用丙肝病毒C+E1区真核表达质粒pcDNA-HCV/C+E1按400ug/只剂量免疫BALB/C小鼠,14周后同一剂量再加强免疫一次。加强免疫后2周,在50%PEG1450介导下将脾细胞与SP2/0小鼠骨髓瘤细胞(51)融合。实验结果融合率达54.3%(313/576)。阳性率为5.4%(17”313)。克隆化后得到6株稳定分泌抗丙肝病毒C区单克隆抗体杂交瘤细胞株。这6株杂交瘤均产生IgM抗体,接种BALB/C小鼠后产生腹水的效价为164-1320(ELISA)。  相似文献

以草决明无菌苗子叶为外植体,接种于7类诱导愈伤组织的培养基上:A.MS+2.02,4-Dmg/L(以下单位省略)+0.3BA+0.2NAA.B.MS+0.22,4-D十0.2BA+2.0NAA.C.MS+1.02,4-D+0.5BA+0.2KT.D.MS+0.7BA+1.5NAA+0.1KT.E.MS+1.52,4-D+0.7BA十0.2MAA.F.MS+0.42.4-D+1.0NAA+0.1KT.G.MSB(MS的无机成份和B5有机成分)+0.15NAA+BA,KT和ZT各0.5。8~15天后分别有90~99.6%的子叶片被诱导出愈伤组织,并且F.与C.类培养基对诱导愈伤组织比较理想,放于G培养基上的愈伤组织有9.2~30.2%的芽分化率。芽在生根培养基1/2MS+0.2IBA中、98%生根,形成完整的再生植株。  相似文献

将构建好的可表达GST融合蛋白的重组病毒AcMNPV-OCC^--GST-6xHis-Etp28感染Sf9细胞,一定时间后取感染了病毒的细胞裂解物上清液进行SDS-PAGE分析,结果显示53kDa的融合蛋白(GST-6xHis-Etp28)呈不溶状态。在原有裂解液的基础上,加固体十二烷基肌氨酸钠致终浓度1.5%,并将Triton X-100的比例由1%提高到2%。SDS-PAGE结果显示至少有1/  相似文献

用浸泡法得到了(E160A)天花粉蛋白(trichosanthin, TCS),(E160D)TCS与Ade 和(E160A)TCS与FMP复合物的晶体.在Mar Research 面探测器系统上分别收集了0.20 nm ,0.19nm 和0.205 nm 分辨率的X 射线衍射数据,数据处理用Mar Scale 程序系统完成.用同晶差值Fourier法解析了(E160A)TCS-Ade,(E160D)TCS-Ade 和(E160A)TCS-FMP的晶体结构,结构修正利用X-PLOR程序,修正结果,晶体学R因子分别为0.166,0.176,0.179.键长和键角的RMS偏差分别为0.0010 nm 和2.503°,0.0013 nm 和2.665°,0.0012 nm 和2.676°.在这三个结构中均未见到Glu189侧链方向的改变.Ade 或FMP仍结合在N-糖苷酶活性口袋之中,它夹在Tyr70和Tyr111两个侧链环之间,与Tyr70环近乎平行.这一结果表明:TCS中的Glu160分别突变成Ala 和Asp,仍能与AMP发生N-糖苷酶反应,但是活性降低了一些.可见Glu160对TCS与AMP的作用是重要的,但不是必要的.  相似文献

将马铃薯Y病毒普通系(PVY0)的外壳蛋白基因克隆到表达质粒pMALc2中,构建这一基因在大肠杆菌中的表达载体pMALc2PVY0CP。SDSPAGE及Westernbloting检测结果表明,这一表达栽体在E.coliDH5α中经IPTG诱导可表达分子量为71.8kDa的特异性融合蛋白。以amyloseresin亲合柱层析纯化这一融合蛋白为抗原,免疫家兔制备了效价为1∶1024的特异性抗血清。用该抗血清可通过对流免疫电泳、免疫双扩散及Westernbloting对PVY进行检测  相似文献

血管内皮细胞和血管平滑肌细胞在结构和功能上关系密切,二者的相互关系在血管舒缩和血管壁结构的调节中起重要作用。本文观察了培养的小牛肺动脉内皮细胞(PAEC)和肺动脉平滑肌细胞(PASM)在细胞增殖方面的相互调节作用。混合培养的PAEC和PASM细胞的3H-TdR参入明显降低(P<0.001,与对照组相比)。无论向培养的PAEC和PASM中分别加入PASM和PAEC的条件培养基还是二者共培养时,均发现PAEC的3H-TdR参入明显降低,而PASM的3H-TdR明显升高(P<0.05,与对照组相比)。流式细胞测定也发现共培养时PAEC的G1期细胞增多,G2/M期细胞减少;而PASM的G1期细胞减少,G2/M期细胞增多。共培养的PASM细胞内cAMP增加,cGMP含量降低;而PAEC细胞的cAMP和cGMP含量均降低(P<0.01,与对照组相比)。上述结果提示,PAEC和PASM相互作用可能通过第二信使而调节它们本身的增殖  相似文献

Synexin enhances the aggregation rate but not the fusion rate of liposomes   总被引:3,自引:0,他引:3  
The effect of synexin on the calcium-induced fusion of large unilamellar liposomes was studied by using two assays for the mixing of aqueous contents. The results were analyzed in terms of the mass action kinetic model, which describes the overall fusion reaction as a two-step sequence consisting of a second-order process of liposome aggregation followed by a first-order fusion reaction. By using several different lipid compositions and varying the electrolyte composition, it was possible to select the rate-limiting step of the overall fusion process. When aggregation was the rate-limiting step, as in the case of Ca2+-induced fusion of phosphatidylserine (PS), phosphatidate (PA)/phosphatidylethanolamine (PE) (1:3), and PS/PE (1:3) liposomes, synexin increased the overall fusion kinetics by increasing the aggregation rate constant (up to 100-fold). When aggregation was rapid compared to destabilization of apposed membranes, i.e., fusion was rate limiting, synexin either had no effect or reduced the overall fusion kinetics. In one such case involving liposomes composed of PA/PS/PE/phosphatidylcholine (PC) (10:15:65:10), synexin reduced the fusion rate constant by 50%. The effect of calcium-induced synexin polymerization was investigated by preincubation of synexin with calcium prior to addition of liposomes. Prepolymerization by Ca2+ always decreased the activity of synexin such that it was less than the activity of an equal amount of untreated monomers. However, it was found that the activity of synexin monomers polymerized to an average hexameric size was greater than that of one-sixth as many untreated monomers, with respect to the liposome aggregation rate constant. Synaptosomal fractions isolated from rat cerebral cortex were incubated in the absence and presence of phosphatidylserine (PS) liposomes. The PS treated synaptosomes yielded ligher bouyant bands when centrifugated on discontinuous sucrose gradients. In these bands an increase in the lipidic phosphorus/protein ratio together with a massive incorporation of PS and a relative decrease of phosphatidylcholine was observed. Besides, in the synaptosomes treated with PS (0.5–0.7 μg/mg protein). Na+, K+-ATPase activity was 30–50% higher than in the controls.

We have employed an assay for lipid mixing based on the self-quenching relief of octadecylrhodamine B fluorescence. The decrease in the self-quenching of the probe in the gradient bands of liposomes treated synaptosomes, besides the chemical analysis data, indicated that a fusion process between synaptosomes and liposomes has taken place. It is the first time that a fusion process is proved between native synaptosomes and an artificial lipidic membrane.  相似文献

The initial kinetics of divalent cation (Ca2+, Ba2+, Sr2+) induced fusion of phosphatidylserine (PS) liposomes, LUV, is examined to obtain the fusion rate constant, f11, for two apposed liposomes as a function of bound divalent cation. The aggregation of dimers is rendered very rapid by having Mg2+ in the electrolyte, so that their subsequent fusion is rate limiting to the overall reaction. In this way the fusion kinetics are observed directly. The bound Mg2+, which by itself is unable to induce the PS LUV to fuse, is shown to affect only the aggregation kinetics when the other divalent cations are present. There is a threshold amount of bound divalent cation below which the fusion rate constant f11 is small and above which it rapidly increases with bound divalent cation. These threshold amounts increase in the sequence Ca2+ less than Ba2+ less than Sr2+, which is the same as found previously for sonicated PS liposomes, SUV. While Mg2+ cannot induce fusion of the LUV and much more bound Sr2+ is required to reach the fusion threshold, for Ca2+ and Ba2+ the threshold is the same for PS SUV and LUV. The fusion rate constant for PS liposomes clearly depends upon the amount and identity of bound divalent cation and the size of the liposomes. However, for Ca2+ and Ba2+, this size dependence manifests itself only in the rate of increase of f11 with bound divalent cation, rather than in any greater intrinsic instability of the PS SUV. The destabilization of PS LUV by Mn2+ and Ni2+ is shown to be qualitatively distinct from that induced by the alkaline earth metals.  相似文献

The synaptic vesicle protein synaptotagmin I (Syt I) binds phosphatidylserine (PS) in a Ca2+-dependent manner. This interaction is thought to play a role in exocytosis, but its precise functions remain unclear. To determine potential roles for Syt I-PS binding, we varied the PS content in PC12 cells and liposomes and studied the effects on the kinetics of exocytosis and Syt I binding in parallel. Raising PS produced a steeply nonlinear, saturating increase in Ca2+-triggered fusion, and a graded slowing of the rate of fusion pore dilation. Ca2+-Syt I bound liposomes more tightly as PS content was raised, with a steep increase in binding at low PS, and a further gradual increase at higher PS. These two phases in the PS dependence of Ca2+-dependent Syt I binding to lipid may correspond to the two distinct and opposing kinetic effects of PS on exocytosis. PS influences exocytosis in two ways, enhancing an early step leading to fusion pore opening, and slowing a later step when fusion pores dilate. The possible relevance of these results to Ca2+-triggered Syt I binding is discussed along with other possible roles of PS.  相似文献

In the present study we characterized the capacity of zinc to protect lipids and proteins from Fe2+-initiated oxidative damage. The effects of zinc on lipid oxidation were investigated in liposomes composed of brain phosphatidylcholine (PC) and phosphatidylserine (PS) at a molar relationship of 60:40 (PC:PS, 60:40). Lipid oxidation was evaluated as the oxidation of cis-parinaric acid or as the formation of 2-thiobarbituric acid-reactive substances (TBARS). Zinc protected liposomes from Fe2+ (2.5-50 microM)-supported lipid oxidation. However, zinc (50 microM) did not prevent the oxidative inactivation of glutamine synthetase and glucose 6-phosphate dehydrogenase when rat brain supernatants were oxidized in the presence of 5 microM Fe2+ and 0.5 mM H2O2. We also studied the interactions of zinc with epicatechin in the prevention of lipid oxidation in liposomes. The simultaneous addition of 0.5 microM epicatechin (EC) and 50 microM zinc increased the protection of liposomes from oxidation compared to that observed in the presence of zinc or EC separately. Zinc (50 microM) also protected liposomes from the stimulatory effect of aluminum on Fe2+-initiated lipid oxidation. Zinc could play an important role as an antioxidant in biological systems, replacing iron and other metals with pro-oxidant activity from binding sites and interacting with other components of the oxidant defense system.  相似文献

The interaction of spermine with acidic phospholipids was investigated for its possible relevance to membrane fusion. Equilibrium dialysis was used to measure the binding of spermine and calcium to large unilamellar vesicles (liposomes) of phosphatidate (PA) or phosphatidylserine (PS). Spermine bound to isolated PA and PS liposomes with intrinsic association constants of approximately 2 and 0.2 M-1, respectively. Above the aggregation threshold of the liposomes, the binding of spermine increased dramatically, especially for PA. The increased binding upon aggregation of PA liposomes was interpreted as evidence for the formation of a new binding complex after aggregation. Spermine enhanced calcium binding to PA, while it inhibited calcium binding to PS, under the same conditions. This difference explained the small effect of spermine on the overall rate of calcium-induced fusion of PS liposomes as opposed to the large effect on PA liposomes. The rate increase could be modeled by a spermine-induced increase in the liposome aggregation rate. The preference for binding of spermine to PA over PS suggested a preference for accessible monoesterified phosphate groups by spermine. This preference was confirmed by the large effects of spermine on aggregation and overall fusion rates of liposomes containing phosphatidylinositol 4,5-diphosphate. The large spermine effects on these liposomes compared with phosphatidate- or phosphatidylinositol-containing liposomes suggested that spermine has a strong specific interaction with phosphatidylinositol 4,5-diphosphate. Clearly, phosphorylation of phosphatidylinositol can lead to a large change in the spermine sensitivity of membrane fusion.  相似文献

The effects of different types of lipid membrane defects on Ca2+-induced fusion of liposomes containing phosphatidylserine (PS) were investigated using fluorescent probes. Teleocidin enhanced the fusion of phospholipid vesicles in an assay system using terbium/dipicolinic acid during mixing of internal aqueous phases of vesicles upon fusion. 12-O-Tetradecanoylphorbol-13-acetate (TPA) suppressed the fusion. This latter phenomenon was also observed by measuring the excitation energy transfer. The promotion of membrane fusion by teleocidin was ascribed to dehydration of the membrane surface, the suppressive effect of TPA to desorption of Ca2+ from the membrane surface. Thus, Ca2+-induced fusion of PS vesicles was shown to be sensitive to defects of the membrane surface, but insensitive to defects of the hydrophobic core of the lipid membrane.  相似文献

How the lipid composition of liposomes determines their ability to fuse with Sendai virus membranes was tested. Liposomes were made of compositions designed to test postulated mechanisms of membrane fusion that require specific lipids. Fusion does not require the presence of lipids that can form micelles such as gangliosides or lipids that can undergo lamellar to hexagonal phase transitions such as phosphatidylethanolamine (PE), nor is a phosphatidylinositol (PI) to phosphatidic acid (PA) conversion required, since fusion occurs with liposomes containing phosphatidylcholine (PC) and any one of many different negatively charged lipids such as gangliosides, phosphatidylserine (PS), phosphatidylglycerol, dicetyl phosphate, PI, or PA. A negatively charged lipid is required since fusion does not occur with neutral liposomes containing PC and a neutral lipid such as globoside, sphingomyelin, or PE. Fusion of Sendai virus membranes with liposomes that contain PC and PS does not require Ca2+, so an anhydrous complex with Ca2+ or a Ca2+-induced lateral phase separation is not required although the possibility remains that viral binding causes a lateral phase separation. Sendai virus membranes can fuse with liposomes containing only PS, so a packing defect between domains of two different lipids is not required. The concentration of PS required for fusion to occur is approximately 10-fold higher than that required for ganglioside GD1a, which has been shown to act as a Sendai virus receptor. Supramolecular aggregates containing cationic lipids have been widely used as transfection mediators due to their ability to interact with negatively charged DNA molecules and biological membranes. First steps of the process leading to transfection are partly electrostatic, partly hydrophobic interactions of liposomes/lipoplexes with cell and/or endosomal membrane. Negatively charged compounds of biological membranes, namely glycolipids, glycoproteins and phosphatidylserine (PS), are responsible for such events as adsorption, hemifusion, fusion, poration and destabilization of natural membranes upon contact with cationic liposomes/lipoplexes. The present communication describes the dependence of interaction of cationic liposomes with natural and artificial membranes on the negative charge of the target membrane, charges which in most cases were generated by charging the PS content or its exposure. The model for the target membranes were liposomes of variable content of PS or PG (phosphatidylglycerol) and erythrocyte membranes in which the PS and other anionic compound content/exposure was modified in several ways. Membranes of increased anionic phospholipid content displayed increased fusion with DOTAP (1,2-dioleoyl-3-trimethylammoniumpropane) liposomes, while erythrocyte membranes partly depleted of glycocalix, its sialic acid, in particular, showed a decreased fusion ability. The role of the anionic component is also supported by the fact that erythrocyte membrane inside-out vesicles fused easily with cationic liposomes. The data obtained on erythrocyte ghosts of normal and disrupted asymmetry, in particular, those obtained in the presence of Ca2+, indicate the role of lipid flip-flop movement catalyzed by scramblase. The ATP-depletion of erythrocytes also induced an increased sensitivity to hemoglobin leakage upon interactions with DOTAP liposomes. Calcein leakage from anionic liposomes incubated with DOTAP liposomes was also dependent on surface charge of the target membranes. In all experiments with the asymmetric membranes the fusion level markedly increased with an increase of temperature, which supports the role of membrane lipid mobility. The decrease in positive charge by binding of plasmid DNA and the increase in ionic strength decreased the ability of DOTAP liposomes/lipoplexes to fuse with erythrocyte ghosts. Lower pH promotes fusion between erythrocyte ghosts and DOTAP liposomes and lipoplexes. The obtained results indicate that electrostatic interactions together with increased mobility of membrane lipids and susceptibility to form structures of negative curvature play a major role in the fusion of DOTAP liposomes with natural and artificial membranes.  相似文献
