不同激素对花生离体分化的影响   总被引:8,自引:0,他引:8  
对TDZ和2,4-D等激素在花生成熟胚外植体分化中的影响进行了研究.结果表明,花生成熟胚3~5 d龄实生苗的幼叶和胚轴在低浓度TDZ的诱导下,可分化产生高频不定芽和少量体细胞胚,转到无激素MS培养基或MS BA 0.5 mg/L NAA 0.4 mg/L的培养基后形成丛生苗.丛生苗分离后转入含1/2 MS(大量元素) IBA 0.4 mg/L的培养基中诱导生根,可形成完整的再生植株.幼叶分化率高于胚轴,但胚轴分化成苗速度快.无菌水浸泡16~24 h的胚轴在5~ 30 mg/L 2,4-D的诱导下,分化产生低频不定芽;而胚叶则产生高频体细胞胚,但畸形较严重.  相似文献

本文报道利用花生成熟胚幼叶为外植体获得高频植株再生的方法,为花生转基因提供有效的受体系统。通过诱导培养基TDZ、BA、NAA的浓度以及种子萌发时间、继代培养基种类五个因素不同水平的正交试验,筛选出了分化高频发生的最佳组合为:MS培养基中应含有TDZ 1.0 μmol/L、BA 0.4 μmol/L、NAA 5.0 μmol/L,种子萌发4 d,继代培养基为MS0。本研究表明,五因素中诱导培养基TDZ浓度为诱导花生幼叶分化的主要影响因素,其次为继代培养基、种子萌发时间,而诱导培养基中BA和NAA的浓度作用较小。试管苗生根后移栽田间,可正常开花结果。  相似文献

本研究以粗肋草‘Red Valentine’带侧芽的根茎为外植体,研究不同消毒时间、不同种类外源激素对其根茎侧芽萌发诱导、愈伤组织诱导、丛生芽诱导、丛生芽增殖和生根的影响。结果表明:最佳灭菌时间为18 min,污染率为20.0%。MS+6-BA 2.0 mg/L+IBA 0.2 mg/L为最佳侧芽萌发诱导培养基,MS+TDZ 0.5 mg/L+2,4-D 2.0 mg/L为最优愈伤诱导培养基,1/2MS+TDZ 0.5 mg/L为最佳丛生芽诱导培养基,最佳丛生芽增殖培养基为MS+6-BA 3.0 mg/L+NAA 0.1 mg/L或MS+KT 4.0 mg/L+NAA 0.1 mg/L,最佳生根培养基为1/2MS+NAA 0.2 mg/L。本研究通过对粗肋草‘Red Valentine’进行离体培养,初步得出了适合外植体各时期生长的最佳培养基配方,为粗肋草试管苗的商业化、工厂化及规模化生产提供理论和技术指导。  相似文献

以登峰芥蓝为试材,取无菌苗的茎段、下胚轴为外植体分别进行组织培养研究。结果表明:MS+6-BA 3.0 mg/L+NAA 0.2 mg/L为下胚轴最适诱导培养基,MS+6-BA 5.0 mg/L+NAA 0.2 mg/L为带腋芽茎段最适诱导培养基;最适宜的不定芽增殖培养基为MS+6-BA 3.0 mg/L+NAA 0.2 mg/L;试管苗在1/2MS+NAA 0.1 mg/L+IBA 0.2 mg/L培养基中进行生根培养效果最好;经炼苗后组培苗移栽成活率达96.6%。  相似文献

以云南野生碧玉兰(Cymbidium lowianum)种子无菌萌发的试管苗为试验材料,进行其丛生芽诱导增殖、生根的组培快繁技术体系研究。结果表明:适宜的增殖培养基配方为1/2MS+6-BA 2.0 mg/L+KT 0.5 mg/L+NAA 0.2 mg/L,增殖率可达300%;生根培养基的配方为1/2MS+NAA 1.5 mg/L+6-BA 0.3 mg/L,其根数多,健壮。  相似文献

本研究以粗肋草‘Red Valentine’带侧芽的根茎为外植体,研究不同培养基类型、不同外源激素及浓度、不同转接周期对其愈伤组织及丛生芽诱导的影响,进而优化如意愈伤组织和丛生芽诱导培养基。结果表明:最佳愈伤组织诱导培养基为MS+0.5 mg/L TDZ+2.0 mg/L 2,4-D和1/2MS+0.5 mg/L TDZ+2.5 mg/L 2,4-D;最佳愈伤组织分化培养基为1/2 MS+0.4 mg/L TDZ,最佳转接周期为30 d;最佳丛生芽诱导培养基为5.0 mg/L6-BA+0.2 mg/L NAA和1/2 MS+0.5 mg/L TDZ,最佳转接周期为30 d。优化培养基后愈伤组织及丛生芽诱导效率更高。本研究通过设计优化试验,筛选出了愈伤组织诱导及丛生芽诱导的最佳培养基,为粗肋草属植物的快速繁殖及规模化生产提供了一定的理论指导。  相似文献

以江西铅山红芽芋(Colocasia esculenta L.Schott var.cormosus‘Hongyayu’)试管苗为材料,建立了芋球茎片两步法离体快繁体系,并对其再生苗的形态指标、染色体数目、生理和光合特性以及叶绿素荧光特性进行了检测。结果表明:(1)红芽芋球茎片单芽诱导的最佳培养基为MS+KT 2 mg/L+6-BA 1 mg/L+NAA0.1mg/L,诱导培养30d后将单芽从球茎片上分离,再接种到生根培养基(MS+KT 2mg/L+NAA 0.1mg/L)上培养30d即可形成完整植株,移栽成活率高达98%;(2)由球茎片单芽、丛生芽、不定芽离体快繁获得的红芽芋再生苗在形态指标、叶下表皮气孔参数、染色体数目、生理生化指标以及叶片光合特性参数和叶绿素荧光特性方面均无显著差异。说明红芽芋球茎片两步法离体培养的再生苗繁殖系数高、染色体数目稳定,该离体快繁体系可应用于江西铅山红芽芋的工厂化生产。  相似文献

以徐长卿子叶为材料,进行愈伤组织诱导、愈伤组织和不定芽分化、试管苗生根、移栽及移植等研究。结果表明,MS+6-BA 0.3 mg/L+NAA 0.2 mg/L+2,4-D 0.4 mg/L是子叶愈伤组织诱导和继代培养的理想培养基;MS+6-BA 0.8 mg/L+NAA 0.2 mg/L是愈伤组织分化培养的理想培养基;MS+6-BA 0.6 mg/L+NAA 0.2 mg/L是不定芽分化继代培养的理想培养基;炉灰渣是试管苗移栽的理想基质,移栽成活率可达97%。试管苗移植后长势旺盛,根系发达,当年开花。根据本试验结果可建立徐长卿子叶诱导再生体系。  相似文献

花生实生苗幼叶接种于MS TDZ 0.2mg/L NAA0.4mg/L诱导培养基上经诱导培养,继而转移到无激素培养基MS可获得不定芽和体细胞胚。组织学观察表明,花生不定芽和体细胞胚均起源于愈伤组织表层,不定芽为多细胞起源,而体细胞胚起源于单个胚性原始细胞。体细胞胚的发育经历多细胞原胚、球形胚、心形胚、鱼雷胚和子叶胚等时期发育成小植株。  相似文献

In 1996, the first documented outbreak of salmonellosis associated with the consumption of peanut butter was reported. This study was undertaken to determine survival characteristics of high (5.68 log10 cfu g(-1)) and low (1.51 log10 cfu g(-1)) inocula of a five-serotype mixture of Salmonella in five commercial peanut butters and two commercial peanut butter spreads. Populations in samples inoculated with 5.68 log10 cfu g(-1) and stored for 24 weeks at 21 or 5 degrees C decreased 4.14-4.50 log10 cfu g(-1) and 2.86-4.28 log10 cfu g(-1), respectively, depending on the formulation. The order of retention of viability was: peanut butter spreads > traditional (regular) and reduced sugar, low-sodium peanut butters > natural peanut butter. Differences in rates of inactivation are attributed to variation in product composition as well as size and stability of water droplets in the colloidal matrix, which may influence nutrient availability. With the exception of natural peanut butter, products initially inoculated with 1.51 log10 cfu of Salmonella g(-1) (32 cfu g(-1)) were positive for the pathogen after storage for 24 weeks at 5 degrees C. At 21 degrees C, however, with the exception of one peanut butter spread, all products were negative for Salmonella after storage for 24 weeks. Post-process contamination of peanut butter and spreads with Salmonella may to result in survival in these products for the duration of their shelf life at 5 degrees C and possibly 21 degrees C, depending on the formulation.  相似文献

Visual selection occurs when predators preferentially remove conspicuous varieties from prey populations and thus confer a selective advantage on inconspicuous varieties. In previous papers we have described a general method for simulating such natural selection, and we here give details of an improvement that again uses wild birds as predators, green and brown pastry ‘baits’ as prey, and trays containing coloured stones as the backgrounds. We used backgrounds of three different colours: green (on which brown baits were conspicuous), brown (on which browns were inconspicuous), and white (on which both prey types were conspicuous). The important difference from the previous design was that these backgrounds were presented simultaneously on the same bird table. We placed equal numbers of green and brown prey on each background and recorded the numbers eaten by wild birds. We did six experiments to test the design. Two different methods of measuring selection showed that the results were generally in the direction predicted from the hypothesis that conspicuous prey are more likely to be eaten by predators.  相似文献

Three distinct N-malonyltransferases were purified from peanut seedlings, accepting either anthranilic acid, D-tryptophan, or 3,4-dichloroaniline, respectively, as a substrate. Partially purified malonyl-CoA:D-tryptophan malonyltransferase also catalyzed the formation of the corresponding malonic acid conjugate when 1-aminocyclopropane-1-carboxylic acid was employed as a substrate. These N-malonyltransferases were clearly distinguished from several O-malonyltransferase activities also present in the same seedlings. N-Malonic acid conjugates have been previously isolated from peanut either as a natural constituent or after feeding with xenobiotics. By analogy to the results reported with cultured parsley cells, multiple malonyltransferases in peanut may have a role in vacuolar transport. Crude extracts of young peanut seedlings were incapable of hydrolyzing the respective N-malonic acid conjugates. However, dialyzed extracts of older plants released malonic acid from malonyl-1-aminocyclopropane-1-carboxylic acid but not from malonyl-3,4-dichloroaniline, suggesting that some N-malonic acid conjugates may be metabolized in plants which are approaching senescence.  相似文献

Peanut allergy is one of the most severe food allergies. One effort to alleviate this problem is to identify peanut germplasm with lower levels of allergens which could be used in conventional breeding to produce a less allergenic peanut cultivar. In this study, we identified one peanut line, GT-C9, lacking several seed proteins, which were identified as Ara h 3 isoforms by peptide sequencing and named iso-Ara h 3. Total seed proteins were analyzed by one-dimensional (SDS-PAGE) and two-dimensional gel electrophoreses (2-D PAGE). The total protein extracts were also tested for levels of protein-bound end products or adducts such as advanced glycation end products (AGE) and N-(carboxymethyl) lysine (CML), and IgE binding. Peanut genotypes of GT-C9 and GT-C20 exhibited significantly lower levels of AGE adducts and of IgE binding. This potential peanut allergen iso-Ara h 3 was confirmed by peptide sequences and Western blot analysis using specific anti-Ara h 1, Ara h 2, and Ara h 3 antibodies. A full-length sequence of iso-ara h 3 (GenBank number DQ855115) was obtained. The deduced amino acid sequence iso-Ara h 3 (ABI17154) has the first three of four IgE-binding epitopes of Ara h 3. Anti-Ara h 3 antibodies reacted with two groups of protein peptides, one with strong reactions and another with weak reactions. These peptide spots with weak reaction on 2-D PAGE to anti-Ara h 3 antibodies are subunits or isoallergens of this potential peanut allergen iso-Ara h 3. A recent study suggested that Ara h 3 basic subunits may be more significant allergenicity than the acidic subunits.  相似文献

The dependence upon solution conditions of the quaternary structure and gross conformation of peanut agglutinin was examined by sedimentation equilibrium, sedimentation velocity, gel chromatography, and circular dichroism. At pH 8, the protein exists as a compactly folded tetramer of molecular weight 98,000. Between pH 4.75 and pH 3.0, the molecular reversibly dissociates to a (still globular) dimer. In the presence of denaturants such as SDS or guanidinium chloride, the protein dissociates to its four equal-sized constituents polypeptide chains. The circular dichroic spectrum of peanut lectin exhibits changes in the near ultraviolet upon binding of lactose, whereas the far ultraviolet spectrum remains unchanged. Dissociation to the dimeric state produces subtle changes in both the near and far ultraviolet circular dichroic spectrum.  相似文献

The protocol for obtaining a high frequency of plant development via somatic embryogenesis from mature zygotic embryo-derived leaflets of peanut (Arachis hypogaea L.) involves multiple stages; these include the induction of embryogenic masses, development of embryos, radicle emergence/conversion of embryos and the development of plants from rooted abnormal embryos. Sixteen genotypes were subjected to this protocol by exposing mature zygotic embryo-derived leaflets to the common media sequence and comparing responses. Although the protocol was effective for all the genotypes, variation in frequency of response at each stage of development indicated that, with the exception of root meristem differentiation and subsequent radicle emergence, the whole process of somatic embryogenesis depended on the genotypic constitution of the original plant. The failure of somatic embryos to undergo conversion to plantlets could be a genotype-dependent characteristic.
