为了表达马尾松毛虫质型多角体病毒(DpCPV)多角体蛋白基因(S10片段)并探讨多角体蛋白在真核细胞中的定位,从DpCPV中分离出S10,与pET-28a载体连接成重组表达质粒pET28-S10;将S10克隆到杆状病毒转座载体pFASTBACHTb中,依次筛选出重组转座质粒pFASTBACS10,重组穿梭质粒BacmidS10,重组杆状病毒AcS10。多角体蛋白基因表达后,用SDS-PAGE、Western-blot和免疫荧光技术对表达产物进行了检测。结果表明:S10原核表达质粒、重组杆状病毒成功获得;在昆虫细胞中表达的质型多角体蛋白主要定位于细胞质,同时有少量产物定位于细胞核。  相似文献

通过对马尾松毛虫质型多角体病毒的增殖、纯化,获得一株单一类型的质型多角体病毒。提纯的病毒粒子经SDS-酚抽提,琼脂糖凝胶电泳分离基因组dsRNDA,回收纯化第十片段S10。S10经DMSO变性,逆转录合成cDNA第一链,PCR扩增后,克隆在pGEM-T载体上,对重组子进行限制性内切酶分析及序列测定。结果表明,克隆片段全长763bp,起始密码AUG位于3-5残基,终止密码UGA位于747-749残基。推测DpCPV多角体蛋白基因编码248个氨基酸的多肽,分子量28kD。和家蚕质型多角体病毒(BmCPV)多角体蛋白基因相比较,核苷酸和编码氨基酸序列同源性分别为89.3%和97.6%。  相似文献

现行的杆状病毒表达外源基因的方法是将外源基因取代病毒中的多角体基因,因而得到的重组杆状病毒感染活体时不能经口感染,只能进行针刺注射,效率低且易引起活体感染其他疾病。将家蚕核型多角体病毒(Bombyx mor inucleopolyhedrovirus,BmNPV)中的多角体基因(polyhedrin,poly)及其启动子片段克隆到转座子载体pigA3GFP中,将其与辅助质粒pHA3PIG利用脂质体介导法导入家蚕细胞中,经过多次筛选获得稳定的转基因家蚕细胞。之后先将BmPAK6(含LacZ)及BmGFP(含GFP)重组病毒分别感染转基因细胞,再将得到的重组病毒经口感染5龄家蚕幼虫。结果显示,重组杆状病毒可以经口感染家蚕幼虫。这些研究表明来自于转基因家蚕细胞的poly基因表达产物可以提高重组杆状病毒经口感染家蚕率,为解决杆状病毒表达系统中重组病毒不能经口感染家蚕幼虫的问题提供新思路。  相似文献

为探寻马尾松毛虫质型多角体病毒(DpCPV 1)p44蛋白的功能,构建了DpCPV 1基因组S8片段的原核表达体系,表达纯化蛋白后免疫家兔制备了多克隆抗体。利用Bac-to-Bac杆状病毒表达系统,构建了3种重组的杆状病毒质粒(Bacmid-p44、Bacmid-p44-e GFP和Bacmid-e GFP)。转染昆虫细胞Sf9进行表达,通过Western blot检测和蛋白的亚细胞定位观察。Western blot检测结果显示,Bacmid-S8在昆虫细胞Sf9中表达实际蛋白的大小为35 kD,比在原核系统中表达的蛋白(44 k D)略小;利用激光共聚焦显微镜观察p44-e GFP的融合蛋白的亚细胞定位发现,融合p44的绿色荧光蛋白(eGFP)主要聚集在细胞质中,而未融合的eGFP则分布于整个细胞,说明DpCPV 1的p44蛋白定位于细胞质中。  相似文献

异源多角体蛋白对家蚕核型多角体病毒粒子的包装   总被引:1,自引:0,他引:1  
利用PCR方法从AcMNPV基因组DNA中分离出多角体蛋白基因 ,将该扩增片段克隆到转移载体pBacPAK8中 ,得到重组转移载体pOAc。将该质粒DNA与线性化的Bm BacPAK6病毒基因组DNA共传染BmN细胞 ,得到了能形成多角体且不产生蓝色空斑的重组病毒hp BmNPV。纯化该重组病毒的多角体颗粒 ,并对多角体蛋白、病毒核酸及多角体病毒颗粒进行分析 ,发现AcMNPV的多角体蛋白能在家蚕细胞中大量表达且能在细胞内识别家蚕核型多角体病毒并组装成多角体颗粒 ;病毒基因组DNA因部分交换 ,其酶切行为发生了相应的变化 ;电镜观察发现经AcMNPV多角体蛋白包装的家蚕核型多角体病毒的多角体颗粒大小为1 2 μm~ 2 9μm ,明显小于野生型家蚕核型多角体病毒的多角体颗粒  相似文献

肌动蛋白抑制了杆状病毒多角体蛋白基因的转录与表达   总被引:2,自引:0,他引:2  
为进一步研究肌动蛋白在杆状病毒感染晚期的作用,利用Bac-to-Bac系统构建了表达多角体基因、肌动蛋白与绿色荧光蛋白融合基因的重组病毒vAc-ph/70GA,同时构建对照病毒vAc-ph.实验发现,重组病毒vAc-ph/70GA感染Sf9细胞后能持续表达肌动蛋白,但不能形成多角体,vAc-ph则能形成多角体.sDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)显示,vAc-ph/70GA感染晚期,细胞内没有多角体蛋白的表达;RT-PCR的结果进一步表明多角体基因的转录被抑制.肌动蛋白的表达并没有影响vAc-ph/70GA对细胞的感染力.结果表明,晚期表达的外源肌动蛋白抑制了多角体基因的转录和表达,从而导致多角体不能正常产生.  相似文献

大尺蠖核型多角体病毒(BsNPV)的多角体蛋白基因(ocu)定位在2.3kb的BamHl—H片段上,用xho1、sma1、HindIII和PstI构建了H片段的物理图谱,并测定了两端636bp序列。在5’端发现了杆状病毒ocu基因共有的典型特征,即:ATG起始区;启动子区(14bp保守序列);TATA box和CATA box区等。单一xhoI位点在ATG上游-15bp处,适于作为构建ocu基因转移载体的插入位点。在这一基因5’端序列中发现了五个反转重复单元CGAGC GCTCG,讨论了这一单元在杆状病毒ocu基因高效表达中的调控功能。  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.
