参照GenBank中Purdue株序列对TGEVTS株聚合酶基因(ORF1)设计特异性引物,经RT-PCR扩增获得了20054bp的片段,与预期大小相符。TS株与Pur46-MAD株的ORF1核苷酸和氨基酸序列同源性分别为98.8%和99.0%,与FIPV、PEDV、HCV299E及SARS氨基酸同源性分别为88%、57%、57%和45%。不同冠状病毒比较结果显示RdRp有很高的保守性而且有重要的作用,序列分析标明TS株在ORF1a和ORF1b重叠区有核糖体剪切位点UUUAAAC,且相应区域RNA二级结构形成3个茎环结构。  相似文献

猪Ⅱ型圆环病毒豫A株的全基因组克隆与序列分析   总被引:13,自引:0,他引:13  
参照国外发表的猪Ⅱ型圆环病毒(porcine circovirus type 2,PCV-2)全基因组序列,设计一对PCV-2特异性引物,用该室分离的PCV-2豫A株感染PK-15细胞,从中提取PCV-2复制型基因组DNA,并以之为模板进行PCR扩增.回收PCR产物,构建重组测序质粒T-PCV-2.测序结果表明,猪Ⅱ型圆环病毒豫A株的全基因组为1767bp,与GenBank收录的PCV-2国外分离株核苷酸的同源性可高达97%.序列分析表明,复制型豫A株的基因组包含10个读码框架,其中ORF1、ORF2是其两个最主要的读码框架,分别编码314、234个氨基酸.豫A株和PCV-1间的ORF1、ORF2的氨基酸序列同源性分别为85%、66%,与其它PCV-2毒株间的ORF1氨基酸同源性均在98%以上,而ORF2的氨基酸同源性为92%~97%.  相似文献

应用RT-PCR方法扩增了冠状病毒鸡传染性支气管炎中国分离毒株LX4的mRNA1 ORF1,并将其进行了克隆、序列测定和分析.表明LX4mRNA1基因包含2个ORF,其中ORF1a由11 916个核苷酸组成,编码一条3 971个氨基酸残基组成的多肽.ORF1b由7 950个核苷酸组成,编码2 649个氨基酸残基组成的多肽.与美国Beaudette株对应的ORF1a核苷酸及推导的氨基酸序列同源性均为85%,与对应的ORF1b的同源性分别为89%和95%.二者ORF1a和ORF1b重叠区推测的"滑脱序列"(slippery sequence)核苷酸序列一致,"假结"(pseudoknot)基本结构相似.不同之处在于"假结"第2环中LX4的第7位为A,而Beaudette株在该位置为G.与Beaudette株比较,LX4 ORF1a核苷酸变异最频繁的区域集中在第2 656~3 098位碱基处,且在该区域有60个碱基的插入,导致推导的氨基酸序列插入了20个氨基酸残基.LX4 ORF1b序列缺失9个核苷酸,主要集中在第5 168~5 181位之间,导致对应的氨基酸缺失3个.但这些差异是否与病毒的生物学特性有关不明.  相似文献

设计8对引物分片段扩增桃拉综合征病毒中国分离株ZHZC3全基因组,病毒两末端序列采用末端快速扩增方法(RACE)获取.扩增产物克隆到pMD18-T载体并测序,用DNAstar软件拼接全序列及同源性比较.结果显示ZHZC3全序列除去3' poly (A)尾,由10202个碱基组成,有两个开放阅读框,分别编码2107和1011个氨基酸的聚蛋白.与美国参考株HI94相比,在编码区没有核苷酸的缺失和插入,但在5' UTR缺失3个A,两者整体核酸同源性达97.9%.ORF1 中ZHZC3与HI94及巴西株(BLZ01)的核酸同源性分别为97.6%、97.7%,在ORF2中ZHZC3与HI94、BLZ01的核酸同源性则分别为98.3、97%.与国外株ORF2的部分序列比较发现ZHZC3和中国台湾株均与美国株HI94同源性最高.克隆分析6株TSV中国大陆株主要结构蛋白CP2基因,发现其编码的氨基酸存在三个高变区,中国大陆株更有其独特的氨基酸变异模式,312 (S), 449 (A), 451 (Q) 和468 (H).表明该病毒的整体变异性不高,但中国的流行株已形成其自己的遗传演变特征.在此基础上,利用生物学软件对CP2蛋白功能域和三维结构进行了预测,为进一步分析CP2蛋白结构与功能关系奠定了基础.ZHZC3株是第一个测定全序列的TSV中国株.  相似文献

三株散发性戊型肝炎病毒变异株的部分序列比较   总被引:1,自引:0,他引:1  
对三株散发性戊型肝炎病毒Ch-T11、Ch-T21、Ch-T40的部分基因组做克隆测序,经比较发现与其它国内外HEV株ORF2部分相应核苷酸序列同源性在78.3-81.3%,Ch-T21与Ch-T40的核苷酸同源性为98.8%,而Ch-T11与前两者的同源性则分别为89.8%和90.2%.Ch-T11、Ch-T21、Ch-T40与其它HEV株相应氨基酸序列同源性在95.8-97.9%,它们之间的氨基酸同源性则为100%.系统进化树分析显示,这三株HEV可能代表着一新型HEV.  相似文献

PMWS病猪猪圆环病毒2型全基因组序列分析   总被引:7,自引:0,他引:7  
根据GenBank中发表的猪圆环病毒2型(PCV2)全基因序列,设计一对特异性引物,用PCR方法直接从5省病料中分别扩增出5个PCV2毒株的全基因组,分别命名为FujianPCV、GuangxiPCV、HainanPCV、HunanPCV和ShandongPCV.将扩增片段克隆于pMD 18-T载体,进行序列测定,结果表明,除FujianPCV株核苷酸长度为1768bp,其余四株均为1767 bp.应用DNAstar序列分析软件分析表明,本实验的5个PCV2分离株与世界上其它地区的PCV2分离株密切相关,核苷酸序列同源性达93%~98.8%,与PCV1毒株的序列同源性只有68.4%~70%.其中ORF1和ORF2所编码的氨基酸序列与国内外PCV2毒株比较,同源性也很高,分别为98.1%~99.4%和88%~99.1%.  相似文献

对猪伪狂犬病毒鲁A株(PRV LA株)TK基因进行了克隆和序列测定,并分析比较了该序列与PRV NIA-3株、Ea株、SH以及HSV-1和VZV的同源性,结果表明:在全长1048bp的DNA序列中,包括着一个963bp的开放阅读框(ORF),可编码320个氯基酸组成的多肽;在整个TK基因的ORF内,PRV LA株与PRV NIA-3株、PRV Ea株、PRV SH株、HSV-1、VZV的TK基因比较,核苷酸的同源性分别为98.9%、99.5%、99.3%、36.4%、39.1%,氨基酸的同源性分别为98.4%、99.7%、98.7%、36.6%、37.2%.PRV LA株TK具有疱疹病毒胸苷激酶催化结构域的保守氨基酸共有序列和亚结构域特征序列.将PRV-LA TK、人类和小鼠的胸苷酸激酶、人类脱氧胞苷激酶、人类腺苷酸激酶的对应于这两个亚结构域的氨基酸用DNA Star分析的进化树表明,疱疹病毒的TK与人类和小鼠的胸苷酸激酶的亲缘关系比与人类脱氧胞嘧啶激酶的亲缘关系更近,因此疱疹病毒的TK基因在进化上可能起源于宿主细胞的胸苷酸激酶基因.  相似文献

为进行猪戊型肝炎病毒(HEV)ORF2基因特征研究,参照GenBank中已发表的戊型肝炎病毒(HEV)核酸序列,设计了一对扩增HEV ORF2基因的引物,利用RT-PCR等方法克隆出了一株猪戊型肝炎病毒甘肃分离株GS189的ORF2基因cDNA片段.序列测定结果表明,swCH-GS189株的ORF2基因长2 025 bp,编码674个氨基酸,与GenBank中公布的其它毒株间的核苷酸序列同源性为79.1%~91.8%,推导的氨基酸序列同源性为89.5%~98.8%.系统发育进化树结果表明,该分离株为基因IV型.  相似文献

为阐明分离自云南省的蚊媒阿卡斑病毒(Akabane virus,AKV)DHL10M110株(简称云南株)的分子生物学特征,采用RT-PCR和核苷酸序列测定方法,对云南株进行全基因组序列测定和分析。结果获得了云南株全基因组核苷酸序列,该病毒株的L、M和S基因核苷酸序列全长分别为6 869bp、4 309bp和856bp,其中开放读码框(Open reading frame,ORF)核苷酸序列依次为6 756bp、4 206bp和702bp,并分别编码2252、1402和234个氨基酸。这三个基因片段ORF的系统进化分析表明,云南株L基因与AKV标准株OBE-1(日本)和AKVS-7/SKR/2010株(韩国)处于同一进化分支;在M和S基因进化树中,云南株与日本、韩国和台湾地区的AKV流行株亲缘关系较近,但云南株单独处于一个小的进化分支。同源性分析表明,云南株L、M和S片段ORF与OBE-1株的核苷酸(氨基酸)同源性分别为92.6%(98%)、88.5%(94%)和96.4%(99.1%)。云南株与OBE-1株的L、M和S片段分别有45、84和2个氨基酸位点的差异。此外,无论L、M和S基因核苷酸序列的同源性或系统进化分析均表明,云南株与肯尼亚和澳大利亚AKV分离株进化关系较远,同源性较低。本研究进一步通过病毒全基因组序列分析证实DHL10M110病毒株为AKV,属亚洲进化群,但与日本和韩国株相比仍有一定差异,云南株地域特征明显。这是首次在中国大陆地区测定到该病毒的全基因组序列。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

对我国花型Hemibarbuspattern鱼类作了骨骼系统比较,结果表明,此类型鱼类脑颅较长,副蝶骨平直或稍弯曲,眶蝶骨腹纵嵴发达(铜鱼Coreius septentrionalis例外),下颞窝和咽突中等大,基枕骨后突发达;脑颅中的上筛骨的后突、侧筛骨的外筛突,蝶耳骨的外突、上耳骨的后突、围眶骨和后颞窝等均有明显的差异;咽颅中的舌颌骨、尾舌骨、鳃盖骨和下咽齿的列数等又有显著的区别;附肢骨骼中的腰带骨、脊椎骨中的复合神经骨和第4椎骨腹侧的悬器等也有不同之处。据此,这些差异和区别可作为属间或种间的分类依据。  相似文献
