通过三亲本杂交将携带豌豆根瘤菌吸氢基因的质粒pAL618转移到台湾毛豆根瘤菌292-C中,经抗性筛选、质粒检测和吸氢活性测定,得到能稳定遗传的结合株292-C2和292-C3。在自生条件下结合株均表现较高的吸氢活性,而受体株292-C不吸氢,在共生条件下结合株表现为高吸氢,而受体株表现为放氢。  相似文献

通过三亲本杂交将含有花生根瘤菌吸氢基因的质粒pZ 55(Tcr)转入不吸氢的花生根瘤菌Ra 34等菌株 (Hup- ,Nif+,Apr)中 ,筛选到既具有吸氢又具有固氮能力的花生根瘤菌结合株Rz34 2。在自生和共生条件下 ,结合株均可表达高吸氢和高固氮活性。以结合株Rz34 2接种的花生植株叶片的干重比不接种的、接种受体株Ra34(Hup- )和接种对照菌株L8 3(Hup+,Nif+)的分别高 6.2 %、7.6%和 6.3% ;种子的含氮量分别高 8 9%、1.00 %和 6.0 % ;产量分别高 188%、1 0 5%和 1 0 7%。研究结果表明 ,以含吸氢基因的结合株接种花生能提高根瘤菌与花生的共生固氮效率 ,增加作物的产量。  相似文献

紫云英根瘤菌109菌株完整细胞的吸氧活性,在空气气相中衰减快,加氢或降低温度,衰减延缓。完整细胞的吸氢反应受体,包括甲基蓝、氧、铁氰化钾、TTC、甲基紫精、Cyt C_3、硝酸钾、DCPIP和反丁烯二酸盐的表现不相同。当氧为受体时,气相氧浓度为5%,吸氢值最大,甲基蓝为受体时,吸氢活性高。另外,氧为受体的吸氢受到碳水化合物抑制,并与基础吸氧速率相关。紫云英根瘤菌的氢氧化与碳水化合物氧化竞争电子传递途径。  相似文献

以质粒pRK404为载体亚克隆含大豆根瘤菌吸氢酶结构基因(hup)的片段,构建成嵌合质粒pHR11、pHR4和pHR10。通过三亲本杂交将这些嵌合质粒导入无吸氢活性的Rhodobactersphaeroides241菌株(Hup-),均获得Hup+的接合子。利用启动子检测质粒pMP220证明,在hup对结构基因上游1.2kb内存在hup启动基因片段。以pRK2013为助质粒可将pHR11导入Enterobactercloacae和Klebsiellaoxytoca等土壤固氮菌株。本文以接合子E.cloacaeEH1113为例,通过对基因组DNASouthern杂交分析证明,嵌合质粒pHR11在EH1113中稳定贮存和复制。H2诱导接合子EH1113吸氢酶活性高表达,吸氢活性与放氢活性比值约为对照的两倍。当以延胡索酸为电子受体时,吸氢酶的吸氢作用支持菌株固氮酶活性的提高。  相似文献

应用含豌豆根瘤菌部分吸氢基因的探讨及PCR分析,从花生根瘤菌基因文库中筛选到含有吸氢基因的重组质粒pZ-55。用BamHⅠ、EcoRⅠ和KpnⅠ等内切酶对pZ-55进行酶切分析,构建了pZ-55的酶切物理图谱。经三亲本杂交分析,pZ-55能互补诱变株Ln-1(Hup^-),获得在自生条件下表达吸氢活性。  相似文献

核苷酸和烟酸的添加,使紫云英根瘤菌109氢酶吸氢活性表达增加。cAMP(1 mmol/L),烟酸(70 mmol/L)的存在,缓解了葡萄糖酸钠或果糖引起的吸氢活性阻遏,cAMP的解阻遏效应在年轻的菌体(48 h)表现较为明显。但以MB为受体的破碎细胞吸氢活性则未见增加,烟酸的促进效应受到氯霉素(40μg/ml)的抑制。其他核苷或核苷酸,如腺嘌呤,尿嘧啶,ATP,ADP,AMP,UMP,UTP都能促进吸氢活性的表达。诱导氢酶前,细胞ATP库已处于低水平,并保持稳定,添加琥珀酸盐后,ATP库水平提高,吸氢活性表达受抑。  相似文献

紫云英根瘤菌氢酶表达依赖于H_2并受碳底物和高O_2浓度的阻遏及cAMP的显著促进。整体细胞的吸氢活性对O_2不敏感,受碘乙酸(50mmol L~(-1))的强烈抑制。少数氧化还原电位为正值的人工电子受体可支持吸氢活性。与紫云英根瘤菌不同,巴西固氮螺菌氢酶表达并不依赖于H_2,受碳底物阻遏及cAMP促进的效应均不显著,而对O_2敏感。整体细胞吸氢活性受碘乙酸的抑制作用不明显。无论正、负值氧化还原电位人工电子受体均可支持吸氢活性。在经饥饿的静止细胞中,H_2可支持固氮活性并增强固氮酶对O_2的耐受能力。  相似文献

自生条件下,测定准噶尔盆地南部的68株根瘤菌吸氢活力,获得一株Hup~+的冬箭筈豌豆根瘤菌C_(48)。经与紫云英根瘤菌株109及89比较,两者氢酶表达的最适pH相同;温度分别以20或30℃为宜;Ni~(2+)显著促进吸氢表达,但C_(48)还受Co~(2+ )、Mg~(2+)、Cu~(2+)的促进;紫云英根瘤菌的氨酶表达受碳水化合物抑制较冬箭筈豌豆根瘤菌明显。此外,自生条件下生长的Hup—菌株,经与宿主共生后,Hup~+的百分率大为增加。  相似文献

检测了四株大豆根瘤菌在不同的大豆品种上形成根瘤的放氢、吸氢、固氮活性及豆血红蛋白的含量;同时测定了植株干物质的积累。结果表明,所有固氮根瘤都放氢,自生条件下Hup~-根瘤菌形成的根瘤仍不具吸氢活性,相对固氮率在0.75左右。而Hup~(?)菌株根瘤的相对固氮率在0.91~1之间。寄主植物对Hup~(?)菌株的吸氢活性有影响。相关分析表明,根瘤的豆血红蛋白与吸氢活性呈负相关。干物质积累与固氮酶活性关系最密切,氢酶活性的影响是次要的。  相似文献

以质粒pKT230为栽体,亚克隆大豆根瘤菌吸氢酶结构基因(hupSL)片段,构建成嵌合质粒pKH1。将质粒分配系统基因(parDE)片段和吸氢酶结构基因(hup)片段插入载体质粒pRK415,构建成质粒pRKBH。质粒pKH1、pRKBH和载体pRK415经转化和三亲本杂交,得到DH5α/pHR11、DH5α/pRKBH、E1201/pKH1、NG13/pKH1(NGH999)、NG1390/pRK415、NG1390/pHR11、NG1390/pRKBH和NG1390/pKH1(NGH982)等接合子。稳定性分析发现,质粒pKH1在催娩克氏杆茵中传80代后仍有92%以上的菌株含此质粒,说明质粒pKH1有较高的稳定性。吸氢酶活性分析表明,H  相似文献

HRu2Fe2PdC(CO)123-ß-C10H15) cluster was prepared in the reaction of (Et4N) [HFe2Ru2C(CO)12] with [Pd(η3-ß-C10H15)Cl]2. X-ray structural study of HRu2Fe2PdC(CO)123-ß-C10H15) (where ß-C10H15 is ß-pinenyl) revealed a wing-tip butterfly geometry of the metal core and (1R, 2S, 3S, 5R) absolute configuration for both crystallography independent molecules in the crystal. Chiroptical properties of this cluster are compared with other clusters containing a Pd(η3-ß-C10H15) fragment and discussed.  相似文献

探讨不同保鲜剂对非洲菊切花的瓶插寿命、观赏品质以及生理生化指标的影响。结果表明,在供试7组保鲜剂中,以处理7(T7):20g/L Suc+200mg/L 8-HQC+150mg/L CA+75 mg/L KH2PO4·3H2O+1g/L CaCl2的保鲜效果最佳,可显著延长非洲菊切花瓶插寿命;T7处理有利于减缓花瓣组织pH上升、花青素含量下降及花瓣细胞膜相对透性的上升,从而延长瓶插寿命。  相似文献

Occurrence and activity of the hydrogen uptake enzyme were studied in root nodule homogenates made from plants of Alnus incana (L.) Moench collected from field sites in the northern part of Sweden. Nitrogenase (EC 1.7.99.2) activity (estimated by acetylene reduction) and hydrogen evolution were studied in excised nodules. All Frankia sources showed acetylene reduction activity, and possessed a hydrogen uptake system. Hydrogen uptake in nodule homogenates from the Frankia sources measured at 23.8 μM H2 ranged from 0.04 to 5.0 μmol H2 (g fresh weight nodule)−1 h−1. The H2 uptake capacity of nodule homogenates from one of the Frankia sources was almost 8 times higher than the hydrogen evolution from nitrogenase, both expressed on a nodule fresh weight basis. Frankia sources from field sites 6 and 11 showed Km for H2 of 13.0 and 23.6 μM H2, respectively. This indicates similarities in the hydrogen uptake enzymes in the two Frankia sources. It is concluded that hydrogen uptake is a common characteristic in Frankia.  相似文献

Tantalaalkylidene compounds, CHR=TaCl3L2 (R=tBu or CMe2Ph, L=THF or 1/2dimethoxyethane) mixed with the cyclopalladated dimer [Pd(2-C6H4CH2NMe2)(μ-Cl)]2, 1, afford good yields of heterodimetallic complexes [Pd(2-C6H4CH2NMe2)(μ-Cl)(μ-CHR=TaCl3L], 3a, 3b, in which the Ta=C unit is η2-interacting with the palladium atom, while a chloride ligand is bridging the tantalum and the palladium atoms. These compounds are fairly stable in air in the solid state and also in solution at RT. The interaction of the Ta=C unit with Pd in these bimetallic compounds is weak as shown by the ready formation of [Pd(2-C6H4CH2NMe2)PyCl] and CHR=TaCl3Py2 upon treatement with pyridine. Compounds analogous to 3a, b can also be obtained with 12 electrons tantalum complexes. Thus treating the same cyclopalladated dimer 1 with CHR=Ta(OAr)3 (OAr=2,6-diisopropylphenyloxy) led to a much more stable though electron deficient species: [Pd(2-C6H4CH2NMe2)(μ-Cl)(μ-CHtBu=Ta(OAr)3], 3c. Substitution in 3a of one chloride ion by an alkyl group occurred at the tantalum metal via reaction with ZnR2 (R=---CH2CMe2Ph) leading to [Pd(2-C6H4CH2NMe2)(μ-Cl)(μ-CHtBu=TaCl3(CH2CMe2Ph)], 4 for which there is no free rotation around the new Ta---C bond and in which one of the methylene protons is strongly interacting with the palladium centre. This compound is believed to mimic an intermediate to the formation of tantalacarbyne derivative, which was obtained earlier via reaction of the uncomplexed tantalacarbene compound with dialkylzinc compounds.  相似文献

目的: 用心肺运动试验(CPET)定量评估化疗对人体整体心肺功能状态的影响。方法: 本研究选取2006年至2007年在MD Anderson癌症中心进行外科手术前化疗的6例食管癌患者。对该6例患者分别行化疗前后最大症状限制性CPET的客观定量评估。结果: ①新辅助化疗前后CPET核心指标的变化:显著降低的核心指标有峰值摄氧量(Peak VO2)(P<0.05),无氧阈(AT)、峰值氧脉搏(Peak VO2/HR)、摄氧通气效率峰值平台(OUEP)(P<0.01),核心指标显著上升的有VE/VCO2最小值、VE-vs-VCO2斜率(P<0.05)。2化疗前后CPET循环指标的变化:患者静息状态及热身状态的VO2无明显变化,AT (P<0.01)及Peak VO2 P<0.05)均有下降。除静息及极限状态的下降不显著外,热身状态的摄氧通气效率(OUE)(P<0.05)及OUE@AT (P<0.01)呈现显著下降。四个状态的VE/ VCO2均呈现上升趋势,但仅在热身状态的VE/ VCO2P<0.05)及VE/ VCO2@AT(P<0.01)有统计学意义。四个状态的VO2 /HR均呈下降趋势,除VO2/HR@AT(P<0.05)显著下降外,其他三个状态的变化尚无统计学差异。③化疗前后CPET通气指标的变化在整个运动过程中,除热身状态的PETCO2P<0.05)显著下降外,其余状态的PETCO2呈现下降趋势,但尚无统计学差异。呼吸频率、每分通气量几乎无变化。患者四个状态的潮气量、VO2/RF及VCO2/RF均呈现下降趋势,但均无统计学差异。四个状态的PETO2均呈现上升趋势,但无明显统计学差异。结论: 心肺运动试验可以从呼吸、循环、代谢等方面客观定量评估化疗前后机体功能状态的变化。  相似文献

花生根瘤菌类菌体含有吸H2酶,以O2或亚甲蓝为电子受体均表现吸H2活性。类菌体还原减氧化的吸收差示光谱在424、500、520、550、560、585及595 nm处呈现吸收峰,表明细胞色素c、b、和o参与H2的氧化过程。CN-明显影响520、550、560 nm处的吸收峰,意味着有些细胞色素c和b对CN-敏感,在H2代谢中充当末端氧化酶的作用。抗霉素A、HONO、CN-、N3-和DBMIB均强烈抑制吸H2活性,但鱼藤酮不抑制吸H2活性。说明细胞色素b、c、a和泛配参与H2氧化的电子传递,而NADH脱氢酶不参与。花生根瘤菌类菌体的H2氧化系统是个复杂具分支的电子传递体系。  相似文献

目的: 观察饱和氢气生理盐水对盲肠结扎穿孔(CLP)大鼠肺组织的作用。方法: 将24只健康雄性SD大鼠(体重250~300 g)随机分成4组(每组6只):①假手术对照组(Sham组)+生理盐水组:盲肠根部穿过丝线,不行结扎和穿孔,手术前10 min腹腔注射生理盐水(10 mg/kg);②CLP组+生理盐水组:结扎盲肠根部并用18号针头穿刺2个孔,2个孔相距约1cm,手术前10 min腹腔注射生理盐水(10 mg/kg);③Sham+H2组:手术前10 min腹腔注射饱和氢气生理盐水(10 mg/kg); ④CLP+ H2组:手术前腹腔注射饱和氢气生理盐水(10 mg/kg)。各组于手术后8 h进行观察:采用生物化学和RT-PCR的方法分别检测大鼠肺组织中CSE/H2S体系的变化。采用H2S供体硫氢化钠(NaHS)诱导的肺组织损伤动物模型,另取32只健康雄性SD大鼠(体重250~300 g),随机分为4组(每组8只):①生理盐水组:腹腔注射生理盐水(10 mg/kg);②H2S组:腹腔注射H2S供体NaHS(56 μmol/kg);③H2S+H2组:腹腔注射NaHS前10 min注射饱和氢气生理盐水(10 mg/kg);④H2组:腹腔注射生理盐水前10 min注射饱和氢气生理盐水(10 mg/kg)。于给药后8 h测定肺系数,检测肺组织中MDA含量、MPO活性及细胞因子TNF-α、IL-6和IL-10含量,观察肺组织形态学变化。结果: 饱和氢气生理盐水可抑制CLP大鼠肺组织中CSE/H2S体系:减少CLP大鼠肺组织中H2S的生成,抑制H2S的合成酶CSE的活性及mRNA表达 (P均<0.05);外源性给予H2S(NaHS)可造成肺组织损伤,饱和氢气生理盐水可明显减轻H2S所致的肺组织损伤:大鼠肺系数明显减小(P< 0.05),MDA含量下降(P<0.05);肺组织MPO活性下降(P<0.05);大鼠肺间质和肺泡中PMN的浸润程度明显减轻,肺组织形态及IQA接近正常(P<0.05)。结论: 饱和氢气生理盐水可通过抑制CLP大鼠肺组织中CSE/H2S体系,发挥其改善CLP大鼠肺组织损伤的作用。  相似文献

UV photolysis of Ru3(CO)12 physisorbed onto porous Vycor glass leads to the oxidative addition product (μ-H)Ru3(CO)10(μ-OSi). The latter reacts thermally with 1-pentene to form a stable adduct, HRu3(CO)10(OSi)(1-C5H10), and photolysis of the adduct results in isomerization of the alkene. HRu3(CO)10(OSi)(1-C5H10) + hv → (μ-H)Ru3(CO)10(μ-OSi) + 2-pentene As with other photoactivated hybrid systems, the cis-/trans-2-pentene product ratio changes during photolysis. Unlike the other systems, where light generates a thermal catalyst, the data gathered here indicate a photoassisted catalytic process in which photoactivation of HRu3(CO)10(OSi)(1-C5H10) leads to an excited state similar to a π-allyl complex.  相似文献

Five free-living Frankia strains isolated from Casuarina were investigated for occurrence of hydrogenase activity. Nitrogenase activity (acetylene reduction) and hydrogen evolution were also evaluated. Acetylene reduction was recorded in all Frankia strains. None of the Frankia strains had any hydrogenase activity when grown on nickel-depleted medium and they released hydrogen in atmospheric air. After addition of nickel to the medium, the Frankia strains were shown to possess an active hydrogenase, which resulted in hydrogen uptake but no hydrogen evolution. The hydrogenase activity in Frankia strain KB5 increased from zero to 3.86 μ mol H2 (mg protein)−1 h−1 after addition of up to 1.0 μ M Ni. It is likely that the hydrogenase activity could be enhanced even more as a response on further addition of Ni. It is indicated in this study that absence of hydrogenase activity in free-living Frankia isolated from Casuarina spp. is due to nickel deficiency. Frankia living in symbiosis with Casuarina spp. show hydrogenase activity. Therefore, the results also indicate that the hydrogenase to some extent is regulated by the host plant and/or that the host plant supplies the symbiotic microorganism with nickel. Moreover, the result shows that this Frankia is somewhat different from Frankia isolated from Alnus incana and Comptonia peregrina ., i.e., Frankia isolated from A. incana and C. peregrina showed a small hydrogen uptake activity even without addition of nickel.  相似文献

采用正交试验、单因子试验和植物组织培养方法,探讨几种因子对野葛块根组织脱分化与再分化的影响。结果表明,野葛块根愈伤组织诱导的最佳培养基为MS+NAA 0.5mg/L+6-BA 1.0mg/L+2,4-D 2.0mg/L,暗培养更有利于愈伤组织的诱导;野葛块根愈伤组织的最佳出芽培养基为MS+NAA 0.5mg/L+6-BA 3.0mg/L或MS+NAA 0.5mg/L+KT 2.0mg/L,光照培养更有利于愈伤组织芽的再分化;野葛块根愈伤组织再生芽生根最佳培养基为MS+NAA 0.5mg/L+PP333 3.0mg/L+蔗糖30g/L;PP333 3.0mg/L和蛭石:珍珠岩(2:1)基质能显著提高再生苗的移栽成活率。  相似文献
