通过比较6种不同型号的大孔吸附树脂对家蝇蛋白的吸附解吸特性,发现D101大孔吸附树脂的性能优于其他5种,吸附流速、浓度影响大孔吸附树脂的动态吸附性能。D101大孔吸附树脂对未经诱导的家蝇蛋白的吸附量可达217.18mg/g(以干树脂总量为基准),洗脱率为76.48%。吸附后的大孔吸附树脂用15%、35%、55%的乙醇溶液阶段洗脱,各洗脱组分的疏水性逐渐增大,蛋白质含量也明显增加。用E.coli、S.aureus和B.subtilis对各洗脱组分进行抑菌活性测定,抑菌活性随洗脱组分的疏水性增加而增大。测得55%乙醇洗脱组分的抑菌活性最大,其中对E.coli的抑菌圈直径达5.8mm。  相似文献

本论文通过研究大孔树脂对构树叶黄酮吸附过程中动力学与热力学的变化,寻找大孔吸附树脂对该体系吸附能力的规律。结果表明,大孔吸附树脂对构树叶黄酮的吸附在9 h后达到平衡,且吸附过程符合二级动力学方程;不同温度下该大孔吸附树脂对构树叶黄酮的吸附等温线采用Freundlich和Langmuir等温吸附方程进行拟合,结果与Freundlich等温吸附模型拟合度较高,说明该吸附为优惠吸附,同时吸附焓变ΔH0、吸附自由能ΔG0、吸附熵变ΔS0,说明大孔吸附树脂对构树叶黄酮的吸附过程是放热的,该吸附过程为自发不可逆过程,且吸附有明显的物理吸附特性。  相似文献

利用大孔吸附树脂提取蜀葵花色素的研究   总被引:15,自引:0,他引:15  
研究利用大孔树脂吸附和分离蜀葵(Althaearosea(L.)Cavan)花红色素,比较了D-072、D-401、D-301-G、D-101、NKA-9、D-290、D-110七种树脂对该色素的静态吸附情况以及不同极性解吸剂对吸附色素的树脂洗脱的效果,从中选择出吸附和解吸效果最佳的树脂以及较适的解吸剂。结果表明:用D-401大孔吸附树脂作吸附剂,色素吸附率达91%;解吸剂用含0.1%HCl的60%酸化乙醇,色素可被充分洗脱下来,解吸效果较好;树脂通过回收再生后可重复利用。大孔吸附树脂法精制蜀葵花色素工艺相对简单,原料、试剂利用率较高。  相似文献

为了研究大孔吸附树脂纯化山茱萸总皂苷的动态吸附工艺,运用静态吸附与解吸试验对大孔吸附树脂进行筛选,然后通过单因素试验、正交试验和方差分析确定了大孔吸附树脂吸附山茱萸总皂苷的最佳操作条件.结果表明,HPD-300树脂对山茱萸总皂苷的吸附和解吸性能较好.确定的最佳吸附条件为料液浓度3.5 m g.mL-1,上柱速度3.5 BV.h-1,pH值为7.0.HPD-300大孔吸附树脂可较好地纯化山茱萸总皂苷.  相似文献

以苹果渣多酚类化合物为吸附模型,制备一种可有效吸附分离苹果渣多酚的磁性树脂,研究其吸附、解吸效能,并就吸附和解吸效能、使用次数与普通大孔树脂进行比较。结果显示,5种普通树脂中D280、H103、AB-8对苹果渣多酚的吸附率均高于80%,AB-8、D3520树脂对苹果渣多酚的解吸率均高于90%,AB-8树脂的吸附率和解吸率最好;磁性大孔树脂吸附0.5 h后即可达到吸附平衡,吸附平衡后剩余液多酚质量浓度低于AB-8树脂吸附平衡后剩余液多酚质量浓度,吸附效能明显优于AB-8树脂;磁性大孔树脂解吸效能也略优于AB-8树脂;使用4 次后,磁性大孔树脂的吸附率仍然高于70%,稳定性优于AB-8树脂。可见,磁性大孔树脂是一种良好的苹果渣多酚吸附剂,有较好的工业化应用前景。  相似文献

目的:研究大孔吸附树脂纯化鸡枞皂苷的方法.方法:采用分光光度法测定鸡枞皂甙的含量,分别考察了树脂种类、样品液浓度、pH值、吸附流速、洗脱剂浓度对鸡枞皂甙分离纯化的影响.结果:HPD-450树脂最适合鸡枞皂苷的纯化.工艺条件为:洗脱剂乙醇的体积分数为40%,吸附流速为1~2mL/min,样品液浓度为2.5~4.0mg/mL,样品液pH值为5~7.采用HPD-450大孔吸附树脂对鸡枞皂甙进行纯化效果最优.结论:在上述条件下,大孔树脂可用于鸡枞皂甙的分离纯化.  相似文献

利用11种大孔吸附树脂对乌饭树树叶黑色素进行纯化分析,AB-8、NKA-9和S-8型大孔吸附树脂的静态吸附量和解吸率较高。由吸附与时间的关系曲线选择AB-8型大孔吸附树脂进行动态分析,得出最佳的纯化条件为:pH为5.05左右,溶液浓度为0.68~1.33 mg/mL,吸附流速为3 mL/min。使用7倍体积的95%乙醇解吸率可以超过95%。树脂使用三次后必须进行再生处理。  相似文献

采用大孔吸附树脂分离纯化人工蛹虫草培养基残基中的虫草素,以HPLC法测定样品中虫草素含量,筛选出了适宜的大孔树脂NKA-II。研究了pH、洗脱剂乙醇体积分数等因素对该树脂吸附性能及解析性能的影响。结果表明,NKA-II型树脂纯化虫草素的最佳吸附条件为pH9,吸附流速2BV/h,该树脂对虫草素的吸附量可达到16.5mg/g。洗脱工艺条件为2.5倍树脂柱体积的50%乙醇,NKA-II大孔树脂对虫草素的解析率可达到95%以上。  相似文献

目的:为了找到方便、准确、快捷的米根霉RL6041菌株发酵液中乳酸含量的检测方法。方法:同时采用EDTA定钙法、高效液相色谱(HPLC)法和生物传感仪法三种方法测定米根霉RL6041菌株发酵液中的乳酸含量。结果:米根霉RL6041菌株三批次发酵的测试结果的差异显著性分析表明,在同一发酵批次的乳酸含量测试中,EDTA定钙法与HPLC法、生物传感仪法有显著差异(p<0.05),而HPLC法与生物传感仪法无显著性差异(p>0.05)。EDTA定钙法平均误差7.50g/L;而HPLC法和生物传感仪测定法的平均误差分别为2.84g/L和2.39g/L。结论:EDTA定钙法简便快速,但重复性差;HPLC法准确、灵敏;生物传感仪测定法比较准确和方便。  相似文献

Oral mannitol 200 g. was given on 24 occasions to patients with intractable oedema (cardiac, renal, or hepatic) and produced a mean weight loss of 3 kg. The procedure was found to be a safe, rapid, and effective method of treating oedema, and it should be used when diuretic therapy proves ineffective, particularly in the presence of hyponatraemia.  相似文献

The resin competition method for determining the dissociation constants of metal ion-nucleotide complexes was modified to take into account the fact that some metal nucleotide complexes are anionic and thus, like the free nucleotide, will bind to the resin. A simple, rapid spectrophotometric titration procedure is given for the determination of both the metal ion-nucleotide complex dissociation constant and the ratio of the affinities of the nucleotide and its complex for the resin.  相似文献

Liquid chromatography/mass spectrometry using electrospray ionisation was used to analyse rhamnolipids produced by a Pseudomonas aeruginosa strain with mannitol or naphthalene as carbon source. Identification and quantification of 28 different rhamnolipid congeners was accomplished using a reverse-phase C(18) column and a 30 min chromatographic run. Isomeric rhamnolipids that were not chromatographically resolved could be identified by interpretation of their mass spectra and their relative proportions estimated. The most abundant rhamnolipid produced on mannitol contained two rhamnoses and two 3-hydroxydecanoic acid groups. The most abundant rhamnolipid produced from naphthalene contained two rhamnoses and one 3-hydroxydecanoic acid group.  相似文献

Aim of this study was to set up a method by capillary electrophoresis to detect lactulose and mannitol in urine after an oral load, and to estimate the intestinal permeability in controls and in type I diabetes patients. The underivatized carbohydrates were monitored by indirect UV detection using sorbate, cetyltrimethylammonium bromide and LiOH as background electrolyte. Urines were purified by solid phase extraction, shaken with cation exchange resin, filtered and analysed. Carbohydrates migrated in <10 min in relation to their pK(a) and M(r). Controls (n = 33) and patients (n = 23) had an excretion ratio lactulose/mannitol 0.025 (0.018-0.051) and 0.067 (0.050-0.127), respectively (p < 0.01, median, interquartile range).  相似文献

A convenient solid phase synthesis of a Thrombin Receptor Glycopeptide Mimetic analogue namely, 1-O-Methyl-2-N-{1'-(argininocarbonyl)-4'-[(4'-fluoro)-benzylamido]-cyclohexane}-glucosamine using Fmoc/tBu methodology and the 4-Methoxybenzhydryl bromide resin is described. The synthesized analogue was purified by Reverse Phase High Performance Liquid Chromatography (RP-HPLC) and was identified by Electron Spray Ionization-Mass Spectrometry (ESI-MS) and Nuclear Magnetic Resonance (NMR). The synthetic protocol introduced for the first time successfully the acid sensitive 4-Methoxybenzhydryl bromide resin as a scaffold for the synthesis of glycopeptides resulting in high yield reactions. This synthetic procedure could be a general one for the convenient synthesis of such glyco compounds as the method was used for the first time to glycosylate a non peptide mimetic of an important protein sequence, in particular of the thrombin receptor active site S42FLLR46.  相似文献

Liquid Na+ resin microelectrodes were used to measure intracellular Na+ activities (alpha iNa+) in the mammalian tight epithelium, rabbit urinary bladder. alpha iNa+ averaged 7 +/- 1 mM and was independent of Na+ transport rate over the range of 2 to 8 muA/muF. (1 mF congruent to 1 cm2 apical membrane area). After Na+ loading the cells the Na+ pump in the basolateral membrane was measurably electrogenic. A method for shielding the Na+-sensitive microelectrodes is described which increases the response time and eliminates an electrical shunting artifact.  相似文献

A procedure is described for the extraction of diethylstilbestrol (DES) from animal tissue for quantitative capillary gas chromatography/mass spectrometry (GC/MS). The procedure is based upon use of a strong anion exchange polystyrene divinylbenzene resin for sample purification. The recovery of DES from the resin clean up was 88% in the high parts per trillion (ppt) range. Criteria for identification of DES using selected ion monitoring (SIM) GC/MS are presented. Liquid chromatography/mass spectrometry (LC/MS) was used to investigate altered DES cis/trans ratios observed in biological extracts.  相似文献

Osmotic titration of ATPase activity in rat liver mitochondria was consistent with enhanced porosity of the mitochondrial inner membrane to mannitol due to ATP hydrolysis even when endogenous respiration was inhibited by rotenone. The occluded ATPase activity, which exhibits osmotic activation with an optimum near isotonicity, depends both on the ATPase activity per se and on the activity of the ADP/ATP carrier. Purified ADP/ATP carrier incorporated into small, unilamellar liposomes was critically shown to exhibit dependence of its activity on the osmotic pressure differences across the membrane, with maximal activity corresponding to isotonicity, regardless of the actual internal tonicity.  相似文献

Oral cholera chemical vaccine in the doses tested (2 and 3 tablets) proved to be areactogenic, harmless and immunologically effective in a controlled limited trial in 150 volunteers.
