genetic diversity using Random amplification of polymorphic DNA (RAPD) markers. Out of 20 primers screened, 14 primers yielded completely identical fragments patterns. The other 7 primers gave highly polymorphic banding patterns among the radiomutants. All the cultivars were identified by using only 7 primers. Moreover, individual mutants were also distinguished by unique RAPD marker bands. Based on the presence or absence of the 48 polymorphic bands, the genetic variations within and among the 18 cultivars were measured. Genetic distance between all 18 cultivars varied from 0.40 to 0.91, as revealed by Jaccard’s coefficient matrix. A dendrogram was constructed based on the similarity matrix using the Neighbor Joining Tree method showed three main clusters. The present RAPD analysis can be used not only for estimating genetic diversity present in gamma ray-induced mutants but also for correct identification of mutant/new varieties for their legal protection under plant variety rights.  相似文献

A procedure which involves the use of RAPD markers, obtained from bulked genomic DNA samples, to estimate genetic relatedness among heterogeneous populations is demonstrated in this study. Bulked samples of genomic DNA from several alfalfa plants per population were used as templates in polymerase chain reactions with different random primers to produce RAPD patterns. The results show that the RAPD patterns can be used to determine genetic distances among heterogeneous populations and cultivars which correspond to their known relatedness. The results also indicate that, by using ten primers with bulked DNA samples from ten individuals, 18–72 populations or cultivars can be distinguished from each other on the basis of at least one unique RAPD marker. We anticipate that DNA bulking and methods for comparing RAPD patterns will be very useful for identifying cultivars, for studying phylogenetic relationships among heterogeneous populations and for selecting parents to maximize heterosis in crosses.  相似文献

Two PCR-based techniques, RAPD and ISSR, were utilized for determination of genetic relationship of 24 strawberry cultivars used in breeding program at the Research Institute of Pomology and Floriculture in Poland. Polymorphism of investigated genotypes was observed in reactions with 23 out of 48 tested RAPD primes and 41 from 90 tested ISSR primers. Relationship, determined on the base of polymorphic products analysis and showed in the form of dendrograms (UPGMA percent method), was generally similar for both techniques, although similarity values based on ISSR data were higher than those based on RAPD. The parallel use of two data sets seems to allow for precise estimation of cultivars relationship and diminishing mistakes connected with methods' technical limitations.  相似文献

Ginger (Zingiber officinale Roscoe) is an economically important plant, valued all over the world. The existing variation among 16 promising cultivars as observed through differential rhizome yield (181.9 to 477.3 g) was proved to have a genetic basis using different genetic markers such as karyotype, 4C nuclear DNA content and random amplified polymorphic DNA (RAPD). The karyotypic analysis revealed a differential distribution of A, B, C, D and E type of chromosomes among different cultivars as represented by different karyotype formulas. A significant variation of 4C DNA content was recorded in ginger at an intraspecific level with values ranging from 17.1 to 24.3 pg. RAPD analysis revealed a differential polymorphism of DNA showing a number of polymorphic bands ranging from 26 to 70 among 16 cultivars. The RAPD primers OPC02, OPA02, OPD20 and OPN06 showing strong resolving power were able to distinguish all 16 cultivars. The extent of genetic diversity among these cultivars was computed through parameters of gene diversity, sum of allele numbers per locus and Shannon's information indices. Cluster analysis, Nei's genetic similarity and genetic distances, distribution of cultivars into special distance classes and principal coordinate analysis and the analysis of molecular variance suggested a conspicuous genetic diversity among different cultivars studied. The genetic variation thus detected among promising cultivars of ginger has significance for ginger improvement programs.  相似文献

The genetic distance of 11 cotton genotypes varying in heat tolerance was studied using RAPD markers. Fifty-three random decamer primers were used for the estimation of genetic distance. Among the 53 RAPD primers, which were custom synthesized by GeneLink Inc., UK, 32 were polymorphic and 21 were monomorphic. The 32 polymorphic primers produced 273 fragments, with a mean of 8.3 fragments per primer. The number of polymorphic bands produced in the 11 cotton accessions ranged from 1 to 31. Primer GLC-20 produced 31 polymorphic bands, while two primers, GLB-5 and GLC-12, produced one polymorphic band each. A range of 88.89 to 42.48% genetic similarity was observed among the 11 cotton accessions. The highest genetic similarity was observed between FH-945 and BH-160 (88.89%), whereas the lowest value was found between NIAB-801/2 and FH-945 (42.48%). Unique amplification profiles were produced by most of the cultivars; the differences were sufficient to distinguish them from other genotypes. This confirms the efficacy of RAPD markers for the identification of plant genotypes. An accumulative analysis of amplified products generated by RAPDs was sufficient to assess the genetic diversity among the genotypes. This information should be helpful for formulating breeding and genome mapping programs.  相似文献

Optimization of primer screening for evaluation of genetic relationship in 34 cultivars of rose through random amplified polymorphic DNA (RAPD) markers was investigated. Four series of decamer primers were used for screening and optimization of RAPD analysis between which A and N series performed good amplification of fragments as compared with other series. The primers OPN-07 and OPN-15 produced maximum number of DNA fragments in Rosa hybrida cv. Anuraag. Some primer either did not produce amplification or produced very poor amplification. Further, ten selected primers were used for genetic analysis of 34 rose cultivars. The primer OPN-15 amplified 21 fragments in all cultivars tested. A total of 162 distinct DNA fragments (bands) ranging from 100 to 3400 base pairs were amplified by using 10 selected random primers. The cluster analysis indicated that these rose cultivars formed nine clusters.  相似文献

Genetic variation of nine upland and four lowland rice cultivars (Oryza sativa L.) was investigated at the DNA level using the randomly amplified polymorphic DNA (RAPD) method via the polymerase chain reaction (PCR). Forty-two random primers were used to amplify DNA segments and 260 PCR products were obtained. The results of agarosegel electrophoretic analysis of these PCR products indicated that 208 (80%) were polymorphic. All 42 primers used in this experiment were amplified and typically generated one-to-four major bands. Only two primers showed no polymorphisms. In general, a higher level of polymorphism was found between japonica and indica subspecies while fewer polymorphisms were found between upland and lowland cultivars within the indica subspecies. A dendrogram that shows the genetic distances of 13 rice cultivars was constructed based on their DNA polymorphisms. Classification of rice cultivars based on the results from the RAPD analysis was identical to the previous classification based on isozyme analysis. This study demonstrated that RAPD analysis is a useful tool in determining the genetic relationships among rice cultivars.  相似文献

对14个黄皮(Clausena lansium)品种(系)进行RAPD分析,从150个随机引物中筛选出16个能在种质间表现出多态性的引物,共扩增出87条谱带,其中多态性带47条,多态性比例为54.0%。结果表明,除龙川无核黄皮、龙山无核黄皮和冰糖黄皮外,其它11个品种(系)间的遗传距离较近(D≤0.1),说明大多数黄皮品种遗传差异较小,亲缘关系较近。UPGMA聚类分析表明,14个黄皮品种(系)在遗传距离0.15处可划分为3个类群,基本反映了黄皮品种间的遗传多样性。  相似文献

应用RAPD方法对海南60份野生荔枝进行基因组多态性分析,20个随机引物共产生165条RAPD带,DNA片段大小在200~1500bp之间,其中121条为多态性带,占总带数的73.3%,并利用遗传距离进行聚类分析。结果表明,60份野生荔枝可归为6类,表明种群存在一定的遗传变异,也为分析野生荔枝群体间及群体内的遗传多样性探索了有效方法。  相似文献

对取自中国、日本、韩国和朝鲜等东亚地区的24个桔梗种质资源,利用RAPD-PCR标记方法,分析了其遗传变异和遗传关系,旨在为桔梗的品种鉴定和杂交育种的亲本选择提供理论依据。结果表明: (1)用11个引物扩增出131条谱带,平均每个引物扩增出11.9条谱带,并扩增出61个多态性谱带,平均每个引物扩增出5.5条多态性谱带,显示出了相对高的多态率(46.6%)。(2)得到的RAPD数据的遗传相似性范围为0.668~0.994,并以遗传相似系数0.78为标准,将24个桔梗种质资源分为7个类群。  相似文献
