Three new ent-kaurene diterpenoids, oreskaurins A-C (1-3), together with ten known ent-kaurene diterpenoids, enmenin monoacetate (4), effusanin E (5), adenolin B (6), maoecrystal G (7), enmelol (8), trichokaurin (9), sodoponin (10), trichorabdal A (11), nodosin (12), enmein (13), and a flavonoid, vitexin (14), were isolated from Isodon oresbius. Their structures were determined by spectroscopic means. Compound 12 showed inhibitory activity toward K562 cells with IC(50)=1.43 microg/ml.  相似文献

Two new named maoecrystal F(1) and G(2) and seven known ent-kaurene diterpenoids were isolated from the leaves of Rabdosia eriocalyx (Dunn) Hara collected at Xundian County, Yunnan Province, and their structures were determined by spectroscopic analysis. Maoecrystal F and G are elucidated as ent-7α, 15α-dihydrooxyl- lβ,6α-diacetoxyl-7β, 20-epoxy-16-kaurene (1) and ent-7α,15α-dihydroxyl-6α-ace-toxyl-7β, 20-epoxy-16-kaurene(2), respectively.  相似文献

Two new pimarane diterpenoids, momilactone D ( 3 ) and momilactone E ( 5 ), along with three known diterpenoids, momilactone A ( 1 ), sandaracopimaradien‐3‐one ( 2 ), and oryzalexin A ( 4 ) were isolated from Oryza sativa roots. The chemical structures of the compounds were determined by spectroscopic data analysis. The isolated diterpenoids were evaluated for their ability to inhibit NO production and iNOS mRNA and protein expression in LPS‐stimulated RAW264.7 macrophages. Compound 4 showed strong inhibition activity on NO production, and compounds 1 and 4 decreased the expression of iNOS mRNA and protein levels.  相似文献

Two new neo-clerodane diterpenoids have been isolated from the aerial parts of Teucrium oxylepis subsp. marianum. Their structures, 4,18;15,16-diepoxy-6β,12S-dihydroxy-neo-clerado-13(16),14-dien-20,19-olide (teucroxylepin) and 12S-acetoxy-6β,18; 15,16-diepoxy-4,6-dihydroxy-neo-cleroda-13(16),14-dien-20,19-olide (12-O-acetylteugnaphalodin), were established by chemical and spectroscopic means. In addition, 10 already known neo-clerodane diterpenoids were also isolated from the same source.  相似文献

Two novel labdane-type diterpenoids, butanedioic acid mono[(13S)-13-hydroxylabda-8(17),14-dien-19-yl] ester (1) and butanedioic acid bis[(13S)-13-hydroxylabda-8(17),14-dien-19-yl] ester (2), along with the eleven known diterpenoids 3-13 and three other known compounds, were isolated from the bark of Larix chinensis. Their structures were elucidated both spectroscopically and chemically. The major diterpenoids 1-9, 11, and 13, as well as the very abundant phenolic compound 16, were subjected to antibacterial and cytotoxic bioassays. Compounds 7 and 9 showed remarkable in vitro inhibition of Staphylococcus aureus and S. epidermidis (MIC = 37-73 microM).  相似文献

Chemical examination of the ethyl acetate solubles of the CH(3)OH:CH(2)Cl(2) (1:1) extract of the roots of Excoecaria agallocha L. collected from Godavary estuary resulted in the isolation of three more new diterpenoids, agallochins M-O (1-3). The structures of the new diterpenoids were elucidated by a study of their physical and spectral data as methyl ent-13-epi-8,13-epoxy-4,6alpha-dihydroxy-3,4-secolabd-14-en-3-oate (1), methyl ent-13-epi-8,13-epoxy-2,3-secolabd-14-en-2,11-olid-3-oate (2), and methyl ent-17-hydroxy-3,4-secokaura-4(19),15-dien-3-oate (3).  相似文献

Three new 4,5‐seco‐20(10→5)‐abeo‐abietane diterpenoids, 16‐hydroxysalvilenone ( 1 ), 15‐hydroxysalprionin ( 2 ), and 11β,15‐dihydroxysalprionin‐12‐one ( 3 ), and nine known abietane diterpenoids, 4 – 12 , along with one known sempervirane diterpenoid, hispidanol A ( 13 ), were isolated from the aerial parts of Isodon lophanthoides var. graciliflorus. The structures of compounds 1 – 3 were determined on the basis of spectroscopic methods including extensive analysis of NMR and mass spectroscopic data. All diterpenoids were tested for their TNF‐α inhibitory effects on LPS‐induced RAW264.7 cells. Compound 9 (16‐acetoxyhorminone) was the most potent with an IC50 value of 3.97±0.70 μm .  相似文献

Three new ent-kaurane diterpenoids, parvifoline Z (1), parvifoline AA (2), and parvifoline AB (3), together with 14 known compounds, were isolated from the leaves of Isodon parvifolius. The structures of the new compounds were elucidated by 1D- and 2D-NMR spectroscopy and mass spectrometry, and by comparison with known compounds. These three new diterpenoids included three types of ent-kauranoids, namely, C(20)-non-oxygenated-ent-kauranoid, 7,20-cyclo-ent-kauranoid and 6,7-seco-ent-kauranoid-7,20-olide. Compounds 1 and 2 exhibited significant cytotoxicities against A549, HT-29, and K562 cell lines.  相似文献

Ten new cassane-type diterpenoids were isolated from the seeds of Caesalpinia bonduc (L.) Roxb., including 6α-hydroxycaesalpinin P ( 1 ), 14-epi-caesalpinin E1 ( 2 ), 6-deacetylcaesalmin Z ( 3 ), 14-epi-caesalmin Z ( 4 ), caesalpinolides I ( 5 ), K ( 6 ), L ( 7 ), M ( 9 ) and N ( 10 ), and 14-epi-neocaesalpin L ( 8 ). Their planar structures and absolute configurations were fully determined by comprehensive spectroscopic methods, including 2D NMR and electronic circular dichroism spectra. Compounds 1 – 4 are tetracyclic cassane diterpenoids possessing a furan ring, and compounds 5 – 10 are tetracyclic cassane diterpenoids possessing a fused butenolide moiety. The anti-inflammatory and cytotoxic activities of the isolates were evaluated, while none of them showed obvious effects. The current study identified ten new cassane-type diterpenoids from the seeds of C. bonduc (L.) Roxb., which enriched the chemical diversity of the titled herbal medicine.  相似文献

Japodagrin (1) and japodagrone (2), two macrocylic diterpenoids possessing lathyrane and jatrophane skeletons, respectively, have been isolated from the root of Jatropha podagrica Hook. Four other diterpenoids (3-6) were also isolated from this plant. The structures of these compounds were elucidated on the basis of NMR and HRMS analysis, and by spectral comparisons. Four highly acylated diterpenoids, designated as pierisformotoxins A–D ( 1 – 4 , resp.), along with 26 known compounds, were isolated from the flowers of Pieris formosa. Among them, pierisformotoxins A and B ( 1 and 2 , resp.) were new highly acylated grayanane diterpenoids, of which the five‐membered ring A has undergone an oxidative cleavage between C(3) and C(4), followed by lactonization, to give rise to a five‐membered lactone ring between C(3) and C(5), differing from the previously reported grayanane diterpenoids with a 5/7/6/5 ring system. Results of the cAMP‐regulation‐activity assay showed that pierisformotoxin C ( 3 ) at 10 μM (inhibitory ratio (IR): 10.1%) or 2 μM (9.8%), and pierisformotoxin B ( 2 ) at 50 μM (13.9%) significantly decreased the cAMP level in N1E‐115 neuroblastoma cells (p<0.05).  相似文献

ent-Abietane diterpenoids, hebeiabinins A-F (1-5), together with seven known diterpenoids were isolated from leaves of Isodon rubescens var. rubescens. The structures of 1-5 were established on the basis of spectroscopic analyses, including application of 2D NMR spectroscopic techniques. The diterpenoids isolated were evaluated for the cytotoxicity against A549, HT-29, and K562 tumor cells. Compound 5 was the most active with IC(50) value of 0.91 microM against A549 cells.  相似文献

Ten new grayanane diterpenoids, craiobiotoxins I-VIII (1-8) and craiobiosides A (9) and B (10), and five known grayanane diterpenoids, grayanotoxin XVIII, lyoniol A, lyoniol B, pieristoxin H, and grayanoside B were isolated from the leaves of Craiobiodendron yunnanense (Ericaceae). The structures of 1-10 were elucidated by spectroscopic methods including various 2D NMR and X-ray crystal diffraction experiments. AimsCytochrome P450 (CYP) enzymes have been implicated in a large number of preventable drug–herb interactions. Andrographis paniculata Nees, a tropical herb widely used for various health conditions contains two major diterpenoids, andrographolide and 14-Deoxy-11, 12-Didehydroandrographolide. These compounds were evaluated systematically for their effects on CYP1A2, CYP2D6 and CYP3A4 expressions in HepG2 cells.Main methodsQuantitative RT-PCR and Western blot analysis were used to assess the mRNA and protein expression of the three CYPs. CYP3A4 enzyme activity was evaluated using P450-Glo? Assays. The LanthaScreen® TR-FRET PXR (SXR) Competitive Binding Assay was used to determine if the compounds are potential PXR-ligands.Key findingsBoth diterpenoids inhibited the mRNA and protein expressions of CYP1A2, CYP2D6, and CYP3A4. Interestingly, the lowest concentration of both diterpenoids produced a more than 50% reduction in the mRNA and protein expression of CYP3A4 and this reduction was consistent with the enzyme activity. Further experiments revealed that both diterpenoids were also capable of attenuating the ability of dexamethasone to induce CYP3A4 expression, and 14-Deoxy-11, 12-Didehydroandrographolide tended to bind to the PXR-LBD site in a concentration-dependent manner.SignificanceThese diterpenoids are potential CYP3A4 inhibitors and the effects on CYP3A4 may be clinically significant. 14-Deoxy-11, 12-Didehydroandrographolide inhibits CYP3A4 by binding and antagonizing PXR function.  相似文献
