选择合适的微载体浓度、细胞接种密度以提高微载体利用率,优化微载体培养体系猪睾丸细胞(Swine testicle cells)的贴附生长与维持。使用DMEM补加10%血清、LSM(Low serum medium)两种培养基考察微载体浓度、细胞接种密度对细胞生长维持的影响,进而比较ST细胞在不同条件下对Cytodex1微载体的利用率。结果显示,使用LSM在T150方瓶中连续传代培养30d,平均比生长速率为0.626d~(0-1),是DMEM补加10%FBS培养基的1.15倍。选择10×10~5cells/mL细胞接种3g/LCytodex1搅拌瓶体系,最大细胞密度为38.3×10~5cells/mL,微载体利用率上升到58.8%。在灌注培养体系中培养ST细胞15d,最终细胞密度达到36.6×10~5cells/mL,扩增了13.6倍。微载体悬浮培养的使用一方面有利于ST细胞的贴附与生长,实现高密度生长,另一方面增加了微载体的使用成本,选择合适的微载体浓度、细胞接种密度,能够最大化利用微载体与培养基中的营养物质实现细胞的最优生长。  相似文献

利用7.5 L生物反应器篮式贴壁培养和全悬浮批式培养CHO工程细胞株表达可溶性肿瘤坏死因子受体Ⅱ-脂联素球部(sTNFRⅡ-gAD)融合蛋白,比较这两种培养方法的产率,以便优化高效表达sTNFRⅡ-gAD融合蛋白的制备工艺.篮式贴壁培养首先小规模培养CHO工程细胞株,待细胞增殖到一定密度后以3× 105~4× 105 cells/mL密度接种生物反应器贴壁培养3d,调换成不含血清的LK021培养基继续培养4d.而全悬浮无血清批式培养则以3×105~4×105 cells/mL密度的CHO工程细胞株接种于生物反应器,连续培养7d.培养过程实时监测培养条件,维持pH和DO的稳定.分别收集细胞上清,离心去细胞后用Pellicon切相流超滤系统对蛋白进行浓缩,并通过DEAE离子交换柱进行纯化.结果显示,篮式贴壁培养和全悬浮批式培养均成功表达了sTNFRⅡ-gAD融合蛋白,产量分别为8.0 mg/L和7.5 mg/L、纯度分别为95%和98%,从而为sTNFRⅡ-gAD融合蛋白的中试工艺研究提供了一定的基础.  相似文献

目的:优化重组抗体在悬浮无血清培养的HEK293 EBNA1瞬时表达,提高重组抗体表达量。方法:将HEK293 EBNA1细胞适应于无血清悬浮培养,筛选适宜的无血清培养基。使用PEI转染质粒进入细胞,瞬时表达重组抗体;使用Modde软件进行实验设计(DOE),优化转染质粒量、轻重链比例、PEI量等影响瞬时表达的条件。培养上清经亲和纯化,获得目标抗体,用快速免疫荧光灶抑制试验(RFFIT)测定抗体活性,用BCA法测定纯化抗体浓度。结果:293SFMII为适宜的HEK293 EBNA1细胞无血清悬浮培养基。对抗体表达影响最大的因素是轻重链比例(P=0.00000),其次为质粒浓度(P=0.00086),最后为PEI(P=0.00257)。优化的转染条件为:质粒用量0.61μg/106细胞,轻重链比例2:1,PEI 2.67μg/106细胞,优化后抗体表达量有显著提高(P=0.007)。结论:通过DOE优化获得了重组抗狂犬病病毒抗体的高水平表达,抗体表达量提高了至少50倍。  相似文献

微囊化重组基因细胞移植治疗肿瘤是一种新兴的肿瘤基因治疗方法,然而由于目前微囊化细胞规模化制备和培养技术还不成熟,阻碍了其在临床治疗中的推广与应用。以重组CHO细胞为模型,考察了不同的微囊制备和培养条件对微囊化细胞生长和内皮抑素表达的影响。实验表明,种子细胞所处的生长阶段和细胞接种密度对微囊化细胞生长和内皮抑素表达的影响较大,对数生长期的细胞进行包囊并且细胞接种密度为1×106~2×106cells/mL微囊时微囊内细胞生长良好、内皮抑素表达量高。微囊制备时间对细胞活性和内皮抑素表达也有较大的影响,制备时间延长对细胞的损伤增大,因此制备时间应控制在5h以内。生物微胶囊在制备过程中会造成细胞损伤,而体外培养是恢复细胞活性的良好方法,在培养过程中微囊接种量为5%时对细胞生长和内皮抑素表达有利。  相似文献

以悬浮适应的表达重组尿激酶原 (Pro-urokinase,pro-UK) CHO工程细胞系11G-S为对象,采用Plackett-Burman实验设计及响应面分析法,设计支持CHO工程细胞 (11G-S) 悬浮生长的无血清培养基。以细胞密度为评价指标,在单因素实验的基础上采用Plackett-Burman实验设计对影响细胞生长的培养基添加成分进行考察,确定了3种对细胞生长明显促进作用的培养基添加成分:胰岛素、转铁蛋白及腐胺。继而利用响应面法分析了这3种添加成分的最佳水平范围,设计了一种适用于CHO工程细胞 (11G-S) 悬浮培养的无血清培养基SFM-CHO-S。11G-S细胞在SFM-CHO-S批次悬浮培养的细胞最大生长密度达到4.12×106 cells/mL,pro-UK的最大累积活性达到5 614 IU/mL,培养效果优于商品化的同类无血清培养基。  相似文献

目的:通过悬浮适应,使中国仓鼠卵巢细胞(CHO细胞)获得悬浮生长的特性,并可在悬浮培养条件下较快地生长。方法:将CHO细胞以3×10^5/mL接种于100mL的三角瓶内,培养时加入1%小牛血清、1g/LPIuronic F-68、25μg/mL硫酸葡聚糖,培养体积35mL,摇床转速90r/min,每24h离心换液,当细胞增殖为2×10^6/mL时传代。结果:经过悬浮适应,细胞的平均比生长速率由适应最初的0.27/d提高为适应后的0.48/d,最大总细胞密度由适应初期的2.5×10^6/mL提高为适应后的6.3×10^6/mL,目的蛋白活性也由适应前的2781U/mL提高为适应后的8878U/mL,适应后细胞的葡萄糖平均比消耗率为1.42μmol/(10^6细胞·d),低于适应前的2.16μmol/(10^6细胞·d)。结论:贴壁生长的CHO细胞经过悬浮适应,不仅可以在悬浮培养条件下快速生长,而且细胞对葡萄糖的利用率也得到提高。  相似文献

近年来,因病毒侵害人类每年都要蒙受巨大的经济损失和社会损失。犬肾细胞MDCK以其具有的培养容易、增殖快、流感病毒感染效率高等特点,成为适用于流感病毒疫苗生产的重要细胞系之一。以MDCK细胞为研究对象,在自制无血清培养基中成功实现了MDCK细胞从有血清培养到无血清培养的驯化;并通过单细胞悬浮培养驯化过程实现了MDCK细胞的无血清单细胞悬浮培养,获得了适于无血清单细胞悬浮生长的ssf-MDCK细胞株,无血清单细胞悬浮批培养最大活细胞密度可达3.81×106 cells/mL,最大比生长速率可达0.056 h?  相似文献

目的:通过对稳定表达抗汉滩病毒鼠/人嵌合抗体的CHO细胞进行驯化,检测其抗体的表达情况,并对所表达抗体的生物学活性进行初步鉴定。方法:将本课题组前期筛选得到的稳定高表达细胞株1-D9进行无血清驯化,使其适合悬浮培养后接种到摇瓶,通过调整不同的培养条件,每天检测细胞的活率和抗体表达量,并对表达的抗体进行生物学活性鉴定。结果:通过驯化得到了适合悬浮培养的CHO细胞株,其抗体表达量高,生产周期短;并优化了其悬浮培养条件,表达的抗体经鉴定其生物学活性稳定。结论:优化了抗汉滩病毒鼠/人嵌合抗体表达条件,为下一步工业化生产提供了实验依据。  相似文献

研究以DMEM/F12(1:1 V/V)培养基为基础,添加不同添加剂优化一种适宜CHO DG44细胞生长的廉价培养基。以细胞密度和细胞活率为主要指标,对DMEM/F12(1:1 V/V)培养基进行了优化。通过正交试验和单因素试验筛选出了CHO DG44细胞生长的最佳培养基。正交试验结果表明添加8mg/L Insulin、10mg/L Transferrin、12mM Glutamine、9mg/L Ethanolamine、9mg/L Sodium selenite、0.5×Lipids、0.5×Vitamin,对细胞生长有较好促进作用,细胞密度从0.6×106 cells/mL上升到1.8×106 cells/mL。在此基础上添加2.5g/L Malt Peptone和2.5g/L YeastExtract可使细胞密度达到2.65×106 cells/mL,基本上达到商业培养基的培养效果,而成本降低了约60%。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
