目的:探讨白细胞介素-17A(IL-17A)对慢性阻塞性肺疾病(COPD)的干预作用及其机制。方法:C57BL/6小鼠随机分为野生型空白对照组、野生型COPD组和IL-7A敲除COPD组,每组20只。野生型空白对照组小鼠不做任何处理,其余两组小鼠暴露于香烟烟雾(1支/次,4次/日,每次45 min,每次间隔时间为1 h,总干预时间为90 d)制作COPD模型。干预结束24 h后,利用动物肺功能检测系统测定小鼠肺功能。收集小鼠支气管肺泡灌洗液(BALF),测定BALF细胞计数和分类。收集小鼠肺组织,采用流式细胞法测定气道上皮IL-17A表达水平,采用酶联免疫吸附法测定肺组织炎症因子水平。采用蛋白免疫印迹法测定小鼠肺组织JNK/AP1信号通路蛋白表达水平。结果:与野生型空白对照组小鼠比较,野生型COPD组小鼠气道上皮IL-17A表达水平明显升高,吸气峰流速(PIF)和呼气峰流速(PEF)明显降低,BALF中性粒细胞、嗜酸性粒细胞、淋巴细胞和巨噬细胞数明显升高,肺组织CXC类趋化因子1(CXCL1)、CXC类趋化因子2(CXCL2)、白细胞介素-1β(IL-1β)和白细胞介素-6(IL-6)表达水平明显升高,JNK、cJun和cFos磷酸化水平及AP1表达水平明显升高(P<0.05);与野生型COPD组小鼠比较,IL-7A敲除COPD组小鼠气道上皮IL-17A表达水平明显降低,PIF和PEF明显升高,BALF中性粒细胞、嗜酸性粒细胞、淋巴细胞和巨噬细胞数明显降低,肺组织CXCL1、CXCL2、IL-1β和IL-6表达水平明显降低,JNK、cJun和cFos磷酸化水平及AP1表达水平明显降低(P<0.05)。结论:香烟烟雾可诱导小鼠气道上皮产生IL-17A,降低(或抑制)IL-17A的产生(或表达或分泌),通过抑制JNK/AP1信号通路,减轻COPD气道炎症反应,改善COPD小鼠肺功能。  相似文献

目的: 研究姜黄素调控Toll-样受体4(TLR4)- p38丝裂原活化蛋白激酶(p38 MAPK)/核因子κB(NF-κB)信号通路缓解过度训练大鼠脾脏炎症反应的作用及其机制。方法: 7周龄SPF级雄性Wistar大鼠分为安静对照组(C组,n=12)、过度训练模型组(OM组,n=11)、姜黄素+模型组(COM组,n=14)。C组不进行任何运动干预,OM和COM组进行8周递增负荷游泳训练。训练期间,COM组以200 mg/(kg·d)、5 ml/kg姜黄素进行灌胃,其他组灌胃等体积0.5 %羧甲基纤维素纳助溶剂。末次训练后24 h,称重计算脾脏指数,光镜观察脾脏组织病理学改变,取血液、脾脏组织检测相关生化指标。结果: 8周递增负荷游泳训练后,光镜下C组大鼠脾脏组织结构正常;OM组较C组脾脏指数极显著降低(P<0.01),并出现典型炎症病理变化;COM组较OM组脾脏指数显著升高(P<0.05),且炎症病理变化有所改善。与C组比较,OM组血清皮质酮(Cor)、NF-κB、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)水平和脾脏单核细胞表面TLR4表达率、TNF-α、IL-6水平均升高(P<0.05或P<0.01),脾脏p38 MAPK、磷酸化p38 MAPK(p-p38 MAPK)和NF-κB蛋白质表达水平均增强(P<0.05或P<0.01);血清睾酮(T)、血清和脾脏白细胞介素-10(IL-10)水平降低(P<0.01)。与OM组比较,COM组血清Cor、NF-κB、TNF-α、IL-6水平和脾脏单核细胞表面TLR4表达率、TNF-α、IL-6水平均降低(P<0.05或P<0.01),脾脏p38 MAPK、p-p38 MAPK和NF-κB蛋白质表达水平均降低(P<0.05);血清T、血清和脾脏IL-10水平升高(P<0.05或P<0.01)。组间T/Cor比值变化趋势与T变化相一致。结论: 8周递增负荷游泳训练引发大鼠过度训练,脾脏组织炎症反应加剧,出现炎症病理变化。训练期间补充姜黄素可以通过下调TLR4-p38 MAPK/NF-κB信号通路相关蛋白质表达,维护促炎/抑炎因子间动态平衡,保护脾脏。  相似文献

目的:探讨6周跑台运动对D-半乳糖致衰老模型大鼠血清炎性因子水平的影响,以此阐明运动延缓衰老的部分机制。方法:将雄性SD大鼠随机分为正常对照组(NC组)、衰老对照组(AC组)、衰老训练组(AE组),每组8只。AC和AE组大鼠每日腹腔注射1次D-半乳糖125 mg/kg,NC组注射等量生理盐水,持续6周。同时,AE组进行6周中小强度跑台训练。6周后,各组大鼠腹主动脉取血,分离血清检测血清中白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)的含量。结果:与正常对照组比较,衰老模型组大鼠血清IL-6、TNF-α水平明显升高(P<0.01);与衰老模型组比较,衰老训练组大鼠血清IL-6、IL-10和TNF-α水平明显减少(P<0.01),表明6周中小强度跑台训练有助于降低衰老机体促炎症因子的分泌,减弱体内炎症反应。结论:6周中小强度跑台训练在一定程度上可以纠正衰老机体内的炎性因子网络紊乱状况,起到干预炎性衰老的目的。  相似文献

目的: 探讨NOD样受体蛋白3(NLRP3)信号通路对非酒精性脂肪肝病(NAFLD)大鼠胰岛素抵抗的影响及乳酸受体G蛋白偶联受体81(GPR81)激动剂的干预作用。方法: 选择清洁级SD雄性大鼠30只,随机分为3组,对照组、NAFLD组、GPR81激动剂组,每组10只。用高脂饮食建立大鼠非酒精性脂肪肝模型;GPR81激动剂组:在非酒精性脂肪肝模型基础上腹腔注射GPR81特异性乳酸激动剂(50 nmol/L),每周1次,其余两组注射等量的生理盐水,共12周。测定肝生化指标、空腹血糖及胰岛素和肝匀浆中炎症因子的含量,观察各组肝组织病理学形态;Western blot检测肝组织中NLRP3、含CARD结构域的凋亡相关斑点蛋白(ASC)、天冬氨酸特异性半胱氨酸蛋白酶1(caspase-1)、胰岛素受体底物-1(IRS-1)、胰岛素受体底物酪氨酸磷酸化(Tyr465-IRS-1)、胰岛素受体底物丝氨酸磷酸化(Ser636-IRS-1)、葡萄糖转运蛋白4(GLUT4)的蛋白表达;qRT-PCR法检测肝组织NLRP3、ASC、caspase-1、IRS-1、GLUT4 mRNA表达水平。结果: 与对照组相比,NAFLD组大鼠血清肝生化指标甘油三酯(TG)、丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)、空腹血糖(FPG)、空腹胰岛素(FINS)和胰岛素抵抗指数(HOMA-IR)值均显著升高(P<0.05);肝组织病理学形态结果表明,NAFLD组大鼠肝组织可见明显的肝脂肪变性,肝细胞有脂肪滴,存在明显的炎性细胞浸润,且NAFLD组肝组织NLRP3、ASC、caspase-1的mRNA和蛋白表达及Ser636-IRS-1的蛋白表达均显著升高,且肝组织及血清中白细胞介素-1β(IL-1β)和白细胞介素-18(IL-18)的含量升高;而IRS-1、GLUT4 的mRNA和蛋白表达Tyr465-IRS-1的蛋白表达显著降低(P<0.05);与NAFLD组相比,GPR81激动剂组上述指标均得到明显改善。结论: NLRP3信号通路活化介导炎症因子产生促进了NAFLD的发生发展,GPR81激动剂可能成为NAFLD潜在的治疗手段。  相似文献

目的: 探究颗粒蛋白前体(PGRN)在过敏性哮喘中的作用及机制。方法: 分别在野生鼠和IL-6 缺陷鼠中设置对照组和哮喘模型组,每组8只。模型组中,在第0日和第7日致敏小鼠(腹腔注射OVA 100 μg),从第14日起连续激发8 d(5%OVA雾化吸入,30 min/d,每日1次),末次激发24 h后取标本;对照组用PBS代替OVA做相同处理。采集支气管肺泡灌洗液(BALF)进行白细胞计数和分类计数;HE染色观察肺组织病理情况;Q-PCR及ELISA检测小鼠肺匀浆、血清和BALF中细胞因子水平。用IL-13刺激A549或BEAS-2B细胞建立体外哮喘炎症模型,每组3个复孔,共4组:PBS处理组、IL-13处理组、IL-13与重组人PGRN蛋白(rhPGRN)共同处理组及p38磷酸化抑制剂(SB203508)处理组。0 min~48 h后收集细胞及上清,用Q-PCR及ELISA检测PGRN和IL-6的表达;Western blot检测p38的磷酸化。结果: 与对照组相比,哮喘组小鼠肺匀浆和BALF中PGRN均显著降低(P< 0.01),血清PGRN有降低的趋势,然而哮喘小鼠BALF中IL-6显著升高(P<0.05)。与野生鼠哮喘组相比,IL-6缺陷鼠哮喘组BALF中白细胞总数降低(P<0.05),中性粒细胞数降低(P<0.05),PGRN显著升高(P<0.05),肺部病理损伤也减轻。体外实验中,IL-13处理组与PBS处理组相比,PGRN显著降低(P<0.05),IL-6显著增高(P< 0.05),p38的磷酸化增加;p38抑制剂处理组比未处理组中IL-6水平降低(P<0.05)。IL-13与rhPGRN共同处理组的IL-6显著低于IL-13处理组(P<0.05),p38的磷酸化降低(P<0.05)。结论: PGRN通过抑制p38磷酸化降低IL-6水平从而减轻哮喘小鼠气道炎症。  相似文献

目的:研究纳米卟啉金属有机骨架(NPMOF)对斑马鱼幼鱼神经系统发育的作用。方法:斑马鱼胚胎在发育6 h(hpf)后随机分为两组:对照组(n=500)和暴露组(n=500),对照组斑马鱼孵化于E3溶液中,暴露组于100 mg/L的NPMOF-E3溶液中,持续暴露至28、48、72、96或120 hpf。以20条斑马鱼为一批,在28、48、72、96和120 hpf分别提取3批实验组和对照组的总RNA用于RT-PCR;在120 hpf,实验组和对照组分别取20条PTU处理过的斑马鱼用于整体胚胎原位杂交,同样是在120 hpf,每组取150条用于免疫荧光染色,30条用于行为学测试。NPMOF的形状和尺寸用透射电镜测定,其光学特性由紫外分光光度计和荧光光谱仪测定;通过免疫荧光、整体胚胎原位杂交和RT-PCR方法检测各类神经细胞的发育;行为学测试用来监测斑马鱼运动状态的改变。结果:与正常组比较,NPMOF暴露组中神经发育相关基因的表达明显升高(P<0.05),müller细胞和星形胶质细胞的数量明显增加(P<0.05),神经元和少突胶质细胞的分布与形态未显示出差异;行为学测试中,斑马鱼的总运动距离、快速运动时间和运动速度的值显著增加(P<0.05),总静止时间明显降低(P<0.05)。结论:100 mg/L NPMOF溶液的持续暴露对斑马鱼神经系统发育,特别是视网膜中müller细胞和脑中星形胶质细胞的发育有促进作用。  相似文献

目的: 研究黑果枸杞汁对大鼠酒精性肝损伤的保护作用,探讨其中Toll样受体4(TLR4)/p38 丝裂原活化蛋白激酶(p38 MAPK)信号通路的调节机制。方法: 60只雄性SD大鼠随机分为空白对照组(C)、模型组(M)、低剂量黑果枸杞汁组(LLM)、中剂量黑果枸杞汁组(MLM)、高剂量黑果枸杞汁组(HLM),每组12只。M、LLM、MLM和HLM组每天以20 ml/kg(8 g/(kg·d))剂量分2次灌胃400 g/L酒精,C组于相同时间点灌胃等体积蒸馏水。每日首次酒精灌胃前4 h,黑果枸杞汁各剂量组分别以2.4、4.8、9.6 ml/(kg·d)进行灌胃,其他组于相同时间点灌胃等体积蒸馏水。实验周期4周,末次实验结束后24 h处死大鼠,取血液和肝脏,计算肝脏指数,HE染色观察肝脏组织形态,比色法检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,免疫组化法检测肝脏TLR4、p38 MAPK及磷酸化p38 MAPK(p-p38 MAPK)蛋白质表达,酶联免疫吸附试验检测肝脏肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)、白细胞介素-18(IL-18)水平。结果: 与C组比较,M组成功建立酒精性肝损伤模型。与M组比较,黑果枸杞汁各剂量组的相关指标出现改善,其中HLM组肝脏组织形态改善最为显著,其肝脏系数、血清ALT和AST、肝脏TLR4蛋白质表达、p-p38 MAPK/p38 MAPK比值、TNF-α、IL-1β和IL-18水平均显著降低(P<0.05或P<0.01),肝脏IL-10水平显著升高(P<0.01)。黑果枸杞汁各剂量组组间比较,肝脏系数、血清AST、肝脏TLR4蛋白质表达、p-p38 MAPK/p38 MAPK比值、TNF-α和IL-18水平,HLM组较LLM组均显著降低(P<0.05或P<0.01);肝脏IL-10水平,HLM组较LLM和MLM组均显著升高(P<0.05或P<0.01);其余主要指标组间比较均无统计学差异(P>0.05)。结论: 黑果枸杞汁可以通过调控TLR4/p38 MAPK信号通路,改善炎症应激,缓解大鼠酒精性肝损伤,高剂量组效果优于其他剂量组。  相似文献

目的:探讨莪术醇(CC)对非酒精性脂肪性肝(NAFLD)大鼠模型肝功能和肝纤维化的影响及机制。方法:采用高脂饮食构建非酒精脂肪肝炎(NASH)伴肝纤维化的大鼠模型,将60只SD大鼠随机分为:空白对照组、模型组(NASH)、NASH+复方鳖甲软肝片(CBT)组(阳性对照组)、NASH+CC组(25、50、100 mg/kg),每组10只。测量大鼠肝脏占体重的百分比,测量大鼠高密度脂蛋白(HDL)、甘油三酯(TG)、谷丙转氨酶(ALT)、谷草转氨酶(AST)水平,HE染色观察肝纤维化情况,免疫组化检测鼠肝组织α-平滑肌肌动蛋白(α-SMA)表达及肝组织核因子κB p65(NF-κB p65)的阳性染色情况,蛋白印迹(Western blot)检测α-SMA、基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶抑制剂-1(TIMP-1)蛋白表达及Toll样受体-4(TLR4)、转化生长因子激活激酶-1(TAK1)、NF-κB p65、血管细胞粘附分子-1(VCAM-1)蛋白的表达情况,酶联免疫吸附法(ELISA)检测肝组织中白介素(IL-6、IL-10、IL-1β)、肿瘤坏死因子-α(TNF-α)的表达。结果:与空白对照组相比,模型组大鼠HDL、 IL-10含量、MMP-1蛋白表达量显著降低(P<0.05),TG、ALT、AST、肝组织P65阳性率,α-SMA、TIMP-1、TLR4、TAK1、NF-κB p65、VCAM-1表达、IL-6、TNF-α及IL-1β含量显著升高(P<0.05)。与模型组相比,CBT和CC处理后大鼠HDL、 IL-10含量、MMP-1蛋白表达量显著升高(P<0.05),TG、ALT、AST、肝组织P65阳性率,α-SMA、TIMP-1、TLR4、TAK1、NF-κB p65、VCAM-1表达、IL-6、TNF-α及IL-1β含量显著降低(P<0.05),其中模型+CC组以高浓度组改善最显著(P<0.05),但各剂量改善幅度均低于模型+CBT组(P<0.05)。结论:莪术醇通过调节TLR4、TAK1、NF-κB p65信号通路,减轻炎症反应,改善肝功能,从而缓解非酒精性脂肪肝肝肝纤维化,且在一定范围内呈浓度依赖性。  相似文献

目的:探讨在低氧联合脂多糖(LPS)作用下,星形胶质细胞中B淋巴细胞瘤-2/腺病毒E1B 19-kD相互作用蛋白3(BNIP3)的表达和炎症反应变化。方法:将体外培养的原代星形胶质细胞和神经元进行下列分组:常氧组、LPS组、低氧组和LPS+低氧组(每组设置3个复孔)。LPS处理后,低氧组和LPS+低氧组放入低氧细胞孵箱,LPS组和常氧组放入正常的细胞孵箱。LPS浓度:100 ng/ml,氧气浓度为0.3%。处理时间为24 h。原代的星形胶质细胞进行上述的分组,时间点设为6 h、12 h和24 h。Western blot检测BNIP3的表达变化,RT-PCR和ELISA分别检测星形胶质细胞的肿瘤坏死因子-ɑ(TNF-ɑ)、白细胞介素-1β(IL-1β)和白细胞介素6(IL-6)mRNA水平变化和分泌情况。结果:与常氧组比较,低氧组炎症因子的表达没有变化,LPS组和LPS+低氧组的炎症因子TNF-ɑ、IL-1β和IL-6 mRNA水平升高(P<0.01);与LPS组比较,LPS+低氧组炎症因子IL-1β和IL-6 mRNA水平进一步升高(P<0.05,P<0.01)。与常氧组比较,低氧组炎症因子的分泌水平没有变化,LPS组和LPS+低氧组的炎症因子TNF-ɑ和IL-6 分泌水平升高(P<0.01),IL-1β的水平没有变化;与LPS组比较,LPS+低氧组炎症因子TNF-ɑ和IL-6分泌水平没有进一步升高。BNIP3在体外培养的神经元和星型胶质细胞中都有表达;在星形胶质细胞中,与常氧组比较,LPS组BNIP3的表达没有变化,低氧组和LPS+低氧组BNIP3的表达明显增加(P<0.01);在神经元中,与常氧组比较,LPS组BNIP3的表达没有变化,低氧组和LPS+低氧组BNIP3的表达增加(P<0.05,P<0.01);与神经元的低氧组比较,星形胶质细胞的低氧组BNIP3的表达增加更明显(P<0.01)。在星形胶质细胞中LPS联合低氧刺激6、12、24 h后BNIP3蛋白的表达,与常氧组相同时间点比较,LPS组BNIP3的表达没有变化,低氧组和LPS+低氧组BNIP3的表达增加(P<0.05,P<0.01);与低氧组相同时间点比较,6 h和12 h的LPS+低氧组BNIP3的表达增加的更高(P<0.01)。结论:低氧联合LPS刺激可以增强星形胶质细胞的炎症反应,LPS能增加低氧下星形胶质细胞中BNIP3的表达,提示BNIP3在星形胶质细胞的炎性反应中可能具有一定的调节作用。  相似文献

目的: 探讨miR-520a-3p调控宫颈癌细胞因子分泌的分子机制。方法: 通过TargetScanHuman分析miR-520a-3p与NF-κB复合体亚基RELA的匹配情况,然后通过荧光素酶报告系统检测miR-520a-3p是否靶向NF-κB复合体亚基RELA;使用LPS刺激宫颈癌HELA细胞后,将miR-520a-3p mimics与转染试剂混合后滴入HELA细胞中,此为过表达组;将miR-520a-3p inhibitor与转染试剂混合后滴入HELA细胞中,此为敲低组,通过酶联免疫吸附试验检测过表达组和敲低组GCSF, GM-CSF, IL-2, IL-3, IL-4, IL-5, IL-6, IL-9, IL-10, IL-12 p40, IL-12 p70, IL-13, IL-17, IFN-γ, MCP-1, MCP-5, RANTES, TNFα的表达水平。每次实验重复3次。结果: miR-520a-3p靶向RELA的3’UTR;LPS激活NF-kB信号通路后,宫颈癌HELA细胞分泌的细胞因子GCSF, GM-CSF, IL-2, IL-3, IL-4, IL-5, IL-6, IL-9, IL-10, IL-12 p40, IL-12 p70, IL-13, IL-17, IFN-γ, MCP-1, MCP-5, RANTES, TNFα的蛋白表达水平上升(P<0.05);过表达组中NF-κB复合体亚基RELA的蛋白表达水平下降,宫颈癌HELA细胞分泌的上述细胞因子的蛋白表达水平下降(P<0.05);敲低组中NF-κB复合体亚基RELA的蛋白表达水平上升,宫颈癌HELA细胞分泌的上述细胞因子的蛋白表达水平上升(P<0.05)。结论: miR-520a-3p通过靶向NF-κB信号通路的关键分子RELA抑制宫颈癌HELA细胞的细胞因子分泌。  相似文献

Edwardsiella tarda is an important Gram-negative bacterium that causes systemic infections in a wide range of hosts including fish. The pathogenic mechanisms in this disease are still poorly understood in fish. Indian major carp, Labeo rohita were intraperitoneally challenged with a pathogenic isolate of E. tarda to measure sequential changes in immunity level. A significant decrease in the superoxide production, myeloperoxidase, alternative complement activity, total protein levels and antiprotease activity of serum was marked in the infected fish. However, the serum lysozyme activity and haemagglutination titre were raised in the infected fish. Similarly, a significant rise in specific antibody titre was noticed on and after 10 days post-challenge. This study also elucidates the changes in the relative expression of some immune-related genes viz., interleukin 1-beta (IL-1β), inducible nitric oxide synthase (iNOS), complement component C3, β2-microglobulin, CXCa, tumor necrosis factor-alpha (TNFα), and C-type and G-type lysozymes during the infection. Significant up-regulation of IL-1β, iNOS, C3, CXCa and expression of both types of lysozyme genes was noticed at 6–12 h post-challenge (h.p.c.) whereas down-regulation of β2-microglobulin and TNFα genes was observed after 48 h p.c. The results obtained here strengthen the understanding on molecular pathogenesis of edwardsiellosis in L. rohita.  相似文献

In this study, the acute toxicity of paraquat (PQ) in common carp was determined. Then, the contents and mRNA levels of the cytokines interleukin‐1β (IL‐1β), interferon‐γ (IFN‐γ), and tumor necrosis factor‐α (TNF‐α) were evaluated following subacute exposure to PQ. The results show that the LC50 of PQ for common carp at 72 and 96 h was 15.962 and 15.106 mg/L, respectively. Moreover, after 7 days of subacute PQ exposure, the IL‐1β content in the fish liver and kidney increased, although it decreased in spleen. However, changes in the IFN‐γ content showed an irregular trend. The TNF‐α content increased in the liver and spleen but decreased in the kidney. Additionally, PQ exposure also induced alterations in the mRNA levels of IL‐1β, IFN‐γ, and TNF‐α. These results suggest that PQ exposure may result in suppression or excessive activation of the immune system in treated fish and lead to immune dysfunction and reduced immunity.  相似文献

IPNV is the agent of a well-characterized acute disease that produces a systemic infection and high mortality in farmed fish species and persistent infection in surviving fish after outbreaks. Because modulation of the host expression of pro and anti-inflammatory cytokines can help establish persistence, in this study, we examined the expression of IL-1β, IL-8, IFNα1 and IL-10 during acute and persistent IPNV infection of Atlantic salmon. Results showed that IPNV infection induces an increase of the IFNα1 and IL-10 mRNA levels in the spleen and head kidney (HK) of fish after acute experimental infection. Levels of the pro-inflammatory cytokines IL-1β and IL-8 did not rise in the spleen although an increase of IL-1β, but not of IL-8, was observed in head kidney. In carrier asymptomatic salmon, cytokine gene expression of IFNα1 in the spleen and IL-10 in head kidney were also significantly higher than expression in non-carrier fish. Interestingly, a decrease of IL-8 expression was also observed. IPNV infection of SHK-1, which is a macrophage-like cell line of salmon, also induced an increase of expression of the anti-inflammatory cytokine IL-10 with no effects on the expression of IL-1β and IL-8. The effects are induced by an unknown mechanism during viral infection because poly I:C and the viral genomic dsRNA showed the opposite effects on cytokine expression in SHK-1 cells. In summary, IPNV always induces up-regulation of the anti-inflammatory cytokine IL-10 in Atlantic salmon. As this is accompanied by a lack of induction of the pro-inflammatory cytokines IL-1β and IL-8, the anti-inflammatory milieu may explain the high frequency, prevalence and persistence of IPNV in salmon. We report results of a comprehensive analysis of inflammatory gene expression during the course of infection of Trypanosoma carassii in the goldfish. We observed significant increases in mRNA levels of genes encoding pro-inflammatory cytokines IFN-γ, TNFα1 and TNFα2; IL-1β-1 and IL-1β-2; IL-12-p35 and IL-12-p40; CCL1; CXCL8, anti-inflammatory cytokines IL-10 and TGFβ and iNOS A and iNOS B, using quantitative PCR. Expression levels and profiles of these cytokines and iNOS isoforms varied in the different tissues (kidney, spleen, liver) of goldfish during the course of T.?carassii infection. The expression of majority of genes that encode pro- and anti-inflammatory cytokines were up-regulated during the acute phase of infection (days 7-21 post-infection). The mRNA levels of these cytokines returned to normal levels or were down-regulated during the elimination phase of infection (days 28-56), with exception of IL-10 in the spleen and liver of infected fish. A parallel up-regulation of IFN-γ and IL-10 mRNA levels were observed in all tissues of infected fish during the acute phase of the infection. The expression of iNOS genes (iNOS A and B) was significantly delayed (day 14?pi) in the kidney, liver and spleen of infected fish. These results provide insights into the interaction between T.?carassii and goldfish, and suggest that Th1/Th2-like responses may be important for controlling T.?carassii infection in the goldfish.  相似文献

黄颡鱼饲料中添加谷胱甘肽降低藻毒素毒性作用的研究   总被引:1,自引:0,他引:1  
通过60d的生长实验研究饲料中不同含量的谷胱甘肽(GSH)对黄颡鱼(Pelteobagrus fulvidraco Richardson)摄食含蓝藻粉饲料后的毒性保护作用。共配制6种近似等氮等能(粗蛋白45%,总能19.5kJ/g)的实验饲料。以不添加蓝藻粉、不添加GSH的饲料组作为正对照,其他5种饲料均添加7.5%的藻粉,微囊藻毒素(Microc ystins,MCs)含量约为58μg/g,GSH添加量分别为:0(负对照)、200、500、800和1400mg/kg。实验结果表明,第1至第30天时,负对照组与正对照组实验鱼的摄食率无显著性差异(P>0.05)。第31至第60天时,负对照组实验鱼的摄食率显著高于正对照和其他GSH添加组(P0.05),但各GSH添加组肾脏中GSH显著低于正对照而高于负对照(P<0.05)。200、500和800mg/kg GSH添加组肝脏和肌肉中丙二醛(MDA)含量显著低于负对照(P<0.05)。在整个实验期间,随着饲料中GSH含量的增加,肝脏、肾脏、全肠和肌肉中微囊藻毒素含量先显著下降,当GSH添加量为500和800mg/kg时达到稳定,直至GSH添加量为1400mg/kg时显著上升。根据对黄颡鱼增重,肝脏和肌肉中MDA的含量以及组织中毒素的分析,在饲料中添加500-800mg/kg GSH可以有效缓解黄颡鱼饲料中微囊藻毒素的毒性。    相似文献

According to our previous results the gastropod Lymnaea stagnalis exposed to MC-producing cyanobacteria accumulates microcystins (MCs) both as free and covalently bound forms in its tissues, therefore representing a potential risk of MC transfer through the food web. This study demonstrates in a laboratory experiment the transfer of free and bound MCs from L. stagnalis intoxicated by MC-producing Planktothrix agardhii ingestion to the fish Gasterosteus aculeatus. Fish were fed during five days with digestive glands of L. stagnalis containing various concentrations of free and bound MCs, then with toxin-free digestive glands during a 5-day depuration period. MC accumulation was measured in gastropod digestive gland and in various fish organs (liver, muscle, kidney, and gills). The impact on fish was evaluated through detoxification enzyme (glutathion-S-transferase, glutathion peroxydase and superoxyde dismutase) activities, hepatic histopathology, and modifications in gill ventilation, feeding and locomotion. G. aculeatus ingestion rate was similar with intoxicated and toxin-free diet. Fish accumulated MCs (up to 3.96 ± 0.14 μg g−1 DW) in all organs and in decreasing order in liver, muscle, kidney and gills. Hepatic histopathology was moderate. Glutathion peroxydase was activated in gills during intoxication suggesting a slight reactive oxygen species production, but without any impact on gill ventilation. Intoxication via ingestion of MC-intoxicated snails impacted fish locomotion. Intoxicated fish remained significantly less mobile than controls during the intoxication period possibly due to a lower health condition, whereas they showed a greater mobility during the depuration period that might be related to an acute foraging for food. During depuration, MC elimination was total in gills and kidney, but partial in liver and muscle. Our results assess the MC transfer from gastropods to fish and the potential risk induced by bound MCs in the food web.  相似文献

ObjectiveEven though boron (B), as a trace micronutrient, occurs in natural waters and organisms, its high concentration could cause harmful and even toxic for organisms. The aim of present study was to investigate the effects of boric acid (BA) added to feed (0.01%, 0.05%, 0.10%, and 0.20% of B in feed) on Rainbow Trout (Oncorhynchus mykiss) by histopathological methods and compared to the control feed as without B.MethodologyAt the end of the feeding, after weighing the live weight of 9 rainbow trout from each group, tissue fragments were taken from the liver, gill, kidney, skeletal muscle, spleen and brains of fish, which systemic necropsies was done, and were fixed in 10 % buffered formalin solution.ResultsHistopathological examinations revealed degenerative and necrotic changes in the liver, gill, kidney, skeletal muscle, spleen and brain. Hydropic and vacuolar degenerations in liver parenchyma, lamellar edema in the gills, hyaline accumulation in the kidneys, degenerations in the muscles, necrosis in the spleen and hyperemia in the brain were observed in all groups except control group. It was observed that the destruction of boric acid on fish increased depending on the amount of boron supplemented to the feed, and the most effect was in the group fed with 0.20 % boron supplemented feed.ConclusionsThe findings aside from causing pathological changes in all organs in terms of histopathological findings of Boric Acid (BA), the most severe lesions were observed in the liver.  相似文献

The present study investigated the potential changes in hematologic parameters and histology of fish Heteropneustes fossilis when exposed to industrial waste water of different concentrations. The toxicant enter into the fish body from different rout, inhalation, dermal oral, and other several rout for entering in the fish body. The blood parameters of fish H. fossilis control group and treatment were investigated on 1st, 5th, 10th, and 20th days of experiment, hematologic variation was observed in RBC, WBC, MCV, MCH, and MCHC count. Histopathologic changes in liver, intestine, gill, muscle, and heart showed increasing degrees of damage in the tissues in correlation with the accumulation pattern of pesticides, while liver, intestine, gill, muscle, and heart of control group exhibited a normal architecture. Toxic effects of pesticides vary in different organs of the fish H. fossilis significant. The fish exposed to higher concentration showed uncoordinated alterations in behavioral responses, especially erratic and jerky swimming, physiologic, malformation, histologic, hematologic and biochemical changes, frequent surfacing and in gulping, mucus secretion, an increase in opercular movement, and copious secretion of mucus of all over the body.  相似文献

Deoxynivalenol (DON) is one of the most frequently detected mycotoxins in agricultural commodities used as animal feedstuff in Central Europe. This study focuses on determining effects of diets containing DON on oxidative stress markers and detoxifying enzymes in rainbow trout (Oncorhynchus mykiss). The fish were fed with commercial pellets containing DON at a concentration of 2 mg kg?1. Selected enzymes were measured in liver, gill and caudal kidney of the fish after 23 and 32 days of the experiment. Significant differences between the control and experimental groups were observed concerning activities of glutathione peroxidase (GPx) in kidney, glutathione reductase (GR) in gill and kidney, catalase (CAT) in kidney and liver and glutathione S‐transferase (GST) in gill and liver. No significant differences were found for superoxide dismutase (SOD) gene expression, lipid peroxidation (TBARS) and the ferric reducing ability of plasma (FRAP). The data show that DON in the diet at the concentration below EC recommendation (2006/576/EC) induces oxidative stress in the rainbow trout.  相似文献
