用日立835-50型氨基酸分析仪对“增收宝-3”中游离的氨基酸进行了分析。结果表明其中含有15种氨基酸,以脯氨酸含量最高。实验结果揭示,氨基酸可能是“增收宝-3”中抗逆增产的有效成分。  相似文献

首次报道用乙醇系统分析PTC-氨基酸的新方法中各衍生物获得最佳分离的建立过程。PTC-氨基酸衍生后溶于A溶液,然后进样于4μmNovaPakC18柱(3.9mm×150mm)。系统的优化步序包括全面调控流动相的pH值与TEA浓度、乙醇梯度程序、柱温等诸多影响HPLC色谱行为的因素。最适条件为:A溶液含0.14MG酸钠、0.75ml/LTEA、PH6.35;B溶液为100%乙醇;柱温30℃。通过优化的乙醇梯度最终在约44min内将15种PTC-氨基酸很好地分离。此法可用于替代代表新科技水平的PTC-氨基酸乙腈分析系统。  相似文献

从我国东北林区死者脑组织中,分离到蜱传脑炎病毒HLJ-1株,测定了它的E蛋白基因序列和推导的氨基酸序列,以及E蛋白抗原位点反应图谱。将该株与远东亚型Sofyn株和中欧亚型Neuderfl株做了同源性比较,HLJ-1株与Sofyn株E蛋白基因的核苷酸及氨基酸的同源性分别为94.3%和98.8%;与Neudoerfl的核苷酸和氨基酸的同源性分别为83.7%和96%。在E蛋白的核苷酸序列中,HLJ-1和  相似文献

对71例癫痫患者血浆中18种游离氢基酸与36例正常人血浆中游离氨基酸的水平对照。分析表明,抑制性氨基酸(γ-氨基丁酸、甘氨酸)水平明显升高的癫痫患者,其它氨基酸水平绝大多数低于正常人;兴奋性氨基酸(谷氨酸、天门冬氨酸、丙氨酸、谷氨酰胺)水平明显升高的癫痫患者,其它氨基酸水平低于正常人。  相似文献

为研究褪黑素对低氧引起大鼠大脑皮层脑片氨基酸释放变化的影响,利用反相高效液相色谱结合荧光检测法,测定了孵育液中氨基酸类神经递质的含量。低氧条件为通入91.6%N2和8.4%O2的混合气体。低氧30min时,大鼠大脑皮层脑片孵育中,氨基酸类神经递质天冬氨酸、谷氨酸、谷氨酰胺、甘氨酸、牛磺酸和γ-氨基丁酸的含量显著增加,其含量分别是正常氧组的240.4%,334.3%,200.6%,210.4%,16  相似文献

ABSTRACT

The high stereo- and substrate specificities of enzymes have been utilized for micro-determination of amino acids. Here, I review the discovery of l-Phe dehydrogenase and its practical use in the diagnosis of phenylketonuria in more than 5,400,000 neonates over two decades in Japan. Screening and uses of other selective enzymes for micro-determination of amino acids have also been discussed. In addition, novel enzymatic assays with the systematic use of known enzymes, including assays based on a pyrophosphate detection system using pyrophosphate dikinase for a variety of l-amino acids with amino-acyl-tRNA synthetase have been reviewed. Finally, I review the substrate specificities of a few amino acid-metabolizing enzymes that have been altered, using protein engineering techniques, mainly for production of useful chemicals, thus enabling the wider use of natural enzymes.  相似文献

Freshwater pulmonate snails (Biomphalaria glabrata), pre-treated under bacteriostatic conditions, were incubated in 10 ml of standard medium containing various U-14C-labelled amino acids at concentrations of 10 μM. Measurements of mass-specific accumulation rates (MSARs) based on HPLC and the accumulation of U-14C-labelled amino acids into snail tissues have shown unequivocally for the first time that freshwater snails achieved a net accumulation of all the amino acids tested, including aminoisobutyrate (AIB), aspartate, alanine and a mixture of 13 amino acids. There were no significant differences between the MSAR values determined by HPLC from those based on the use of radiolabelled amino acids, whereas MSAR values for control snails were negligible and significantly less. Incubation of snails in media containing radiolabelled aspartate and a mixture of amino acids showed that the accumulated amino acids were readily distributed through the snail’s tissues and then metabolized. The ecological and biochemical questions arising from the fact that freshwater snails are capable of net accumulation of exogenous amino acids at naturally occurring concentrations and subsequent metabolic conversion, contrary to widely held views, are addressed.  相似文献

The mechanism and specificity of amino-acid transport at the plasma membrane of Ricinus communis L. roots was investigated using membrane vesicles isolated by phase partitioning. The transport of glutamine, isoleucine, glutamic acid and aspartic acid was driven by both a pH gradient and a membrane potential (internally alkaline and negative), created artificially across the plasma membrane. This is consistent with transport via a proton symport. In contrast, the transport of the basic amino acids, lysine and arginine, was driven by a negative internal membrane potential but not by a pH gradient, suggesting that these amino acids may be taken up via a voltage-driven uniport. The energized uptake of all of the amino acids tested showed a saturable phase, consistent with carrier-mediated transport. In addition, the membrane-potential-driven transport of all the amino acids was greater at pH 5.5 than at pH 7.5, which suggests that there could be a direct pH effect on the carrier. Several amino-acid carriers could be resolved, based on competition studies: a carrier with a high affinity for a range of neutral amino acids (apart from asparagine) but with a low affinity for basic and acidic amino acids; a carrier which has a high affinity for a range of neutral amino acids except isoleucine and valine, but with a low affinity for basic and acidic amino acids; and a carrier which has a higher affinity for basic and some neutral amino acids but has a lower affinity for acidic amino acids. 野生蔬菜具有独特的口味,丰富的营养,有些还具有防病治病的功能,对人体有良好的保健作用.白子菜[Gynura divaricata (L.) DC.]茎叶有消热、舒筋及止血祛瘀的作用,用于镇咳、风湿性关节痛、骨折、创伤止血,痈肿疮疥等症,在民间还作抗癌草使用[1].红凤菜(Gynura bicolor DC.)茎叶有活血止血,解毒消肿的功效,治痛经,血崩,咳血,创伤出血,溃疡久不收口[2].诸葛菜[Orychophragmus violaceus (L.) Schulz]作为野生蔬菜在江苏、安徽民间有很长的食用历史,测定结果表明,该种含有黄酮类化合物[3].马齿苋(Portulaca oleracea L.)具有清热解毒、消炎、利尿、消肿的功效,民间还用于白发症、糖尿病、心血管疾病的治疗[4],中国卫生部将它列为药食同用的植物.  相似文献

白芷是我国常用中药材的一种,关于白芷镇痛的有效成分,主要认为是异欧前胡素、欧前胡素等香豆素类具有解痉、镇痛等作用的挥发油类化学成分,但白芷镇痛的成分及其机理尚不十分清楚,是否有其他物质也具有一定的药理作用也不得而知,比如说某些药效性氨基酸。该研究采用自动氨基酸分析仪测定了湖南道地茶陵白芷的蛋白质类氨基酸和游离氨基酸的含量,并分析其氨基酸组成。结果表明:总氨基酸检出除Asn以外的其他16种蛋白质氨基酸,含量最高的Arg占总氨基酸的31.21%,接近1/3;必需氨基酸总量达27.01%,其中含量最高的是Leu,占总必需氨基酸的24.14%;药效氨基酸比例较高,在酸水解产物总氨基酸和游离氨基酸中分别达到73.89%和85.78%,其中Arg的含量最高,达1.383g·100g-1,占比为42.24%,游离氨基酸中γ-氨基丁酸、鸟氨酸含量也较高。此外,还含有少量的高半胱氨酸和鹅肌肽等游离非蛋白质氨基酸和短肽;必需氨基酸的组成接近WHO/FAO的建议摄入值,但Met+Cys的RC值最小,为第一限制性氨基酸。这些药效性氨基酸和游离氨基酸可能是茶陵白芷具有良好药效的一个因素,而且茶陵白芷的氨基酸组成和配比较合理、符合人体需要,在强化含硫氨基酸的基础上具有开发成为新型药食同源保健性食品的潜力。研究结果可为进一步研究白芷的药理作用提供依据。  相似文献

The first step of the utilization of the aromatic amino acids as sole nitrogen sources by Brevibacterium linens strain 47 was found to be a transamination. The deaminated metabolites of the amino acids were detected in culture supernatants, and the enzyme activity was identified in cell free extracts. The cells contained increased aromatic amino acid aminotransferase activities on growth on the aromatic amino acids as sole nitrogen sources. Two aromatic aminotransferases (AT-I and AT-II) were separated upon diethylaminoethyl-Trisacryl M column chromatography of cell free extracts. Only AT-I was responsible for the increased level of aromatic amino acid aminotransferase activity of induced cells. In this article, we advocate the radical revision of the 20th-century version of amino acid metabolism as follows. (1) Classic studies on the incorporation of [15N]ammonia into glutamate, once considered to be an epoch-making event, are not distinctive proof of the ability of animals to utilize ammonia for the synthesis of alpha-amino nitrogen. (2) Mammalian glutamate dehydrogenase has been implicated to function as a glutamate-synthesizing enzyme albeit lack of convincing proof. This enzyme, in combination with aminotransferases, is now known to play an exclusive role in the metabolic removal of amino nitrogen and energy production from excess amino acids. (3) Dr. William C Rose's "nutritionally nonessential amino acids" are, of course, essential in cellular metabolism; the nutritional nonessentiality is related to their carbon skeletons, many of which are intermediates of glycolysis or the TCA cycle. Obviously, the prime importance of amino acid nutrition should be the means of obtaining amino nitrogen. (4) Because there is no evidence of the presence of any glutamate-synthesizing enzymes in mammalian tissues, animals must depend on plants and microorganisms for preformed alpha-amino nitrogen. This is analogous to the case of carbohydrates. (5) In contrast, individual essential amino acids, similar to vitamins and essential fatty acids, should be considered important nutrients that must be included regularly in sufficient amounts in the diet.  相似文献

为了弄清不同闽产李(Prunus salicina)果实氨基酸含量、组成及营养价值,采用氨基酸自动分析仪 对不同李果实样品中各种氨基酸的含量进行检测,并对其组成进行比较分析。结果表明,李果实含有18种蛋白质氨基酸和3种非蛋白质氨基酸。田黄李的牛磺酸含量与蛋白质氨基酸含量均最高分别为26.54 mg/100 g和496.14 mg/100 g;皇后李的鸟氨酸、药用氨基酸、酸味氨基酸含量以及特殊功效蛋白质氨基酸的比重均最高分别为0.36 mg/100 g、364.67 mg/100 g、298.28 mg/100 g和73.45%;早熟胭脂李的γ-氨基丁酸含量最高为7.98 mg/100 g;大胭脂李的支链氨基酸含量最高为46.77 mg/100 g。李果实中氨基酸种类齐全、含量存在一定的差异,可根据不同的开发利用目标选择合适的品种。  相似文献
