戊糖乳杆菌(Lactobacillus pentosus)是能利用木质纤维素水解液发酵产乳酸的潜力菌株,发酵条件优化与高产菌株的选育是提高乳酸产量的重要手段。通过单因素试验、Plackett-Burman设计与响应面试验,对戊糖乳杆菌ATCC 8041产乳酸的发酵培养基及发酵条件进行了优化。结果表明,该菌株发酵培养基的最佳组合为葡萄糖93.11 g/L、酵母浸粉5.19 g/L、碳酸钙29.43 g/L、蛋白胨10.00 g/L、Na2HPO4·12H2O 5.00 g/L、Mg SO4 0.20 g/L、Mn SO4 50 mg/L;最佳发酵条件为37℃、p H6.5、接种量6%、装液量80%。在此优化条件下,该菌株发酵产乳酸为54.12 g/L。进一步以戊糖乳杆菌ATCC 8041为出发菌株,通过原生质体进行紫外诱变,经多重筛选,最终获得一株遗传稳定性好的高产乳酸突变株,命名为戊糖乳杆菌Lactic UVC-02,由中国典型培养物保藏中心保存,注册号为CCTCC M 2013209。该突变株Lactic UVC-02经葡萄糖发酵,乳酸产量达64.17 g/L,比出发菌株ATCC 8041(54.12g/L)提高18.6%。  相似文献

目的从生产实际出发,对1株高效乳酸杆菌(Lactobacillus spp)LH进行液体发酵培养基优化及发酵条件研究。方法通过碳源、氮源、无机盐、促生长素等单因子筛选及正交试验设计获得以下最佳培养基:糖蜜12 g/L,酵母膏5 g/L,蛋白胨1 g/L,葡萄糖4 g/L,玉米浆3 g/L,乙酸钠5 g/L,NaC l 5 g/L,K2HPO42.5 g/L,KH2PO42.5 g/L,MgSO40.5g/L,MnSO40.25 g/L。在此培养基上研究了该菌株最佳发酵条件。结果培养基初始pH 6.0,接种量2%(v/v,相对装液量),500 m l三角瓶中装液量为500 m l,发酵温度为30~35℃,静置培养。在最佳培养条件下,LH活菌量达到1.74×10^9CFU/m l。结论通过活菌平板计数法测定了乳酸杆菌LH生长曲线,24 h为最佳种龄,生产收获时间是36 h。  相似文献

从全省各处采集的50多份土样筛选到一株产腈水合酶能力较高的菌株E10a,该菌株产生的腈水合酶为非诱导酶,产酶条件优化实验结果表明,产酶培养基组成:麦芽糖20 g/L,酵母膏5 g/L,尿素7.5 g/L,味精0.75 g/L,K2HPO40.5 g/L,KH2PO40.5 g/L,MgSO40.5 g/L,FeSO4.7H2O 10 mg/L,CoCl210 mg/L,微量元素母液0.8 ml/L;最佳培养条件为:培养温度28℃,摇床转速150 r/min,培养基起始pH 7.0,培养时间5 d,在优化培养条件下,1 h可将1 g/L质量浓度的底物对羟基苯乙腈全部转化为对羟基苯乙酰胺。  相似文献

以生物柴油生产的高浓度副产物甘油为唯一碳源筛选甘油高耐受性1,3-二羟基丙酮(DHA)高产菌株,运用响应面与正交试验优化菌株产DHA条件,提高DHA产量。分子生物学鉴定表明:筛选的高产DHA菌种G40为芽胞杆菌属(Bacillus)菌株,DHA产量为29.46g/L。响应面分析和正交试验优化后,在甘油224.22g/L、K_2HPO_41.60g/L、NaCl0.5g/L、KH_2PO_40.5g/L、(NH_4)_2SO_40.5g/L、酵母膏1.60g/L和pH7.2、35℃、200r/min的条件下,G40菌株发酵60h产生DHA86.84g/L,比优化前提高了194.8%。实验建立了一种利用高浓度甘油高效率发酵生产DHA的方法。  相似文献

目的:对海洋来源的具有产纤溶酶能力的枯草芽孢杆菌(Bacillus subtilis)LC6-1进行紫外诱变,得到高产且稳定的突变株PW6-3,对该突变株发酵产酶的条件进行优化。方法:采用单因素和正交试验进行发酵培养基组分和培养条件的优化。结果:突变株PW6-3的酶活力为(6 960.21 ± 85.51)U/mL,较原始菌株提高了30.48%。以PW6-3为出发菌株,采用单因素及正交试验的方法对菌株进行发酵培养基组分与培养条件优化,最终得到的最佳培养基组分是:玉米淀粉30 g/L,玉米浆干粉40 g/L,CaCl2 3 g/L;最佳发酵培养条件是:32℃,转速200 r/min,接种量3%,pH 6.5,种龄18 h,发酵培养时间66 h,最终菌株的酶活力稳定在(9 203.63 ± 67.85)U/mL。结论:发酵工艺优化后,菌株PW6-3纤溶酶产量较诱变之前的菌株LC6-1提高72.53%,且发酵工艺成本较低,具有较好的经济效益。  相似文献

对拉曼被孢霉突变株F5发酵生产γ-亚麻酸的最适碳源,氮源,发酵时间及湿度,无机盐离子的添加,最适碳源浓度及补加碳源时间等发酵条件进行了研究探讨,最适发酵培养基组成为(g/L):葡萄糖100,酵母浸出粉4,蛋白胨1,K2HPO41,CaCl2 1*10^-2,MgSO4 5*10^-2,FeSO4 1*10^-2,ZnSO4 7.5*10^-3,CuSO40.5*10^-3,MnSO4 2*10^-3,pH6.0,培养湿度为25度,140rpm 振荡培养10天,培养8天后(即收获前2天)补加5%葡萄糖,发酵结果为:DC24.59 g/L,TL10.84g/L,TL/DC 44.09%,GLA/TL10.67%,GLA产量为1156.63mg/L,GLA产量较初始结果提高156.15%,该菌株已达到工业化生产菌株要求。  相似文献

目的:以谷氨酸棒杆菌TQ2223(Phe-/Tyr-/5-MTr/SGr/5-FTr/CINr)为出发菌株,定向选育具有5-甲基色氨酸抗性(5-MTr)、磺胺胍抗性(SGr)、5-氟色氨酸抗性(5-FTr)、8-氮鸟嘌呤抗性(8-AGr)、6-巯基嘌呤抗性(6-MPr)、2-噻唑丙氨酸抗性(2-TAr)等遗传标记的突变株;同时对突变株发酵培养基及条件进行研究,获得最优条件。方法:经硫酸二乙酯诱变处理,测定了诱变时间与致死率的关系,并对发酵培养基中不同氮源、生物素添加量进行了单因素实验,对接种量、发酵培养温度等发酵条件也进行了实验确定。结果:经诱变处理后,定向选育出的菌株TL1105(5-MTr/SGr/5-FTr/8-AGr/6-MPr/2-TAr)在未经优化的摇瓶发酵条件下,L-组氨酸的产量为12.02g/L;而优化培养条件后,L-组氨酸的产量达23~24g/L。结论:确定最佳诱变时间30min,此时致死率为80%。硫酸铵为发酵培养基中最适碳源,生物素添加量为50μg/L,采用5%接种量为宜,组氨酸发酵的最适温度为30℃。  相似文献

在摇瓶发酵条件下,优化提高短短芽胞杆菌ch2-22芽胞浓度和抑菌活性的发酵培养基和培养条件。首先在单因素试验基础上进行响应面设计对ch2-22的发酵培养基进行优化,然后使用单因素试验方法确定最佳发酵条件,得到优化发酵培养基为淀粉35.05 g/L,豆饼粉26.08 g/L,蔗糖10 g/L,鱼粉5 g/L,Na Cl 1 g/L,Mg SO40.3 g/L,(NH4)2SO43 g/L,Mn SO40.1 g/L,K2HPO43 g/L,酵母膏1 g/L,Ca CO32 g/L。培养条件为温度32℃,转速180 r/min,装液量100 m L/500 m L,接种量4%,初始pH为7.0,发酵时间45 h。优化后的芽胞数量达到8.1×109cfu/m L,与优化前的芽胞数量(1.6×109cfu/m L)相比,提高了3.7倍,优化后发酵液效价达到3 350 IU/m L,提高了109%,高于同类菌株的2 000 IU/m L。  相似文献

以已筛选的1株产γ-多聚谷氨酸解淀粉芽胞杆菌C1为出发菌株,对其进行紫外线-亚硝基胍(NTG)复合诱变,并运用单因素试验和正交试验设计对诱变菌株的种子培养工艺进行优化。通过复合诱变选育得到1株能够稳定遗传的正突变菌株C1-6,其摇瓶发酵生产γ-PGA的产量由18.4 g/L提高到24.2 g/L,增加了31.5%,且传代8次后仍能保持稳定。通过单因子试验筛选到玉米粉和黄豆粉作为C1-6生长的C源和N源。正交试验后,C1-6在成分为K2HPO41.0 g/L、Mg SO40.5 g/L、黄豆粉15.0 g/L、玉米粉5.0 g/L,p H 6.5的培养基中,37℃、装液量1/5(150 m L三角瓶装液30 m L)的培养条件下可获得较大的生物量,OD600达到6.31。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献
