在东北红豆杉(taxuscuspidate)细胞培养物的基本培养基中加入30mg@L-12.4-D,可以提高细胞培养物的生长量,其相对最高生长量可以达到85.999mgFW/gFW@d;培养基中加入活性炭粉末lg@L-1,可以克服植物细胞生长过程中的褐化问题;适当种类及含量的生长调节剂的加入:6-BA0.5mg@L-1+KT2mg@L-1,可以使植物细胞合成和积累相对较多的紫杉醇(Taxol)。

以阳桃茎段为材料进行组培快繁。结果表明:阳桃离体培养的细胞分裂素以ZT的效果最好,其次是BA,KT最差;植物生长调节物质是NAA最好,其次是IBA、IAA;较适宜的植物生长调节物质配比为BA 0.5mg L^-1 NAA 0.2mg L^-1 GA 0.2mg L^-1,增殖系数高达3.7,而且畸苗率较低(23.3%)。生根培养基以1/2MS IBA 0.2mg L^-1 IAA 0.1mg L^-1效果好,生根率达87.1%,蔗糖浓度以2.0%一3.0%为宜。 相似文献

We have recently found that about 50% of newly formed oligodendrocytes normally die in the developing rat optic nerve. When purified oligodendrocytes or their precursors are cultured in the absence of serum or added signalling molecules, they die rapidly with the characteristics of programmed cell death. This death is prevented either by the addition of medium conditioned by cultures of their normal neighboring cells in the developing optic nerve, or by the addition of platelet-derived growth factor (PDGF) or insulin-like growth factors (IGFs). Increasing PDGF in the developing optic nerve decreases normal oligodendrocyte death by up to 90% and doubles the number of oligodendrocytes, suggesting that this normally occurring glial cell death might result from a competition for limiting amounts of survival signals. These results suggest that competition for limiting amounts of survival factors is not confined to developing neurons, and raise the possibility that a similar mechanism may be responsible for some naturally occurring cell deaths in nonneural tissues. © 1992 John Wiley & Sons, Inc. 相似文献

钙调素作为Ca ̄(2+)信号的主要胞内受体,在细胞增殖调节中起着重要作用。实验中运用DNA体外重组技术构建了高表达钙调素的真核载体,并将其转染到大鼠正常肾细胞(NRK)中得到钙调素高表达的稳定细胞株。分析表明,高表达钙调素加速细胞生长,促进细胞从G_1期向S期及G_2期向M期的进程,并且使细胞血清依赖性降低,在单层培养中出现接触抑制丧失的岛状生长的现象。基因表达分析表明,原癌基因c-fos、c-myc随钙调素增高而表达加强,这可能是钙调素作用的分子机理之一。 相似文献

A receptor for fibroblast growth factor (aFGF, bFGF) was partially characterized in intact cell cultures, cell plasma membranes, and tissue plasma membrane preparations. Analysis of 24 different cell types from four species identified a 165-kDa FGF receptor present on the cell surface of most mesodermal and neuroectodermal cells. Chemical crosslinking of 125I-aFGF to its cell surface receptor was specifically blocked by a 100-fold molar excess of either aFGF or bFGF. In contrast to the similar molecular weight of FGF receptors, different cell types exhibited significant variations in binding of 125I-aFGF to intact cultures with low values of 8 pM and 700, to high values of 60 pM and 30,000, for the Kd and receptor number per cell, respectively. A binding assay was developed for quantitation of 125I-aFGF binding to cell- and tissue-derived membrane preparations. Membranes prepared from baby hamster kidney cells exhibited a Kd of 55 pM, while a similar Kd of 67 pM was determined for intact baby hamster kidney cells. Although ten different adult bovine tissue membrane preparations and human term placental membranes exhibited no specific binding of 125I-aFGF, FGF receptor was detected in embryonic murine tissues (17 days gestation). These results support the existence, in a variety of cells, of either a common FGF receptor that binds both aFGF and bFGF or closely related FGF receptors that cannot be distinguished by molecular weight. The differential binding of FGF to its receptor in embryonic vs. adult tissues suggests a potentially broad role for FGF in embryonic development and a more restrictive role in the adult. 相似文献

利用基因工程的方法制备了高纯度的人干细胞因子,并对制备的重组人干细胞因子的结构和生物活性进行了研究。结果表明rhSCF在磷酸盐缓冲液中以非共价的二聚体形式存在,其精确分子量、质量肽谱、氨基酸组成及N端、C段序列等均与理论值一致,二硫键位置正确。rhSCF单用或与rhG-CSF合用,对猴外周血造血祖细胞有明显的动员作用。 相似文献

目的:研究食管鳞状细胞癌中肝癌衍生生长因子(HDGF)、血管内皮生长因子(VEGF)的表达及其与微血管形成的关系。方法:通过免疫组化SABC法检测和比较68例食管鳞癌、20例切缘正常组织中HDGF、VEGF的表达和CD34标记的微血管密度(MVD),分析HDGF和VEGF表达之间的关系及其与食管鳞癌患者临床病理因素和食管癌组织MVD值的关系。结果:食管鳞癌组织中HDGF(63.2%)和VEGF(72.1%)的阳性表达率均明显高于切缘正常粘膜组织(15.0%、20.0%)(P0.05),食管鳞癌组织和切缘正常粘膜组织中的MVD值分别为35.48±5.75和13.50±2.1(P0.05)。食管鳞癌组织HDGF的阳性表达率仅与其临床分期明显相关(P0.05),而VEGF的阳性表达率与其淋巴结转移、临床分期均显著相关(P0.05),二者在食管鳞癌组织中的表达呈显著正相关(P0.05)。食管鳞癌组织中HDGF、VEGF阳性表达组MVD值均明显高于HDGF、VEGF阴性表达组(P0.05)。结论:HDGF可能通过诱导VEGF的产生,从而促进血管生成,参与食管鳞癌的发生、发展及转移。 相似文献
