Endophytic fungi from Nyctanthes arbor-tristis were isolated and evaluated for their antimicrobial activity. A total of 19 endophytic fungi were isolated from 400 segments of healthy leaf and stem tissues of N. arbor-tristis. Eighteen endophytic fungi were obtained from leaf, while only ten from stem. Alternaria alternata had the highest colonization frequency (15.0%) in leaf, whereas Cladosporium cladosporioides ranked first in stem with a colonization frequency of 12%. The diversity and species richness were found higher in leaf tissues than in stem. The similarity indices between leaf and stem were 0.473 for Jaccard’s and 0.642 for the Sorenson index, respectively. Of 16, 12 (75%) endophytic fungal extracts showed antibacterial activity against either one or more pathogenic bacteria. The endophytic Nigrospora oryzae showed maximum inhibition against Shigella sp. and Pseudomonas aeruginosa. The leaf endophytes Colletotrichum dematium and Chaetomium globosum exhibited a broad range of anibacterial activity and were active against Shigella flexnii, Shigella boydii, Salmonella enteritidis, Salmonella paratyphi, and P. aeruginosa. Nine out of 16 (56.25%) endophytic fungi exhibited antifungal activity to one or more fungal pathogens. Colletotrichum dematium inhibited 55.87% of the radial growth of the phytopathogen Curvularia lunata. The antimicrobial activity of these endophytic microorganisms could be exploited in the biotechnological, medicinal, and agricultural industries.  相似文献

Fungal endophytes were isolated from surface sterilised leaf segments of five medicinal plants collected from Mandi district, Himachal Pradesh, India. A total of 373 fungal strains belonging to 15 fungal genera and 18 species, Aspergillus niger, A. flavus, A. clavatus, A. variecolor, Penicillium chrsogenum, Alternaria alternata, Curvularia lunata, Haplosporium sp., Phoma sp., Nigrospora sp., Colletotrichum sp., Cladosporium sp., Stemphylium sp., Fusarium sp., Geotrichum sp., Phomopsis sp., Trichoderma sp., Rhizopus sp. and some sterile mycelium were isolated from all the plants. The relative frequency, isolation rate and colonisation rates of endophytes were used to study the endophytic diversity. The results showed that the highest colonisation rate (93.05%) was observed in Adhathoda vasica, while it was 91.66% in Ocimum sanctum, 85% in Viola odorata, 82.81% Cannabis sativa and lowest (61.11%) in Withania somnifera. Moreover, reading the richness and diversity of the endophytic fungi, the highest was obtained for O. sanctum, W. somnifera and C. sativa having eight species each, while lowest (6 and 4) was obtained from A. vasica and V. odorata, respectively. As the role of endophytic organisms in defensive mechanisms of plants is now well established, the present study is an important step to find new and interesting endophytes among the medicinal plants.  相似文献

Seed mycoflora associated with wheat was studied on different media with a particular reference to Blotter and potato dextrose agar (PDA) procedures of ISTA. Seed-borne fungi, viz. Fusarium moniliforme, Rhizopus spp., Mucor spp., Alternaria alternata, Aspergillus niger, Aspergillus flavus, Curvularia lunata, Drechslera spp, Alternaria spp. and Penicillium spp., were isolated from the variety HD264. Blotter method was found to be the best media for the isolation of mycoflora whether borne externally or internally. Total number and frequency of occurrence of fungi were recorded. The effect of seed treatment with different chemicals and eco-friendly botanicals was analysed on germination, and growth, better percentage of seed germination and reduction in fungal pathogen were due to biochemical seed treatment.  相似文献

A total of 1,151 endophytic fungal isolates representing 29 taxa were isolated from symptom-less, surface-sterilized segments of stem, leaf, petiole, and root of Tinospora cordifolia which had been collected at three locations differing in air pollution in India (Ramnagar, Banaras Hindu University, Maruadih) during three seasons (summer, monsoon, winter). Endophytes were most abundant in leaf tissues (29.38% of all isolates), followed by stem (18.16%), petiole (10.11%), and root segments (6.27%). The frequency of colonization (CF) varied more strongly among tissue type and season than location. CF was maximal during monsoon followed by winter and minimal during summer. A species each of Guignardia and Acremonium could only be isolated from leaves, whereas all other species occurred in at least two tissue types. Penicillium spp. were dominant (12.62% of all isolates), followed by Colletotrichum spp. (11.8%), Cladosporium spp. (8.9%), Chaetomium globosum (8.1%), Curvularia spp. (7.6%), and Alternaria alternata (6.8%). Species richness, evenness, and the Shannon-Wiener diversity index followed the same pattern as the CF with the tissue type and the season having the greatest effect on these indices, suggesting that tissue type and season are more influential than geography. Dissimilarity of endophyte communities in regards to species composition was highest among seasons. Colletotrichum linicola occurred almost exclusively in winter, Fusarium oxysporum only in winter and summer but never during monsoon and Curvularia lunata only in winter and during monsoon but never in summer. Emissions of NO(2), SO(2), and suspended particulate matter were negatively correlated with the CF. Ozone did not have any effect. The frequency of most species declined with increasing pollution, but some showed an opposite trend (e.g., Aspergillus flavus). Five unnamed taxa (sterile mycelia) were identified as Aspergillus tubingensis, Colletotrichum crassipes, Botryosphaeria rhodina, Aspergillus sydowii, and Pseudofusicoccum violaceum, using molecular tools. Fifteen of the 29 endophyte taxa exhibited antibacterial activity. B. rhodina (JQ031157) and C. globosum showed activity against all bacterial human pathogens tested, with the former showing higher activity than the latter.  相似文献

Dendrobium is the largest genus of tropical epiphytic orchid, some of which are traditional Chinese medicinal plants. The therapeutic components varied significantly among species. Endophytic microbes (fungi) hidden in medicinal plants may play an important effect on the overall quality of herb. Investigation of fungal composition in host plants is the first step toward elucidating the relationship endophyte-therapeutic content of herbal medicine. In this study, 401 culturable fungal endophytes were isolated and identified from 10 species of medicinal Dendrobium based on morphological and molecular techniques. The results showed that endophytic fungi from Dendrobium plants exhibited high biodiversity (37 genera, about 80 species). Acremonium, Alternaria, Ampelomyces, Bionectria, Cladosporium, Colletotrichum, Fusarium, Verticillium and Xylaria were the dominant fungal endophytes. Tropical epiphytic orchids appear to vary in degree of host specificity in their endophytic fungi.  相似文献

Endophytic fungi were isolated from healthy tissues of Lippia sidoides Cham. (Verbenaceae), a medicinal plant used as an antiseptic in the northeast of Brazil. From 480 fragments of leaves (240) and stems (240), a total of 203 endophytic fungi were isolated, representing 14 species belonging to the groups Ascomycota, Coelomycetes and Hyphomycetes. Endophytic colonization was greater in leaves (50.4%) than in stems (35.4%). Colletotrichum gloeosporiodes had the maximum colonization frequency (12.3%), followed by Alternaria alternata (7.08%), Guignardia bidwelli (6.87%) and Phomopsis archeri (5.41%). Some species showed specificity for the host tissue: Curvularia pallescens, Dreschlera dematioidea, G. bidwellii, Microascus desmosporum, Peacilomyces variotti, Periconia byssoides and Ulocladium oudemansii were only isolated from leaves while Fusarium lateritium and Phoma tracheiphila were only isolated from stems. Through a preliminary screening and fermentation assay, 16 isolates where found to produce antimicrobial metabolites against bacterial and fungi. The diversity and role of endophytes in medicinal plants is briefly discussed. In conclusion, endophytic fungi from L. sidoides have pharmaceutical potential and can be seen as an attractive source of biologically active compounds.  相似文献

制备了欧美107杨树枝条和叶的乙醇粗提物及不同极性溶剂的萃取部分,并测定了它们对植物病原真菌的抑制活性。枝条和叶的乙醇粗提物对棉花枯萎病菌、小麦纹枯病菌、番茄早疫病菌、番茄枯萎病菌、黄瓜枯萎病菌、小麦赤霉以及玉米弯孢等7种植物病原真菌均具有一定的抑制作用。而枝条的乙醇粗提物对杨树溃疡病菌的菌丝生长有一定的促进作用。抗菌活性成分主要集中在乙酸乙酯萃取部分。  相似文献

The isoquinoline alkaloids hunnemanine and norsanguinarine have been isolated from methanolic extract of the whole plant of Eschscholtzia californica. These two alkaloids were checked for their antifungal activity against phytopathogenic fungi Alternaria melongenae, A. brassicola, A. brassicae, Curvularia lunata, C. maculans, Helminthosporium pennisetti, H. oryzae, H. turcicum, Fusarium undum and F. lini. Hunnemanine exhibited 100 % inhibition of spore germination of A. brassicae, H. pennisetti and F. lini at 1000 ppm whereas norsanguinarine exhibited 100 % inhibition of A. brassicicola and C. maculans at this concentration.  相似文献

The effects of biotic stresses on the contents of benzoxazinones (Bxs) were investigated in maize leaves. When the causal agent of southern corn leaf blight, Bipolaris maydis, was inoculated on the third leaf, the amount of 2-hydroxy-4,7-dimethoxy-1,4-benzoxazin-3-one glucoside (HDMBOA-Glc) increased, reaching a maximum level 48 h after inoculation. The inoculation of weakly pathogenic Curvularia lunata and non-pathogenic Alternaria alternata also resulted in accumulation of HDMBOA-Glc, and filtrates of the cultures of B. maydis, C. lunata and A. alternata also showed the accumulation of elicitor-active compounds by the fungi. Furthermore the infection of B. maydis induced formation of dark brown lesions, where most abundant Bx-related compound was 6-methoxy-2-benzoxazolinone (MBOA). The later is formed by degradation of DIMBOA and HDMBOA, whereas HDMBOA-Glc was most abundant in the surrounding green tissues. Among the Bx-related compounds, MBOA exhibited the strongest inhibition of the germination of the conidia and of the growth of germ tubes of B. maydis, C. lunata and A. alternata. In addition to fungal infection, the feeding by rice armyworm larvae resulted in HDMBOA-Glc accumulation. These findings are discussed in relation to the possible ecological relevance of the conversion of DIMBOA-Glc into HDMBOA-Glc.  相似文献

为开发利用刺五加内生真菌资源,寻找新型抗菌物质,从刺五加根茎叶中分离纯化得到内生真菌71株,经菌落形态和显微形态去重后进行ITS序列分子鉴定为18个种,来自于4目、8科、11属,其中链格孢属与镰孢霉属为优势属。根、茎、叶中内生真菌多样性指数分别为1. 112 31、1. 523 658、0. 264 1。18种刺五加内生真菌中,16种内生真菌至少对一种指示菌有抗菌活性。CWJ-5(镰孢霉属Fusarium oxysporum)对金黄色葡萄球菌抑菌效果显著,抑菌直径27. 17±0. 07 mm; CWJ-6(链格孢属Alternaria alternata)对大肠埃希菌抑菌效果显著,抑菌直径18. 07±0. 01 mm。结果表明刺五加内生真菌资源丰富,有很大的开发利用空间,部分菌种抑菌活性显著,值得进一步研究。  相似文献

A systemic study of fungal endophytes associated with different plant parts of Cannabis sativa and their antifungal activity was investigated in the present study. A total of 281 plant segments, including 91 leaves, 93 stem and 97 petioles samples, were screened for the isolation of endophytic fungi. Totally, 212 (77.65%) segments were found colonised by different fungi. Highest colonisation frequency were observed in stem parts (84.94%), then leaves (82.41%) and lowest 59.79% in petiole. Total eight fungal genera belonging to 12 species were isolated. Aspergillus is recorded as the most frequently occurring genera with three species Aspergillus niger, Aspergillus flavus and Aspergillus nidulans followed by Penicillium with two species Penicillium chrysogenum and Penicillium citrinum, while Phoma, Rhizopus, Colletotrichum, Cladosporium and Curvularia with single species. The antifungal potential of A. niger and A. flavus – two most frequently isolated endophytic fungi – was evaluated against two common plant pathogen, Colletotrichum gloeosporioides and Curvularia lunata. Different plant and fungal extracts individually and in combinations showed variations in antifungal activity against both the pathogens. The primary results obtained on antifungal activity of endophytes show their possible role in plant defence mechanism but it is a preliminary approach and more extensive research is still required.  相似文献

Antimicrobial properties of some new carbonyl stabilized sulphonium ylides have been tested against two bacteria Staphylococcus aureus, Escherichia coli and two fungi Alternaria alternata and Curvularia lunata. Eucalyptus citriodora Hook, is frequently cultivated tree in India for its wood and medicinal usages. The endophytic and epiphytic fungi were estimated from healthy leaves of E. citriodora growing in the premise of Banaras Hindu University, Varanasi, India. A total of 33 fungal species were isolated from leaf segments. Of 33 taxa, 20 were recorded as endophytes, while 22 as epiphytes. Nine, out of 33 species were found to be common in leaf tissues and surfaces (Alternaria alternata, Aspergillus fumigatus, A. terreus, Cladosporium cladosporioides, Drechslera rostrata, Humicola grisea, Nigrospora oryzae, Penicillium cristata, and Pestalotia sp.). Out of 478 fungal isolates, 279 were epiphytic while only 199 were endophytic. Most isolates were anamorphic filamentous fungi which often don’t produce sexual spores. The Sorensen’s index of similarity between endophytes and epiphytes (leaf surface colonizers) was found to be at 0.300. Diversity index of fungal species was higher in endophytes than epiphytes. The frequency of colonization differs greatly in both myco-populations. Cladosporium cladosporioides (26%) was dominant species on leaf surfaces while Botrytis cinerea (10.5%) was dominant in leaf tissues. Out of 16 endophytic isolates evaluated for antagonistic test, 8 (50%) gave the antagonistic activity against variety of fungi representing pathogens to both humans and plants.  相似文献

AIMS: The aim of the study was to isolate and characterize the endophytic fungi from the rhizomes of the Chinese traditional medicinal plant Dioscorea zingiberensis and to detect their antibacterial activities. METHODS AND RESULTS: After strict sterile sample preparation, nine fungal endophytes were isolated from rhizomes of the Chinese traditional medicinal plant D. zingiberensis. The endophytes were classified by morphological traits and internal transcribed spacer (ITS) rRNA gene sequence analysis. Their ITS rDNA sequences were 99-100% identical to Nectria, Fusarium, Rhizopycnis, Acremonium and Penicillium spp. respectively. Of these, the most frequent genera were Fusarium and Nectria. One isolate, Dzf7, was unclassified on the basis of its low sequence similarity. The next closest species was Alternaria longissima (c. 92.4% sequence similarity). Endophyte isolate Dzf5 showed the closest sequence similarity (c. 99.5%) to an uncultured soil fungus (DQ420800) obtained from Cedar Creek, USA. Bioassays using a modified broth dilution test were used to detect the antibacterial activity of n-butanol extracts of both mycelia and culture filtrates of D. zingiberensis showed biological activity against Bacillus subtilis, Staphylococcus haemolyticus, Escherichia coli and Xanthomonas vesicatoria. Minimal inhibitory concentration (MIC) values of the extracts were between 31 x 25 microg ml(-1) and 125 microg ml(-1). CONCLUSIONS: Endophytic fungus Dzf2 (c. 99 x 8% sequence similarity to Fusarium redolens) isolated from D. zingiberensis rhizome showed the most potent antibacterial activities. SIGNIFICANCE AND IMPACT OF THE STUDY: Endophytic fungi isolated from D. zingiberensis may be used as potential producers of antibacterial natural products.  相似文献

Herbaceous plants endemic to native Kenyan grassland on the Marula Estate were assessed for their phylloplane fungal occurrence using leaf cultures on tap water agar. From 26 sampled plants, 261 fungal isolates representative of 58 genera and 92 taxa were identified. The principal filamentous fungi were of the genera: Alternaria, Bipolaris, Curvularia, Epicoccum, Fusarium, Nigrospora, Periconia, Phoma and Pithomyces. The fungal species that occurred most frequently were: Alternaria alternata aggreate (73%), Pithomyces chartarum (73%) Nigrospora sphaerica (42%), Torula herbarum (30%) and Microsphaeropsis olivacea (27%). Fungi of limited occurrence were: Didymosphaeria oblitescens, Doliomyces mysoriensis, Endophragmiella dimorphospora, Parapericonia angusii, Podospora minicaudata, Spegazzinia tessarthra, Sporormiella minima, Stagonospora vitensis, Stigmella effigurata and Truncatella conorum-piceae. It is postulated that the phylloplane of these endemic plants provides a niche for endemic fungi. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

采用组织分离法从秦岭药用植物杜仲茎部共分离到内生真菌38株,经形态学鉴定分属于9属,其中青霉属为杜仲内生真菌中的优势菌群,拟青霉属、交链孢属和无孢群属为常见属。对分离得到的杜仲内生真菌发酵液进行了抑菌实验,其中61%的菌株对测试细菌具有抗菌活性,表明杜仲内生真菌的抗菌活性十分普遍;34%的菌株对烟草赤星病菌、苹果炭疽病菌及辣椒炭疽病菌等多种植物病原菌具有较强的抑制活性,表明杜仲内生真菌在植物病害的生物防治领域具有进一步开发的价值。  相似文献

A survey of microorganisms of Striga hermonthica Del. Benth., a root parasite of graminaceous crops, was conducted in northern Ghana in 1992. Thirteen fungal species were isolated from infected S. hermonthica plants. Fusarium spp. were the most prevalent, and were isolated from more than 90% of the S. hermonthica samples collected. Other fungi isolated were Alternaria alternata, Bipolaris specifera, Cladosporium oxysporum, Curvularia falax, Macrophomina phaseolina, Nodulisporium gregarium, Phoma sorghina and Sclerotium rolfsii.  相似文献

The objective of this study was to isolate and identify fungal species found in natural association with adults of Musca domestica. The adult insects were collected from two natural breeding grounds: hog pens and an urban sanitary landfill. The isolated fungi were identified as: Aspergillus flavus (23.8%), A. niger var. niger (14.4%), Penicillium corylophilum (21.4%), P. fellutanum (11.9%), Cladosporium cladosporoides (4.7%), Fusarium sp. (4.7%), Alternaria alternata (11.9%), Curvularia brachyspora (2.4%), Mycelia sterilia (2.4%) and the Mucorales order (2.4%).  相似文献

从云南西双版纳采集7份灯台树样品,经过表面消毒,用4种分离培养基分离得到105株内生放线菌。经16S rRNA基因序列分析鉴定,分布于7个科9个属。利用5种植物病原真菌指示菌对所有菌株的发酵液进行抗菌活性检测。结果显示,有12.4%、14.3%、11.4%、12.4%、8.6%的菌株分别对镰刀霉、疫霉、赤星霉、苹果炭疽、白色念珠菌有抗性。对3株具有广谱抗菌活性的菌株进行再次发酵和抗菌活性复筛,结果显示这3株的抗菌活性稳定,并可能含有生物碱类化合物。  相似文献

从药用植物金荞麦和苦荞麦的根、茎、叶、花中分离到62株内生真菌,并以金黄色葡萄球菌(Staphylococcus aureus)、大肠杆菌(Escherichia coli)、枯草芽孢杆菌(Bacillus subtilis)[CMCC(B)63501]、小麦赤霉病菌(Fusarium graminearum)、黄瓜枯萎病菌(Fusarium oxysporum f.sp.cu-cumerinum)和绵腐病菌(Pythium aphanidermatum)6种微生物为指示菌对分离到的内生真菌进行抗菌活性检测。结果发现,分离的内生真菌菌株KQH-01、KQH-02和JQY-1的发酵醇提取物具有较好的抑菌活性。形态学特征和分子鉴定确定菌株KQH-01为炭角菌属(Xylaria sp.)真菌,菌株KQH-02为球毛壳菌(Chaetomium globosum),菌株JQY-1为葡萄座腔菌(Botryosphaeria dothidea)。  相似文献
