紫云英根瘤菌氢酶表达依赖于H_2并受碳底物和高O_2浓度的阻遏及cAMP的显著促进。整体细胞的吸氢活性对O_2不敏感,受碘乙酸(50mmol L~(-1))的强烈抑制。少数氧化还原电位为正值的人工电子受体可支持吸氢活性。与紫云英根瘤菌不同,巴西固氮螺菌氢酶表达并不依赖于H_2,受碳底物阻遏及cAMP促进的效应均不显著,而对O_2敏感。整体细胞吸氢活性受碘乙酸的抑制作用不明显。无论正、负值氧化还原电位人工电子受体均可支持吸氢活性。在经饥饿的静止细胞中,H_2可支持固氮活性并增强固氮酶对O_2的耐受能力。  相似文献

异养生长的紫云英根瘤菌,其吸氢活性的表达在很大程度上取决于耗氧速率。30个诱导系统的统计和回归分析表明:菌体吸氢活性的表达与耗氧速率之间存在有负相关性。添加碳水化合物,可导致吸氢表达的抑制和推迟,同时引起细胞生长量和耗氧速率的增加。但抑制程度与细胞生长量之间并无明显相关性,而往往与耗氧速率增加相关,氢酶表达时间是在耗氧速率降低到较低水平的时刻。向指数期培养物中添加能量解联剂CCCP(carbonyl cyanide—m—chlorophenylhydrazone)和DNP(2,4—dinitrophenol),降低耗氧速率,氨酶表达可去阻抑。试验还表明:马铃薯汁对吸氢表达有明显的促进效果,可解除某些碳水化合物的阻抑,使某些表型为Hup~-的菌株转变为Hup~+。  相似文献

目的: 探究通过抑制磷酸二酯酶活性促进cAMP发酵合成的工艺方法。方法: 在7 L发酵罐上进行添加氨茶碱的发酵实验,通过对发酵主要参数、关键酶活性、能量代谢水平等进行分析,针对性提出了氨茶碱与柠檬酸盐协同作用促进cAMP合成的发酵工艺。结果: 与对照相比,添加5 mg/L氨茶碱批次的cAMP产量提高25.9%,副产物腺苷浓度减少41.6%,两批次中腺苷酸环化酶和琥珀腺苷酸脱氢酶活性无显著改变,而磷酸二酯酶和5'-核苷酸酶活性明显下降。能量代谢分析结果表明,两批次的胞内ATP/AMP相比于对照批次明显降低,而AMP水平却显著上升,ATP合成水平成为限制产物积累的主要因素。氨茶碱与柠檬酸盐协同添加的cAMP发酵工艺中,cAMP产量达到4.48 g/L,比单独添加柠檬酸钠和氨茶碱分别提高22.1%和13.8%,副产物腺苷浓度仅为0.98 g/L,分别降低51.7%和25.3%。结论: 氨茶碱抑制了磷酸二酯酶和5'-核苷酸酶活性,减少cAMP分解和副产物合成,显著提高cAMP产量,然而ATP合成水平成为产物积累的限制因素。氨茶碱与柠檬酸盐协同添加工艺将抑制磷酸二酯酶活性和提高能量代谢水平相结合,进一步促进了产物发酵合成。  相似文献

根瘤菌自生培养物的吸收分子氢系统,不但可以在自养或异养的固体培养基上表达,而且也可以在异养的液相培养基中表达。其表达受碳水化合物、氧和分子氢浓度诸环境因子调节。诱导氢酶须用指数后期的培养物进行;所有试验过的可利用的碳水化合物均可阻抑氢酶表达;而20m mol/L的KNO_3和NH_4Cl对氢酶表达也有阻抑作用。正交组合试验指出紫云英根瘤菌的高吸氢活性是在40%N_2,14%H_2,4%O_2以及4.2%CO_2的气相条件下表达。Ar取代H_2后,无吸氢活性;氢酶表达后,移去H_2,则表达停止。氧浓度稍高阻抑氢酶表达;氯霉素的阻抑表达效应与氧阻抑类似。  相似文献

对根癌土壤杆菌C58/pGV3850菌株的固氮生理特性研究结果表明,该菌具有自生固氮活性,其固氮活性的最适pH为8.0,温度为30℃.固氮活性在对数生长后期(14h)最高,延缓期和静止期乙炔还原活性较低.该菌株好氧,通过氧呼吸和吸氢酶的作用达到避氧固氮.当通入氧气超过呼吸耗氧时,对固氮活性产生抑制作用.在培养过程中增加NH_4~+浓度,固氮活性会降低,当达到15mmol/L时完全丧失固氮活性,表现NH_4~+阻遏固氮酶蛋白合成.在乙炔还原测定系统中加入NH_4~+并不影响乙炔还原反应,说明没有NH_4~+关闭现象.培养过程中加入MSX(2.5mg/ml)能解除10mmol/L NH_4~+对固氮酶合成的阻遏作用.固定的游离氮不能以NH_4~+形式分泌于胞外.固氮过程中放出大量氢气,培养16小时产氢量达65μmol H_2/mg蛋白.在限碳条件下(0.2%蔗糖)其吸氢酶活力可达520nmol H_2·ml~(-1)·h~(-1).  相似文献

ABA诱导型启动子(rd29A)重组到报告基因(GUS)的上游构建表达载体.通过农杆菌介导转化烟草,获得转基因植株.将转基因植株诱导愈伤组织,建立稳定、均一的转rd29A-GUS融合基因的悬浮培养细胞系.用ABA处理悬浮细胞24 h后GUS活性显著升高,说明外源ABA能够诱导rd29A启动子的表达,获得了用于ABA信号转导研究的实用细胞系.在ABA激活表达的细胞介质中加入尼克酰胺(cADPR合成酶的抑制剂)或U73122(PLC抑制剂)只能部分抑制ABA的效应,但如果加入蛋白激酶抑制剂K252a,抑制效果达95%以上.用可跨膜的cAMP的类似物8-Br-cAMP处理细胞发现,它能代替ABA的作用;当介质中加入1 mmol/L IBMX(磷酸二酯酶的抑制剂)增加cAMP的稳定性,发现低浓度的8-Br-cAMP与ABA相同的效应.以上结果表明cAMP参与了烟草悬浮细胞中ABA信号的传递.  相似文献

在固氮酶能表达的生长条件下,镍可促进浑球红假单胞菌菌株601的氢酶合成,而在含氨培养基上则没有影响。镍促进氢酶合成的最适浓度为10μmol/L,并具有金属专一性,其他二价金属离子对氢酶合成没有作用。镍在促进氢酶吸氢的同时,抑制菌体的放氢,但对固氮酶的乙炔还原活性则几乎没有影响。  相似文献

以虫荧光素酶法检验了四株丝状真菌在葡萄糖—无机盐液体培养过程中的胞内ATP含量。结果表明,只有当胞内ATP浓度低于10~(-S)mg/ml时,真菌才开始合成胞外纤维素酶(FPA)。以不同浓度的各种碳源培养时,菌体胞内ATP含量只要超过10~(-1)mg/ml,FPA的合成即发生阻遏。菌体胞内ATP含量与FPA合成呈显著负相关。以高效液相色谱(HPLC)法检测了菌体培养液中的cAMP含量。在非阻遏条件下,外源cAMP可以提高FPA的合成水平。但外源cAMP不能解除已经发生的酶合成阻遏。菌体ATP和cAMP水平是调节真菌纤维素酶合成的重要因子。  相似文献

在自生条件下,研究了根瘤菌的氢酶与固氮酶的共轭表达(coexpression)。氢酶表达受碳源限制和氧耗速率的调节,并为固氮酶表达条件促进,在固氮酶放氢的诱导下而与固氮酶共轭表达。此外,观察到外源氢支持根瘤菌自生固氮的活性,氢支持效应被葡萄糖酸钠和氧抑制。  相似文献

ABA诱导型启动子 (rd2 9A)重组到报告基因 (GUS)的上游构建表达载体。通过农杆菌介导转化烟草 ,获得转基因植株。将转基因植株诱导愈伤组织 ,建立稳定、均一的转rd2 9A_GUS融合基因的悬浮培养细胞系。用ABA处理悬浮细胞 2 4h后GUS活性显著升高 ,说明外源ABA能够诱导rd2 9A启动子的表达 ,获得了用于ABA信号转导研究的实用细胞系。在ABA激活表达的细胞介质中加入尼克酰胺 (cADPR合成酶的抑制剂 )或U73 12 2 (PLC抑制剂 )只能部分抑制ABA的效应 ,但如果加入蛋白激酶抑制剂K2 5 2a ,抑制效果达 95 %以上。用可跨膜的cAMP的类似物8_Br_cAMP处理细胞发现 ,它能代替ABA的作用 ;当介质中加入 1mmol/LIBMX(磷酸二酯酶的抑制剂 )增加cAMP的稳定性 ,发现低浓度的 8_Br_cAMP与ABA相同的效应。以上结果表明cAMP参与了烟草悬浮细胞中ABA信号的传递。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献

The review deals with study of enzymologic properties of a novel highly specific acetylcholinesterase substrate, N-(β-acetoxyethyl) piperidinium iodomethylate (“piperidylcholine”), and its 30 derivatives that were tested as effectors of cholinesterases of mammals and various species of Pacific squids. It was proven for the first time that responsible for specificity of action was structure of cyclic ammonium grouping of the alcohol part of molecule of the ester substrate. Analysis of specificity is performed based on enzymatic hydrolysis parameters—activity of catalytic center of cholinesterases and bimolecular constant of the reaction rate that are determined at optimal and low substrate concentrations. Among the specially synthesized group of thioester compounds there is revealed one more highly specific acetylcholinesterase substrate—N-(β-acetoxyethyl) piperidinium.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献
