高酯酶活性工程菌的构建及其对有机磷农药的降解   总被引:2,自引:0,他引:2  
将抗性库蚊解毒酶酯酶B1基因片段引入融合表达载体pThioHisA中,转化入大肠杆菌DH5α,在IPTG诱导下,经过8 h,酯酶B1在大肠杆菌中获得融合高效表达,目的蛋白占菌体总蛋白的48.7%.重组质粒pThioHiA-B1表达的酯酶融合蛋白具有较高的酯酶B1活性,能高效降解酯酶的特异性底物β-乙酸萘酯(β-NA),并在37℃、pH7.5的条件下,2h内对1000mg/L的甲基对硫磷降解率达81%.  相似文献

用抗性库蚊酯酶基因,引入原核表达载体pRL439,转化大肠杆菌HB101细胞,获得表达。通过酶切、Southern杂交鉴定重组质粒。研究了重组菌酯酶的活性,重组质粒pRLB1表达的酯酶具有高酶活并能高效降解酯酶的特异性底物α乙酸萘酯(αNA)和β乙酸萘酯(βNA);经对重组菌进行细胞固定化后降解农药三氯杀虫酯(7504),反应时间短,降解效率高  相似文献

为明确不同烤烟品种对烟蚜Myzus persicae(Sulzer)解毒酶活力的影响,比较了9个品种烤烟上的绿色型烟蚜的解毒酶活力,发现烤烟品种对烟蚜α-乙酸萘酯羧酸酯酶、β-乙酸萘酯羧酸酯酶、谷胱甘肽转移酶活力有显著影响,不同品种烤烟上烟蚜的α-乙酸萘酯羧酸酯酶、β-乙酸萘酯羧酸酯酶、谷胱甘肽转移酶均有差异。烤烟品种V2、云烟87和红大上的烟蚜羧酸酯酶活力较高,云烟317、云烟85和K326居中,K346、K358和G28则较低。9个品种烤烟对烟蚜谷胱甘肽转移酶活力有所影响,但差异不大。  相似文献

通过比较云南烟蚜敏感品系和抗性品系的解毒酶(α-乙酸萘酯羧酸酯酶、β-乙酸萘酯羧酸酯酶)和靶标酶(乙酰胆碱酯酶)的活力,研究了烟蚜对有机磷、拟除虫菊酯和氨基甲酸酯类杀虫剂抗性的生化机制,并通过酯酶基因扩增检测和钠离子通道突变检测,研究了其抗性的分子机制。结果表明:α-乙酸萘酯羧酸酯酶活力增强是烟蚜对有机磷类、氨基甲酸酯类杀虫剂及拟除虫菊酯类杀虫剂的抗性机制之一;乙酰胆碱酯酶在烟蚜对有机磷杀虫剂抗性中起重要作用;3个抗性品系烟蚜均没有发生酯酶基因扩增,抗拟除虫菊酯品系烟蚜发生了钠离子通道突变。  相似文献

用抗性库蚊酯酶基因B1的cDNA片段插入质粒pRL-439中的强启动子之后,再与穿梭表达载体pDC-8相连构建成大肠杆菌蓝藻穿梭表达载体pDC-B1,然后通过三亲接合转移法将pDC-B1转入蓝藻Synechococcus sp. PCC7942中,经新霉素筛选获遗传稳定的转基因藻株;纯化单藻落在液体中扩大培养,提取蓝藻质粒,Southern杂交确证B1cDNA已转入受体细胞;用酯酶的特异性底物β-乙酸萘酯(β-NA)检测B1的表达,转基因藻对β-NA的降解明显高于野生藻,证明酯酶B1基因在转基因藻中得到表达。  相似文献

本文对有机磷抗性和感性棉蚜Aphis gossypii三个种群抗性生化机制进行了讲究.首先用解毒酶的抑制剂测定药剂的解毒途径.进一步测定乙酰胆碱酯酶活力及其敏感性和多功能氧化酶、谷胱甘肽s-转移酶、α-乙酸萘酯酶和α-乙酸萘酯羧酸酯酶等解毒酶的活力.结果表明,体内条件下,多功能氧化酶与抗性有关,但在离体条件下,在棉蚜匀浆液中有内源抑制剂存在.α-乙酸萘酯酶和α-乙酸萘酯羧酸酯酶活力的增加,乙酰胆碱酯酶对杀虫剂敏感性的降低也是造成棉蚜对有机磷产生抗性的原因.  相似文献

采用不同浓度的草甘膦0.41 g/L、0.82 g/L、1.23 g/L、1.64g/L、2.05 g/L分别以胃毒和触杀法处理空心莲子草叶甲Agasicles hygrophila成虫,测定其乙酰胆碱酯酶(AChE)、羧酸酯酶(CarE)和谷胱甘肽S-转移酶(GSTs)比活力.试验结果表明:两种处理,草甘膦对AChE活力均有不同程度的抑制作用;对CarE活力影响较为显著,在2.05 g/L浓度下,胃毒处理CarE对α-乙酸萘酯(α-NA)和β-乙酸萘酯(β-NA)水解能力分别是对照组的50%和57%,触杀处理CarE对α-乙酸萘酯(α-NA)和β-乙酸荼酯(β-NA)水解能力分别是对照组的53%和59%;胃毒处理埘酶活力影响大于触杀处理,草甘膦对GSTs的活力影响不明显.  相似文献

用RT-PCR克隆了酯酶B1 5′端B1(a),并对其者了序列测定,将其与3′端B1(b)一起连接到pET-28a中,构了完整融合表达载体pET-ESTB1。转化大肠杆菌BL21,在IPTG诱导下,经过12小时,酯酶B1在大肠杆菌内的融合表达达到27%。通过纯化获得1条带的重组蛋白,用粗酶对马拉硫磷的降解显示,该解毒酶在15分钟即降解22.1%,具有较高降解有机磷酸酯类农药的能力,为利用真核生物的自然资源进行农药污染的生物治理等提供了新途径。  相似文献

研究了表达Cry1Ab杀虫蛋白的转Bt基因抗虫玉米对亚洲玉米螟Ostrinia furnacalis (uenée) 幼虫解毒酶、保护酶和中肠蛋白酶活性的影响,测定比较了取食转Bt基因玉米后幼虫体内α-乙酸萘酯酶、乙酰胆碱酯酶、谷胱甘肽S-转移酶、过氧化氢酶、超氧化物歧化酶、中肠总蛋白酶、类胰蛋白酶和类胰凝乳蛋白酶的活力。结果表明,取食转Bt基因玉米48 h后亚洲玉米螟幼虫体内的α-乙酸萘酯酶、谷胱甘肽S转移酶活力明显低于对照;而乙酰胆碱酯酶活力显著高于对照,在取食48 h、60 h和72 h的活力分别是对照的2.00、1.50和2.50倍。保护酶系、中肠总蛋白酶、弱碱性类胰蛋白酶和类胰凝乳蛋白酶的活性在取食48 h后明显受到抑制;但强碱性类胰蛋白酶的活性显著高于对照,取食48 h、60 h和72 h的活力分别是对照的4.00、1.67和1.33倍。乙酰胆碱酯酶和强碱性类胰蛋白酶可能与亚洲玉米螟对Bt的抗性有关。  相似文献

对东亚飞蝗山西临猗和永济2个地理种群的酯酶特性进行了比较研究。非变性聚丙烯酰胺凝胶电 泳图谱显示:以α-乙酸萘酯为底物染色,2个东亚飞蝗种群谱带差别不明显。但是,酯酶动力 学研究结果表明:以α-乙酸萘酯和α-丁酸萘酯为底物时,永济种群的酯酶活性分别是临猗 种群的1.81倍和1.20倍。永济种群酯酶活性的增高可能与非变性聚丙烯酰胺凝胶电泳图谱显 示出较临猗种群多出的酶带有关。体外酯酶抑制动力学研究表明:永济和临猗2种群所含酯酶大 都为B型酯酶,其含量分别为84.94%和91.47%。永济种群对对氧磷的耐受性要高于临猗种群 ,我们推测可能与2种群马拉硫磷使用背景不同有关。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献
