利用蚊虫连续石蜡切片免疫组织化学技术;对登革Ⅱ型病毒(DEN-2)感染白纹伊蚊Aedes albopictus后的散播时间、程度及组织器官的感染顺序进行监测;以了解DEN-2在媒介白纹伊蚊体内的分布规律。结果表明:大剂量感染登革Ⅱ型病毒后;在蚊虫消化道的主要部位以及大多数组织器官包括神经及内分泌系统在内;如涎腺、脑、神经节等亦检测到病毒抗原。登革Ⅱ型病毒一旦感染并逸出中肠会迅速侵染其它组织。从各组织感染率的高低推断;病毒逸出中肠后通过血淋巴传播到其它组织的顺序通常为:前肠、涎腺、咽部神经节、脑及食管下神经节、后肠及复眼的小眼等。 相似文献

采用C6/36细胞培养分离病毒的方法检测感染登革Ⅱ型病毒的白纹伊蚊Aedes albopictus滞育卵孵化的F1代蚊虫感染率,从第一个生殖营养周期子代蚊虫中未分离到病毒,第二与第三生殖营养周期子代蚊虫最低感染率没有显著性差异(χ2=0.01,P>0.0 5),感染子代的批阳性率为9.1%,最低感染率为1∶330;间接免疫荧光检测结果表明感染登革Ⅱ型病毒的白纹伊蚊滞育卵孵化的子代成蚊能通过叮咬将登革病毒传播给敏感乳鼠。这些研究结果表明登革病毒能在媒介滞育卵内存活并传至子代,子代蚊虫能通过叮咬敏感宿主水平传播病毒。 相似文献

对两个地理品系的白纹伊蚊卵期滞育的实验观察表明:白纹伊蚊宜兴品系(约北纬31°N)不仅亲代受短光照作用后可诱导卵期发生滞育,而且卵期仍具有光照敏感性;海南岛品系(约北纬18°N)中,光期变化不能诱导卵期滞育.宜兴品系卵滞育率随亲代光期缩短而增高的现象只发生在一定范围内(约L:D=9:15到L:D=15:9).卵滞育还受卵期低温、干燥以及孵化时水中溶解氧的影响.卵进入深度滞育是多种环境因素作用的结果. 相似文献

以Balb/C小鼠为实验动物,探讨了埃及伊蚊Aedes aegypti唾液对登革病毒感染宿主的影响。结果显示,在皮下接种剂量相同的情况下,如果Balb/C实验小鼠预先被一定数量的埃及伊蚊叮咬以后,实验小鼠感染病毒的程度有所提高,血清中的抗体滴度明显降低,腹腔巨噬细胞感染登革病毒的阳性率及感染的时间动态曲线也有明显差异,感染高峰期延迟。这些说明实验小鼠被媒介蚊虫叮咬以后变得相对容易被登革病毒感染,可能是媒介蚊虫叮咬小鼠时分泌的唾液对宿主的免疫反应系统有一定作用。因此可以初步肯定埃及伊蚊在登革病毒的感染传播过程中,影响了Balb/C实验小鼠的免疫功能,对登革病毒的感染有一定推动作用。 相似文献

从贵州省独山县麻尾镇白纹伊蚊标本中分离到一株病毒,用抗DEN 14型单克隆抗体经间接免疫荧光法和RTPCR产物酶切分型鉴定为DEN2,并对其 NS1和E基因RTPCR产物进行部分基因序列测定,与DEN2 NGC株比较,麻尾株的NS1基因区有7个碱基发生点突变,E基因区有1个碱基的插入,两个序列被GenBank收录,编号分别为AY277402、AY278226;麻尾分离株与其他9株DEN2型病毒进行系统发育关系的分析,结果显示与D243株系统进化关系最近;证明贵州省存在DEN感染的自然循环。 相似文献

Invasive Aedes mosquito species (Diptera: Culicidae) are of public health concern in Europe because they are either recognized or potential vectors of pathogens. Loop‐mediated isothermal amplification (LAMP) is a rapid and simple method for amplifying DNA with high specificity and efficiency, with the technique having potential for application in the field, including in high‐throughput format. Specific LAMP assays based on rDNA internal transcribed spacers 1 or 2 sequences, considering intraspecies variability at these loci, were developed for Aedes aegypti, Aedes albopictus, Aedes japonicus, Aedes koreicus and the indigenous Aedes geniculatus. No such assays could be developed for Aedes atropalpus and Aedes triseriatus because both loci were too short to serve as target. The assays rely on the clearly visible colour change from violet to sky blue after successful amplification. Sensitivity of egg detection was confirmed with ratios of up to one mosquito egg in 99 other eggs. Simple sample preparation of adults or eggs by mechanical homogenization in water required an additional heat treatment or centrifugation step to avoid non‐specific colour changes. Thus, further technical improvements are needed to render these assays truly field‐applicable, which would greatly facilitate surveillance of these invasive mosquito species and allow for prompt implementation of control measures. 相似文献

Aedes-transmitted arboviruses have spread globally due to the spread of Aedes aegypti and Aedes albopictus. Its distribution is associated with human and physical geography. However, these factors have not been quantified in Cameroon. Therefore, the aim was to develop an Ae. albopictus geo-referenced database to examine the risk factors associated with the vector distribution in Cameroon. Data on the Ae. albopictus presence and absence were collated and mapped from studies in published scientific literature between 2000 and 2020. Publicly available earth observation data were used to assess human geography, land use and climate risk factors related to the vector distribution. A logistic binomial regression was conducted to identify the significant risk factors associated with Ae. albopictus distribution. In total, 111 data points were collated (presence = 87; absence = 24). Different data collection methods and sites hindered the spatiotemporal analysis. An increase of one wet month in a year increased the odds of Ae. albopictus presence by 5.6 times. One unit of peri-urban area increased the odds by 1.3 times. Using publicly available demographic and environmental data to better understand the key determinants of mosquito distributions may facilitate appropriately targeted public health messages and vector control strategies. 相似文献

A significant increase in the number of reported cases of La Crosse virus (LACV) infections in eastern Tennessee has occurred in the last ten years. The objective of this study was to determine the abundance and habitat preferences of the potential vectors of LACV in this region. Adult host‐seeking mosquitoes were collected using CO2‐baited CDC light traps and a series of human‐landing catches in eastern Tennessee from 2004 to 2006. A total of 4,200 female mosquitoes of 23 species was collected by CO2‐baited CDC trapping at ten sites during the study period. Aedes albopictus (Skuse) was the most abundant mosquito collected at all sites and vegetation types, with the ratios of total Ae. albopictus to Ae. triseriatus (Say) females collected being 2.1:1 in 2004, 3.8:1 in 2005, and 4.9:1 in 2006. Ten species were collected during a series of human‐landing catches made at four different sites; one probable and three confirmed case sites of LACV infections, totaling 528 female mosquitoes. Aedes albopictus was the most abundant species collected, with a 4:1 ratio of Ae. albopictus to Ae. triseriatus females. Aedes albopictus exhibited two clear peaks of “landing” activity, one in the early morning and one in the late afternoon or early evening. Simple and multiple regression analyses of the predictors of the number of mosquitoes collected showed that populations of Ae. albopictus were three times more likely to be collected overall than Ae. triseriatus. Species (Ae. albopictus), vegetation (residential), and the previous cumulative precipitation for the four weeks prior to collection were significantly (P < 0.05) associated with the number of mosquitoes collected by CO2‐baited CDC trapping. In Port‐au‐Prince, Haiti, the status of insecticide resistance has not recently been evaluated for Aedes aegypti (L) and Aedes albopictus (Skuse) populations. No prophylactics exist for dengue, so prevention is only through vector control methods. An earthquake occurred in Haiti on January 12, 2010, with a magnitude of 7.0 Mw that devastated the area. Dengue became a major concern for the humanitarian relief workers that entered the country. Bottle bioassays were conducted in the field on adult mosquitoes reared from larvae collected from the grounds of the U.S. Embassy and from an adjacent neighborhood in eastern Port‐au‐Prince, Haiti. At the CDC, Fort Collins, CO, bioassays, molecular, and biochemical assays were performed on mosquitoes reared from field‐collected eggs. A small percentage of the population was able to survive the diagnostic dose in bioassays run in Haiti. Mosquitoes tested at the CDC demonstrated no phenotypic resistance. A variety of factors could be responsible for the discrepancies between the field and lab data, but temperature and larval nutrition are probably most important. Knowledge of localized resistance and underlying mechanisms helps in making rational decisions in selection of appropriate and effective insecticides in the event of a dengue outbreak. 相似文献

Several studies have examined how climatic variables such as temperature and precipitation may affect life history traits in mosquitoes that are important to disease transmission. Despite its importance as a seasonal cue in nature, studies investigating the influence of photoperiod on such traits are relatively few. This study aims to investigate how photoperiod alters life history traits, survival, and blood‐feeding activity in Aedes albopictus (Skuse) and Aedes aegypti (Linnaeus). We performed three experiments that tested the effects of day length on female survival, development time, adult size, fecundity, adult life span, and propensity to blood feed in Ae. albopictus and Ae. aegypti. Each experiment had three photoperiod treatments: 1) short‐day (10L:14D), 2) control (12L:12D), and 3) long‐day (14L:10D). Aedes albopictus adult females were consistently larger in size when reared in short‐day conditions. Aedes aegypti adult females from short‐day treatments lived longer and were more likely to take a blood meal compared to other treatments. We discuss how species‐specific responses may reflect alternative strategies evolved to increase survival during unfavorable conditions. We review the potential impacts of these responses on seasonal transmission patterns, such as potentially increasing vectorial capacity of Ae. aegypti during periods of shorter day lengths. 相似文献

A survey was conducted to evaluate the number of tree-hole breeding mosquito species and their distribution in the six principal woodland types in Israel. Out of approximately 3,000 mature trees examined, only 38 contained holes that retained water for extended periods of time, and breeding mosquitoes were observed in 27 of them. Two specialized tree-hole breeders, Aedes pulchritarsis Rondani and Aedes geniculatus Oliver, were found breeding at several sites in northern Israel, always at locations 500 m above sea level (a.s.l) and with high annual precipitation. Aedes albopictus Skuse which, in Israel, is known as an opportunistic container breeder, was found in this study to have adapted remarkably well to breeding in tree holes and was found in most forest types investigated and in most tree species which had adequate tree holes. Two other species, Culiseta annulata Schrank and Culex pipiens Linnaeus instars, were found in one of the tree holes, but did not survive to reach maturity. 相似文献
