本文通过研究乳酸菌源有机硒干预CCl_4致肝损伤小鼠脾脏NK细胞活性和脂质过氧化反应的变化,探讨该有机硒在抗损伤保护过程中的效应及其机制。分别选用60只健康成年小鼠,雌雄对半,随机分成对照组(C组),有机硒组(Se组),CCl_4组、CCl_4-有机硒保护组(CCl_4-Se组),每组15只。通过腹腔注射CCl_4诱发肝损伤后,分别在第2、4周检测脾脏NK细胞活性及其组织匀浆GSH-Px、CAT、SOD活性和MDA含量变化、结果显示,在整个实验期内,C组、Se组和CCl_4-Se组脾组织匀浆GSH—Px、CAT和SOD活性均高于或明显高于CCl_4组,Se和CCl_4-Se组与C组比较除SOD活性在第4周有明显升高外均差异不显著;CCl_4组小鼠脾脏MDA含量均显著高于C组、Se组和CCl_4-Se组,而CCl_4-Se组与C组接近,Se组较CCl_4-Se组和C组低;Se组NK细胞活性最高,第4周明显高于C组,CCl_4组最低且低于或明显低于CCl_4-Se、Se和C组,CCl_4-Se组与C组无明显差异。结果提示,乳酸菌源有机硒能够提高正常机体抗氧化能力,在干预肝损伤过程中,可以通过改善和提高脾组织抗氧化酶活性及NK细胞活性发挥积极有效的作用。  相似文献

目的:研究中药活性物质蟛蜞菊内酯的保肝作用及其机制。方法:采用小鼠腹腔注射CCl4制作肝损伤模型,测定小鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、丙二醛(MDA),谷胱甘肽(GSH)和超氧化物歧化酶(SOD)指标,进行肝脏的组织病理学检查,观察蟛蜞菊内酯对CCl4所致肝损伤的保护作用。结果:蟛蜞菊内酯能明显降低肝损伤小鼠的血清ALT、AST和肝组织匀浆中MDA含量,SOD活力增强,明显减轻肝组织变性。结论蟛蜞菊内酯对CCl4引起的肝损伤有明显的保护作用,其机制可能与其抗氧化作用有关。  相似文献

为研究维生素C多聚磷酸酯对小鼠抗热应激能力的影响 ,将 4 8只 3~ 4周龄、体重为 16~ 2 9g的健康雄性小鼠随机分为 4组 (对照组、Ⅰ、Ⅱ和Ⅲ组 ) ,在其饵料中分别添加 0、 5 0 0、 2 5 0 0和 5 0 0 0mg/kg的35 %维生素C多聚磷酸酯。喂食 4周后 ,每组取一半小鼠处死取其肝脏 ,另一半置于 (35± 1)℃条件下 ,2 4h后作同样处理。用硫代巴比妥酸分光光度法测肝脏脂质过氧化物 (LPO)的含量 ,用亚硝酸盐形成法测超氧化物歧化酶 (SOD)的活性 ,用分光光度法测过氧化氢酶 (CAT)和谷胱甘肽过氧化物酶 (GSH Px)的活性。经热应激与未经热应激相比 ,LPO :对照组和Ⅰ组显著升高 ,而Ⅱ和Ⅲ组无显著差异。SOD和GSH Px :对照组显著下降 ,其他 3组无显著差异 ;其中SOD :Ⅱ和Ⅲ组显著高于对照组和Ⅰ组 ,GSH Px :Ⅲ组显著高于其他 3组。CAT :对照组和Ⅰ组显著降低 ,而Ⅱ和Ⅲ组无显著差异 ;Ⅱ和Ⅲ组显著高于对照组和Ⅰ组。表明热应激促进了小鼠肝脏LPO的产生 ,抑制了抗化物酶的活性 ;而维生素C多聚磷酸酯对热应激造成的不利影响有缓解作用。  相似文献

本文通过研究CCl4致小鼠肝损伤组织匀浆和血浆一些免疫介质含量的变化以探讨这些免疫介质在CCl4诱发肝损伤过程中作用机制。分别选用30只健康成年小鼠,雌雄各半,随机分成对照组和CCl4负荷组,每组15只。通过腹腔注射CCl4诱发肝损伤后,分别在第2、4、6周检测肝组织匀浆cAMP、cGMP和MDA及血浆IL-2、TNF-α水平的变化。结果显示,在整个实验期内,CCl4组肝组织匀浆cAMP水平均低于或明显低于对照组;cGMP在实验第2周后,高于或显著高于对照组;cAMP/cGMP比值呈现下降趋势,并低于或明显低于对照组;MDA含量明显高于对照组。在整个实验期内,CCl4组血浆IL-2水平下降或显著下降;TNF-α水平则均高于或显著高于对照组。结果提示,CCl4负荷诱发免疫介质cAMP、cGMP、TNF-α和IL-2发生剧烈变化,在介导肝损伤过程中可能起重要作用。  相似文献

探讨禹州漏芦乙醇提取物对四氯化碳(CCl4)诱导小鼠急性肝损伤的保护作用。以CCl4诱导小鼠急性肝损伤模型,检测血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)活性,同时测定肝匀浆中的超氧化物岐化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)的水平。将肝大叶HE染色,观察各组小鼠的肝组织病理改变。结果表明,同模型组比较,禹州漏芦乙醇提取物各剂量组均能降低小鼠血清中ALT、AST及MDA活性,升高肝组织中GSH-Px和SOD的活性,并能明显改善肝组织的病理学损伤。禹州漏芦乙醇提取物对CCl4所致小鼠急性肝损伤具有较好保肝作用,其作用可能与清除体内自由基和抗氧化的作用有关。  相似文献

旨在探究聚乙二醇修饰重组细胞珠蛋白(PEG modified recombinant cytoglobin,PEG-rCygb)对小鼠急性肝损伤的保护作用。采用CCl4诱导KM小鼠急性肝损伤模型,尾静脉注射PEG-rCygb,收集血清及肝脏组织检测各项生化指标及组织病理学变化。结果表明,PEG-rCygb治疗组小鼠肝脏系数减小,血清中AST﹑ALT水平降低,肝组织匀浆中MDA含量减少,GSH含量增加,T-SOD、CAT活性升高。肝组织切片HE染色显示PEG-rCygb可以缓解肝细胞脂肪变性,减少炎症因子,减轻肝细胞损伤。体外细胞学实验表明rCygb经PEG修饰后对H2O2造成的肝星状细胞(HSC)氧化损伤发挥的保护作用增强。研究结果显示PEG-rCygb提高了机体对自由基的清除能力,对CCl4引起的小鼠急性肝损伤具有保护作用。  相似文献

目的:探究菠萝蜜低聚肽(JOPs)对γ射线辐照导致小鼠氧化损伤的保护作用。方法:选取96只SPF级雌性BALB/c小鼠,按体重随机分为空白组、模型组、乳清蛋白组(0.40 g/kg·BW)及3个JOPs干预组(0.20、0.40、0.80 g/kg·BW),每组随机分为2个亚组,8只/亚组。行灌胃干预第14 d除空白组外,小鼠接受60Co γ射线全身辐照,剂量3.5 Gy,剂量率1 Gy/min。两亚组分别在辐射后第3 d和第14 d检测小鼠血清和肝脏超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH Px)活性及丙二醛(MDA)水平。结果:辐照后第3 d及第14 d,相比空白组,模型组血清、肝脏SOD及GSH Px活性均显著降低,MDA水平均显著增高,JOPs各剂量组小鼠血清及肝脏MDA水平均显著低于模型组与乳清蛋白组;辐照后第3 d,相比模型组,JOPs各剂量组小鼠血清及肝脏SOD、GSH Px活性均显著增高,且高剂量组小鼠血清SOD活性及血清、肝脏GSH Px活性与中、高剂量组肝脏SOD活性均显著高于乳清蛋白组;辐照后第14 d,相比模型组,JOPs各剂量组小鼠血清及肝脏SOD、GSH Px活性均显著增高,且JOPs各剂量组小鼠血清SOD活性与高剂量组肝脏SOD、GSH Px活性均显著高于乳清蛋白组。结论: JOPs对γ射线辐照所致氧化损伤具有保护作用。  相似文献

观察柴胡及五味子对CCl4诱导的急性肝损伤的小鼠治疗效果。采用CCl4制备小鼠急性肝损伤模型,测定小鼠血清ALT、AST和肝组织MDA、SOD、GSH水平,并进行肝病理组织学检查。结果:五味子、柴胡、五味子及柴胡连用高、低剂量组与CCl4模型对照组比较ALT、AST、MDA、SOD含量均降低,差异有显著性。结论:五味子和柴胡可降低肝损伤小鼠血浆ALT、AST水平,具有良好的保肝效果。  相似文献

目的比较CCl4和脂多糖诱导的两种急性肝损伤模型中肝细胞凋亡的病理特点。方法雄性BABL/c小鼠随机分为正常组、CCl4模型组与脂多糖模型组。CCl4和脂多糖模型小鼠分别腹腔注射0.03 mL/kg 50?l4/橄榄油混合液和10μg/kg脂多糖与900μg/kg半乳糖胺。9 h后采集标本,检测肝细胞凋亡,血清ALT、AST活性,肝组织SOD活性、MDA含量。结果与正常组比较,CCl4和脂多糖模型小鼠血清ALT与AST活性均明显升高(P<0.01),且脂多糖模型高于CCl4模型(P<0.01);模型小鼠均有明显肝细胞凋亡(P<0.01),且两组间无显著差异,但CCl4模型呈局灶性,以中央静脉周围为主;脂多糖模型呈弥漫性,肝细胞坏死与肝组织炎症更显著(P<0.01)。CCl4和脂多糖模型肝组织均SOD活性升高、而MDA含量下降(P<0.01),但CCl4模型更显著(P<0.05)。结论CCl4与脂多糖诱导的急性肝损伤小鼠模型均有明显的肝细胞凋亡,但CCl4模型以中央静脉区为主,脂质过氧化损伤更为明显;脂多糖模型呈弥漫性,肝细胞坏死与肝组织炎性反应较重。  相似文献

研究了银耳多糖对D-半乳糖致衰老模型小鼠抗氧化能力影响。将ICR小鼠腹腔注射D-半乳糖建立衰老模型,同时给予不同剂量的银耳多糖(TP),8周后获取小鼠心、脑,测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力以及丙二醛(MDA)含量的差异。结果:TP各剂量组SOD和GSH-Px活力高于对照组(P<0.05),且高剂量组SOD、GSH-Px活力均高于低剂量和中剂量组(P<0.05);中、高剂量TP组MDA含量低于对照组(P<0.05)且高剂量组MDA含量低于低剂量组(P<0.05)。结论:银耳多糖对于衰老模型小鼠抗氧化能力具有一定正性调节作用。  相似文献

Selenium (Se) is an essential trace element for humans and animals. Stropharia rugoso-annulata is a nutritional and functional mushroom containing many kinds of bioactive ingredients. The aims of this study were to investigate the Se-enrichment characteristics of S. rugoso-annulata in submerged culture and evaluate the antioxidant activities of Se-enriched mycelia in vivo in terms of the values of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), and malondialdehyde (MDA). The optimum parameters of Se-enrichment under the optimal Se concentration (150 μg/mL) in media were as follows: biomass 8.11 ± 0.25 g/L, Se content in mycelia 4,727.68 ± 13 μg/g, Se-accumulated rate 24.68 ± 1.67%, and percentage of organic Se 96.27 ± 3.26%. The mainly subsistent forms of selenium in Se-enriched mycelia were selenoprotein and selenium-polysaccharide. The contents of total amino acids (TAA) and essential amino acids (EAA) in Se-enriched mycelia were increased by 13.5 ± 1.09% and 12.8 ± 0.89%, respectively. It was efficient for Se-enriched mycelia to elevate GSH-Px and SOD activities and decrease MDA content. These results indicated that Se-enriched mycelia of S. rugoso-annulata represent a novel dietary source of bioavailable supplemental selenium.  相似文献

目的探讨富硒平菇干粉对小鼠的脏器指数与抗氧化能力的影响。方法将60只20g左右的健康昆明种小鼠随机分为6组。1组为对照组;2组为亚硒酸钠组;3组为普通平菇干粉组;4~6组为富硒平菇干粉组,分别在基础日粮中添加低、中、高浓度的富硒平菇干粉。30d后测定小鼠脏器指数与肝脏和血浆中谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)和丙二醛(MDA)含量。结果富硒平菇干粉可显著提高小鼠脾脏、胸腺和肝体指数;可显著提高小鼠肝脏与血浆中GSH-Px、SOD的酶活,降低小鼠肝脏和血浆中MDA含量。结论富硒平菇干粉可显著提高小鼠脏器指数与抗氧化能力。  相似文献

目的:研究酵母多糖(zymosan)对S180荷瘤小鼠抗氧化状态和免疫功能的影响。方法:将昆明小鼠70只随机分为7组,第1组作为正常组,另6组将S180瘤细胞悬液接种于小鼠右前肢腋皮下制备荷瘤小鼠动物模型,分为zymosan低、中、高剂量组和环磷酰胺(Cy)组、肿瘤对照组及zymosan联合Cy治疗组,灌胃第7 d接种S180瘤,第19 d后测定小鼠肝匀浆中丙二醛(MDA)含量、谷胱甘肽过氧化酶(GSH-Px)和超氧化物歧化酶(SOD)活力,评价zymosan对小鼠抗氧化能力的作用。采用逆转录-聚合酶链反应(RT-PCR)检测IL-2、TNF-α、TGF-β1mRNA的表达,评价zymosan对小鼠免疫功能的影响。结果:zymosan各剂量组均能提高肝脏中GSH-Px活性和SOD活力,但zymosan高剂量组显著降低MDA水平(P<0.01)。zymosan各剂量组荷瘤小鼠肿瘤组织内TNF-α、IL-2 mRNA相对表达量明显高于Cy组和肿瘤对照组(P<0.01),同时下调TGF-β1水平。zymosan高剂量对小鼠S180瘤有明显抑制作用,且与Cy合用后有协同作用,并提高了小鼠的TNF-α、IL-2 mRNA的表达。结论:zymosan的抑瘤机制可能主要是抗氧化作用和免疫调节作用,且作用与剂量有关。  相似文献

为了研究铁棍山药(D.oppositacv.Tiegun)多糖对四氯化碳诱导的小鼠急性肝损伤的保护作用,取72只昆明小鼠随机分为对照组,四氯化碳(CCl4)模型组,阳性对照(联苯双酯)组,铁棍山药多糖低、中、高剂量组,每组12只,灌胃处理后使用CCl4制备急性肝损伤小鼠模型,观察各组形态学变化,同时测定生化指标。实验结果显示,经铁棍山药多糖处理的小鼠的肝损伤程度明显轻于模型组,铁棍山药多糖能降低小鼠血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)含量,提高超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性,降低丙二醛(MDA)、一氧化氮(NO)、肿瘤坏死因子-α(TNF-α)的含量。本研究结果表明,铁棍山药多糖对CCl4所诱导的小鼠肝损伤起到一定的保护作用。  相似文献

Thirty-two wether lambs of Tan sheep were randomly assigned into four dietary treatment groups (eight per group) for an 8-wk study and then fed a basal diet deficient in Se (0.06 mg/kg) or diets supplemented to provide 0.10 mg/kg Se from sodium selenite, selenized yeast, and selenium-enriched probiotics, respectively. Blood samples were collected at d 0, 28, and 56 of the experiment and tissue samples were collected at experiment termination. Tissue and blood Se concentrations, blood glutathione peroxidase (GSH-Px) activities, and plasma interleukin levels were analyzed. The results showed that the concentrations of Se in the kidney, liver, and muscle increased in all of the supplemented groups (p<0.01) compared with the control group. However, the Se concentrations in the kidney, liver, and muscle in the groups supplemented with Se yeast and Se-enriched probiotics were higher than those in the group supplemented with sodium selenite (p<0.01). The activities of GSH-Px and the concentrations of Se in blood also increased in all of the supplemented groups during the period of supplementation (p<0.01) compared with the control group. The activities of GSH-Px and the concentrations of Se in the whole blood of the lambs fed with selenized yeast and Se-enriched probiotics were higher than those of lambs fed with sodium selenite (p<0.01 or p<0.05). The concentrations of interleukin-1 and interleukin-2 in plasma significantly increased in all of the supplemented groups during the entire period of experiment (p<0.01) compared with the control group, but had no significant differences among all of the supplemented groups. In conclusion, a diet supplemented with Se for finishing lambs was able to increase the concentrations of Se in tissue and blood, activities of GSH-Px in blood, and levels of interleukins in plasma. Organic Se sources (selenized yeast and Se-enriched probiotics) were more effective than the inorganic Se source (sodium selenite) in increasing tissue and blood Se concentrations and blood GSH-Px activities of lambs. However, there were no significant differences in plasma interleukin levels of lambs between organic and inorganic Se sources.  相似文献

Diabetes mellitus is associated with diabetic impairment of testicular function, ultimately leading to reduced fertility. Its etiology may involve oxidative damage by reactive oxygen substances, and protection against this damage can be offered by antioxidant supplementation. The aim of this study was to investigate the effects of intraperitoneal administration of vitamin C and E, selenium (Se), and vitamin E plus Se (COM) on concentrations of lipid peroxide (as malondialdehyde; MDA), reduced glutathione (GSH), and vitamin E concentrations and glutathione peroxidase (GSH-Px) activity in the testes of rats with diabetes induced by streptozotocin (STZ). Sixty groups were used (10 animals each) and these animals were initially allocated to two groups: control group and diabetic group. The diabetic group was subdivided into five groups as follows: diabetic control (DC), vitamin E, Se, COM, and vitamin C. Animals in the DC group and vitamin C, vitamin E, Se, and COM groups were made diabetic by the injection of STZ on 4 d after an injection of vitamins C and E, Se, and COM. Those vitamins and Se were also administered for 21 consecutive days. The MDA, vitamin E, GSH levels, and GSH-Px activities in testes were determined. Although the vitamin E concentration was higher in the control than in the DC group, the MDA levels were found to be lower in the control than in the DC group. The MDA levels in the testes samples of vitamin C, vitamin E, Se, and COM groups were lower than the DC group. However, GSH-Px activity and GSH levels in the testes were not significantly different between the control and DC groups. Vitamin E concentrations in the vitamin C, vitamin E, Se, and COM groups and GSH levels and GSH-Px activities in the Se, COM, and vitamin C groups were higher than either the control or DC group. The results indicate that reactive oxygen substances may be involved in possible testicular complications in diabetes of rats. Administration of vitamins C and E and Se reduced the testicular lipid peroxidation; these vitamins and Se had significant protective effects on testes of rats against oxidative damage in diabetes.  相似文献

预防给予小鼠松茸多糖(PTM),测定小鼠在受到2 Gy X线照射后,脾和胸腺重量、T淋巴细胞转化能力、外周血中白细胞数和肝组织中SOD、CAT、GSH-Px活性及MDA含量的变化,结果显示PTM处理组小鼠的各项指标与辐射对照组比较均有显著性差别,提示PTM可促进机体自由基的清除,增加机体抗氧化能力,对辐射所致的免疫损伤有明显的保护作用。  相似文献
