为了研究葡萄糖醛酸木聚糖酶在枯草芽孢杆菌中异源表达,笔者从枯草芽孢杆菌中克隆得到带有自身信号肽的葡萄糖醛酸木聚糖酶基因,将其构建到大肠杆菌-枯草芽孢杆菌穿梭质粒中,转化入枯草芽孢杆菌WB800,得到重组菌。通过发酵条件以及培养基成分优化,重组菌中葡萄糖醛酸木聚糖酶酶活达到76.0 U/mL,约为优化前产酶量的5.4倍。葡萄糖醛酸木聚糖酶在枯草芽孢杆菌中实现高效异源表达,为其进一步的实际应用奠定了基础。  相似文献

枯草芽孢杆菌感受态细胞的制备及质粒转化方法研究   总被引:1,自引:0,他引:1  
为便于枯草芽孢杆菌工业化生产应用,对Spizizen创立的枯草芽孢杆菌DNA转化方法进行改进.用GMI和GMII溶液处理枯草芽孢杆菌野生型菌株BS501a、营养缺陷型突变株DBl342和非营养缺陷型突变株WB800,用改进的方法制备感受态细胞,用7.5kb质粒pSBPTQ进行转化,并研究RNA、酵母粉、水解酪蛋白、培养方法对枯草芽孢杆菌质粒转化的影响.结果表明,该方法适用于不同基因型枯草芽孢杆菌的质粒转化,营养缺陷型突变株DBl342的转化率为750 CFU/μg/DNA,非营养缺陷型突变株WB800转化率为1 070 CFU/xg DNA,野生型菌株BS501a转化率为270 CFU/μg/DNA.根据影响转化效率的因素,推测在该方法中,枯草芽孢杆菌质粒转化原理:一定生物量的枯草芽孢杆菌在外界营养条件和钙、镁离子作用下,细胞壁和细胞膜形成缺陷,使外源DNA转入枯草芽孢杆菌细胞内.  相似文献

枯草芽孢杆菌感受态细胞的质粒转化与质粒的分子构型有关。用琼脂糖凝胶电泳法分离纯化的pUB110质粒DNA单体是很少或没有转化活性的。在pUB110质粒DNA上插入一段受体菌染色体DNA,这种杂种质粒的单体可以转化感受态细胞,但受体菌必须是重组型的。转化效率和插入片段的大小有关,片段愈大转化活性愈高,片段小于0.6kb,就和pUB110DNA单体一样,几乎没有转化活性。在pUB110质粒DNA上插入一段解淀粉芽孢杆菌染色体DNA,这种杂种质粒的单体转化效率都很低。本文还讨论了用鸟枪法在枯草芽孢杆菌中进行分子克隆的一些问题。  相似文献

地衣芽孢杆菌感受态细胞的形成及高效电转化   总被引:3,自引:0,他引:3  
芽孢杆菌在营养缺乏的饥饿状态下,细胞易产生感受态因子,处于生长芽孢时期的芽孢杆菌更容易产生感受态。基于此原则利用芽孢杆菌极限营养培养基通过体外处理诱导使地衣芽孢杆菌产生感受态性能,同时调整参数,建立了感受态细胞对质粒pAPR的高效电转化方法。当质粒DNA浓度为1.5μg/ml、转化时电压为1750V的时候,可以得到261个转化子,经鉴定均为阳性克隆子。而常规电转化的最高仅为20个转化子。为以芽孢杆菌为宿主进行高效电转化提供了模型,也为建立适合工业应用的分泌型表达载体的构建打下了一定基础。  相似文献

以质粒pMUTIN-GFP 扩增获得的目的gfp 基因为报告基因,将其克隆到大肠杆菌-枯草芽孢杆菌穿梭载体pBE2,构建成一个具有启动子活性检测功能的重组质粒pBE2-GFP .将组成型启动子P43和诱导型启动子Pspac克隆入pBE2-GFP ,得到重组表达载体pBE-GFP-P43和pBE-GFP-Pspac,转化至大肠杆菌和枯草芽孢杆菌.荧光显微镜检测GFP 蛋白的表达情况.结果 表明,2种不同类型的启动子均能在大肠杆菌BL21和枯草芽孢杆菌1A751中启动gfp 基因的表达.  相似文献

枯草芽孢杆菌是一种广泛应用于基础研究和工业生产的重要模式菌株,具有无致病性、蛋白分泌能力强、遗传背景清晰等多种优势,是生产异源蛋白的理想宿主。目前已有诸多异源蛋白在枯草芽孢杆菌中实现表达和分泌,其中包括淀粉酶、β-半乳糖苷酶和蛋白酶等有价值的工业酶。本文从异源蛋白表达和分泌的关键步骤出发,总结了枯草芽孢杆菌生产异源蛋白的传统策略和最新技术。除此之外,分析了当前研究存在的瓶颈并对如何提高异源蛋白产量提出了新的建议和策略。  相似文献

【背景】碱性蛋白酶是工业用酶中占比最大的酶类,广泛应用于清洁、食品、医疗等行业。近期研究发现碱性蛋白酶在生产生物活性肽方面有巨大潜力,这将进一步拓宽其在保健食品领域中的应用。【目的】利用枯草芽孢杆菌异源表达地衣芽孢杆菌来源的碱性蛋白酶SubC。【方法】通过筛选3种枯草芽孢杆菌宿主菌株(Bacillus subtilis 1A751、MA07、MA08)和6种信号肽(AmyE、AprE、NprE、Pel、YddT、YoqM),同时优化诱导剂浓度、发酵培养基和发酵时长,最终得到最优重组菌株MA08-AmyE-subCopt。【结果】重组菌株MA08-AmyE-subCopt的胞外酶活力为3.33×103 AU/mL,胞外蛋白分泌量为胞内可溶蛋白表达量的4倍,与携带野生型信号肽的对照组菌株WT相比,酶活提高了73.4%。【结论】异源碱性蛋白酶SubC在枯草芽孢杆菌中成功表达,为碱性蛋白酶SubC的表达和在保健食品领域的工业化应用提供了理论基础。  相似文献

携带穿梭质粒的大肠杆菌与作为受体的枯草芽孢杆菌分别培养至不同生长阶段混合均匀后静置40min,涂布选择性平板,37℃培养30h后得到一定数目的转化子,DNaseⅠ敏感实验证实质粒是通过自然遗传转化而非其它形式发生转移。实验发现大肠杆菌可以在特定生长时期向胞外分泌DNA,并且在对数期具有最高的提供质粒的能力,而生长后期的细胞因为体系中DNase量的增加转化频率下降。进一步的研究发现枯草芽孢杆菌在营养丰富的LB培养基中也具有与基本培养基中相当的转化能力,并且在对数生长前期具有较高的转化频率。  相似文献

【目的】凝结芽孢杆菌(Bacillus coagulans)是一种非常有工业应用前景的微生物,但遗传转化的困难是限制对其进行代谢工程改造的关键因素。本研究主要考察生长培养基、电击缓冲液、复苏培养基中添加高渗剂如山梨醇、甘露醇等对凝结芽孢杆菌转化效率稳定性的影响,并对凝结芽孢杆菌电击转化条件进行优化。【方法】用穿梭质粒p NW33N电转化凝结芽孢杆菌P4-102B,系统地考察高渗条件下细胞生长阶段、感受态菌体浓度、电击缓冲液组分和复苏培养基组成等因素对转化效率的影响。【结果】同一电场压力下高渗体系转化效率较低渗体系明显提高且稳定性较好,菌体在含有0.5 mol/L山梨醇的LB培养基中生长到OD600为0.8时收集,用SMG[0.5 mol/L山梨醇,0.5 mol/L甘露醇,10%(质量体积比)甘油]电击缓冲液洗涤菌体4次制备感受态,在固定电场强度14 k V/cm、脉冲时间5 ms、1 mm规格的电转杯进行电击转化,电转化后立即加入含有0.5 mol/L山梨醇和0.38 mol/L甘露醇的LB复苏培养基培养,能够获得最佳的转化效率2.7×102 CFU/μg DNA。【结论】使用高渗电击转化法能够提高电击转化的稳定性和重复性,并且可以获得较高的转化效率。  相似文献

枯草芽孢杆菌(Bacillus subtilis)具有很强的分泌能力,产酶能力强,属于食品级安全微生物.以枯草芽孢杆菌为宿主菌异源表达木聚糖酶,采用同源重组的方法将木聚糖酶基因连接到载体pWB980上,构建表达载体pWB980-xynZF-2,电击转化枯草芽孢杆菌WB600,获得重组工程菌WB600-pWB980-xynZF-2.对重组工程菌进行单因素发酵条件优化和正交试验.结果表明:重组枯草芽孢杆菌摇瓶发酵的最适接种量1%、种龄14 h、装液量50 mL、温度34℃,转速160 r/min、发酵时间192 h.在摇瓶发酵条件下,木聚糖酶产量可达到0.168 U/mL.  相似文献

Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Rasmussen’s encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and 16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis, intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8+ T cells. Our results suggest that EBV and HHV6 infection and the activation of CD8+ T cells are involved in the pathogenesis of RE.  相似文献

正Dear Editor,Chikungunya virus (CHIKV), an arbovirus in the family of Togaviridae, genus Alphavirus, is transmitted by the A.aegyptii or A. albopictus mosquito, and causes disease in humans characterized by fever, rash, and arthralgia (Silva and Dermody 2017; Suhrbier 2019). It was first reported in 1953 in Tanzania, and caused only a few outbreaks and sporadic cases in Africa and Asia in last century. However, in the epidemic in 2004, CHIKV acquired mutations that conferred enhanced transmission by the A. albopictus mosquito(Schuffenecker et al. 2006). Since then, it has successively caused outbreaks in Africa, the Indian Ocean, South East Asia, the South America, and Europe (Zeller et al. 2016).  相似文献

In conclusion, the novel visual RT-LAMP assay is a simple, rapid, and sensitive approach for detection of SARS-CoV-2, and it is ready for application in primary care and community hospitals or health care centers, and even patients' own houses in response to the current SARS-CoV-2 epidemic because the assay does not require sophisticated equipment and skilled personnel. Furthermore, it is also ready to be used in fields for screening samples from wild animals and environments to facilitate the identification of potential intermediate hosts that mediate the cross-species transmission of SARS-CoV-2 from bats to humans.  相似文献
