不同基因型对南阳牛生长发育性状的影响   总被引:2,自引:0,他引:2  
以南阳牛为实验动物,利用PCR-SSCP和PCR-RFLP技术研究了南阳牛生长激素(GH)基因,IGF-I基因以及IGF-IBP3基因的遗传多态,并分析了遗传多态位点对南阳牛0月龄、6月龄、12月龄、18月龄、24月龄以及36月龄不同生长时期生长发育性状的影响.结果表明: 在6月龄~18月龄,GH基因的GH-P5位点的BB基因型对南阳牛的体长、体高显著的正效应;在生长的后期(24月龄~36月龄),IGF-IBP3-P5基因位点对南阳牛的后躯发育起主要调控作用,BB基因型个体的尻宽显著高于AA型.这表明基因的正效应不是在所有的生长周期表现,而是在特定的时期内表现对南阳牛生长发育的显著影响.  相似文献

阿黑皮素原(Pro-opiomelanocortin, POMC)在动物采食和能量平衡调控中发挥重要作用, 文章对绵羊POMC基因外显子3进行扩增和测序, 筛选多态性位点, 并分析多态位点与湖羊和东弗里生×湖羊杂种羊生长性状的相关性。测序后发现湖羊POMC基因外显子3有2个单碱基突变(g.273 T/C和g.456 G/A), 根据273位点处发生的T/C突变, 建立PCR-RFLP分析方法, 并对162只湖羊和130只东湖杂种羊进行检测分析。结果发现, 在湖羊群体中检测到TT(0.469)、TC(0.438)和CC(0.093)3种基因型, 而在东湖杂种羊群体中仅检测到TT(0.754)和TC(0.246)两种基因型。POMC基因外显子3的273位点多态性与生长性状的相关性研究结果显示:湖羊群体中CC基因型个体的2月龄断奶重、4月龄尻高及TC基因型个体4月龄体长和管围均显著高于TT型个体(P<0.05); CC基因型个体的4月龄重、6月龄重极显著高于TT和TC基因型个体(P<0.01); CC基因型个体的4月龄体高和体长极显著高于TT型个体(P<0.01), 且显著高于TC基因型个体(P<0.05)。此外, CC型个体的管围极显著高于TT基因型个体(P<0.01)。东湖杂种羊群体中TC基因型个体的2月龄断奶重、4月龄重及4月龄体高、体长、胸深和管围都显著高于TT型个体(P<0.05), TC型个体的6月龄重极显著高于TT型个体(P<0.01)。研究结果表明, POMC基因外显子3与绵羊生长性状相关, C等位基因对体重及体尺性状的增加更有利。该结果为进一步探讨POMC基因作为绵羊生长性状的辅助选育标记奠定了基础。  相似文献

利用PCR—SSCP技术研究了南阳牛、秦川牛、郏县红牛、西镇牛、鲁西牛和荷斯坦奶牛6个牛品种539个个体leptin基因的遗传多态性。结果表明,PCR扩增产物大小为330bp,PCR—SSCP分析表现出多态。南阳牛、秦川牛、郏县红牛、西镇牛、鲁西牛和荷斯坦奶牛的A等位基因频率分别为0.558,0.492,0.571,0.658,0.591,0.615;B等位基因频率分别为0.442,0.508,0.429,0.342,0.409,0.385。不同基因型与体重、体尺等生长性状指标相关性分析的结果表明:南阳牛群体内除12月龄的体高和日增重、18月龄的坐骨端宽和日增重外,BB型个体的六月龄、十二月龄、十八月龄、二十四月龄体斜长、胸围、体重、坐骨端宽、体高和日增重均显著的大于AB和AA型个体(P〈0.05);秦川牛群体内BB基因型个体十字部高上显著高于群体AA、AB型个体(P〈0.05),即BB〉AA、AB,可作为秦川牛体尺指标(十字部高)候选基因之一,但在体重、胸围、体长指标上均无显著差异(P〉0.05),所以不宜作为体重、胸围、体长指标候选基因;郏县红牛群体内AB与BB基因型个体在十字部高和坐骨端宽上显著高于群体AA型个体(P〈0.05),而群体内不同基因型在体重和体尺指标(体高、体斜长、胸围)上无显著差异(P〉0.05)。序列分析表明,leptin基因多态是第66位发生G→T、第67位发生A→C及299位发生新的单核苷酸突变C→T所造成。  相似文献

以131头纯种南阳牛为研究材料, 利用PCR-RFLPs对二酰甘油酰基转移酶2(DGAT2)基因的第6内含子和第7内含子的多态性及其与生长发育的相关性进行了分析。结果表明: 南阳牛在该位点分别检测到两种等位基因A/B和N/M, 频率分别为0.875/0.125和0.971/0.029。A和N等位基因是群体中的优势等位基因。该基因内含子6对南阳牛6月龄的体高、2岁体重、6月龄到两岁的胸围和体斜长都有显著的影响, 内含子6的AA基因型的6月龄的体高比杂合型高3.8 %, 两岁体重高3.9%, 6月龄到两岁的胸围分别高3.8%、3.4%、3.7%、4.3%; 6月龄到两岁的体斜长分别高3.8%、3.6%、3.8%、3.1%。内含子7对18月龄和两岁的坐骨端宽有极显著的影响(P<0.01); 对两岁的胸围有显著的影响(P<0.05)。  相似文献

POU1F1基因的遗传变异对南阳牛生长发育性状的影响   总被引:8,自引:0,他引:8  
利用PCR-RFLP技术首次研究了南阳牛群体100个个体POU1F1基因多态性及其与体重、体尺等生长性状指标之间的相关性。结果表明,南阳牛群体POU1F1基因座的451bp的PCR产物被限制性酶Hinf Ⅰ消化后表现多态性,它们的等位基因A/B频率为:0.465/0.535,且处于Hardy-Weinberg平衡状态。同时,南阳牛群体POU1F1-Hinf Ⅰ基因座不同基因型与体重、体尺等生长性状指标相关分析的结果表明:南阳牛群体内BB与AB基因型个体在初生重、断奶前平均日增重、六月龄体重、体斜长和胸围以及十二月龄的体重、体高、体斜长和胸围指标上有显著差异,且BB〉AB(P〈0.05);群体内BB型个体在十二月龄体重指标上显著高于群体的AA型个体,即BB〉AA(P〈0.05),在其他各年龄段的各项体重和体尺指标上同样呈现出B等位基因高于A等位基因的一种趋势。初步认为BB基因型为优势基因型,相应地B为优势等位基因,对选择有正向效应,提示POU1F1基因的占等位基因可能与高生长发育性状有关。  相似文献

文章采用DNA测序、PCR-RFLP和CRS-PCR技术对979头中国荷斯坦牛POU1F1基因与PRL基因进行研究,发现了3个新SNPs,分别是POU1F1基因第二外显子G1178C、PRL基因5侧翼区A906G和A1134G。采用SAS统计软件GLM程序,利用最小二乘法拟合线性模型,分析基因多态性与产奶性状的关系。结果表明:POU1F1基因1178位点GC基因型在产奶量、乳蛋白量、乳脂量方面均为优良基因型。PRL基因5侧翼区906位点AG基因型在产奶量方面为优良基因型,1134位点不同基因型产奶性状差异不显著。对PRL基因5侧翼区的906位点和POU1F1基因的1178位点进行基因互作分析,结果在乳脂率、乳蛋白率、产奶量、乳蛋白量和乳脂量方面各基因型组合之间均未观察到显著差异,说明基因聚合效应并不是单基因效应的简单相加,基因聚合效应在分子育种中具有更重要的意义。  相似文献

运用同源序列克隆技术结合反转录PCR技术和3′,5′cDNA末端快速扩增技术得到了牛FABGL基因的完整CDS,3′非翻译区和部分5′非翻译区.序列分析和生物信息学研究表明,所获得的牛FABGL基因的cDNA包含994个核苷酸和780 bp的开放阅读框及198 bp的完整的3′非翻译区.该基因编码260个氨基酸残基蛋白,在氨基酸水平上与人的同源基因具有高度的相似性(88%).采用PCR-SSCP方法,在递交的包含完整CDS的长为1 925 bp的该基因的基因组DNA序列(GenBank接受号DQ409814)1 065 bp 和1 792 bp处,分别发现了两个单核苷酸碱基突变Y=C/T,R=A/G;它们分别位于该基因的第五和第八内含子.对包含这两个多态位点的3个品种(安格斯、海福特和西门塔尔)牛的共179个个体等位基因频率与部分肉质及生长性状进行了关联分析,结果发现,在第八内含子内具有GG基因型的个体的肉用性能指数(4.283±.0.475kg/cm)较具有AA基因型个体的(4.008±0.465kg/cm)高(P≤0.01);而且同一位点具有GG基因型的个体的眼肌面积(73.380±13.005 cm2)显著高于具有AA基因型的个体(67.744±12.777 cm2)(P≤0.05).在第五内含子内,具有CC、CT、TT3种不同基因型的个体之间,平均日增重差异均达到极显著水平(P≤0.01),以具有TT基因型的个体平均日增重最高(0.652±0.330kg/d),CC基因型的最低(0.421±0.178kg/d).  相似文献

利用PCR-SSCP结合测序技术对405头24月龄秦川牛脂联素基因SNPs位点进行检测, 运用SPSS统计程序中的GLM模型将检测到的SNPs位点与部分胴体及肉质性状的相关性进行了分析。结果检测到AA、AB、BB、CC、CD 5种基因型, 其中AB、BB型个体在脂联素基因第2外显子 64 bp处发现G→C突变, CD型个体第3外显子50 bp处发现C→T的突变, G→C导致谷氨酸(GGA)转化为谷氨酰胺(GCA), C→T导致丝氨酸(TCA)转化为亮氨酸(TTA)。方差分析结果表明: AA型个体的宰前活重、胴体重、眼肌面积显著高于BB型(P<0.05), 而在胴体腿臀围方面, AA型个体极显著高于AB型、BB型个体(P<0.01)。CD型个体的宰前活重、胴体腿臀围、皮下脂肪厚、背膘厚、嫩度都显著优于CC型个体(P<0.05)。脂联素基因该位点可能是影响秦川牛胴体及肉质性状的主效QTL或与之紧密连锁, 可作为秦川牛高档牛肉生产的候选分子标记。  相似文献

利用PCR-SSCP技术对南阳牛、利木赞、盖洛威共100头牛的生长激素受体基因的外显子10(GHR10)部分序列进行单核苷酸多态性研究, 并分析了该基因的不同基因型与3个品种牛生产性状的关系。结果表明: 南阳牛、利木赞、盖洛威3个品种中共存在6种基因型(AA、BB、CC、AB、AC、BC), c2检验表明该实验群体在这一位点上处于Hardy-Weinberg平衡状态(P > 0.05), 利木赞和盖洛威的PIC表现为中度多态,而南阳牛表现高度多态。测序结果显示: 所扩增GHR10部分片段共有5处碱基突变, 分别是495 bp (A/T), 622 bp(C/T), 650 bp(A/C), 702 bp(T/C), 730 bp(A/G); 并导致3处氨基酸替代: 其中622 bp Pro/Ser(脯氨酸/丝氨酸), 650 bp Asn/Thr(天冬酰胺/苏氨酸), 730 bp Ser/Gly(丝氨酸/甘氨酸)。最小二乘分析表明: 基因型AB、BC所对应的12月龄体重最小二乘均值显著高于基因型CC所对应的最小二乘均值(P < 0.05, P < 0.01); 18月龄体重BC基因型显著高于CC基因型(P < 0.01), 基因型AB所对应的18月龄胸围最小二乘均值显著高于基因型CC所对应的最小二乘均值(P < 0.05)。  相似文献

Transmembrane protein 18 (TMEM18) gene is expressed in the central nervous system that has recently been linked to human obesity and body mass index (BMI) in genome wide association studies (GWAS). In this study, two novel single nucleotide polymorphisms (SNPs), g.3835 G > A (aa. Gly > Ser) and g.3865 A > G were detected in cattle TMEM18 gene by DNA sequencing. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and forced PCR-RFLP methods were used to genotype the two SNPs in 1218 Chinese native cattle individuals. The association between novel SNPs and growth traits (especially on body weight and body height) in Nanyang cattle showed that individuals with GG genotype was higher than those with AA genotype at g.3835G>A locus (P < 0.05) in Nanyang cattle. Significant association also revealed that individuals of AA genotype at g. 3865 A>G locus were higher in terms of body weight, average daily gain, chest circunmference, body length (P < 0.05), body height and hucklebone width (P < 0.01) than GG genotypes in Nanyang cattle. Moreover, the effect of XspI-MluI combinations with growth traits in Nanyang cattle revealed that AAGG type had higher body weight, body length, chest circumference and average daily gain (P < 0.05) than GGAG in Nanyang cattle aged at 6 months.  相似文献

GH secretagogue receptor (ghrelin receptor, GHSR) is known to be involved in the control of GH release by mediating the strong stimulatory effect of the endogenous ligand, ghrelin, on GH secretion. Associations between the GHSR gene polymorphism and the growth traits were revealed in Nanyang cattle. The mutations at nt456(G > A) and nt667(C > T) were complete linkage and located in exon 1 of the coding region of the GHSR gene. Least squares analysis revealed a significant statistical effect (P < 0.05) of the GHSR gene different genotypes on body weight and average daily gain at 6 months of age in Nanyang cattle. Individuals with GHSR-MM genotype showed higher body weight and average daily gain than individuals with GHSR-MN genotype.  相似文献

PCR-SSCP and DNA sequencing methods were employed to screen the genetic variation of VEGF gene in 671 individuals belonging to three Chinese indigenous cattle breeds including Nanyang, Jiaxian Red and Qinchuan. Three haplotypes (A, B and C), four observed genotypes (AA, AB, BB and AC) and three new SNPs (6765T>C ss130456744, 6860A>G ss130456745, 6893T>C ss130456746) were detected. The analysis suggested that one SNP (ss130456744) in the bovine VEGF gene had significant effects on birth weight, body weight and heart girth at 6 months old in the Nanyang breed (P < 0.05). The results showed that the SNP (ss130456744) in intron 2 of the VEGF gene is associated with early development and growth of Chinese cattle. These findings raise hope that this polymorphism can be a molecular breeding marker in breeding strategies through marker assisted selection (MAS) in Chinese domestic cattle.  相似文献

Monoacylglycerol acyltransferase 2 (MGAT2), as a candidate gene for quantitative traits, relates to dietary fat uptake, lipids synthesis and storage, which plays a major role in the absorption of dietary fat by catalyzing the resynthesis of triacylglycerol in enterocytes. In this study, based on DNA pool sequencing and PCR-RFLP methods, polymorphisms of the MGAT2 gene were detected in 1145 Chinese indigenous cattle. The results revealed two novel mutations located on exon 1 and exon 5 (NM_001099136.1:m.84G>T and 756A>G). Hence, we described the HaeIII forced PCR-RFLP method in exon1 and a MluI PCR-RFLP method in exon5 to detect them. In addition, the associations of these polymorphisms with growth traits were evaluated in Nanyang cattle. The results showed that only HaeIII locus was associated with body weight and average daily gain aged 6 months, and individuals with genotype TT showed significantly higher body weight and average daily gain than those with genotype GG.  相似文献

The objective of this study was to estimate the allele and genotype frequencies of the IGF-IR/TaqI, m-calpain/HhaI, and UCP-3/BglI polymorphisms and to determine associations between these polymorphisms and growth traits in Chinese indigenous cattle breeds. Genotyping was performed on 321 animals including 135 Nanyang, 80 Qinchuan, and 106 Jiaxian cattle. No significant differences in growth traits were observed between the genotypes of IGF-IR/TaqI polymorphism in Nanyang cattle. The m-calpain/HhaI, and UCP-3/BglI polymorphisms were associated with body weight, withers height, and body length of 6 months (P < 0.05 or P < 0.01), body length of 18 months (P < 0.05), and body length and heart girth of 24 months (P < 0.01) in Nanyang cattle. In addition, Nanyang cattle aged 12 months with AB genotype of m-calpain/HhaI polymorphism had higher body length and heart girth than those with BB genotype (P < 0.01). The withers height were greater (P < 0.01) in Nanyang cattle aged 12 months with genotype AB of UCP-3/BglI polymorphism than those with genotype BB. The Nanyang cattle aged 24 months with genotype AA had higher withers height than those with genotype BB (P < 0.05). In conclusion, the m-calpain/HhaI and UCP-3/BglI polymorphisms may be used as DNA markers for selection in the breeding process of Nanyang cattle.  相似文献

In this study, polymorphism in the exon 1, 2, 3, 4, 5, 6, 7, 8, 9, 10 and 11 of bovine NUCB2 gene was detected by PCR-SSCP and DNA sequencing methods in 686 individuals from three Chinese cattle breeds. Two haplotypes (M and N), three observed genotypes (MM, MN and NN) and two SNPs (NC_007313: g. 27451G>A, NC_007313: g. 27472T>C) were detected. The frequencies of haplotypes M and N in inland Chinese three breeds were 0.531–0.721 and 0.279–0.469 respectively. The studied showed that Nanyang, Jiaxian Red and Qinchuan cattle populations were in Hardy–Weinberg equilibrium at SNPs locus of NUCB2 gene (P > 0.05). Polymorphism of the NUCB2 gene was shown to be associated with growth traits in Qingchuan and Nanyang cattle breed. The linkage of two mutant sites in the bovine NUCB2 gene had significant effects on body length, body weight, heart girth, and average daily gain at 24 months (P < 0.05). Results of this study suggested that the NUCB2-gene-specific SNP may be a useful marker for growth traits in future marker-assisted selection programmes in inland Chinese cattle.  相似文献

Retinoic acid receptor responder 2 gene (RARRES2) encodes a novel adipokine protein that plays a crucial role in regulating several biological processes, including immune responses, adipocyte differentiation, type 2 diabetes and metabolic syndrome. In this paper, polymorphisms of the bovine RARRES2 gene were detected in 1300 individuals from six breeds by DNA pooling, CRS?CPCR?CRFLP and DNA sequencing methods. The results showed that NC_007302:g.117035859A>G, 117035706G>A and 117034290A>G were in the coding region, which resulted in three synonymous mutations and only 117033779C>G was in the 3?? UTR. Additionally, associations of the four novel SNPs with growth traits were analyzed in Nanyang cattle up to 2?years of age. In P1-PvuII locus, individuals with genotype BC had greater body height and hucklebone width than those with genotype AA, AC and AB at the age of 24?months. In P3-BamHI locus, individuals with genotype AG had higher hucklebone width than those with genotype GG at the age of 24?months. However, no statistically significant differences were observed in P5-SmaI locus. These results indicated that RARRES2 gene might be a potential candidate gene for marker-assisted selection (MAS).  相似文献
