Houttuynia cordata Thunb., traditionally used as a therapeutic plant in folk medicine, has shown antioxidant and anticancer activities. The species, as a core component of paleoherbs, is normally characterized based on the presence of different types of secretory tissue: oil cells, three types of secretory cells and glandular hairs. The aim of this work was to study the structural, componential, and the functional characteristics of the secretory tissues in both the floral and vegetative parts. The results indicate that oil cells and secretory cells are distributed in all organs of the plant, while glandular hairs are situated on the aerial stems and leaves. Both oil cells and glandular hairs initiate from the protoderm, but their developmental processes are different. Although three types of secretory cells initiate from different primary meristems, the developmental patterns of different secretory cells are the same. Also, although the origins of secretory cells are different from oil cells, their early developmental processes are the same. Histochemical results show that oil cells, secretory cells and glandular hairs produce flavonoids, phenolic compounds, tannins, lipids, aldehyde and ketone-compounds. In addition, there are terpenoids and pectic-like substances in oil cells, alkaloids in secretory cells of aerial stems, and terpenoids and alkaloids in glandular hairs. These compounds play very important roles in protecting plants from being eaten by herbivores (herbivory) and infected by microbial pathogens. The oil cell and secretory cell, as unicellular secretory tissues, are intermediates between the primitive surface glandular and secretory cavity and canal during the evolution of secretory structures.  相似文献

Traditional Chinese medicine(TCM) preparations are widely used for healthcare and clinical practice. So far, the methods commonly used for quality evaluation of TCM preparations mainly focused on chemical ingredients. The biological ingredient analysis of TCM preparations is also important because TCM preparations usually contain both plant and animal ingredients,which often include some mis-identified herbal materials, adulterants or even some biological contaminants.For biological ingredient analysis, the efficiency of DNA extraction is an important factor which might affect the accuracy and reliability of identification. The component complexity in TCM preparations is high, and DNA might be destroyed or degraded in different degrees after a series of processing procedures. Therefore, it is necessary to establish an effective protocol for DNA extraction from TCM preparations. In this study, we chose a classical TCM preparation,Liuwei Dihuang Wan(LDW), as an example to develop a TCM-specific DNA extraction method.An optimized cetyl trimethyl ammonium bromide(CTAB) method(TCM-CTAB) and three commonlyused extraction kits were tested for extraction of DNA from LDW samples. Experimental results indicated that DNA with the highest purity and concentration was obtained by using TCM-CTAB. To further evaluate the different extraction methods, amplification of the second internal transcribed spacer(ITS2) and the chloroplast genome trnL intron was carried out.The results have shown that PCR amplification was successful only with template of DNA extracted by using TCM-CTAB. Moreover, we performed high-throughput 454 sequencing using DNA extracted by TCM-CTAB. The factors which effected the extraction of total flavones from Polygonum aviculare L.var.aviculore were discuseed with respects to solvent,concentration and the extracting temperature.The rutin was used as a standard compound,the total flavones extracted by the different extracting procedure were quantitated by measurement of absorbance at 510 nm.The results show that 80℃ and 35% alcohol are optimal and the extracting rate of the total flavones in as high as 4.349%.  相似文献

Fungal endophytes have been isolated from almost every plant, infecting their hosts without causing visible disease symptoms, and yet have still proved to be involved in plant secondary metabolites accumulation. To decipher the possible physiological mechanisms of the endophytic fungus-host interaction, the role of protein phosphorylation and the relationship between endophytic fungus-induced kinase activity and nitric oxide (NO) and brassinolide (BL) in endophyte-enhanced volatile oil accumulation in Atractylodes lancea plantlets were investigated using pharmacological and biochemical approaches. Inoculation with the endophytic fungus Gilmaniella sp. ALl2 enhanced the activities of total protein phosphorylation, Ca2~-dependent protein kinase, and volatile oil accumulation in A. lancea plantlets. The upregulation of protein kinase activity could be blocked by the BL inhibitor brassinazole. Furthermore, pretreatments with the NO-specific scavenger cPTIO significantly reduced the increased activities of protein kinases in A. lancea plantlets inoculated with endophytic fungus. Pretreatments with different protein kinase inhibitors also reduced fungus-induced NO production and volatile oil accumulation, but had barely no effect on the BL level. These data suggest that protein phosphorylation is required for endophyte- induced volatile oil production in A. lancea plantlets, and that crosstalk between protein phosphorylation and the NO pathway may occur and act as a downstream signaling event of the BL pathway.  相似文献

The lesser grain borer, Rhyzopertha dominica is a major insect pests of stored grain in the tropics. Vegetable oils (chamomile, sweet almond and coconut) at 2.5, 3.5, 5.0, 7.0 and 10.0 mL/kg were tested against Rhyzopertha dominica (F.) in wheat grain. All bioassays were conductr, d at 30℃ and 65% + 2% RH. Treatments with vegetable oils at high dose (10.0 mL/kg) achieved over 95% control within 24 h of exposure to freshly treated grain, There was little difference between the three oils in their effect. Persistence of oils in grains was tested at short-term storage intervals (48, 72 and 96 h) and intermediate-term intervals (10, 20 and 30 days) after treatments. The activity of all products decreased with storage period. Seed viability was reduced by the high dose rate (10.0 mL/kg) of oil treatments. The potential use of vegetable oils as supplementary or alternative grain protectants against insect damage in traditional grain storage in developing countries is discussed.  相似文献

This paper presents a molecular dynamics simulation of the behaviours of non-polar droplets merging and also the fluid molecules interacting with a hydrophobic surface. Such behaviours and transport phenomena are popular in general microchannel flow boiling and two-phase flow. The droplets are assumed to be composed of Lennards-Jones type molecules. Periodic boundary conditions are applied in three coordinate directions of a 3-D system, where there exist two liquid droplets and their vapour. The two droplets merge when they come within the prescribed small distance. The merging of two droplets apart from each other at different initial distances is tested and the possible larger (or critical) non-dimensional distance, in which droplets merging can occur, is discussed. The evolution of the merging process is simulated numerically by employing the Molecular Dynamics (MD) method. For interactions with hydrophobic solid wall, a system with fluid confined between two walls is used to study the wetting phenomena of fluid and solid wall. The results are compared with those of hydrophilic wall to show the unique characteristics of hydrophobic interactions by microscopic methods.  相似文献

Aims The vulnerability of xylem embolism is one of the key physiological factors that are related to plant mortality. Vulnerability curves are typically used for determining the vulnerability of xylem embolism. However, the shapes of vulnerability curves vary with the methods of assessment, especially in plant species with long xylem vessels. This study aims to investigate the feasibility of using different methods for establishment of vulnerability curves. Methods Robinia pseudoacacia branches, with long xylem vessels, were used as plant materials for comparison of three different methods in establishing vulnerability curves, including bench top dehydration, Cochard Cavitron centrifugation and Sperry centrifugation. In the Sperry centrifugation method, rotors of two different sizes were used to test the ‘open vessel artifact’ hypothesis. Important findings The vulnerability curve established by the bench top dehydration method displayed an “s” shape, while both the Cochard Cavitron centrifugation and Sperry centrifugation methods produced “r” shape curves. Vulnerability curves derived from the bench top dehydration method and the centrifugation methods were significantly different. Using the Sperry centrifugation method, the R. pseudoacacia branch samples in the 14.4 cm rotor had a higher proportion of open vessels, while the embolic vulnerability curves established on the 27.4 cm and 14.4 cm long stem segments were similar, indicating that the Sperry centrifugation method does not produce “open vessel artifact”. © Chinese Journal of Plant Ecology.  相似文献

With three different methods, the total flavonoids from the roots of cultivated Gtycyrrhiza pallidiflora Maxim. were determined and compared by ultraviolet-visible spectrophotometry. The results showed that the extract yield of total flavonoids in crude extracts was 4.54% by direct determination method, 4.27% by AI( NO3)3 colorimetry method and 2.09% by AlCl3 colorimetry method. The content of total flavonoids in roots of cultivated G. pallidiflora was only 0.05%. The cultivated G. pallidiflora in Lanzhou is not unfit for the medicine source of flavonoids.  相似文献

B8Gly is absolutely conserved in insulins during evolution. Moreover, its corresponding position is always occupied by a Gly residue in other members of insulin superfamily. Previous work showed that Ala replacement of B8Gly significantly decreased both the activity and the foldability of insulin. However, the effects of substitution are complicated, and different replacements sometimes cause significantly different results. To analyze the effects of B8 replacement by different amino acids, three new insulin/single-chain insulin mutants with B8Gly replaced by Ser, Thr or Leu were prepared by protein engineering, and both their foldability and activity were analyzed. In general, replacement of B8Gly by other amino acids causes significant detriment to the foldability of single-chain insulin: the conformations of the three B8 mutants are essentially different from that of wild-type molecules as revealed by circular dichroism; their disulfide stabilities in redox buffer are significantly decreased; their in vitro refolding efficiencies are decreased approximately two folds; the structural stabilities of the mutants with Set or Thr substitution are decreased significantly, while Leu substitution has little effect as measured by equilibrium guanidine denaturation. As far as biological activity is concerned, Ser replacement of B8Gly has only a moderate effect: its insulin receptor-binding activity is 23% of native insulin. But Ttir or Leu replacement produces significant detriment: the receptorbinding potencies of the two mutants are less than 0.2% of native insulin. The present results suggest that Gly is likely the only applicable natural amino acid for the B8 position of insulin where both foldability and activity are concerned.  相似文献

Analysis of fish meat using gas chromatography is described. Flavor is the sensation arising from the interplay of the signals of sensing smell, taste and irritating stimuli from food stuff. For human, flavor and nutrition are inseparable. In fish, trace amount of volatile organic compounds (VOCs) are the major compounds to affect consumer’s preference, which are produced during storage and spoilage. In the present study, volatile compounds were extracted by Likens-Nickerson concurrent distillation apparatus from wild and farmed Cirrhina mrigala and Cyprinus carpio. The quantitative and qualitative estimation of volatiles was made by gas chromatography. Wild and farmed fish of different fish sizes were compared for these compounds (appearing in the form of peaks), which were identified from their retention time by comparing with the standards. Fifteen major VOCs were found in these species which included hexadecane, 3-octanol, hexanal, decane, 3-hexene-1-ol, 2-undecanone, 2-heptanone, butanal, 2-nonanone, 1- heptanal, furaldehyde, 3-methyl-1-butanal, trans-3-hexene-1-ol, octanal and decanal. These compounds varied qualitatively and quantitatively in both wild and farmed fish of different fish sizes.  相似文献

利用GC-MS对石香薷挥发油进行了定性、定量分析。采取超临界CO2萃取、水蒸气蒸馏和有机溶剂石油醚萃取这三种方法提取石香薷挥发油。这三种方法提取石香薷挥发油的主要成分基本相似,主要为含氧化合物(香薷酮、百里香酚和香荆芥酚)等,采取超临界CO2萃取和水蒸气蒸馏的石香薷挥发油品质较优。超临界CO2萃取法为提取石香薷挥发油的理想方法。  相似文献

通过超临界CO2萃取均匀设计实验和微波辅助萃取艾叶挥发油的正交实验比较,考察影响提取的主要因素,寻求最佳萃取工艺。超临界CO2萃取最佳工艺条件为:萃取压力16MP,萃取温度31℃,CO2流量20kg/h和时间80min,得率3.75%;微波萃取最佳工艺条件为:辐射功率720w,辐射时间200s,溶剂量400mL,洗涤剂量50mL,得率4.85%。水蒸馏法提取率为1.87%。结果表明超临界CO2和水蒸馏法萃取艾叶挥发油品质最好;微波萃取收率最高,但品质较差。  相似文献

用GC/MS方法,定性定量地分析了用3种不同方法提取的姜油的化学成分,分别鉴定了46,50,61个化合物。蒸馏油主要含有单萜、倍半萜烯类化合物,未见生姜特有的辣味成分。冷榨油和超临界CO2萃取油除倍半萜类化合物外,主要含有辣味成分,含量分别为18.61%,23.09%。冷榨油及超临界CO2萃取油较好地保持了生姜的特征辛香气及辣味成分  相似文献

From the rose hip seed, which is generally a waste material, valuable oil can be obtained for medicinal use. Various extraction methods have been compared: traditional solvent extraction with ultrasound-, microwave-, sub- and supercritical fluid extraction (SFE). Unsaturated fatty acid (UFA: oleic-, linoleic- and linolenic acid; 16.25-22.11%, 35.94-54.75%, 20.29-26.48%) and polyunsaturated fatty acid (PUFA:linoleic- and linolenic acid) content were over 90% and 60% in the recovered oils. The oils contained different amounts of metals. The concentration of some metals, particularly iron in microwave oil (27.11 microg g(-1)) is undesirable from the aspect of stability. By traditional solvent extraction, oil was obtained in 4.85 wt/wt%. Subcritical FE appeared to be the best method for the recovery of rose hip oil with highest oil yield (6.68 wt/wt%), carotene- (145.3 microg g(-1)) and linoleic acid content (54.75%). Supercritical FE without organic solvent is suitable for mild recovery of oil. The oil was rich in UFA and PUFA (92.7% and 76.25%) and contained the lowest amount of carotene and pheophytin (36.3 and 45.8 microg g(-1)). Oil yield in most new extraction methods (microwave extraction, super- and subcritical FE) was higher than in the case of traditional Soxhlet extraction. The main benefit of supercritical FE with CO2 is the solvent free oil while in the case of other extractions evaporation of the solvent is needed. Although the content of bioactive compounds in oils was different, all oils may be appropriate for medicinal use.  相似文献

采用水蒸气蒸馏法、有机溶剂萃取法、CO2超临界萃取法3种提取方法提取组培百里香精油,比较分析精油得率、精油化学成分以及相对含量,以期得出最佳提取方法。结果表明:水蒸气蒸馏法提取的百里香精油得率为O.21%,主要化学成分为:百里酚(36.53%)、间伞花烃(14.13%)、松油烯(8.09%)和石竹烯(4.14%);有机溶剂萃取法提取的精油得率为0.19%,主要化学成分为:1,2-苯二甲酸-单-2-乙基己基酯(55.23%)、百里酚(873%)、松油烯(5.23%);CO2超临界萃取法提取的精油得率为0.27%,主要化学成分为:百里酚(26.68%)、3-苯基-2-丙烯酸-甲酯(21.55%)、间伞花烃(9.69%)。从精油得率、精油质量以及精油主要化学成分综合比较3种方法,水蒸气蒸馏法是提取百里香精油的最佳方法。  相似文献

采用超临界流体萃取法(SFE)和水蒸气蒸馏法分别对北细辛根及根茎进行提取分离,分别得5.5%SFE萃取物和2.8%挥发油。应用GC-MS分析,从北细辛SFE萃取物中鉴定出7种化学成分,占萃取物总量的72.70%,其中甲基丁香酚为44.62%;从细辛挥发油中鉴定出19种化学成分,占挥发油总量的88.53%,其中甲基丁香酚为43.02%。两种方法有6种成分完全相同,其中致癌物质黄樟油素SFE法比蒸馏法低2.8倍。北细辛根及根茎超临界萃取物与水蒸汽蒸馏挥发油主要成分相同,但其化学组成存在较大差异,提示药效亦不相同。超临界流体萃取法是剂型改革的有效方法之一。  相似文献

用超临界CO2、超声波、微波和机榨4种方法提取茶油,利用GC-MS分析茶油成分。结果表明:超临界CO2萃取的茶油与超声波和微波提取的主要化学成分几乎无差异,机榨茶油的化学成分与前3种的有一些差异,未测出维生素E和角鲨烯。超临界CO2萃取的茶油已达到国家成品茶油的一级标准,超临界CO2萃取法为提取茶油的较理想方法。  相似文献
