5种常见植物DNA提取效率的比较   总被引:3,自引:0,他引:3  
以小麦、玉米、甘蓝、花生、菠菜的幼嫩叶片为实验材料,采用高盐低pH值法、SDS法和CTAB法3种不同的DNA提取方法,提取其总DNA,用琼脂糖凝胶电泳检测和紫外分光光度法对所得DNA进行比较分析.结果表明:玉米和菠菜采用SDS法提取基因组DNA效果明显优于CTAB和高盐低pH法.CTAB法对高脂肪花生的DNA提取,所得浓度较高.而高盐低pH值法建议减少使用.  相似文献

比较不同DNA检材的提纯方法,便于法医的实际应用选择。采集饮用过的饮料瓶或矿泉水瓶、废弃的烟蒂、使用过的手套口罩及衣物各90份,随机分为Chelex组、磁珠组和有机组,各组检材分别使用相应的方法进行DNA的提纯,对提纯后三组检材的DNA提取液的STR分型、DNA提取量与紫外线分光光度计下OD值水平进行比较。由磁珠法提纯得到的DNA超过9个基因座的例数最多;提取量与DNA提取纯度显著优于其他二组,差异具有统计学意义(p0.05)。对于微量接触或污染严重的检材,可使用DNA IQ磁珠法进行提纯,可优化DNA的提取纯度,利于法医工作的进行。  相似文献

采用PEG介导的原生质体转化方法,研究了将含有潮霉素抗性基因的质粒pMP-Hygro转入红曲菌9903A的影响因素。实验结果表明,原生质体转化最佳条件为:1.0mol/L的山梨醇为渗透压稳定剂;以含有50mmol/L的Ca2 和最终质量分数为40%的PEG4000为转化介质;最佳原生质体浓度和载体DNA用量分别为1×108个/mL、1μg/100μL原生质体;原生质体再生培养基采用不含无机盐的培养基,再生方式采用原生质体液涂布单层再生培基平板法。得到的平均DNA转化率可达160个/μgDNA。本文所研究的PEG介导的原生质体转化方法可以较好的向红曲菌细胞导入外源DNA,并使外源DNA在红曲菌细胞内大量表达。  相似文献

玉米大斑病是严重危害玉米生产的一个世界性真菌病害。由于玉米大斑病菌(Exserohilum turcicum)在无性生长过程中迅速产生黑色素,致使原生质体难以分离。测试了包括Fungase、Funcelase、Novozyme、Glucanex、Driselase、Uskizyme、Kitalase在内的7种细胞壁降解酶及其组合、病原菌菌株和培养基对原生质体分离效果的影响。结果表明菌株的培养形态和菌丝的生长状态显著影响原生质体的分离效率;酶组合Kitalase+Glucanex+Driselase, Kitalase+Glucanex和Kitalase+Uskizyme能够有效地分离玉米大斑病菌的原生质体。初步的转化试验表明,质粒pAN71可以用于该病原菌的转化。这些结果将为E.turcicumExserohium属其它真菌的基因克隆提供一些有用的信息。  相似文献

以拟南芥原生质体为实验体系,研究不同浓度的3种重金属离子对拟南芥原生质体的毒性和DNA损伤的差异。结果表明,用1-5mmol·L^-1的Zn^2+、Cd^2+和Cu^2+分别处理的拟南芥原生质体,2小时内活力逐渐下降,并表现出明显的浓度依赖性:与相同浓度的Cd^2+和Cu^2+相比,Zn^2+对拟南芥原生质体活力的影响程度较小,表现出较低的毒性。单细胞凝胶电泳检测发现,用0.1-0.8mmol·L^-1的Zn^2+、Cd^2+和Cu^2+分别处理拟南芥原生质体30分钟,以OTM值表示的原生质体DNA损伤量随重金属离子浓度的增加而递增:相同浓度(0.5mmol·L^-1)的3种重金属离子相比,Zn^2+对原生质体的遗传毒性明显低于Cu^2+和Cd^2+。综合原生质体活力和DNA损伤的单细胞凝胶电泳检测结果,发现ZnO^2+对拟南芥原生质体的遗传毒性较低,而CdO^2+和Cu^2+的遗传毒性较高。本研究建立的拟南芥原生质体实验体系,结合运用单细胞凝胶电泳技术,能够快速、灵敏地检测重金属对植物细胞的遗传毒性。  相似文献

以烟草原生质体为材料,采用彗星电泳检测用0.5W·m^-2紫外线以不同时间(0、5、10、30、60和120s)诱导的烟草原生质体中DNA的损伤。结果表明,在0~10s的时间内代表DNA损伤程度的尾矩、Olive尾矩等参数与紫外线照射时间具有良好的时间依赖关系。本文建立的烟草原生质体体系采用彗星电泳技术,可以快速而灵敏地检测紫外线对植物细胞的损伤程度。  相似文献

利用原生质体瞬间表达系统证实,即使用大量蓖麻毒素A链基因DNA导入诸葛莱(Ory-chophoragmus violaceus)原生质体,其蛋白质合成仍不被抑制。但是诸葛菜和灰叶烟草(Nicotiana plumbagenifolia)原生质体的蛋白质合成却容易被表达的白喉毒素A链所抑制。用CaMV 35 S启动子和Kozak序列控制的白喉毒素A链嵌合基因DNA在3h内能完全中止灰叶烟草原生质体蛋白质合成,此期间蛋白合成总量仅为正常时5%左右。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献
