低强度He-Ne激光对红细胞变形性的影响已得到广泛的认可和应用,但其具体调节机制尚不明确,通过研究低强度He-Ne激光对红细胞胞浆内钙离子浓度的影响,探讨其对红细胞变形性影响的机制。采用A23187处理过的红细胞,分别用5 mW和9 mW激光照射后,观察红细胞胞浆内钙离子浓度变化。结果显示:低强度He-Ne激光照射后的红细胞与无照射组的红细胞相比,红细胞胞浆内钙离子浓度显著降低,但在本实验中钙离子浓度的降低与激光照射剂量无显著相关。由此得出结论:降低红细胞胞浆内钙离子浓度可能是低强度He-Ne激光调节红细胞变形性的重要机制。  相似文献

分别采用5 mW/mm2 He-Ne激光辐照、10.08 kJ/m2·d 增强UV-B辐射及二者组合对"临优2018"小麦幼苗进行处理,通过紫外吸收法测定各处理组幼苗中微管结合蛋白MAP65s的含量,并且采用SDS-聚丙烯酰胺凝胶电泳对MAP65s蛋白进行鉴定,进一步分析He-Ne激光对增强UV-B辐射引起的小麦幼苗微管骨架损伤的修复效应.结果显示:经UV-B处理(B)后,小麦幼苗中的MAP65s蛋白含量低于对照组(CK)(P<0.01);单独激光(L)处理,MAP65s蛋白含量高于对照组(CK)(P<0.05);经He-Ne激光和UV-B复合处理(BL)后,MAP65s蛋白含量高于UV-B处理组,低于CK组.该结果表明:一定剂量的He-Ne激光辐照能够促进小麦幼苗微管结合蛋白MAP65s的合成,并且可能在一定程度上修复增强UV-B辐射对小麦幼苗微管骨架系统造成的损伤.  相似文献

Influence of light intensity and wavelength on the yield of free radicals in seeds of tomato and carrot.
Seeds of tomato ( Lycopersicum esculentum L. cv. Gribovsky-1180) and carrot ( Daucus carota L. cv. Santanet) were irradiated by continuous light of different wavelengths (white, blue, green, red, far-red and He-Ne laser) and different power density. Free radical output was measured using Electron Spin Resonance spectra (ESR). The dependence of the content of free radicals (FR) on the light power density was established. The yield of FR in the seeds irradiated with monochromatic light of the same power density was inversely proportional to the wavelength. The ESR spectra showed maximum absolute number of FR for white light. Successive irradiations gave cumulative effects on the output of free radicals in both directions: increase by the action of white light and decrease by the action of red and far-red light. The contact of irradiated seeds with steam decreased the amount of FR.  相似文献

In this study, Aspergillus terreus was irradiated by a 7.3 mW He–Ne laser in the presence of crystal violet, toluidine blue O and hematoporphyrin as photosensitizers. Xylanases recovered from non-irradiated and irradiated fungi were purified and characterized. The maximum production of xylanase (42.2 U/ml) was obtained after 5 min of laser irradiation in the absence of the photosensitizer. The irradiation of the sensitized fungus diminished the production of xylanase. On purification using G-100, the specific activity of xylanase recovered from the irradiated fungus was 292 U/mg protein representing a 37-fold purification over the crude extract compared with 95.6 U/mg protein representing the 12.8-fold for the enzyme recovered from the non-irradiated fungus. The enzyme recovered from the irradiated fungus had lower molecular weight as compared with that recovered from the non-irradiated one. Characterization of the purified enzymes revealed that the enzyme recovered from the irradiated fungus was more thermostable and had a wider range of optimum reaction temperature (60–70°C) and pH (4.0–12.0), compared to the non-irradiated one.  相似文献

Mitochondrial alterations were monitored after low power green laser (532 nm, 30 mW) irradiation in the case of whole cells (B-14) and isolated mitochondria (from Wistar rat heart). 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium-bromide (MTT) assay products were significantly higher (by 8%) in irradiated B-14 cells as compared to non-irradiated controls. Mitochondrial transmembrane potential of B-14 cells, measured by means of a fluorescent probe 3,3'-dihexyloxacarbocyanine iodide (DiOC6(3)), significantly increased (by 13%) after exposure to green laser irradiation. Another MTT assay was used for isolated mitochondria suspensions in order to examine the effect of green laser irradiation on stimulation of processes related to oxidative phosphorylation. It revealed 31.3%-increase in MTT assay products in irradiated mitochondria as compared to controls. Laser irradiation of isolated mitochondria suspension did not significantly change 1,6-diphenyl-1,3,5-hexatriene (DPH) fluorescence anisotropy, indicating that mitochondrial membrane fluidity was not affected by laser light. Fluorescence emission spectra of irradiated as well as non-irradiated mitochondria suspensions showed fluorescence maximum at 635 nm, corresponding to emission of Protoporphyrin IX, which was significantly lower (by 20.7%) in irradiated sample.  相似文献

研究不同功率的低强度He-Ne激光对正常人体红细胞流变学特性影响。以正常人体红细胞为研究对象,测量了低强度He-Ne激光在不同照射时间、不同功率条件下红细胞的变形、取向、膜流动性、膜的微粘度和渗透脆性的变化情况。结果表明:照射后红细胞的变形性和膜流动性增强、渗透脆性下降。照射对红细胞流变学特性影响显著,其中激光能量为0.24 J、照射血样为2 mL时取得的照射效果最佳。  相似文献

The effect of He-Ne laser radiation (632.8 nm; 0.3 mW/cm parallel to oxygenation on metabolism indices of blood, taken in early postresuscitation period after discontinuation of hemorrhagic shock was studied in vitro. Compared to oxygenation alone, combined therapy resulted in more marked oxygen consumption of the blood, normalization of plasma antioxidant total activity, superoxydismutase erythrocyte catalase activity, the number of double connections in fatty acid chain of general lipids and membrane viscosity were elevated. Regular dependence of lipid peroxidation on a number of metabolic blood parameters is revealed.  相似文献

本文应用HeNe激光对甘蔗糖蜜工业性生产用酿酒酵母菌SaccharomycescerevisiaeAS2.1189进行辐照处理,经酵母菌糖蜜酒精发酵试验,对产乙醇含量进行了气相色谱分析,发现HeNe激光对酿酒酵母菌具有明显的生物刺激效应和可能的诱变作用,并初步筛选到产乙醇含量有较大变化的辐照变异菌株;同时,通过对这些辐照变异菌株的乙醇脱氢酶同工酶的分析,进一步证实了HeNe激光对酿酒酵母菌的诱变作用。这就为工业上利用He-Ne激光对酿酒酵母菌进行诱变育种展现新的前景。  相似文献

The seed embryos of Isatis indigotica Fort were exposed to He-Ne laser (5.23 mW/mm2, radiated for 5 min) and microwave (1.26 mW/mm2, radiated for 8 s) irradiation to determine the effects of microwave and He-Ne laser pretreatment on enzyme activities, and biophoton emission of cotyledon. Then: (i) changes in the activities of enzymes in I. indigotica cotyledon (such as amylase, transaminase, and proteinase) were measured to investigate the effects of He-Ne laser and microwave pretreatment; and (ii) biophoton emission was measured to determine the speed of cell division and metabolism. Results from these experiments indicated that: (i) the activities of amylase, transaminase, and proteinase of the cotyledon pretreated by HeNe laser and microwave were significantly increased; and (ii) the intensity of biophoton emission was enhanced significantly by He-Ne laser and microwave irradiation. These changes suggest that He-Ne laser and microwave pretreatment can improve the inner energy of seeds, lead to an enhancement of cotyledon enzymes, and speed up the metabolism of the cell, resulting in significantly increased biophoton emission.Moreover, the mechanism of action of the effects of laser and microwave radiation on the microcalorimetric parameters, enzyme activities, and biophoton emission of seeds is discussed on the basis of the results obtained.  相似文献

为了解低强度氦氖激光对心肌细胞超微结构、超氧化物歧化酶(SOD)和丙二醛(MDA)的影响,采用2.87J/cm^2,7.17J/cm^2和28.66J/cm^2三种剂量氦氖激光照射大鼠心前区,电镜观察心肌超微结构,测定心肌组织SOD和MDA的含量。结果发现心肌线粒体有轻微损伤,心肌组织SOD含量在7.17J/cm^2剂量显著下降(P<0.05),而在28.66J/cm^2剂量显著升高(P<0.05),心肌组织MDA含量在7.17J/cm^2和28.66J/cm^2两种剂量下降有显著升高(P<0.01),在2.87J/cm^2剂量,心肌组织SOD和MDA含量均无显著变化(P<0.05)。结果提示心肌SOD含量和脂质过氧化随不同的激光剂量而变化。  相似文献

采用He-Ne激光(5 mW/mm2)辐照增强UV-B辐射(10.08 kJ/m2.d)的晋麦8号小麦幼苗,通过测定小麦幼苗叶片细胞质膜透性、丙二醛(MDA)的含量以及脂氧合酶(LOX)、过氧化氢酶(CAT)、抗坏血酸过氧化物酶(APX)和谷光苷肽过氧化物酶(GPX)的活性变化,研究He-Ne激光对增强UV-B辐射的小麦幼苗膜脂过氧化的影响。结果显示,He-Ne激光辐照可使经UV-B辐射后小麦幼苗叶片质膜相对透性、MDA含量、LOX活性降低,而使CAT、APX和GPX的活性均升高。分析表明UV-B辐射增强可导致膜脂过氧化加剧,而一定剂量的He-Ne激光能够通过促进酶促抗氧化系统酶的活性来缓解紫外线辐射下小麦幼苗的膜脂过氧化作用。  相似文献

The biostimulating activity of low level laser radiation of various wavelengths and energy doses is widely documented in the literature, but the mechanisms of the intracellular reactions involved are not precisely known. The aim of this paper is to evaluate the influence of low level laser radiation from an multiwave locked system (MLS) of two wavelengths (wavelength = 808 nm in continuous emission and 905 nm in pulsed emission) on the human erythrocyte membrane and on the secondary structure of human serum albumin (HSA). Human erythrocytes membranes and HSA were irradiated with laser light of low intensity with surface energy density ranging from 0.46 to 4.9 J cm?2 and surface energy power density 195 mW cm?2 (1,000 Hz) and 230 mW cm?2 (2,000 Hz). Structural and functional changes in the erythrocyte membrane were characterized by its fluidity, while changes in the protein were monitored by its secondary structure. Dose-dependent changes in erythrocyte membrane fluidity were induced by near-infrared laser radiation. Slight changes in the secondary structure of HSA were also noted. MLS laser radiation influences the structure and function of the human erythrocyte membrane resulting in a change in fluidity.  相似文献

在临床应用中,低功率He-Ne激光(632.8 nm)能促进骨骼肌修复,加速创伤愈合,降低牙齿的超敏感性,减缓疼痛等.大量研究表明:低功率He-Ne激光能调节细胞的众多行为,如细胞增殖、分泌、迁移、粘附、蛋白质合成和基因表达等.但低功率He-Ne激光调节细胞行为的分子机制并未阐明,考察低功率He-Ne激光照射后细胞内活性氧自由基的产生水平和游离ca2 浓度是否会发生变化,通过激光扫描共聚焦显微镜,分别利用H:DCFDA和Fluo-3/AM这两种荧光探针,检测到经He-Ne激光照射后,肺腺癌细胞内活性氧自由基的水平上调以及游离Ca2 浓度增加.该研究为低功率He-Ne激光的生物光刺激效应提供了可能的分子机理.  相似文献

The effects of low laser irradiation on angiogenesis in injured rat tibiae   总被引:4,自引:0,他引:4  
The influence of He-Ne laser radiation on the formation of new blood vessels in the bone marrow compartment of a regenerating area of the mid-cortical diaphysis of the tibiae of young adult rats was studied. A small hole was surgically made with a dentistry burr in the tibia and the injured area received a daily laser therapy over 7 or 14 days transcutaneously starting 24 h from surgery. Incident energy density dosages of 31.5 and 94.5 Jcm(-2) were applied during the period of the tibia wound healing investigated. Light microscopic examination of histological sections of the injured area and quantification of the newly-formed blood vessels were undertaken. Low-level energy treatment accelerated the deposition of bone matrix and histological characteristics compatible with an active recovery of the injured tissue. He-Ne laser therapy significantly increased the number of blood vessels after 7 days irradiation at an energy density of 94.5 Jcm(-2), but significantly decreased the number of vessels in the 14-day irradiated tibiae, independent of the dosage. These effects were attributed to laser treatment, since no significant increase in blood vessel number was detected between 8 and 15 non-irradiated control tibiae. Molecular mechanisms involved in low-level laser therapy of angiogenesis in post-traumatic bone regeneration needs further investigation.  相似文献

目的:探讨低强度氦氖激光对临床中风等缺血性疾病患者红细胞变形能力影响情况。方法:临床采集不同缺血性疾病患者新鲜血样,检测各组红细胞变形能力情况。对中风患者血样进一步分组,给予不同剂量的氦氖激光处理,讨论临床低强度氦氖激光量效关系,探讨具有临床疗效的有效剂量情况。结果:各疾病病组红细胞变形能力较正常对照组红细胞变形能力为低;缺血疾病组样品经低强度氦氖激光照射后,其红细胞变形能力较未经激光照射组有显著不同。结论:显示体外人红细胞变形能力因患者疾病情况、红细胞状态、低强度氦氖激光临床治疗输出功率等因素有不同。  相似文献

采用He-Ne激光辐照对增强UV-B辐射后小麦幼苗ROP GTPase损伤修复作用进行了研究。采用了SDS-PAGE电泳法检测各组ROP GTPase的含量和激光共聚焦显微镜对小麦微丝进行FITC荧光强度标记的测定。研究结果表明:经增强UV-B辐射后,小麦幼苗的Rop GTPase含量降低,LCSM扫描细胞原生质体形状发生改变,细胞骨架受到破坏,其被标记的荧光变暗,强度减弱,在整个UV-B处理期间均低于对照组(CK),再以He-Ne激光处理后,其含量,形状和强度均有所提高,但仍低于对照组。由此说明,UV-B辐射能使小麦幼苗的Rop GTPase含量下降,微丝骨架受到破坏,一定剂量的激光对UV-B辐射后小麦的蛋白含量和细胞骨架有一定修复作用。Rop GTPase参与了微丝骨架重组的过程。  相似文献

采用功率为3.50 mW/mm2的He-Ne激光处理不同时期蚕豆,研究其四叶期叶片中丙二醛(MDA)含量,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性及同工酶谱的变化。结果表明,与对照相比,He-Ne激光处理不同时期蚕豆,MDA含量均显著降低,且各处理间没有显著差异;SOD、POD、CAT酶活性有不同程度提高。SOD、POD同工酶的酶谱在浸种24小时和胚芽露头期经激光处理后改变,在一叶期处理后没有改变;CAT同工酶谱没有变化;在一叶期处理的三种同工酶谱都没有改变。  相似文献

采用经紫外线(UV)、^60Co、亚硝基胍(NTG)复合诱变得到的黄曲霉曲酸产生菌(UCN7—12),进行激光诱变处理。研究证实在经过UV、^60Co、NTG诱变处理后,黄曲霉突变株用He—Ne激光与YAG激光进行诱变处理仍能提高产酸率,其中He—Ne激光辐照处理20min,正变率为12.1%,产量提高约13%。YAG激光辐照处理300sec,正变率16.7%,产量提高18.3%。说明上述两种激光对黄曲霉曲酸产生菌有一定的诱变效应。  相似文献
