目的比较腹部和颈部小鼠心脏移植模型的优缺点,探讨二次器官移植或双心脏移植研究的可行性和实用性动物模型。方法参照Ono法和Chen法并加以改进,建立了同系或者同种小鼠腹部和颈部异位心脏移植模型,同时对两种手术方式的成功率、手术时间、移植心脏存活时间和病理组织学进行了比较。结果腹部和颈部异位心脏移植手术成功率分别为86.7%和83.3%,两种手术方式在总手术时间、移植心脏存活时间和病理组织形态学检查上均无明显差异(P0.05)。结论在熟练掌握显微外科技术的基础上,两种异位心脏移植模型都能顺利建立,两者既可分开选用,又可结合运用,以适应不同实验需求。  相似文献

目的探讨小鼠心脏移植模型的建立与改良。方法供体采用BLAB/c小鼠,受体采用C57小鼠。用24GACuff管法套扎供心肺动脉与颈外静脉,用自制Cuff管套扎供心升主动脉与颈总动脉。结果供心冷缺血时间少于15min,手术时间大大缩短,成功率明显提高。结论改进后的模型显著降低了手术难度,可用于移植免疫的各种研究。  相似文献

目的通过显微外科技术建立小鼠原位肺移植模型,为肺移植研究提供动物模型。方法采用C57BL/6小鼠作为供、受体,行同基因小鼠原位左肺移植,使用Cuff套管法进行气管及血管吻合。术后7、14、21、28 d取移植肺及原肺,行HE染色,评价肺移植后效果。结果学习曲线后,共30例小鼠移植,手术成功率89%,小鼠成活率100%。供体手术时间:(35.2±9.81)min,受体手术时间:(24.6±7.42)min,冷缺血时间是:(46.6±8.92)min,热缺血时间是:(17.2±3.08)min。同基因移植物大体及病理无明显改变,病理显示与原肺无差别。结论本技术能够方便快捷建立小鼠肺移植模型,成功率高,可重复性强,符合原位肺移植临床生理,是研究肺移植发病机制和治疗的良好动物模型。  相似文献

目的构建大鼠腹腔内工作型心脏移植模型,总结影响模型成功率的因素。方法 Brown Norway到Lewis大鼠心脏移植90例,其中预实验50例,正式实验40例。采用肺动脉和左房吻合、主动脉和受体腹主动脉吻合的方法建立工作型心脏移植,统计手术存活率和死亡原因,分析确保成功率的关键因素。结果术者通过练习,手术成功率稳定77.5%,供心冷缺血时间为(34±5)min,整个手术时间为(71±11)min。HE染色显示移植后发生了免疫反应,移植模型可靠。结论决定手术成功率的影响因素众多,其中主要因素有:合格的实验动物、供体心脏的保护、快速有效的血管缝合和术后动物护理。  相似文献

目的:建立一种快速有效的大鼠腹腔异位心脏移植模型。方法:采用SD大鼠作为受体,Wistar大鼠作为供体,行同种异位腹腔心脏移植,术后给以CsA5 mg/kg/d灌服,心脏移植手术方法采用改良的Ono术式,观察改良的腹腔异位心脏移植各步骤所需时间、术后成功率及主要并发症发生率。结果:共建立40只大鼠异位心脏移植模型,手术成功率92.5%。动脉吻合时间12.5±2.3min,静脉吻合时间12.3±1.5 min,供心缺血时间37±3.5 min,受体血管阻断时间34.2±2.6 min,总手术时间90.2±4.8 min,出现的主要并发症为出血和供心复跳失败(各占5%、2.5%)。结论:改进的大鼠腹部异位心脏移植技术是一种简便、快速、有效、成功率高的模型制作方法。  相似文献

目的建立并改进小鼠腹部异位心脏移植模型,为器官移植研究提供技术支持。方法SPF级近交系小鼠112只进行心脏移植手术,在传统方法的基础上进行改进,观察改进后手术效果并分析其优势。结果手术成功率为89.28%。总手术时间(103.9±16.5)min,受体手术时间(73.0±7.9)min。改进的方法简化了操作步骤,降低了手术难度。结论改进的小鼠腹部异位心脏移植技术是一种简便、有效、成功率高的模型制作方法。  相似文献

目的:探讨供体特异输注(donor-specific transfusion,DST)及不同剂量FK506对同种异体小鼠心脏移植的影响.方法:应用显微外科技术制作颈部移植心脏急性排斥反应小鼠模型,将移植受体小鼠分为4组:对照组(单纯移植组,未加DST),DST 移植组,DST 移植 FK506(2meg/(kg·d))组,DST 移植 FK506(0.3mg/(kg·d))组,比较各组移植心脏生存时间,心肌病理改变及外周血血清细胞因子水平:血清IL-2,IL-4,IL-10和IFN-γ水平.结果:术前1天应用DST与连续应用较小剂量FK506可显著延长移植物存活.术后第七天病理检查发现联合应用DST和FK506的两组移植物急性免疫排斥反应明显比其他两组减轻.血清中IL-2和IFN-γ水平在联合应用DST和FK506的两组明显低于其它两组,IL-4和IL-10水平在联合应用DST和FK506的两组高于对照组和DST 移植组.结论:术前DST及持续应用较小剂量FK506可有效抑制同种异体小鼠颈部心脏移植术后急性排斥反应,显著延长移植物的生存时间.  相似文献

目的采用腹部套管法建立大鼠心脏移植模型,并分析该方法优劣,为器官移植研究提供合适的动物模型。方法SD大鼠60只,参照Baxter的报道进行同种腹部异位心脏移植,成功制作了该模型并总结了经验。结果共实施手术30例,成功27例,成功率90%;移植心存活30 d以上。结论新型的大鼠心脏移植模型简单易行,成功率高,适合用于器官移植研究,值得推广应用。  相似文献

目的 建立大鼠腹腔异位工作型心脏移植动物模型.方法 Wistar大鼠30只,将供心主肺动脉与左房吻合,再将供心移植到受体腹部,将升主动脉与供心腹主动脉端侧吻合,使得左心房获得前负荷,成为工作型心脏.结果 成功建立大鼠腹腔异位工作型心脏移植模型,15例成功11例,手术成功率为73%.手术平均时间为(38.8±5.7)min.移植心脏获得长期存活,在28 d后通过超声观察可见供体心脏工作正常.结论 手术的关键是提高供心的摘取技术及受体的血管吻合技术,同时术后给与供心适当的按摩辅助也非常必要.此模型可以更好的用于移植免疫学的研究.  相似文献

目的 研究石韦对小鼠免疫功能和同种异基因皮片移植的影响.方法 分别给小鼠灌胃高、中、低3个剂量石韦水煎液,研究其对正常小鼠和由左旋咪唑引起的免疫亢奋模型小鼠脾脏重量指数、巨噬细胞的吞噬活性、淋巴细胞转化、IgM分泌量的影响;观察石韦对小鼠同种异基因皮片移植的影响.结果 石韦可抑制正常小鼠巨噬细胞吞噬活性、T淋巴细胞转化率和IgM的分泌量(P<0.05).3个剂量的石韦均可调节免疫亢奋小鼠脾脏重量指数、巨噬细胞吞噬活性、T淋巴细胞转化率恢复至正常水平,中剂量组可以显著降低IgM的分泌量至正常水平.小鼠移植皮片的存活时间:石韦低剂量组为(16.13±1.13)d,中剂量组为(15.62±1.01)d,高剂量组为(14.89±2.01)d,显著长于对照组的(9.75±0.89)d.结论 石韦可抑制正常小鼠的免疫功能,调节免疫亢奋小鼠免疫功能恢复至正常水平,减轻机体对同种异基因皮片移植的排斥反应.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
