拮抗菌BS—98分泌抗菌蛋白的条件及其发酵液特性   总被引:7,自引:2,他引:5

以上流式厌氧污泥床(UASB)反应器中的反硝化颗粒污泥为样品,研究了颗粒污泥的基本特性。测定出颗粒污泥中的优势无机元素为Ca、P.颗粒表面以球菌和短杆菌为主。反硝化菌是颗拉中的优势菌群,数量可达6.5x10-1.5x1010个/ml颗粒污泥。初步鉴定了两株脱氮菌Micrococcussp.strainNPseudomonasaeruginosastrain.  相似文献

将1989-1991年采集的宁夏枸杞根腐病标样进行分离、接种后证明,致病菌有:前类镰刀菌[Fusariumsolani(Mart.)Sacc.],尖孢镰刀菌[F.Oxysporumschl.],同色镰刀菌(F.concolorReinking),串珠镰刀菌(F.Moniliformesheldon)。其中后三种为国内首次报道,并对致病力最强的尖孢镰刀菌作了培养特性、形态特征、寄主范围方面的研究。同色镰刀菌和串珠镰刀菌为弱寄生菌。  相似文献

根据酵母属( Saccharomyces Meyen ex Reess) 分类学研究最新进展,核实并更新了保藏于中国普通微生物菌种保藏中心的该属菌株的种类归属。在形态和生理生化性状,包括对6 种糖的发酵能力、对18 种碳源和3 种氮源化合物的同化能力、在无维生素培养基中和37 ℃下的生长情况、对放线菌素酮的抗性等常规分类学研究的基础上,对部分疑难菌株进行了脉冲电泳核型比较分析。酿酒酵母( Saccharomycescerevisiae) 、贝酵母( S.Bayanus) 和巴氏酵母( S.Pastorianus) 三者与少孢酵母( S.Exiguus) 在电泳核型上具有明显的差异,主要表现在前三者染色体DNA 分子的大小范围均为225 ~2200 kb ,而S.Exiguus 缺少小于365 kb 的染色体DNA 分子。S.Cerevisiae 的模式和权威菌株具有12 ~14 条染色体DNA 带;S.BayanusS.Pastorianus 的模式菌株均有17 条带,但在带型上存在一定差异。原归于S.Cerevisiae 的株菌AS2-100 具有16 条带,与S.Cerevisiae 区别明显而与S.…  相似文献

铜绿假单胞菌(Pseudomonasaeruginosa)PIC-N对萘、邻苯二甲酸、水杨酸等有较强的氧化能力。发现该菌株以禁为底物可诱导产生芳香烃分解酶系。菌株中存在一个57.4kb的质粒,经限制性内切酶HindⅢ处理可产生7个片段,用限制性内切酶EcoRⅠ处理可产生8个片段。将以限制性内切酶HindⅢ部分酶切的片段克隆至大肠杆菌质粒pMFY43上,获得29个克隆株。通过对含有菌株PIC-N质粒HindⅢ片段的7个重组质粒进行限制酶分析,绘出了该质粒HindⅢ内切酶7个切点的酶切图谱。  相似文献

以质粒Puc18为载体,从枯草杆菌(Bacilussubtilis)DB104基因组中克隆到一个内切葡聚糖酶基因。限制酶切分析表明重组质粒中的插入片段为3.5kb,其中各含有一个EcoRI,HindⅢ和PvuⅡ位点。该插入片段含有一完整的内切葡聚糖酶基因,其自身启动子能被大肠杆菌转录系统所识别。当加入lacZα基因启动子的诱导物IPTG后,内切葡聚糖酶表达量提高2.8倍。加入0.5%葡萄糖能抑制该基因的表达。该基因在大肠杆菌DH5αF′中表达的内切葡聚糖酶分布为:胞外67.3%,胞间周质3.9%,胞内28.8%。Southern杂交证实了该插入片段来自供体菌B.SubtilisDB104。  相似文献

Some characteristic features of the lipid metabolism of Bacillus brevis var. G.-B. natural variants and Bacillus brevis mutant 101 were studied. The authors found that upon submerged cultivation gramicidine S-producing P+-variant and B. brevis mutant 101 synthesized higher amounts of tocopherols as compared to other colonial-morphological variants. The highest tocopherol content was observed in P+-variant, whose cells contained the highest amount of total lipids as compared to other gramicidine S-producers.  相似文献

Identification of thermophilic bacteria in solid-waste composting   总被引:14,自引:0,他引:14  
The thermophilic microbiota of solid-waste composting, with major emphasis on Bacillus spp., was examined with Trypticase soy broth (BBL Microbiology Systems) with 2% agar as the initial plating medium. Five 4.5-liter laboratory units at 49 to 69 degrees C were fed a mixture of dried table scraps and shredded newspaper. The composting plants treating refuse at Altoona, Pa., and refuse-sludge at Leicester, England, were also sampled. Of 652 randomly picked colonies, 87% were identified as Bacillus spp. Other isolates included two genera of unidentified nonsporeforming bacteria (one of gram-negative small rods and the other of gram-variable coccobacilli), the actinomycetes Streptomyces spp. and Thermoactinomyces sp., and the fungus Aspergillus fumigatus. Among the Bacillus isolates, the following, in order of decreasing frequency, were observed: B. circulans complex, B. stearothermophilus, B. coagulans types A and B, B. licheniformis, B. brevis, B. sphaericus, Bacillus spp. types i and ii, and B. subtilis. About 15% of the Bacillus isolates could be assigned to species only by allowing for greater variability in one or more characteristics than has been reported by other authors for their strains. In particular, growth at higher temperatures than previously reported was found for strains of several species. A small number of Bacillus isolates (less than 2%) could not be assigned to any recognized species.  相似文献

Aim:  To isolate and identify black pepper ( Piper nigrum L) associated endophytic bacteria antagonistic to Phytophthora capsici causing foot rot disease.
Methods and Results:  Endophytic bacteria (74) were isolated, characterized and evaluated against P. capsici . Six genera belong to Pseudomonas spp (20 strains), Serratia (1 strain), Bacillus spp. (22 strains), Arthrobacter spp. (15 strains), Micrococcus spp. (7 strains), Curtobacterium sp. (1 strain) and eight unidentified strains were isolated from internal tissues of root and stem. Three isolates, IISRBP 35, IISRBP 25 and IISRBP 17 were found effective for Phytophthora suppression in multilevel screening assays which recorded over 70% disease suppression in green house trials. A species closest match (99% similarity) of IISRBP 35 was established as Pseudomonas aeruginosa ( Pseudomonas EF568931), IISRBP 25 as P. putida ( Pseudomonas EF568932), and IISRBP 17 as Bacillus megaterium ( B. megaterium EU071712) based on 16S rDNA sequencing.
Conclusion:  Black pepper associated P. aeruginosa , P. putida and B. megaterium were identified as effective antagonistic endophytes for biological control of Phytophthora foot rot in black pepper.
Significance and Impact of the Study:  This work provides the first evidence for endophytic bacterial diversity in black pepper stem and roots, with biocontrol potential against P. capsici infection.  相似文献

The decolourisation of synthetic melanoidins (i.e., GGA, GAA, SGA, and SAA) by three Bacillus isolates (Bacillus thuringiensis (MTCC 4714), Bacillus brevis (MTCC 4716) and Bacillus sp. (MTCC 6506)) was studied. Significant reduction in the values of physicochemical parameters was noticed alongwith the decolourisation of all four melanoidins (10% v/v). B. thuringiensis (MTCC 4714) caused maximum decolourisation followed by B. brevis (MTCC 4716) and Bacillus sp. (MTCC 6506). A mixed culture comprised of these three strains was capable of decolourising all four melanoidins. The medium that contained glucose as a sole carbon source showed 15% more decolourisation than that containing both carbon and nitrogen sources. Melanoidin SGA was maximally decolourised (50%) while melanoidin GAA was decolourised least ( approximately 06%) in the presence of glucose as a sole energy source. The addition of 1% glucose as a supplementary carbon source was essential for co-metabolism of melanoidin complex. The decolourisation of synthetic melanoidin by three Bacillus spp. significantly reduced the toxicity to the tubificid worm (Tubifex tubifex, Müller).  相似文献

One hundred nineteen isolates from a commercial zucchini purée stored at 4, 10, and 20 to 25 degrees C were fingerprinted using repetitive sequence-based PCR (REP-PCR) and classified into 35 REP types. One representative isolate of each REP type was subsequently identified by API50CHB/20E profile and partial rrs gene sequence analysis. Nine REP types were misidentified by the API system. Strains were misidentified as being in the Bacillus circulans (group 2) API taxon or in taxa with a low number of positive API characters such as Brevibacillus brevis. A phylogenetic analysis pointed to one new species of Bacillus and three new species of Paenibacillus among the misidentified REP types. Bacterial components in zucchini purée were compared phenotypically with those obtained in previous work on broccoli, carrot, leek, potato, and split pea purées, based on simple matching coefficient and unweighted pair group method with averages cluster analysis. Out of 254 strains, 69 strains previously identified as B. circulans (group 2) or B. circulans/B. macerans/B. polymyxa were assigned to a new Paenibacillus taxon phylogenetically related to P. azotofixans. Storage conditions at 4 degrees C favored the development of "B. macroides/B. maroccanus" and Paenibacillus spp. in zucchini purées and Paenibacillus spp. in other purées. Storage conditions at 20 to 25 degrees C favored the development of B. subtilis group (B. licheniformis and B. subtilis) and B. cereus group strains. At 10 degrees C, Paenibacillus spp. were always present at high frequencies, whereas the occurrence of B. macroides/B. maroccanus (in zucchini purées), B. cereus, and B. pumilus varied with the experiment.  相似文献

The structural organisation of the flagellum basal body was studied in Bacillus brevis var. G.-B. P+ by electron microscopy. It was compared with that of Escherichia coli MS 1350. The basal body of a B. brevis flagellum contains, in addition to two pairs of rings on a rod, another ring-like structure (d = 13.6 nm, h = 4.3 nm) which we referred to as a "collar". The collar makes the basal body of B. brevis different from that of B. subtilis, another Gram-positive bacterium. The collar seems to fasten the flagellum of B. brevis to the cell wall. We have concluded that the basal body can differ not merely among bacterial systematic groups, but also among bacteria belonging to one and the same genus. The role of individual elements in the structure of the basal body of bacterial flagella is discussed.  相似文献

Ten years-long survey on pathogen status of mouse and rat breeding colonies   总被引:1,自引:0,他引:1  
Eleven pathogens including P. aeruginosa, Salmonella spp., E. coli O115a, c: K(B), P. pneumotropica, B. bronchiseptica, C. kutscheri, Tyzzer's organism, M. pulmonis, Sendai virus, MHV and Syphacia spp. were surveyed in 217 mouse and rat breeding colonies during 1972-1981. In conventional animals, P. pneumotropica and/or Syphacia spp. were detected in nearly 90% of 89 mouse and 64 rat colonies. Sendai virus, M. pulmonis, P. aeruginosa and MHV were positive in 51.7 to 23.6% of the colonies, and Tyzzer's organism, B. bronchiseptica and probably SDA virus were also detected in more than 10% of the rat colonies. Salmonella spp., E. coli O115a, c: K(B) and C. kutscheri were found in a few colonies. In SPF animals, P. aeruginosa was isolated from about one third of 33 mouse and 31 rat colonies, and P. aeruginosa was isolated from about one third of 33 mouse and 31 rat colonies, and P. pneumotropica was also positive in 3 rat colonies. Infection rates of P. pneumotropica, M. pulmonis, Sendai virus and Syphacia spp. were usually higher than 40% of animals sampled from colonies contaminated with them. Accidental contaminations of SPF colonies were usually caused by P. pneumotropica and Syphacia spp.  相似文献

Bacillus subtilis and Bacillus brevis 47-5, carrying the Bacillus stearothermophilus alpha-amylase gene on pUB110 (pBAM101), synthesized the same alpha-amylase as the donor strain as determined by the enzyme's thermal stability and NH2-terminal amino acid sequence. Regardless of the host, the 34-amino acid signal peptide of the enzyme was processed at exactly the same site between two alanine residues. B. brevis 47-5(pBAM101) secreted the enzyme most efficiently of the hosts examined, 100, 15, and 5 times more than B. stearothermophilus, Escherichia coli HB101(pH1301), and B. subtilis 1A289(pBAM101), respectively. The efficient secretion of the enzyme in B. brevis 47-5(pBAM101) was suggested to be due to the unique properties of the cell wall of this organism.  相似文献
