He-Ne激光和增强UV-B辐射对小麦幼苗类囊体捕光色素的影响   总被引:2,自引:1,他引:1  
采用5mW.mm-2He-Ne激光辐照、10.08kJ.m-2d-1UV-B辐射及二者组合对冬小麦幼苗进行处理。通过测定叶绿体捕光色素含量和色素蛋白组成的变化,进一步探讨He-Ne激光对增强UV-B辐射后小麦幼苗类囊体捕光色素损伤的修复效应。循环处理小麦幼苗4d,利用90%乙醇和80%丙酮分别提取各处理组小麦幼苗叶片中的叶绿素,通过纸层析和分光光度法检测捕光色素含量的变化,并探讨不同处理对叶绿素与蛋白质结合牢固性的影响。利用柱层析法测定色素蛋白的主要成分。研究表明:与对照组相比,增强UV-B辐照后小麦幼苗捕光色素总含量降低了17.76%,叶绿素和蛋白质结合牢固度显著降低,色素蛋白的组成也发生变化,D1和D2蛋白质条带消失;而一定剂量He-Ne激光辐照可使增强UV-B辐射后的叶绿体色素含量增加约10.64%,但仍低于ck组约8.12%,叶绿素和蛋白质结合牢固度也显著高于B组,色素蛋白的组成与对照组相似。因此,低剂量的He-Ne激光辐照对增强UV-B辐射后小麦幼苗类囊体捕光色素的损伤具有促进修复效应。  相似文献

在临床应用中,低功率He-Ne激光(632.8 nm)能促进骨骼肌修复,加速创伤愈合,降低牙齿的超敏感性,减缓疼痛等.大量研究表明:低功率He-Ne激光能调节细胞的众多行为,如细胞增殖、分泌、迁移、粘附、蛋白质合成和基因表达等.但低功率He-Ne激光调节细胞行为的分子机制并未阐明,考察低功率He-Ne激光照射后细胞内活性氧自由基的产生水平和游离ca2 浓度是否会发生变化,通过激光扫描共聚焦显微镜,分别利用H:DCFDA和Fluo-3/AM这两种荧光探针,检测到经He-Ne激光照射后,肺腺癌细胞内活性氧自由基的水平上调以及游离Ca2 浓度增加.该研究为低功率He-Ne激光的生物光刺激效应提供了可能的分子机理.  相似文献

The aim of this study was to investigate the effects of melatonin on rat gastric fundus smooth muscle. Melatonin (10(-4) to 10(-3) M) had no effect on the basal tone of gastric smooth muscle. After precontraction with carbachol (10(-6) M) or serotonin (10(-7) M), melatonin caused a concentration dependent inhibitory action. The half maximal effect on serotonin-induced contraction was found with 1.12 +/- 0.86 x 10(-5) M of melatonin. Increasing concentrations of melatonin (10(-5) to 10(-3) M) resulted in a right shift of the serotonin concentration response curve (10(-10) to 10(-5) M). This inhibitory effect of melatonin was partially blocked in the presence of apamin (10(-10) to 10(-7) M), a specific blocker of the small conductance calcium-dependent potassium channel, but not in the presence of other potassium channel blockers like charybdotoxin (10(-8) M), glibenclamide (l0(-5) M), or tetraethylammonium (ODQ, 10(-4) M). The inhibitory effect was not changed in the presence of the neuronal blocker tetrodotoxin (10(-6) M), the selective P2-receptor antagonist pyridoxalphosphate-6-azophenyl-2',4'-disulphonic acid (3 x 10(-5) M), the nitric-oxide synthase inhibitor N-nitro-L-arginine (3 x 10(-4) M), or the guanylate cyclase inhibitor 1H-[1,2,4]oxadiazolo[4,3-a]-quinoxalin-1-one (10(-4) M), suggesting that neither the purinergic, nitrergic, nor guanylate cyclase pathways were involved. We further investigated inhibitory responses to electrical field stimulation (EFS) at different frequencies under non-adrenergic, non-cholinergic (NANC) conditions on a serotonin-induced contraction in the presence of melatonin (10)-5 to 10(-4) M). Melatonin significantly reduced these inhibitory NANC responses in higher (8-32 Hz), but not lower (05-4 Hz), frequencies (16 Hz without melatonin, 103 +/- 6.3%; melatonin 10(-5) M, 80.4 +/- 7.5%; melatonin 10(-4) M, 39.1 +/- 17.1%). Melatonin had no effect on contractile responses induced by EFS under basal tone. These results demonstrate that the inhibitory effect of melatonin in rat gastric fundus smooth muscle is apamin sensitive, but is not affected by other potassium channel blockers. This suggests that melatonin may be another transmitter candidate for the apamin sensitive responses within the gastrointestinal tract.  相似文献

A monolayer of HeLa cells, at the stationary phase of growth, exposed to He-Ne laser radiation (632.8 nm; 100 J/m2) either 5 min or 60 min prior to gamma irradiation (0.1-10 Gy; 6.75 Gy/min), or 5 min after irradiation has been investigated. With a 5-min interval between irradiation sessions (both sequences) the survival curves are virtually the same as those for gamma-irradiated cells only. With He-Ne laser radiation delivered 60 min before gamma irradiation with doses exceeding 5 Gy, a fraction of radioresistant cells is identified whose D0 is almost twice as high as D0 of basic cell mass (3.6 and 1.7 Gy respectively. The survival curve becomes a two-component one. A hypothesis is proposed that He-Ne laser radiation activates, in some cells, the processes that promote the repair of radiation damages.  相似文献

为研究弱激光照射对人血液携氧能力的影响及机制,我们用荧光仪分别测量了He-Ne激光照射前后正常血液及其组分(血浆、红细胞)的荧光光谱,研究了激光照射导致的光谱变化,并分析了光谱变化与血液携氧能力改变的关系。实验结果显示:全血液标本在490nm及614nm附近有荧光峰值;血浆的荧光则主要分布在420-500nm之间;红细胞在500nm及614nm附近有荧光。He-Ne激光照射后,全血液及红细胞在614nm处的荧光谱都有较明显的变化,且较相似。由此可得出结论,He-Ne激光照射可影响血液的携氧能力。  相似文献

Photoinduced changes of human ceruloplasmin (hCp) under laser and ultraviolet (UV) radiation have been studied. A complex character of hCp spectral modifications points to a different degree of the protein molecule folding. After the influence of UV radiation (240-390 nm) in a dose range 906-4530 J/m2 the decrease of oxidase hCp activity was registered. These changes are thought to be connected with modification of the conformation of hCp active centre or copper reduction that was confirmed by the decrease of optical density at 610 nm. An inactivation constant (kf = 3.8 x 10(-4) 1/s) and a photoinactivation quantum yield (1.4 x 10(-4)) were calculated. Kinetic dependencies of o-phenilen-diamine oxidation by intact and UV-modified glycoprotein do not correspond to Michaelis-Menten equation. UV radiation in a dose range 151-4530 J/m2 induced increasing of ceruloplasmin superoxiddismutase activity. The influence of He-Ne laser radiation (1.3 mW; lambda = 632.8 nm, exposure time from 1 up to 30 min) leads to the raise of the hCp functional activity; superoxiddismutase activity changes more significantly. Possible photophysical and photochemical processes in hCp molecule, leading to photomodification and changes of protein functional activity have been represented on the scheme.  相似文献

本研究观察到10-7~10-5kg/L脂多糖(lipopolysacharide,LPS)可显著促进血管平滑肌细胞(VSMC)的增殖及DNA的合成(P<005)。5×10-4~10-3kg/LLPS却抑制VSMC的增殖及DNA的合成,降低其活力(P<001),并呈时间依赖效应。一氧化氮合酶抑制剂NNitroLArginine(LNNA)可拮抗LPS的抑制作用。大剂量LPS作用组VSMC上清液中一氧化氮(NO)代谢产物NO-3和NO-2的含量与对照组相比显著增加(P<001),48h组比24h组增加91%,72h组比48h组增加45%;同时,诱导性一氧化氮合酶(inductivenitricoxidesynthase,iNOS)免疫组化染色呈阳性。结果表明,低浓度LPS促进VSMC增殖和DNA合成,而高浓度LPS却明显抑制VSMC增殖和DNA合成,降低其活力。这种抑制作用可能与LPS诱导VSMC产生的NO有关。  相似文献

Candida tropicalis isolated from acclimated activated sludge was used in this study. Cell suspensions with 5 x 10(7) cells ml(-1) were irradiated by using a He-Ne laser. After mutagenesis, the irradiated cell suspension was diluted and plated on yeast extract-peptone-dextrose (YEPD) medium. Plates with approximately 20 individual colonies were selected, and all individual colonies were harvested for phenol biodegradation. The phenol biodegradation stabilities for 70 phenol biodegradation-positive mutants, mutant strains CTM 1 to 70, ranked according to their original phenol biodegradation potentials, were tested continuously during transfers. Finally, mutant strain CTM 2, which degraded 2,600 mg liter(-1) phenol within 70.5 h, was obtained on the basis of its capacity and hereditary stability for phenol biodegradation. The phenol hydroxylase gene sequences were cloned in wild and mutant strains. The results showed that four amino acids were mutated by irradiation with a laser. In order to compare the activity of phenol hydroxylase in wild and mutant strains, their genes were expressed in Escherichia coli BL21(DE3) and enzyme activities were spectrophotometrically determined. It was clear that the activity of phenol hydroxylase was promoted after irradiation with a He-Ne laser. In addition, the cell growth and intrinsic phenol biodegradation kinetics of mutant strain CTM 2 in batch cultures were also described by Haldane's kinetic equation with a wide range of initial phenol concentrations from 0 to 2,600 mg liter(-1). The specific growth and degradation rates further demonstrated that the CTM 2 mutant strain possessed a higher capacity to resist phenol toxicity than wild C. tropicalis did.  相似文献

采用He-Ne激光(5 mW/mm2)辐照增强UV-B辐射(10.08 kJ/m2.d)的晋麦8号小麦幼苗,通过测定小麦幼苗叶片细胞质膜透性、丙二醛(MDA)的含量以及脂氧合酶(LOX)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)和谷光苷肽过氧化物酶(GPX)的活性变化,研究He-Ne激光对增强UV-B辐射的小麦幼苗膜脂过氧化的影响。结果显示,He-Ne激光辐照可使经UV-B辐射后小麦幼苗叶片质膜相对透性、MDA含量、LOX活性降低,而使CAT、APX和GPX的活性均升高。分析表明UV-B辐射增强可导致膜脂过氧化加剧,而一定剂量的He-Ne激光能够通过促进酶促抗氧化系统酶的活性来缓解紫外线辐射下小麦幼苗的膜脂过氧化作用。  相似文献

The effect of He-Ne laser (632.8 nm) pre-irradiation on UVA (343 nm)-induced DNA damage in the human B-lymphoblast cell line NC37 was investigated using the comet assay. He-Ne laser pre-irradiation was observed to result in a dose-dependent decrease in UVA-induced DNA damage. This effect was also found to be dependent on the incubation period between He-Ne laser pre-irradiation and the UVA exposure. Whereas the control cells with a higher DNA damage point to an initial ability of faster repair, both the control and the He-Ne laser pre-irradiated cells subsequently show the same rate of DNA repair. The results suggest that He-Ne laser irradiation protect the cells from UVA-induced DNA damage primarily through an influence on processes that prevent an initial DNA damage. 1. Porcine cardiac native tropomyosin was phosphorylated by bovine cardiac 3':5'-cyclic AMP-dependent protein kinase. Most of the phosphate incorporation was observed in troponin I, the maximum of which was 0.7 mol of Pi per mol of troponin I. 2. In the presence of phosphorylated native tropomyosin, actomyosin ATPase activity was 15-40% lower than that in the presence of the unphosphorylated preparation at all calcium ion concentrations (1.5 x 10(-8) M-2.4 x 10(-5) M). Half-maximum activation of ATPase was obtained with a concentration of 7 x 10(-7) M Ca2+ (unphosphorylated) and 1.3 x 10(-6) M Ca2+ (phosphorylated), respectively. Maximum ATPase activity was reached with 3 x 10(-6) M Ca2+ (unphosphorylated) and 1.0 x 10(-5) M Ca2+ (phosphorylated). 3. Porcine cardiac troponin I isolated by affinity chromatography inhibited ATPase activity of desensitized actomyosin in the presence of tropomyosin. There was little difference between phosphorylated troponin I and a control preparation with regard to the inhibitory effect of ATPase activity. 4. Troponin C from rabbit skeletal muscle neutralized the inhibitory effect of troponin I. The minimum amount of troponin C required for complete neutralization was approximately equimolar to troponin I. The inhibitory effect of phosphorylated troponin I was neutralized by troponin C less effectively than that of unphosphorylated preparation.  相似文献

用He-Ne激光(5.23×10-3W.mm-2)处理经不同浓度PEG6000胁迫的小麦幼苗,研究水分胁迫条件下激光辐照对小麦幼苗生理特性的影响。结果表明,He-Ne激光辐照可显著提高水分胁迫下小麦幼苗的根系活力以及抗坏血酸过氧化物酶(APX)和谷胱甘肽还原酶(GR)活性(P<0.05),并使其脯氨酸含量显著增加(P<0.05),而对其超氧化物歧化酶(SOD)的活性没有显著影响。可见,He-Ne激光可通过提高根系活力、抗氧化酶活性和脯氨酸含量来显著增强小麦幼苗的抗旱性。  相似文献

Pulmonary veins (PV) make a significant contribution to total pulmonary vascular resistance. We investigated the cellular mechanisms by which the intravenous anesthetics propofol and thiopental alter adenosine triphosphate-sensitive potassium (KATP+) channel relaxation in canine PV. The effects of KATP+ channel inhibition (glybenclamide), cyclooxygenase inhibition (indomethacin), nitric oxide synthase inhibition (L-NAME), and L-type voltage-gated Ca2+ channel inhibition (nifedipine) on vasorelaxation responses to levcromakalim (KATP+ channel activator) alone and in combination with the anesthetics were assessed. The maximal relaxation response to levcromakalim was attenuated by removing the endothelium and by L-NAME, but not by indomethacin. Propofol (10(-5), 3x10(-5), and 10(-4) M) and thiopental (10(-4) and 3x10(-4) M) each attenuated levcromakalim relaxation in endothelium-intact (E+) rings, whereas propofol (3x10(-5) and 10(-4) M) and thiopental (3x10(-4) M) attenuated levcromakalim relaxation in endothelium-denuded (E-) rings. In E+ rings, the anesthesia-induced attenuation of levcromakalim relaxation was decreased after pretreatment with L-NAME but not with indomethacin. In E-strips, propofol (10(-4) M) and thiopental (3x10(-4) M) inhibited decreases in tension and intracellular Ca2+ concentration ([Ca2+]i) in response to levcromakalim, and these changes were abolished by nifedipine. These findings indicate that propofol and thiopental attenuate the endothelium-dependent component of KATP+ channel-induced PV vasorelaxation via an inhibitory effect on the nitric oxide pathway. Both anesthetics also attenuate the PV smooth muscle component of KATP+ channel-induced relaxation by reducing the levcromakalim-induced decrease in [Ca2+]i via an inhibitory effect on L-type voltage-gated Ca2+ channels.  相似文献

分别采用5 mW/mm2 He-Ne激光辐照、10.08 kJ/m2·d 增强UV-B辐射及二者组合对"临优2018"小麦幼苗进行处理,通过紫外吸收法测定各处理组幼苗中微管结合蛋白MAP65s的含量,并且采用SDS-聚丙烯酰胺凝胶电泳对MAP65s蛋白进行鉴定,进一步分析He-Ne激光对增强UV-B辐射引起的小麦幼苗微管骨架损伤的修复效应.结果显示:经UV-B处理(B)后,小麦幼苗中的MAP65s蛋白含量低于对照组(CK)(P<0.01);单独激光(L)处理,MAP65s蛋白含量高于对照组(CK)(P<0.05);经He-Ne激光和UV-B复合处理(BL)后,MAP65s蛋白含量高于UV-B处理组,低于CK组.该结果表明:一定剂量的He-Ne激光辐照能够促进小麦幼苗微管结合蛋白MAP65s的合成,并且可能在一定程度上修复增强UV-B辐射对小麦幼苗微管骨架系统造成的损伤.  相似文献

The mammalian Kv4.3 potassium channel is a fast activating and inactivating K+ channel widely distributed in mammalian tissues. Kv4.3 is the major component of various physiologically important currents ranging from A-type currents in the CNS to the transient outward potassium conductance in the heart (I(to)). Here we show that the KCNE3 beta-subunit has a strong inhibitory effect on current conducted by heterologously expressed Kv4.3 channels. KCNE3 reduces the Kv4.3 current amplitude, and it slows down the channel activation and inactivation as well as the recovery from inactivation. KCNE3 also inhibits currents generated by Kv4.3 in complex with the accessory subunit KChIP2. We find the inhibitory effect of KCNE3 to be specific for Kv4.3 within the Kv4 channel family. Kv4.3 has previously been shown to interact with a number of beta-subunits, but none of the described subunit-interactions exert an inhibitory effect on the Kv4.3 current.  相似文献
