微悬臂列阵传感器在生物检测方面具有快速、痕量和非标记的特性. 我们以镀金并在其上固定了 DNA 探针的微悬臂为正极,在靶杂交液槽内引入另一电极作为负极,构成电场驱动微悬臂 DNA 生物传感器. 对该传感器系统施加静电场,驱动 DNA 分子朝正极迁移,使溶液中的 DNA 分子富集在微悬臂上,促进 DNA 分子的杂交. 结果表明: a. DNA 在微悬臂上的杂交时间仅需 3 min,加快了微悬臂生物传感器对 DNA 分子的检测速度; b. 提高了微悬臂生物传感器的灵敏度,可以检测到皮克级的 DNA 分子.  相似文献

为了基于羧基化石墨烯/半胱胺修饰金电极建立更为先进的多巴胺生物传感器,以定量检测儿茶酚胺类神经递质多巴胺,利用自组装技术将半胱胺修饰于金电极上,再利用1-乙基-\[3-二甲基氨基丙基\]碳酰二亚胺盐酸化物/N-羟基琥珀酰亚胺(EDC/NHS)交联剂将羧基化石墨烯固定在修饰后的金电极上制成多巴胺电化学传感器。先对修饰电极进行表征以检验其灵敏度,再利用循环伏安法研究该电极在多巴胺溶液中的电化学行为,包括检测条件的优化和传感器性能的测定。修饰电极表征结果表明,羧基化石墨烯/半胱胺修饰金电极提高了电极传递电子的能力,具有较高的灵敏度。经单因素实验得出,最佳检测条件为利用pH 6.00的0.30 mol·L-1磷酸盐缓冲溶液(PBS)配制多巴胺溶液,扫描速率设定为200 mV·s-1。在最佳检测条件下,制备的多巴胺电化学传感器电流的大小随着多巴胺浓度的增大而增大,在1.0×10-3~3.5×10-3 mol·L-1范围内呈现良好的线性关系,线性回归方程为I=8.120 6C+7.017,相关系数R2为0.999 5。且该传感器精密度好,稳定性强,具有一定的抗干扰能力。研究结果为药物盐酸多巴胺注射液中多巴胺含量测定提供了支撑。  相似文献

目的:了解毛细管内核酸偶联和游离辣根过氧化物酶(HRP)催化化学发光差异。方法:不同浓度HRP和经核酸杂交固定于毛细管内壁HRP催化化学发光反应。结果:①游离HRP催化化学发光线性检测范围窄(2.7×10-5-1.3×10-6mg/ml,R2〉0.96),下限为1.0×10-7mg/ml;②2.0×1014-2.0×106copies/ml的5μl单链DNA杂交后,1.0-10min时DNA浓度M对数(lgM)与化学发光I值线性相关(R2〉0.99),且大于阴性I值平均数+3倍标准偏差(s.d);③5.0×1011-5.0×106copies/ml的5μl PCR产物杂交后,10min内PCR产物对数lgM与I值线性相关(R2〉0.97),且大于阴性I值平均数+3倍标准偏差(s.d)。4.0-7.0min内lgM与I值的R2〉0.99,3次平行检测标准偏差〈5.0%。结论:毛细管内核酸杂交的HRP催化化学发光检测线性范围宽、灵敏度高、底物用量少,有望用于临床核酸分子杂交检测。  相似文献

目的:为研制检测H5亚型禽流感的压电免疫传感器。方法:用巯基丙酸在镀银电极石英晶体自组装巯基丙酸单分子膜再通过N-乙基-N’-(3-二甲氨基)丙基碳化二亚胺盐酸(EDC)和N-羟基琥珀酰亚胺(NHS)偶联抗H5亚型禽流感病毒的特异性单抗构建传感器芯片,建立了可以检测H5亚型禽流感病毒的免疫传感器。结果:结果表明,该法具有较好的特异性,不与H9亚型流感病毒和NDV反应;检测灵敏度达到10—50个EID50。结论:本文结果为检测禽流感病毒免疫传感器的进一步深入研究奠定了基础,这为其它相关病毒的监测提供了一种新途径。  相似文献

基于生物催化和免疫原理的生物传感器在环境领域中获得了广泛应用.近年来,随着分子生物学和生物技术的发展,人们开发了以核酸探针为识别元件,基于核酸相互作用原理的DNA生物传感器.该传感器可用于受感染微生物的核酸序列分析、优先控制污染物的检测以及污染物与DNA之间相互作用的研究,在环境污染监测中具有潜在的巨大应用前景.简要介绍了核酸杂交生物传感器的基本原理及其在环境微生物和优先控制污染物(priority pollutant)检测中的应用研究进展.  相似文献

目的:建立化学发光Southern blot检测细胞内HBV DNA的方法,同时检测3种不同靶点抗乙肝药物的体外作用。方法:用地高辛标记HBV探针,优化杂交条件,检测来自HepG2及HepG2.2.15 HBV DNA复制中间体;利用建立的化学发光Southern blot检测HBV DNA的方法检测经拉米夫定、Bay41-4109、α-Galcer以不同药物浓度处理的HepG2.2.15HBV DNA复制中间体的水平。结果:(1)标记的HBV探针的检测灵敏度为0.1pg,杂交系统的检测灵敏度为1pg,可检测到HepG2.2.15细胞内的HBV DNA特异性信号;(2)以该法检测胞内HBV DNA可见3种药物都有明显的抑制作用,其半数有效量(IC50)分别为1.53μmol/L、0.41μmol/L、0.01μmol/L。结论:胞质HBV DNA的水平能准确地反映不同靶点抗HBV药物的抗病毒效果,建议在观察药物特别是中药抗病毒研究中采用。  相似文献

建立了显微分离黄鳝单条染色体及检测其特异性的方法;在Olympus倒置显微镜下用毛细玻璃微针手工分离黄鳝减数分裂Ⅰ终变期3号染色体,将期DNA作模板进行DOP-PCR扩增后,分别以α^-32P-dCTP和Biotin-11-dUTP标记的单条染色体DNAPCR扩增产物及Biotin-11-dUTP标记的大豆18SrRNA基因作探针进行Southern杂交,FISH和Dot杂交来检测其特异性,结果表明:(1)减数分裂Ⅰ终变期染色体标本是进行染色体显微操作的理想材料;(2)DOP-PCR扩增产物片段在200-1000bp之间,平均600bp左右;(3)杂交结果显示,本研究所获得的单条染色体是黄鳝3号染色体;(4)与显微操作仪和微激光分离相比较。该方法不需要昂贵仪器,在常规实验室即可操作,具有广泛的普及应用意义。  相似文献

依据晶体谐振频率是其表面沉积物的函数和模板-引物杂交后引物链延伸的酶促反应原理,构建了引物链延伸反应性基因传感器技术.该技术的主要特点是以快速、敏感的频变信息作为基因杂交与引物链延伸的显示系统.研究结果提示,本项技术可用于核酸同源性分析、微量DNA检测、DNA整合性的确定,也可用于目的基因的分离、特定条件下的基因突变分析及推断某基因在其基因组中的位置.  相似文献

根据荷斯坦牛SRY基因设计一对引物,采用聚合酶链式反应(PCR)技术,以中国沼泽型水牛(Swamp Buffalo)基因组DNA为模板,扩增得到SRY(Sex Deterimation region of Y chromosome)全序列约2005bp,其中1-504bp为5’启动子区,1196-2005bp为3’侧翼序列,在505-1195bp为SRY的外显子,编码229个氨基酸。在SRY HMG box区域设计探针,用地高辛标记后分别与雄性、雌性水牛基因组DNA进行Southern 杂交,结果显示该段序列只在雄性DNA样本中有杂交信号,证明SRY基因为雄性特异。BLAST比对结果显示与牛属动物SRY基因的同源性为96%,其中SRY基因HMG box区域同源性高达99%,说明SRY基因具有高度的进化保守性  相似文献

目的:建立化学发光法检测体外HBV复制水平的方法,研究其灵敏度和稳定性.方法:用HBV DNA重组质粒pCH9转染到人肝癌细胞株HepG2和Huh7中,5d后收集细胞并抽提其HBV复制中问体DNA,转印后以地高辛标记HBV DNA为探针进行杂交,用化学发光法检测杂交结果,同时进行探针灵敏度的检测.结果:转染后HepG2和Huh7细胞提取HBV DNA中检测出较强的复制中间体的信号,分别为松散环状DNA(rcDNA),双链线性DNA(dslDNA),单链DNA(ssDNA),探针检测的灵敏度可达到lpg,接近同位素法检测的灵敏度.整个实验重复3次获得同样结果.结论:成功建立了稳定的化学发光法检测体外HBV复制水平的方法.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

The fossil record of bryophytes is generally poor and infertile plants attributable to liverworts and mosses could also be thalloid vascular plant gametophytes or herbaceous lycophytes respectively. The paucity of the bryophytic record could be the result of relatively rapid degradation of bryophytic material in comparison to that of vascular plants, the absence of lignified cells in bryophytes sustaining this belief. However, certain organs of bryophytes are as robust as those of vascular plants (e.g. spores) and the hydroxybenzofuran polymers produced by some mosses could be of similar preservational resistance to lignins. A simplistic experiment to test the relative resistance to decay of bryophytic material has been undertaken. Its findings suggest that bryophytic plant material has a similar preservational potential to that of a selected vascular plant, particularly in organic rich sediments. This result may be construed to imply that bryophytes were indeed less abundant in the preservational environments of the Coal Measures than might be expected on the basis of current palaeoenvironmental reconstructions.  相似文献

In December 2019, SARS-CoV-2 was first detected in the samples obtained from three adult patients who suffered from an unknown viral pneumonia in Wuhan (Li et al. 2020). This unknown viral pneumonia is further named as coronavirus disease 2019 (COVID-19) by the World Health Organization. To date, the number of new COVID-19 cases has continued to skyrocket and the impact of SARS-CoV-2 on humans is far greater than any pathogen of this century in both breadth and depth. Previous studies have shown that adults with COVID-19 have symptoms of fever, dry cough, dyspnea, fatigue and lymphocytopenia. Moreover, COVID-19 is more likely to cause death in the elderly, especially those with chronic comorbidities (Huang et al. 2020). In Wuhan, more than 50, 000 COVID-19 cases have been confirmed, including over 780 pediatric patients, and only one child death case (Lu et al. 2020). Although the number of children cases was far fewer than that of adults, COVID-19 might endanger children's health and the information on children remains limited, especially in serological study. In the retrospective study, the investigators analyzed the epidemiological, clinical and serological characteristics of children with COVID-19 in Wuhan in the early stages of the outbreak, which might provide theoretical and practical help in controlling COVID-19 and similar emerging infectious diseases in the future.  相似文献
