In this paper, Psammosilene tunicoides, an endemic species to China, was cytologically studied for the first time. The morphology of the nuclei at resting stage was categorized to be simple chromocentre type. The morphology of mitofic-pmphase chromosomes was categorized to be the interstitial type.28 chromosomes were observed at the mitotic metaphase, and 14 bivalent chromosomes were observed at diakinesis. So, the basic chromosome number was confirmed to be x = 14. Psomzr~silene tunicoides is different from Silene rubicunda in the basic chromosome number and the morphology of the nuclei at resting stage and mitotic-prophase chromosomes, because Sgene rub/cund has the basic chromosome number of x= 10 and 12, and its nuclei at resting stage and mitotic-prophase chromosomes is sparsely diffuse type and continuous type respectivrly.  相似文献

Right before the excystment of an Euplotes encysticus sawtooth-like folds appeared among the pellicle plasmalemma,the inner and outer alveolar membranes were still sticking together,and were not distinguishable.Microtubular layers already formed at the sites beneath the dorsal cortical pellicle corresponding to vegetative cells,but they still proceed to be organized on the ventral structures.Cristae,highly-tangled with tubular-type structures,appeared on the mitochondria,and were morphologically similar to that of vegetative cells.In the cortical ciliatures,such as ciliary shafts,kinetosomes,surrounding fibrillar cirral baskets,and attached structures of ciliatures,etc.,they are different from those in resting cysts which are degenerated or lost.All the ciliature microtubules of ciliary shafts are of the 9 2 pattern,but the microtubule-like structure aggregates at tripletmicrotubule centers of many kinetosmes,are still under various stages of differentiation.Microtubules beneath the kinetosomal rows are of a developmentally elongated stage;crowded chromatins of various shapes and sizes are found in macronucleus,but there are no nuclear pores (formed by nuclear membrane as in resting cysts) on the nuclear membrane where these chromatins attached.  相似文献

This study presents the results of a demographic analysis of 20 years of birth and death records of red gorals kept at the Shanghai Zoo.The age of the first successful breeding,sex ratio at birth,seasonality of reproduction and mortality rate are presented.The females or males of red goral born in captivity can give their first birth at the age of 2 or 3 separately.The births mainly occurred from May to July and peaked in June.Ratio of females to males at birth was 2.36:1.1 The mortality rate for infants and adults were 23.1% and 11.8%,respectively while there was no sub-adult death recorded during the 20 years period.There was a definite relation between mortality rate and seasons and mainly in July.  相似文献

The seed cotyledons of a euhalophyte, Suaeda physophora, were found to be dark green. The pigment extracted from the cotyledons was proved to be chlorophyll for the absorption spectra curve of extracts the same as that for leaves. Photosynthetic oxygen-exchanging could be detected after the seeds were moistened for 6 h. Microstructure of organelles in cotyledons of ungerminated seeds was detected by transmission electron microscopy. Histochemical pigmentation was used to investigate the degree of damage on the membrane of radicles. A xerophyte, Haloxylon persicum, whose cotyledons of desiccated seeds also contain chlorophyll was used as a comparative species. The results showed that S. physophora maintained the ultra-structure of chloroplasts, the integrity of plasma membranes of radicles kept much better than that of H. persicum, which showed the great adaptability to salinity of the euhalophyte even at the seed-germination stage. Seeds were incubated in 0 and 700 mmol/L NaCl for 10 days in darkness at 20 ℃ , then ungerminated seeds in NaCl solution were transferred to deionised water and reincubated for another 8 days and was recorded the germination recovery. Dry seeds moistened with deionised water germinated gradually in the dark and germination was maximal after 3 or 4 d. Seeds at 700 mmol/L NaCl for 10 d and transferred back to deionised water germinated abruptly, most seeds germinated in one day and photosynthesis was also detected. It is concluded that recovery germination of S. physophora ensured the seeds could germinate rapidly after salinity declines, for example under the mild but transitory favorable edaphic condition in early spring. Photosynthesis function in seeds might also promoted radicle growth and seedling establishment.  相似文献

In this work the susceptibility of mobile and cystic forms of Borrelia burgdorferi to hydroxychloroquine (HCQ) was studied. The minimal bactericidal concentration (MBC) of HCQ against the mobile spirochetes was >32 μg/ml at 37 °C, and >128 μg/ml at 30 °C. Incubation with HCQ significantly reduced the conversion of mobile spirochetes to cystic forms. When incubated at 37 °C, the MBC for young biologically active cysts (1-day old) was >8 μg/ml, but it was >32 μg/ml for old cysts (1-week old). Acridine orange staining, dark-field microscopy and transmission electron microscopy revealed that the contents of the cysts were partly degraded when the concentration of HCQ was ≥MBC. At high concentrations of HCQ (256 μg/ml) about 95% of the cysts were ruptured. When the concentration of HCQ was ≥MBC, core structures did not develop inside the cysts, and the amount of RNA in these cysts decreased significantly. Spirochetal structures inside the cysts dissolved in the presence of high concentrations of HCQ. When the concentration of HCQ was ≥MBC, the core structures inside the cysts were eliminated. These observations may be valuable in the treatment of resistant infections caused by B. burgdorferi, and suggest that a combination of HCQ and a macrolide antibiotic could eradicate both cystic and mobile forms of B. burgdorferi. Effects of phosphorus limitation on the physiological and biochemical changes of the freshwater bloom alga Microcystis aeruginosa Qutz. are reported in the present study. As a result of phosphorus limitation, biomass was controlled to some extent and the protein content per cell in vivo decreased. However,the carbohydrate content per cell was higher in phosphorus limitation over the 8d of cultivation. Soluble proteins were distinct in the media, whereas phosphorus deficiency induced the presence of a unique protein (16.2 kDa). Under conditions of phosphorus limitation, the activities of both superoxide dismutase and peroxidase per cell in vivo were lower than under normal conditions in the last cultivation. The in vivo absorption spectra of cells showed chlorophyll absorption peaks at 676 and 436nm, over 10nm red-shifted from the normal position; cells showed an absence of a chlorophyll c with an in viva absorption peak at 623nm and an extraction absorption peak at 617nm. The chlorophyll a/carotene and chlorophyll a/xanthophylls ratios decreased under conditions of phosphorus limitation, photosynthetic efficiency (Fv/Fm) was clearly lower, and the low-temperature fluorescence emission spectra indicated a higher peak at 683nm and a lower peak at 721nm relatively, with the 721nm peak drifting slightly to the red and the 683 nm peak strengthened with a weakened 692nm shoulder peak.  相似文献

Many animal species migrate over long distances, but the physiological challenges of migration are poorly understood. It has recently been suggested that increased molecular oxidative damage might be one important challenge for migratory animals. We tested the hypothesis that autumn migration imposes an oxidative challenge to bats by comparing values of 4 blood-based markers of oxidative status (oxidative damage and both enzymatic and nonenzymatic antioxidants) between Nathusius' bats Pipistrellus nathusii that were caught during migration flights with those measured in conspecifics after resting for 18 or 24 h. Experiments were carried out at Pape Ornithological Station in Pape (Latvia) in 2016 and 2017. Our results show that flying bats have a blood oxidative status different from that of resting bats due to higher oxidative damage and different expression of both non enzymatic and enzymatic antioxidants (glutathione peroxidase). The differences in oxidative status markers varied betwee n sampli ng years and were in depende nt from in dividual body con dition or sex. Our work provides evidence that migratory flight might impose acute oxidative stress to bats and that resting helps animals to recover from oxidative damage accrued en route. Our data suggest that migrating bats and birds might share similar strategies of mitigating and recovering from oxidative stress.  相似文献

Electrofusion between blastula cells and unfertilized eggs in loach were investigated usingdielectrophoretic field where, under alternating sinusoidal electric field, blastula cells formed beads-like chain in close contact with the unfertilized egg and cell fusion occurred between eggs and thecells in tight contact with them. The nuclei ofblastula cells were brought into the cytoplasm of therecipient eggs, where they promoted the development of the fused eggs just like the zygote nuclei.But the development of the fused eggs was different from that of zygotes. Several nuclei might enterone and the same egg simultaneously and all of them could undergo division, resulting in severalblastomere after the first cleavage of the recipient egg. Before blastula stage, the embryo developingfrom the fused egg showcd irregular shape, but it was soon regulated and developed to a normalblastula which often continued its development into a normal individual. Cell/egg electrofusion cameto its highest fosion rate (80%) 8nd hatching rate (20%), with cell density at 1×10~3 cells/ml, Ca~(++)concentration at 10 mM, mannitol at 0.2 M and when the blastula cells were digested with 100μg/ml pronase E for 6-10 min at 20℃. The mechanism underlying development of electrofused eggsis discussed. As the result indicates, electrofusion might prove to be a promising biotechnology justas nuclear transplantation.  相似文献

In glacial lakes on an alpine pasture in Switzerland, benthic cyanobacteria produced microcystin, a cyclic hepatotoxic heptapeptide. The cyanobacteria formed dense mats on sediments and submerged stones. The mats consisted mainly of Oscillatoria limosa, Phormidium konstantinosum (= Oscillatoria tenuis) and Tychonema granulatum (= Oscillatoria granulata). In order to characterize the ecological conditions of these cyanobacteria, nutrient concentrations were determined, and an automatic data acquisition station was installed in one of the lakes. It continuously recorded air temperature, global irradiance, precipitation, atmospheric pressure, wind speed and direction; as well as temperature, pH, oxygen content and conductivity of the lake water. The nutrient situation in the lakes was mainly influenced by the erosion of the gneissic catchment by glacial meltwater and by precipitation. In the glacial lakes, the concentrations of calcium, iron, magnesium, sodium and sulphate increased throughout the summer season. Conductivity values of 4–110 μS cm-1 represent generally low nutrient concentrations. Nevertheless, iron concentrations of up to 20 μM occurred. Biomass, expressed as protein concentration, as well as the microcystin content of the cyanobacterial mats varied within one season and between different years (1994 and 1995). In one cyanobacterial mat community, biomass and microcystin concentrations were highest at the same time, in an other one the microcystin content was maximal three weeks after the highest biomass concentration was reached. Our observations suggest that biomass and toxin production in the mats were strongly influenced by mechanical stress, temporary desiccation and high irradiation. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
