新疆青海中度嗜盐放线菌生物多样性初步研究   总被引:9,自引:4,他引:5  
从我国新疆、青海等地采集数份盐碱土样或泥样,采用淀粉-酪素琼脂培养基、甘油天门冬酰胺琼脂培养基、土壤浸汁琼脂培养基分别从中分离到8株、32株中度嗜盐放线菌菌株。经形态、生理学特性与全细胞壁氨基酸组分分析结果比较,选取其中的14株进行16S rDNA序列分析。就物种多样性而言,新疆、青海分离到中度嗜盐放线菌分布至少有3个科,5个属。其中有拟诺卡氏菌科(Nocardiopsaceae)的拟诺卡氏菌属(Nocardiopsis)和链单孢菌属(Streptomonospora);假诺卡氏菌科(Pseudonocardiaceae)的普氏菌属(Prauserella)和糖单孢菌属(Saccharomonospora);链霉菌科(Streptomycetaceae)的链霉菌属(Streptomyces)。就地区分布来讲,新疆分离到的中度嗜盐放线菌种类要远高于青海。  相似文献

从广西地区的土样中,分离到4株细胞壁Ⅳ型,糖型A, 无枝菌酸的假诺卡氏菌科的放线菌菌株,编号分别为191、221、202、和212。根据4株 菌的形态学特征和细胞化学特征,将其归入糖单孢菌属。与该属5个已知种的7个代表株进行 的rDNA的BamHI酶切片段长度类型分析(Ribotyping)的结果表明:191为青绿色糖单孢菌(S.viridis),202为青灰色糖单孢菌(S.caesia),221和212为相同的与青绿色糖单孢菌(S .viridis)的亲缘关系最近的种,但不同于已知的任何一个种。  相似文献

从山东荣成养鱼场发病牙鲆分离到一株病原菌M3,革兰氏阴性,杆状,能运动,菌落半透明,用BIOLOG细菌鉴定系统不能鉴定。通过16S rDNA序列分析和同源性检索发现M3菌株与弧菌属的同源性较高,为94%~98%。系统发育学分析表明菌株M3与鳗弧菌关系最近,相似性为996%,其生化性状也和鳗弧菌的特征相似,故可把M3定为鳗弧菌(Vibrio anguillarum)。  相似文献

从江苏无锡土壤中分离到两株玫瑰小双孢菌SIPI226和SIPI207,经形态、化学分析、Ribotyping及16S rRNA分析,两菌株细胞壁含meso\|DAP、磷酸类脂PIV、无枝菌酸,醌为MK9(H0,H2,H4),G+C mol%分别为683和694。经初步鉴定为玫瑰小双孢菌的两个新亚种:玫瑰小双孢菌无锡亚种(Microbispora rosea subsp. wuxiensis)和玫瑰小双孢菌鼋头渚亚种(Microbispora rosea subsp. yuantouzhuensis)。菌株SIPI226和SIPI207分别为玫瑰小双孢菌无锡亚种和玫瑰小双孢菌鼋头渚亚种的典型菌株。  相似文献

一株茶碱降解菌的分离和鉴定   总被引:6,自引:0,他引:6  
从制药废水生物处理系统的活性污泥中经富集分离到一株能以茶碱作为唯一碳、氮源生长的茶碱降解菌Tcn3,该菌株可以利用茶碱的最高浓度为3 000 mg/L。当茶碱浓度为1000 mg/L时被彻底降解的时间仅需48 h。Tcn3菌株降解茶碱的最适pH为80。K+是该菌株降解茶碱的必需元素。采用16S rDNA序列分析法及传统的生理生化特征鉴定法对该菌株进行鉴定,结果表明,Tcn3的16S rDNA的核苷酸序列与善变副球菌(Paracoccus versutus) ATCC 25364的同源性为997%,在细菌系统发育分类学上属于变形菌α亚类,Rhodobacter组:副球菌属,善变副球菌。  相似文献

将耐辐射奇球菌(Deinococcus radiodurans)的recA基因克隆到表达质粒pET15b中,并在Escherichia coli HMS(DE3)中高效表达了可溶性的RecA重组蛋白。同时将recA基因通过穿梭质粒pRADZ3导入recA缺损E. coli TG2细胞中,Western印迹实验显示RecA蛋白能够在不需要诱导剂IPTG的条件下稳定表达。辐射抗性实验表明,D. radiodurans的recA基因在E. coli细胞中的表达能够完全补偿recA缺损E. coli辐射抗性能力。  相似文献

辐射过程中耐辐射奇球菌蛋白酶变化的检测与分析   总被引:3,自引:2,他引:1  
采用明胶和酪蛋白底物酶谱法以及荧光酪蛋白底物对紫外线以及γ射线辐射后恢复期耐辐射奇球菌R1(Deinococcus radiodurans R1,DRR1)的蛋白酶变化进行了检测。结果发现,DRR1存在高活性大分子量组成性表达蛋白酶,与Karlin等[16]提出的DRR1蛋白酶为预测高表达蛋白(PHX)的设想一致。DRR1包含大量分子量大于140kD 的明胶降解酶和分子量大于120kD的酪蛋白降解酶,其中活性最高的174kD明胶酶在经SDS变性处理后仍有较高活性,该蛋白酶在DRR1受紫外线辐射和电离辐射后恢复期的表达模式存在差异,在γ射线电离辐射过程中以及电离辐射后恢复的晚期活性较高。此外,还发现一些蛋白酶特异性由辐射所诱导,表明这些蛋白酶可能参与细胞信号通路中蛋白的顺序降解,也提示DRR1损伤修复过程中细胞内存在一个精确的蛋白酶系统。这些蛋白酶的表达与细胞的营养状态相关。同时对一株由本实验室从北京地区土壤中分离到的杆状耐辐射菌RR533.2的明胶和酪蛋白蛋白酶谱进行了测定,结果发现其蛋白酶谱与DRR1相类似。  相似文献

从深海样品ES0109中分离到一株具有高内切葡聚糖酶活力的细菌DY3, 16SrDNA序列分析表明该菌与交替假单胞菌属(Pseudoalteromonas sp.)的Pseudoalteromonas citreaPseudoalteromonas elyakovii的同源性为99%。PCR扩增DY3的内切葡聚糖酶基因cel-X全长1479bp,编码一个492AA的蛋白质。酶的氨基酸序列分析表明CelX与Pseudoalteromonas haloplanktis的内切葡聚糖酶CelG有95%的相似性,包括一个糖基水解酶家族5的催化结构域,一个连接序列和位于C端的的CBM5结构域。对酶性质的初步研究发现,CelX的最适温度为40 ℃,酶的最适pH在6~7之间。  相似文献

本文鉴定了一株可完全降解直链烷基苯磺酸盐(Linear Alkylbenzene Sulphonate,简称LAS)的菌株GZ6。革兰氏染色阴性,细胞为杆状或短杆状,大小为0.5μm~0.8 μm×1.0μm~2.0 μm,其生长pH范围为pH6.0~10.0,最适生长pH为7.0,生长温度范围为4℃~40℃,最适生长温度为30℃。生化特征测定除过氧化氢酶、尿酶、精氨酸脱羧酶反应为阳性,其它均为阴性。可利用Chloridazon、安替比林(antipyrin)以及LAS等为碳源。不能利用大多数糖醇。醌组分以泛醌Q10为主。菌体脂肪酸主要为C18∶1、C16∶0及C16∶1。DNA中G+C mol%含量为7010。16S rRNA 序列分析表明菌株GZ6 与其亲缘关系最近的菌株Phenylobacterium immobile DSM1986T序列相似值为9749%,DNADNA杂交率为40%。菌株GZ6具极生鞭毛,可运动,两者在细胞形态有很大差异。故将菌株GZ6定为苯基杆菌属的新种可动苯基杆菌(Phenylobaterium mobile) GZ6。  相似文献

从南极乔治王岛土壤分离到一株诺卡氏菌形放线菌C3905菌株。其气生菌丝白色,基内菌丝无色至乳脂或浅粉;菌丝直径0.5~0.8μm,断裂为杆状和球状体,表面光滑。胞壁化学I型;无枝菌酸;磷酸类酯PI型;优势甲基萘酯为MK9(H4)。DNA中G+C含量为68.3~68.9mol%。兼性嗜冷,生长适温为15℃~20℃。产生抗肿瘤抗生素sandramycin。基于以上特征及分子遗传分类的研究结果,我们建议C3905菌株作为白色类诺卡氏菌的一个变种,命名为白色类诺卡氏菌南极变种,Nocardioides albus var.antarcticus。  相似文献

A gram-negative, rod-shaped, gas-vacuolate, aerobic, heterotrophic bacterium was isolated from Spirit Lake, Washington. It was further characterized by phase-contrast and electron microscopy and phenotypic tests. The isolate does not utilize carbohydrates. Deoxyribonucleic acid-ribosomal ribonucleic acid (rRNA) hybridizations showed that this bacterium is a member of rRNA superfamily IV where it occupies a separate position on the Azorhizobium caulinodans rRNA sub-branch which, is part of an rRNA cluster containing also the Beijerinckia, the Bradyrhizobium japonicum and the Rhodopseudomonas palustris sub-branches. Its relationship to other gas-vacuolated genera within rRNA superfamily IV is discussed. A new genus Aquabacter with one species Aquabacter spiritensis is proposed. The type strain is strain SPL-1 (=ATCC 43981=LMG 8611).  相似文献

A red-pigmented, radiation-resistant, Gram-negative, rod-shaped bacterium isolated from irradiated pork is described. The D10 values in buffer solution and on pork mince are 3·45 and 5·05kGy respectively. The strain has been identified as a Deinobacter species.  相似文献

The isolation and characterization of a denitrifying bacterium that is both moderately halophilic and alkaliphilic is described. The organism was isolated for use in the development of a bioprocess that could potentially reduce the costs of ion exchange resin regenerant disposal. The process of ion exchange, after resin regeneration, produces a briny, alkaline waste that is difficult and expensive to dispose. The biological removal of nitrate and subsequent reuse of these brines can potentially provide a cost-saving alternative to disposing of this waste product. To achieve our objective, a moderately halophilic, alkaliphilic bacterium was isolated from sediment samples taken from the salt plain of Alkali Lake in Washington State (USA). The haloalkaliphilic bacterium, designated strain 4A, is motile with rod-shaped cells that are 3 to 5 microm long and 1 microm wide. Electron acceptors used include oxygen, nitrate, and nitrite. In addition, it has similar specific nitrate reduction rates and biomass yields as non-halophilic denitrifying bacteria. It is capable of using a variety of electron donors. This organism can grow at NaCl concentrations ranging from 0.2 to 4.5 M with optimum growth occurring at 1.5 M and pH values ranging from 6 to 12 with 9.5 being the optimum pH. The temperature range for growth of strain 4A is 4-50 degrees C with optimal growth occurring at 30 degrees C. The G + C content is 66 mol%. Phylogenetic analyses based upon 16S rDNA gene sequence placed isolate 4A in the genus Halomonas. In addition, DNA-DNA hybridization experiments clearly indicate that it is a unique species. Phenotypic and phylogenetic studies indicate that isolate 4A represents a new species. We propose the name Halomonas campisalis for this species and strain 4A (ATCC 700597) as the type strain. Due to its denitrification ability, broad carbon utilization range and its high salinity and pH tolerance this organism, and similar ones, hold promise for the treatment of saline, alkaline waste.  相似文献

A Gram-negative, strictly aerobic, coccoid to short rod-shaped marine bacterium strain MKT107(T) was isolated from the molluscan top shell Omphalius pfeifferi pfeifferi collected on the coast of Japan. Phylogenetic analysis based on the 16S rRNA gene sequence showed that strain MKT107(T) constitutes a new lineage in alpha-Proteobacteria related to the genera Nereida, Roseobacter, Staleya, Oceanibulbus and Sulfitobacter. Strain MKT107(T) was found to require salt for its growth and to be mesophilic. It contained 18:1omega7c and 16:0 as major cellular fatty acids and 3-OH 10:0 and 3-OH 12:0 as hydroxy fatty acids. The DNA base composition of the isolate was 61.6 mol% G+C. The major quinone was Q-10. Sufficient differences existed to distinguish this strain from currently recognized bacterial genera. Therefore, the isolate is classified as representing a new genus and species, Tateyamaria omphalii gen. nov., sp. nov. (type strain MKT107(T) =IAM 15108(T) =KCTC 12333(T); GenBank accession no. A thermophilic facultative bacterial isolate was recovered from 3.2km depth in a gold mine in South Africa. This isolate, designated GE-7, was cultivated from pH 8.0, 50 degrees C water from a dripping fracture near the top of an exploration tunnel. GE-7 grows optimally at 65 degrees C and pH 6.5 on a wide range of carbon substrates including cellobiose, hydrocarbons and lactate. In addition to O(2), GE-7 also utilizes nitrate as an electron acceptor. GE-7 is a long rod-shaped bacterium (4-6microm longx0.5microm wide) with terminal endospores and flagella. Phylogenetic analysis of GE-7 16S rDNA sequence revealed high sequence similarity with G. thermoleovorans DSM 5366(T) (99.6%), however, certain phenotypic characteristics of GE-7 were distinct from this and other previously described strains of G. thermoleovorans.  相似文献

A crescentic Gram-negative rod-shaped bacterium motile by a laterally inserted tuft of flagella was isolated from a boggy ditch water habitat. Cells occurred usually singly or in pairs, but sometimes short chains, long helical cells or spheroplasts with flagella still attached were observed. Its metabolism was obligate fermentative. The fermentation of glucose yielded mainly acetate and propionate. It grew with a generation time of 1 h 50 min. The DNA base ratio was found to be 51.6 mol % G+C. The characteristics of this organism indicated that it belongs to the genus Selenomonas closely similar to and by its main characteristics identical with the rumen bacterium Selenomonas ruminantium. The differing characteristics — production of catalase and lower temperature optimum (25°C) — interpretable as the result of adaptation to the specific environmental conditions may justify classification of the isolate into a new subspecies of S. ruminantium named Selenomonas ruminantium subsp. psychrocatalagenes. Additional information on the DNA base composition in strains of Selenomonas ruminantium (GA 192 and HD 1) was obtained.  相似文献

Cecal microbiota of chicken was screened for bacteria involved in the biotransformation of isoflavones. A new facultative anaerobic bacterium, capable of deglycosylation of the isoflavone genistin, was isolated and identified as a Lactobacillus delbrueckii -like strain. The isolate MF-07 was Gram-positive, facultatively anaerobic, catalase negative, non-spore-forming, nonmotile and a straight rod. The polyphasic taxonomic data, along with 16S rRNA gene sequence comparison, demonstrated that the isolate MF-07 was most closely related to L. delbrueckii group of the Lactobacillus genus. Considerable amounts of genistein were accumulated with genistin as a substrate within the first 12 h of fermentation. Formononetin and daidzein were not metabolized. The influence of several carbon sources on the growth of the isolate MF-07 and biotransformation of genistin was also investigated. This is the first study in which an anaerobic Lactobacillus bacterium from the chicken intestinal tract that metabolizes genistin to produce its bioactive metabolite was identified and characterized.  相似文献

A study of the ultrastructural characteristics of an intracellular bacterium infecting the redclaw crayfish, Cherax quadricarinatus, a pathogen referred to previously as a rickettsia-like organism (RLO), revealed the presence of different developmental stages. These included a rod-shaped and uniformly electron-dense elementary body (EB) and an intermediate body (IB). The length of the EB varied between 0.48 and 0.6 microm, and the diameter was 0.3 microm. The IB was 0.75 to 1.1 microm long by 0.36 to 0.44 microm in diameter. Although the EB of this bacterium has ultrastructural characteristics similar to those of Rickettsiella, no information is available regarding its genetic relationship to this genus, and the intracellular bacterium should continue to be referred to as a rickettsia-like organism. The hemocytes had different levels of infection, and the RLO proliferated inside these cells. The EB appeared to be free in the cytoplasm of infected hemocytes and other cells; however, this might be a fixation artifact. The EB was also contained in membrane-bound vacuoles along with the IB. RLO colonies were observed inside small granular cells. No large granular cells were observed in the sections examined; therefore, no data were obtained regarding infection of this type of hemocyte. The fixed phagocytes on the external side of the terminal hepatic arterioles had an activated interrupted layer containing RLO bacteria. Stem cells in the hematopoietic tissue were also infected, and some cells were apparently being released into circulation.  相似文献

A novel type of purple sulfur bacterium was isolated from a hypersaline sulfur spring on the shore of the Dead Sea. The cells of the isolate are irregularly rod-shaped or curved, and motile by means of a tuft of polar flagella. The photosynthetic system, containing bacteriochlorophyll a and carotenoids of the spirilloxanthin series, is located on stacks of lamellar membranes in the cell cytoplasm. The organism can grow either photoautotrophically with sulfide as electron donor, which is oxidized via extracellular sulfur to sulfate, or photoheterotrophically, using acetate, succinate, fumarate, malate or pyruvate as carbon sources. The bacterium is obligately anaerobic, and requires a source of reduced sulfur for growth. The isolate is moderately halophilic, and grows optimally at NaCl concentrations between 3 and 8%, temperatures between 30 and 45°C, and neutral pH. 16S ribosomal RNA oligonucleotide cataloging suggests a close relationship to purple sulfur bacteria of the genus Ectothiorhodospira. As the isolate differs greatly from the described members of the genus Ectothiorhodospira, we describe the isolate as a new species, and propose the name Ectothiorhodospira marismortui sp. nov.  相似文献
