目的:研究枳实提取物及其药效组分橙皮苷和新橙皮苷对氧化低密度脂蛋白(oxidized lowd ensity lipoprotein,Ox—LDL)损伤的人脐静脉内皮细胞(human umbilical vein endothelial cells line,HUVEC)细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)表达和一氧化氮(nitric oxide,NO)释放的影响。方法:体外培养HUVEC,50μg/mLOX—LDL制造HUVEC损伤模型。以MTS染色法检测细胞毒性确定用药浓度。细胞ELISA法测定细胞表面ICAM-1的含量,试剂盒测定细胞培养上清液中NO含量。结果:①枳实提取物小于等于2mg/mL时,橙皮苷浓度小于等于0.03125mg/mL时,新橙皮苷浓度小于等于0.25mg/mL时,HUVEC存活率分别大于80%。②2.0mg/mL和1.0mg/mL两个浓度的枳实提取物、15.625μg/mL的橙皮苷和0.2500mg/mL新橙皮苷对OX—LDL诱导的HUVEC的ICAM-1表达有显著抑制作用。③2.0mg/mL枳实提取物显著提高OX—LDL诱导的HUVEC和正常HUVEC培养液中的NO含量;7.813ixg/mL、15.625μg/mL和31.250μg/mL 3个浓度的橙皮苷能显著提高OX—LDL诱导的HUVEC培养液中的NO含量,31.250μg/mL的橙皮苷能促进正常HUVEC的NO释放;0.2500mg/mL和0.1250mg/mL 2个浓度的新橙皮苷能显著提高OX—LDL诱导的HUVEC培养液中的NO含量。结论:枳实提取物及其药效组分橙皮苷、新橙皮苷能抑制Ox-LDL诱导的HUVEC的ICAM-1表达,促进Ox-LDL诱导的HUVEC的NO释放。  相似文献

目的:研究沼泽红假单胞菌中类胡萝卜素的提取工艺条件。方法:对细胞收集、前处理、色素初浸、二次萃取,皂化和溶剂回收等方面进行了系统的研究,并通过紫外分光光度法和高效液相色谱法对所提取的类胡萝卜素进行了分析。结果:实验结果表明,类胡萝卜素的最佳提取工艺条件为:碱性氯化钙絮凝法收集细胞,乙醇浸泡法进行前处理,49℃下丙酮初浸提浸提4h(搅拌并加抗氧化剂),丙酮用量为5mL/g菌泥,回收丙酮,乙醚二次萃取,30℃下皂化1.5h,洗涤,浓缩并回收乙醚,在此提取工艺条件下不仅能够有效地提取红螺菌中的类胡萝卜素,而且还能分离细菌叶绿素。结论在本实验条件下,根据标准曲线计算出类胡萝卜素的含量高达6.148mg/L菌泥。通过HPLC分析表明:类胡萝卜素提取液中主要成分至少有10种。  相似文献

从青蒿水提物中分离得到一种缩合鞣质(CTA),在体外进行了抗HSV-2和HBV活性研究。结果表明,CTA具有显著抗HSV-2活性,与阳性对照药物阿昔洛韦(ACV)比较,OC50分别为6.84mg/mL和3.69mg/mL,IG50分别为0.162mg/mL和0.138mg/mL,表明CTA的细胞毒性比ACV小,最低抗病毒活性浓度相当。用含CTA的培养基培养HepC2.2.1.5细胞,结果表明在浓度为2.5mg/mL时有轻微细胞毒性,在浓度为2.5~0.156mg/mL对HepG2.2.1.5细胞表达的HBeAg具有显著的抑制作用,0.625mg/mL浓度档培养第12d对HBeAg抑制率达到90.45%,提示CTA具有潜在的抗HBV活性。  相似文献

讨论了从烟草废料中提取绿原酸的优势和甲醇、乙醇、丙酮、水等不同溶剂经超声波辅助提取绿原酸的效果:研究结果表明,用体积分数40%的甲醇得到的浸提液中,绿原酸质量浓度为2.11mg/mL,比以水为溶剂时高出近50%.不同浓度的甲醇溶液中,体积分数50%的甲醇提取绿原酸的浓度最高。对树脂的吸附动力学分析表明,大孔树脂CN-101对烟草浸提液中绿原酸的吸附遵循Freundlich等温方程,吸附和解析分离所得的绿原酸收率为87.6%.在超声辅助条件下,利用甲醇等有机溶剂提取烟草中的绿原酸,进而用大孔树脂进行吸附分离的方法可行。  相似文献

本文采用薄层层析法分析了成熟辣椒果实中类胡萝卜素的组成及主要类胡萝卜素的相对含量。用溶剂油从红辣椒干果皮中抽提类胡萝卜素,经除辣味处理后得到类胡萝卜素混合物,并对其进行皂化。以硅胶G为固定相,正己烷/乙酸乙脂/丙酮/甲醇(80/10/5/5)为展开剂,对类胡萝卜素皂化后混合物进行层析,依此法可分离出清晰的13个有色斑点。对这些斑点作紫外可见光谱分析.由最大吸收峰和极性判断各斑点的成分,并由吸光植计算其相对含量。  相似文献

为探究香蕉皮类胡萝卜素提取及综合利用的可能性,采用单因素试验与正交试验相结合的方法优化香蕉皮类胡萝卜素的提取工艺条件。结果表明:其最佳提取工艺条件为:以丙酮为提取溶剂、料液比1∶25、提取温度45℃、提取时间75min,在此工艺条件下可获得最佳提取效果,类胡萝卜素提取量平均值为12.541 mg/g。另用超声波辅助提取法,得到香蕉皮中类胡萝卜素提取量平均值为13.740mg/g,在提取时间仅为20min的情况下,比普通浸提法的提取量增加了9.56%,提取效率大大提高。  相似文献

碱性普鲁兰酶产生菌的原生质体制备与诱变选育   总被引:1,自引:0,他引:1  
研究报道了Bacillus sp SX—12原生质体制备与再生最佳条件。实验表明,在液体完全培养基中加入2%的甘氨酸培养10h,原生质体制备最佳条件为:溶菌酶浓度0.5mg/mL,酶解温度37℃,酶解时间1.5h时原生质体形成率为93.8%。原生质体形成最佳高渗稳定剂为甘露醇,再生率26.4%。在原生质体制备的最佳条件下,用紫外线诱变技术选育产碱性普鲁兰酶的高产菌株。筛选到1株高产菌株SX—12C87,酶活由出发菌株的2.42μ/mL提高到6.87μ/mL,提高了约1.8倍。  相似文献

N-乙酰鸟氨酸脱酰基酶可在重组菌BL21(DE3)-pET22b-argE中表达。首先确定了该酶的细胞表达定位,再研究了诱导温度、诱导剂种类及浓度、诱导起始菌体密度、诱导时间等因素对重组菌生长及目的蛋白表达活性的影响。结果表明,IPTG和乳糖皆可诱导目的蛋白表达,乳糖的诱导效果优于IPTG。在诱导起始0D600为0.46时加入15g/L乳糖,20℃诱导18h最适于目的蛋白的活性表达。表达条件优化后,酶活从1.68U/mL提高至282.99U/mL,约为原来的168倍。  相似文献

在锁掷酵母(Sporidioboluspararoseus)发酵产类胡萝卜紊的过程中,发酵产物中类胡萝卜紊种类繁多,而且性质相似,加大了不同色素分离纯化的难度。为定向积累不同种类的类胡萝卜素,以本实验室保藏锁挪酵母JD-2为出发菌,研究了氮源种类和浓度及溶氧对锁掷酵母产类胡萝卜素的影响,并在7L发酵罐中进行了补料分批发酵试验。发现培养基中同时添加有机氮源和无机氮源且溶氧控制较低(5%)时有利于β-胡萝卜素的大量积累,最佳有机氮源和无机氮源分别为玉米浆(20g/L)、硫酸铵(5g/L)。补料分批发酵时β-胡萝卜素产量达到31.28mg/L,红酵母烯12.38mg/L。培养基中只添加有机氮源且相对溶氧控制相对较高(30%)时有利于红酵母烯的大量积累,最佳有机氮源为酵母膏(20g/L)。补料分批发酵时红酵母烯产量达到38.96mg/L,8.胡萝卜素12.36mg/L。  相似文献

The effect of simultaneous administration of vitamin C (ascorbic acid), L-cystein (Cys) and vitamin E (tocopherol) on the melanogenesis in vivo and in vitro was studied. Forty-eight brownish guinea pigs were divided into 4 groups as follows: VC group, VC+Cys group, VC+Cys+VE group and control group. They were given these vitamins by oral administration every day. UV-B exposure (0.384 J/cm2) on their depleted back skin was done at the day 8, 10, 12, 15 17 and 19. After UV-B irradiation, vitamins were administrated further 3 weeks. The luminosity score was measured using a Color Reader CR-11 (Minolta, Co) and the numbers of DOPA-positive melanocytes of their back skin were counted. B16 melanoma cells were incubated with VC, N-acetyl cystein (NAC) and VE. After 4 days of incubation, cells were harvested. The melanin contents and the tyrosinase activities in cells were measured. The luminosity score in the VC+VE+Cys group was higher than those in the other groups. The numbers of DOPA-positive melanocytes of guinea pigs treated with VC, VE and Cys were significantly decreased compared with those in VC group. In B16 melanoma cells, simultaneous treatment of VC, VE and NAC was the most effective to decrease the melanin contents and to inhibit tyrosinase activity.  相似文献

The aim of this research was to examine the effects of a triple antioxidant combination (vitamins E (VE) and C (VC) plus alpha-lipoic acid (LA)) on the total lipid and cholesterol levels and the fatty acid composition of brain tissues in experimental diabetic and non-diabetic rats. VE and LA were injected intraperitoneally (50 mg/kg) four times per week and VC was provided as a supplement dissolved in the drinking water (50 mg/kg). In addition, rats in the diabetes 1 and D+VELAVC groups were given daily by subcutaneous insulin injections (8 IU/kg), but no insulin was given to rats in the diabetes 2 group. The results indicate that the brain lipid levels in the D+VELAVC, diabetes 1 and diabetes 2 groups were higher than in the control group (P<0.01). Total lipid was also higher in the non-diabetic rats treated with LA and VC. Total cholesterol was higher in the diabetes 1 and diabetes 2 groups (P<0.05) than in controls. Cholesterol levels were similar in the D+VELAVC and LA groups but lower in the VC, VE and VELAVC groups of non-diabetic rats (P<0.05 and P<0.01). In respect of fatty acid composition, palmitic acid levels were lower in the diabetes 2 and non-diabetic VE groups than the control group (P<0.05), but higher in the non-diabetic LA group (P<0.05). Oleic acid (18:1 n-9) levels were lower in the diabetic and non-diabetic groups than the control group (P<0.01), but higher in the non-diabetic LA group. Arachidonic acid (20:4 n-6) levels were similar in the diabetes 1, D+VELAVC and control groups (P>0.05) but higher in the non-diabetic VE, VC, LA and VEVCLA groups (P<0.05) and lower in the diabetes 2 group (P<0.05). Docosahexaenoic acid (22:6 n-3) was elevated in the diabetes 2 and VEVCLA groups (P<0.01, P<0.05). In conclusion, the current study confirmed that treatment with a triple combination of VE, VC and LA protects the arachidonic acid level in the brains of diabetic and non-diabetic rats.  相似文献

探讨不同维生素对P. y17XL感染BALB/c小鼠的免疫调节作用。将6~8周龄,雌性BALB/c小鼠,随机分为维生素( V)处理组和对照组。 V处理组小鼠分别经50 mg/kg VA、600 mg/kg VE 或1 g/只VC连续10 d灌胃,0.2 mL/只;对照组小鼠分别给予相同剂量的溶剂(大豆油或生理盐水)处理。之后,各组小鼠分别经腹腔接种1&#215;106 P. y17XL寄生的红细胞,动态观察感染小鼠原虫血症水平和生存率;流式细胞术检测感染后第0、3和5天小鼠脾细胞树突状细胞( DCs)亚群( pDCs与mDCs)百分比及功能分子( TLR9和MHC域)的表达。与对照组相比,VA和VE处理组小鼠原虫血症水平升高,生存率降低;感染后第5天脾细胞中pDCs与mDCs亚群水平以及DCs表面受体TLR9和表面分子MHC域的表达水平显著下降。相反,VC处理组小鼠原虫血症水平降低,生存率延长,pDCs与mDCs亚群以及TLR9和MHC域的表达水平显著升高。结果表明,不同维生素对疟疾感染产生不同的调控作用,VA和VE通过抑制DCs数量和功能,加重疟疾感染,而VC则促进DCs数量和功能,推迟感染进程。  相似文献

以初始体质量(7.270.40) g的青鱼为研究对象, 采用维生素E(VE)有效含量分别为14.36(对照组)、25.14、37.66、62.97、113.92和210.45 mg/kg 6种等氮等能的实验饲料, 饲养青鱼幼鱼8周后, 根据生长情况选取对照组、62.97和210.45 mg/kg VE组进行24h氨氮胁迫(20 mg/L), 研究VE对青鱼幼鱼生长、免疫及抗氨氮胁迫能力的影响。结果表明: 以特定生长率为指标, 折线模型分析表明青鱼有效维生素E需要量为45.00 mg/kg。肌肉、肝脏和血清VE含量与饲料中VE含量呈明显正相关, 当饲料VE含量超过113.92 mg/kg时, 肌肉和肝脏VE含量均达到饱和。VE对鳃丝Na+/K+-ATP酶活性(NKA)和血清皮质醇(COR)无显著影响, 但随着饲料VE含量的升高, 过氧化氢酶(CAT)和总超氧化物歧化酶活性(T-SOD)呈上升趋势, 丙二醛含量(MDA)呈下降趋势。氨氮胁迫对各处理组肌肉VE含量和血清CAT活性无影响, 但肝脏VE含量均显著降低(P0.05), 且62.97和210.4 5 mg/kg VE组血清VE水平有所升高。在胁迫后, 对照组血清T-SOD、鳃丝NKA活性显著降低, 皮质醇含量显著增加(P0.05)。与对照组相比, 62.97和210.45 mg/kg VE组T-SOD、NKA活性和皮质醇含量在胁迫前后无显著变化。各处理组MDA含量在胁迫后虽均显著升高, 但210.45 mg/kg VE组在胁迫后MDA含量仍显著低于对照组(P0.05)。以上结果说明, 青鱼幼鱼获得最大生长的有效维生素E需求量为45.00 mg/kg, 且较高VE能有效提高青鱼机体免疫力, 缓解氨氮胁迫对青鱼机体的负面影响。    相似文献

This study was designed to examine the effects of dietary vitamin E (VE) on modulation of immune responses when supplied with two levels of n-3 highly unsaturated fatty acids (n-3 HUFA) in rainbow trout, Oncorhynchus mykiss. Six semipurified diets were prepared containing three levels of dietary VE (0, 100 or 1000 mg alpha-tocopheryl acetate kg(-1) diet) and n-3 HUFA either at 20 or 48% of dietary lipid provided from fish oil or docosahexaenoic acid (DHA) concentrated fish oil respectively. The diets were fed to rainbow trout (100 g initial mean weight) for 15 weeks. The VE, vitamin C (VC) content in plasma and tissues and the nonspecific immune responses, both humoral (alternative complement activity, total immunoglobulin) and cellular (phagocytosis, nonspecific cytotoxicity) were examined. VE contents in the kidney reflected the dietary input but were lower in fish fed 48% n-3 HUFA diets, and could have impaired some of immune responses compared to fish fed 20% n-3 HUFA. VC contents in kidney followed the same pattern as VE. Both humoral and cellular immune functions deteriorated in fish fed VE deficient diets whereas improvement in most of the parameters corresponded to its supplementation. However, the higher dose of dietary VE did not substantially enhance the responses assayed compared to the 100 mg dose. Besides clearly indicating the role of VE in maintaining the immune functions in fish in relation to dietary n-3 HUFA, this study has revealed that optimum health benefits could be achieved when VE is maintained slightly above the levels generally recommended for normal growth.  相似文献

-The genotoxic effect of rifampicin (RMP), one of the most active antituberculosis agents is studied. Also, the possible protection provided by the natural antioxidant vitamins C (VC) and E (VE) against the genotoxic effect of RMP is assessed.Mice were orally treated by gavage with 10, 50, 150 and 300 mg RMP kg(-1) body weight (bw). Also, oral treatment was conducted with RMP plus the vitamins. Mice received 300 mg RMP kg(-1) bw plus 100, 200 and 400mg VC or VE kg(-1) bw. Samples were taken 24h after the treatment. Repeated treatments with: (1) the therapeutic dose of RMP (10 mg kg(-1) bw); (2) RMP plus a dose of 25, 50 and 75 mg VC kg(-1); (3) RMP plus 10, 20 and 40 mg VE kg(-1) bw for 30 consecutive days were conducted.The tested doses of RMP induced a significant increase in the percentage of chromosome aberrations. However, a lower percentage of chromosome aberrations was observed when animals were treated with the therapeutic dose for 30 consecutive days.The obtained results revealed that chromosome aberrations induced by RMP decreased to a significant extent when mice were treated with RMP plus VC. The repeated doses of VC reduced the percentage of chromosome aberrations induced by RMP in a significant and dose-dependent manner. On the other hand, repeated doses of VE were not very effective in reducing the percentage of chromosome aberrations induced by RMP. Only the highest dose (3 x 40 mg kg(-1) bw) showed a significant effect (P<0.01).The results on the induction of chromosome damage clearly show that only VC appears able to efficiently protect the bone-marrow cells when given together with RMP, while no significant reduction in the yield of chromosome aberrations was observed for VE in combination with the antituberculosis drug.  相似文献

以黑灵芝为研究对象,采用不同方法提取黑灵芝中的有效成分,以BHT和抗坏血酸为对照品,采用DPPH.法研究各种提取物对自由基的清除作用。结果表明:微波萃取法是最适合提取黑灵芝中有效成分的现代方法,不仅产率高,而且速度快,节能;不同溶剂提取物对DPPH自由基清除率强弱依次为95%乙醇>丙酮>水>氯仿,提取物用量与清除能力成量效关系;黑灵芝的丙酮提取物浓度达到1 mg/mL时已超过10 mg/mL BHT的抗氧化效果,相当于0.5 mg/mL浓度抗坏血酸的抗氧化效果。  相似文献

We examined the effects of dietary vitamin E (VE) on oxidative damage to DNA and lipids in the liver a few days after total body irradiation (TBI). ODS rats, which lack vitamin C synthesis, were fed either a low VE diet (4.3 λmg λVE/kg) or a basal VE diet (75.6 λmg λVE/kg) for 5 weeks while vitamin C was supplied in the drinking water. The VE level in the liver of the low VE group was lower and the levels of lipid peroxides were higher compared to those of the basal VE group: the relative levels in the two groups were 1:30 for VE, 18:1 for 4-hydroxynonenal (HNE), and 10:1 for hexanal (HA). The level of 8-hydroxydeoxyguanosine (8OHdG), a marker of oxidative DNA damage, did not differ between the low VE and the basal VE groups. When the rats received TBI at the dose of 3 λGy and were killed on day 6, the levels of HNE, HA and 8OHdG increased by 2.2-, 2-, and 1.5-times, respectively, in the low VE group, but TBI did not cause such increases in the basal VE group. Changes in antioxidative enzymes (glutathione peroxidase, catalase, and Cu/Zn-SOD) in the liver could not explain the different responses of the two diet groups to TBI-induced oxidative damage. The concentrations of vitamin C and glutathione in the liver did not differ between the two groups. These results suggest that dietary VE can prevent the oxidative damage to DNA and lipids in the liver which appear a few days after TBI at dose of 3 λGy.  相似文献

Almond skin polyphenolics (ASP) and vitamin C (VC) or E (VE) inhibit the Cu2+-induced generation of conjugated dienes in human low-density lipoprotein (LDL) in a synergistic manner. However, the mechanism(s) by which this synergy occurs is unknown. As modification of apolipoprotein (apo) B-100 is an early, critical step in LDL oxidation, we examined the effects of combining ASP or quercetin and antioxidant vitamins on the oxidation of this moiety as well as on the alteration of LDL conformation and electronegativity (LDL−). In a dose-dependent manner, ASP (0.12–2.0 μmol/L gallic acid equivalents) decreased tryptophan (Trp) oxidation by 6.7–75.7%, increased the generalized polarity (Gp) of LDL by 21.0–81.5% at 90 min and reduced the ratio of LDL− to total LDL (tLDL) by 38.2–83.8% at 5 h. The actions of ASP on these parameters were generally additive to those of VC and VE. However, a 10–25% synergy of ASP plus VC in protecting apo B-100 Trp against oxidation may result from their synergistic interaction in prolonging the lag time to oxidation. ASP and VE acted in synergy to reduce LDL−/tLDL by 24–43%. Quercetin's actions were similar to ASP, though more effective at inhibiting Trp oxidation. Thus, ASP and quercetin reduce the oxidative modification of apo B-100 and stabilize LDL conformation in a dose-dependent manner, acting in an additive or synergistic fashion with VC and VE.  相似文献
