以白洛克肉鸡 (EE)、中国丝羽乌骨鸡 (CC)、农大褐 (DD)和白来航 (AA) 4个纯种鸡为材料 ,进行 4× 4完全双列杂交 ,共得到 16种杂交组合。应用mRNA差异显示技术 (DDRT PCR)检测了 8周龄纯种和杂种鸡之间肝脏组织基因的差异表达。结果表明 ,在纯种和杂种间共有 8种 15类基因差异表达模式 ,杂种和纯种之间基因表达存在明显的差异。对各种基因差异表达模式与 10个肉用性状的杂种优势率进行相关分析发现 ,表达一致型P8(t1111)与肉用性状的杂种优势率相关不显著 (P >0 0 5) ,这说明杂种优势的形成与某些基因的差异表达有关 ;正交或反交特异表达型P4(t0 10 0、t0 0 10 )与 8周龄个体重、腿肌重、半净膛重、全净膛重相关显著 (P <0 0 5) ,与胸肌重相关极显著 (P <0 0 1) ;单亲特异表达型P1(t10 0 0、t0 0 0 1)与腹脂重相关显著 (P <0 0 5) ,与体斜长相关极显著 (P <0 0 1) ;双亲特异表达型P7(t10 0 1)与腿肌重、翅重、半净膛重、肌间脂宽的杂种优势率相关显著 (P <0 0 5) ;正交或反交单亲表达一致型P2 (t110 0、t0 0 11、t10 10、t0 10 1)与肌间脂宽的杂种优势率相关显著 (P <0 0 5) ;单亲表达一致型P5(t1110、t0 111)胫骨长的杂种优势率相关显著 (P <0 0 5)。  相似文献

肝脏脂肪酸结合蛋白(L-FABP)与脂肪运输及沉积关系密切。文章以8周龄丝羽乌鸡(CC)、农大褐蛋鸡(DD)及其正反杂交组合鸡(CD和DC)为试验材料,利用mRNA差异显示技术,在肝脏组织中获得一条阳性差异表达片段。通过片段回收、测序及序列比对发现,该差异表达片段为鸡L-FABP基因的全长cDNA编码序列(NCBI登录号:AY321365)。Northern杂交和半定量RT-PCR结果显示,该基因在正反杂交组合CD及DC的肝脏组织中表达量均明显高于亲本CC和DD,与杂种鸡的高腹脂及较大肌间脂宽表型趋势相同。鉴于L-FABP基因的高表达可能导致杂种鸡的脂肪沉积高于亲本,有必要针对鸡L-FABP基因进行深入的功能研究。  相似文献

三个品种家鸡催乳素基因cDNA的克隆及序列分析   总被引:28,自引:0,他引:28  
从粤黄鸡、毛丝乌骨鸡及伊莎蛋鸡的垂体中快速抽提总RNA,根据国外已发表的肉用仔鸡乳素基因cDNA的序列,设计并合成了能与特定载体末端互补的1对引物,经反转录-聚合酶链式反应(RT-PCR)方法扩增获得了特异性片段。将扩增片段与线性化质粒pBSSK连接,克隆后进行序列分析,与已报道的肉用仔鸡、矮脚鸡和火鸡催乳素基因的核苷酸序列及推导的氨基酸序列进行了比较。结果表明,不同品种间核苷酸同源性介于93.97%-99.87%之间,其中丝毛乌骨鸡与矮脚鸡间的同源性最高为99.87%。推导的相应氨基酸序列的同源性在98.25%-100%之间,也是丝毛乌骨鸡与矮脚鸡间的同源性最高,为100%。在粤黄鸡、丝毛乌骨鸡和伊莎蛋鸡中,发现前两者的催乳素前体的cDNA片段推导的 氨基酸序列中信号肽裂触位点跟肉用仔鸡,矮脚鸡及火鸡的一样,为Leu-Pro-IIe-Cys,而伊莎蛋鸡的信号肽裂解位点则不一样,为Pro-Pro-IIe-Cys。此位点的差异可能导致催乳素前体翻译加工的不同,使伊莎蛋鸡无就巢性。这3个家鸡品种与国外的肉用仔鸡、矮脚鸡催乳素氨基酸序列还在以下位置出现差异:71、141、150、175。在肉用仔鸡、丝毛乌骨鸡催乳素氨基酸序列中还发现了一个肝素结合位点175-181(L-R-R-D-S-H-K)。  相似文献

鸡白细胞介素 2 (chIL 2 )是近年来新发现的一类细胞因子 .根据Sundick等发表的鸡IL 2基因序列设计一对特异性引物 ,从ConA体外激活的脾淋巴细胞中提取mRNA ,通过RT PCR方法分别扩增和克隆了我国仙居鸡、丝羽乌骨鸡两个地方品种和艾维茵商品肉鸡IL 2cDNA .仙居鸡、丝羽乌骨鸡和艾维茵商品肉鸡IL 2基因的编码区均由 42 9nt组成 ,编码一个由 143个氨基酸组成的前体蛋白 .基因 5′端含有 17个核苷酸 ,3′端含有 2 85个核苷酸组成的非编码区 ,3′ UTR中含有 5个重复的“ATTTA”序列 .编码蛋白氨基酸与来自GenBank的Kestrel、Obese和SC品系的来杭鸡比较 ,氨基酸的突变主要发生在 2 8~ 3 2位 .而这一区域仙居鸡和丝羽乌骨鸡的编码氨基酸序列与Kestrel来杭鸡相同 ,艾维茵商品肉鸡与Obese和SC来杭鸡相同 .基因系统进化树分析表明 ,仙居鸡、丝羽乌骨鸡和Kestrel来杭鸡具有很近的亲缘关系 ,艾维茵商品肉鸡与Obese和SC来杭鸡具有很近的亲缘关系 .中国的药用鸡品种———丝羽乌骨鸡在非常保守的 13 3位发生了氨基酸突变  相似文献

以明星肉鸡和丝毛乌骨鸡杂交产生的F2代鸡群为实验材料,以鸡细胞外脂肪酸结合蛋白基因为影响鸡脂肪性状的候选基因,应用PCR-SSCP(单链构象多态)法对该基因5′调控区近端1.6 kb的序列进行单核苷酸多态性检测.其中第1对引物扩增的片段有2处碱基突变,第2对引物扩增的片段有1处碱基突变和1处碱基插入.经过与鸡腹脂性状进行最小二乘法分析,结果表明,第2对引物扩增的BB型基因片段(突变型)与AA型(野生型)和AB型(杂合型)相比有较高的腹脂重和腹脂率(P<0.001),表明该基因对于鸡腹脂蓄积具有大的影响或与控制腹脂性状的主基因连锁.  相似文献

“乌色”是乌骨鸡的一个主要性状,乌骨鸡的经济价值与药用价值关键在于乌骨鸡的“乌色”性状。为筛选与乌骨鸡“乌色”性状可能相关的基因,应用δ差异显示方法,系统比较全同胞武定乌骨鸡与非乌骨鸡的基因表达情况,回收、再扩增与纯化差异片段,并通过Northern blot验证、克隆和测序后,获得29个ESTs。序列分析结果表明,有8个ESTs分别与鸡已知基因骨骼肌-β原肌球蛋白基因(A17)、贲门肌球蛋白碱轻链基因(B57)、插入激活ras致癌基因(A36)、fra-2致癌基因(B36)、16S rRNA基因(A35)及线粒体基因组序列(D36、C39和D9)同源,序列相似性分别为97%、100%、98%、98%、98%、99%、99%和97%。5个ESTs分别与其他生物已知基因同源,它们是人TTN基因转录本变异novex-2(B53)、人磷酸葡糖变位酶5基因(B109)、人信号识别物54kD基因(B20)、人与鼠核糖核酸酶/血管形成抑制因子基因(C26、D31),差异片段与它们的序列相似性分别为82%、82%、87%、99%和99%,这些基因在鸡中还没被克隆。13个ESTs属于新基因片段,其中9个与数据库中ESTs序列相似性较高,4个属于全新基因片段,在数据库中没有与它们序列相似性较高的ESTs。选择16条差异片段,根据测序结果设计特异性引物,利用12个随机采集的武定乌骨鸡或非乌骨鸡样本,进行基因表达分析,结果表明,1个新基因(A7)片段、2个已知基因(插入激活ras致癌基因A36、信号识别物54kD基因B20)片段具有乌骨鸡或非乌骨鸡表达特异性,从而初步确定它们与武定乌骨鸡“乌色”性状相关。  相似文献

为探讨小麦杂种优势形成的分子机理,以一套双列杂交组合的苗期叶片为材料,利用mRNA差异显示技术分析了杂种及其亲本间MADS-box、G- box、Ser/Thr蛋白激酶、EIF-4A、ARF1基因家族共5类家族基因在杂交种和亲本之间的表达差异。并与杂种性状表现和杂种优势进行了相关分析。结果发现,除ARF1家族基因外,其余家族基因在杂种和亲本间存在显著的表达差异,差异表达类型可概括为4种:(1)双亲共沉默;(20单亲表达沉默;(3)杂种特异表达;(4)单亲表达一致。分析发现,MADS-box、G-box和EIF-4A家族基因在杂种和亲本间的差异表达模式相似,均以单亲特异表达和种特异表达类型所占比例最高。相关分析结果表明,以上所有家族基因的总体差异表达程度与所有性状的杂种表现均不相关,MADS-box家族基因中杂种特异表达类型与小穗数、单株产量和单穗产量杂种优势呈显著正相关,双亲共沉默类型与小穗数、千粒重和单穗产量杂种优势呈显著负相关,另外,EIF-4A家族基因中单亲表达一致型与单穗产量杂种优势呈显著正相,但双亲共沉默类型与小穗数和单穗产量杂种优势呈显著负相关,对于G-box基因家族而言,仅小穗数杂种优势和双亲共沉默类型成显著负相关,而蛋白激酶家族基因的各种差异类型与性状杂种优势的相关分析均不显著。这些研究表明,调控基因的差异表达与杂种优势形成有密切关系。  相似文献

以红色原鸡(Red Jungle Fowl, RJF)、丝羽乌骨鸡(Taihe silk Chicken, TS)、隐性白洛克鸡(White Recessive Rock, WRR)为资源群, 在鸡一号染色体的Contig.060226.1上选取了一个200 kb的区域, 比较研究了3个群体的SNP (single nucleotide polymorphism)多样性、估算了鸡的初始群体有效规模大小(effective size of population, Ne)。红色原鸡、丝羽乌骨鸡、隐性白洛克鸡3个群体的平均杂合度分别为0.28533±0.03475、0.32926±0.03919、0.30168±0.04038。显著性检验差异不显著(P=0.2368>0.05)。根据Latter 和Nei的方法对鸡的群体有效规模进行了估算, 鸡的初始群体有效规模大小为20 000~150 000。鸡在驯养的早期阶段经历过严厉的瓶颈效应, 但瓶颈效应对鸡各品种SNP的多样性并未产生显著影响。笔者认为, 鸡在驯养的早期阶段群体有效规模足够大, 品种分化过程中群体迅速扩张, 品种间的广泛杂交(特别是和红色原鸡之间)以及鸡基因组的高重组率等因素是导致家鸡和原鸡以及各家鸡品种间SNP多样性没有显著差别的重要原因。  相似文献

以小麦幼苗为材料 ,采用mRNA差异显示方法和银染技术 ,对经过用 16 % (- 0 .5MPa)PEG - 6 0 0 0溶液处理不同时间而诱导表达的小麦基因进行分离 ,共得到cDNA差异片段 5 2条。经ReverseNorthern验证 ,检出阳性表达片段 15个 ,克隆并测序。经GenBank查询 ,11个片段序列与已知序列有较高的同源性 ,4个片段同源性非常低 ,可能为新基因。  相似文献

采用mRNA差异显示技术,分析了水稻幼苗受低剂量UV-B辐射的mRNA表达丰度差异,获得了18个差异片段,其中15个差异片段在表达水平上有差别,1条为被抑制表达,而UV—B诱导特异表达的为2条.将这2条片段进一步回收、重扩增发现确为特异带型.对其中的一个差异片段RUVB2进行序列测定和国际互联网查询结果表明,该序列位于Oryza sativa第10染色体的OSJNBb0091N21BAC上(登录号:AC091122),且该cDNA片段与水稻的3个EST序列有95%以上的同源性,而与其它序列同源性较低.  相似文献

No differences exist between basal plasma corticosterone concentrations of broilers ferals and first generation large crossbreds (White leghorn x New Hampshire). Crossbreds show near double adrenal responsiveness to synthetic ACTH administration, indicating a heterosis effect for this characteristic. Extraction of chicken plasma with 30% aqueous polyethylene glycol solution eliminates the need for tedious and expensive pre-assay purification of plasma.  相似文献

DNA fingerprints of Japanese quail male and female pure line breeders were obtained with probes 33.6, 33.15, and R18.1 and they yielded a total of 59 scoreable bands. Bandsharing (0 < BS < 1) was calculated within and between six quail lines of two origins, and under reciprocal recurrent (AA and BB), within-line (DD and EE) or no (PP and FF) selection. Twenty one pair types were compared. BS was 0.30 higher within line than between lines. BS with the control line was smaller for reciprocal recurrent selection lines than for lines under individual selection. Bandsharing between the two reciprocal recurrent selection lines was 0.19 lower than between lines under individual selection. These results indicate that the two selection methods had different effects on the genetic constitution of the lines, in agreement with previous observations made from the analysis of biochemical polymorphisms with the same set of birds. Egg production and weight traits of pure and crossbred progeny from fingerprinted quail were obtained and compared, and a linear relationship with the measure of bandsharing was estimated. No significant regression coefficient of performance on BS was found over all progeny genetic types. Heterosis from individual matings could also be estimated under the two selection methods since the same birds were parents of both pure and crossbred performance-tested quail. The association of heterosis with the difference between BS of parents of the purebreds and BS of parents of their half-sib crossbreds was favourable and significant for early production traits in lines DD and EE, but no relationship was found in lines AA and BB. These results indicate that the high level of heterosis obtained through reciprocal recurrent selection, and the heterosis observed under within-line selection may have, partly at least, a different genetic determinism. Therefore, the relationship of heterosis with BS may also depend on the past history of selection in the lines.  相似文献

We have isolated and characterized a cDNA encoding a chicken beta homolog of c-erbA, or thyroid hormone receptor (TR). Chicken liver cDNA libraries were screened with a rat TR beta-1 cDNA probe, and several cDNA inserts were isolated and characterized. The sequence of one cDNA predicts a 369-amino-acid open reading frame (ORF), with a protein sequence that possesses 96% identity with that of rat TR beta-1, but only 88% identity with chicken TR alpha. These data indicate that the cDNA likely encodes a beta form of TR that has the expected putative DNA and T3 binding domains. The chicken TR beta (chTR beta) in vitro translated protein binds T3 with high affinity, and binds both the thyroid hormone response element (TRE) from the rat growth hormone gene and the Xenopus vitellogenin A2 gene estrogen response element (ERE), similarly to that of the rat TR beta-1. Northern blot analysis revealed the expression of a 7.0-kb RNA in several tissues including cerebellum, pituitary, kidney, and liver. This chicken liver TR beta cDNA sequence varies in both the 5' and 3' untranslated regions from the chicken kidney TR beta cDNA sequence recently reported (Forrest et al., 1990). The 5' untranslated cDNA sequence divergence occurs near a potential splice site junction of the human TR beta gene, suggesting that this chicken liver cDNA may represent an alternatively spliced RNA product of the chicken TR beta gene.  相似文献

配套系在动物育种中能最大程度地利用品系的杂种优势,以获得最大的经济效益,配套系的选育是今后肉兔遗传改良的发展趋势。肉兔父系以选择平均日增重和上市体重为主,断奶后生长性状一般与料肉比呈有利的负遗传相关,可作为料肉比的间接选择标准。而母系以选择断奶数和产仔数为主。大多数繁殖性状的遗传力较低,在进行遗传评估时应当尽可能全面地收集个体和相关亲缘关系个体的记录。重复观测值动物模型下的BLUP估计是进行肉兔专门化品系选育的主要方法。虽然窝内个体数性状的直接选择效率低于生长性状,但其杂种优势一般高于生长性状。通过计算杂交参数和比较同代杂种与纯种间的生产性能可以进行杂种优势的估计。文章对肉兔专门化品系的选育、杂种优势的利用和良种繁育体系的建立进行了综述,总结了肉兔专门化品系的培育方法,性状选择的标准以及杂种优势的估计结果。  相似文献

Best-hybridized crossing should ideally result in optimal exploitation of heterosis of lines and capitalize on expressed heterosis. Rabbit breeding is heading in the direction of breeding for best-hybridized crossing of meat rabbits. Most special sire lines are selected for post-weaning average daily gain and marketing weight. Post-weaning growth has a negative and favorable genetic correlation with the feed conversion ratio, which is used in indirect selection for feed conversion ratio. The most common selection criteria for special maternal lines are related to litter size at birth or at weaning. Since the heritability of most reproductive traits is low, we must collect as many individual and relative records as possible in the genetic evaluation of rabbits. The BLUP procedure under an animal repeatability model is the most common procedure used for evaluation of animals in selection programs for special lines of meat rabbits. Direct selection for litter size is less efficient than selection for post-weaning growth, but the estimation of heterosis is generally higher for litter size than that for the post-weaning growth. Evaluation of heterosis could be performed by estimating crossbreeding parameters in the cross or comparing contemporary productivity among purebreds and crossbreds. Here, we reviewed breeding of special lines, exploitation of heterosis of crossbreds, and establishment of crossbreeding system of hybrid meat rabbits and summarized the methodologies of breeding special lines, criteria in selection programs, and the result of heterosis estimates.  相似文献

以丝羽乌骨鸡和隐性白洛克正反交产生的F2代为实验群体, 采用PCR-SSCP和DNA测序的方法检测鸡载脂蛋白A5(apoA5)基因的单核苷酸多态性(SNPs), 并将所发现的SNPs与体重、胸肌重、腿肌重、心脏重、肝脏重和腹脂重等屠体性状进行关联分析。结果发现, 鸡apoA5基因5′-调控区C-169T, 外显子2 C600T、T635C, 外显子3 C841G、C914T、C1142G、C1394T共7个突变位点。其中外显子2突变位点C600T、T635C对12周龄腹脂重、腹脂率、肝脏重和心脏重有显著影响(P<0.05), 根据PCR-SSCP的结果将其分为6种基因型(AA, AB, AC, BB, BC, CC): 其中CC型个体的腹脂重和腹脂率显著高于AA型、AB型、AC型、BB型、BC型 (P<0.05); AC型个体的肝脏重显著低于AA型、AB型、BB型、BC型和CC型的肝脏重(P<0.05); BC型个体的心脏重显著低于BB型的心脏重(P<0.05)。  相似文献

Rabbits are very sensitive to heat stress because they have difficulty eliminating excess body heat. The objective of the current study was to evaluate the effects of heat stress on slaughter weight, dressing percentage and carcass and meat quality traits of rabbits from two genetic groups. Ninety-six weaned rabbits were used: half were from the Botucatu genetic group and half were crossbreds between New Zealand White sires and Botucatu does. They were assigned to a completely randomized design in a 2 × 3 factorial arrangement (two genetic groups and three ambient temperatures: 18°C, 25°C and 30°C) and kept under controlled conditions in three environmental chambers from 5 to 10 weeks of age. Slaughter took place at 10 weeks, on 2 consecutive days. Meat quality measurements were made in the longissimus muscle. Actual average ambient temperature and relative humidity in the three chambers were 18.4°C and 63.9%, 24.4°C and 80.2% and 29.6°C and 75.9%, respectively. Purebred rabbits were heavier at slaughter and had heavier commercial and reference carcasses than crossbreds at 30°C; however, no differences between genetic groups for these traits were found at lower temperatures. No genetic group × ambient temperature interaction was detected for any other carcass or meat quality traits. The percentages of distal parts of legs, skin and carcass forepart were higher in crossbred rabbits, indicating a lower degree of maturity at slaughter in this group. The percentage of thoracic viscera was higher in the purebreds. Lightness of the longissimus muscle was higher in the purebreds, whereas redness was higher in the crossbreds. Slaughter, commercial and reference carcass weights and the percentages of thoracic viscera, liver and kidneys were negatively related with ambient temperature. Commercial and reference carcass yields, and the percentage of distal parts of legs, on the other hand, had a positive linear relationship with ambient temperature. Meat redness and yellowness diminished as ambient temperature increased, whereas cooking loss was linearly elevated with ambient temperature. Meat color traits revealed paler meat in the purebreds, but no differences in instrumental texture properties and water-holding capacity between genetic groups. Purebred rabbits were less susceptible to heat stress than the crossbreds. Heat stress resulted in lower slaughter and carcass weights and proportional reductions of organ weights, which contributed to a higher carcass yield. Moreover, it exerted a small, but negative, effect on meat quality traits.  相似文献

Upon stimulation with lipopolysaccharide (LPS) the chicken macrophage cell line HD-11 secretes factors with cytokine activity. To characterize these molecules, representational difference analysis with RNA of LPS-induced and uninduced HD-11 cells was performed. Two cDNA clones were isolated that code for polypeptides with structural features of chemokines. cDNA K60 codes for a novel CXC chemokine of 104 residues including a putative signal peptide of 20 amino acids at the N-terminus. It is 67% identical to the previously cloned chicken chemokine 9E3/CEF4. K60 exhibits a similar degree of sequence identity to human interleukin 8 and other related CXC chemokines (about 50%), rendering straight-forward predictions of its biological properties difficult. cDNA K203 codes for a novel CC chemokine of 89 amino acids including a putative N-terminal signal peptide of 21 residues. It is 43% identical to a previously characterized chicken protein with homology to mammalian macrophage inflammatory protein 1beta (MIP-1beta). K203 exhibits about 50% sequence identity to human MIP-1beta and other related CC chemokines.  相似文献

During the domestication of farm animals, humans have manipulated genetic variation for growth and reproduction through artificial selection. Here, data are presented for growth, reproductive, and behavior traits for the red junglefowl, a line of White Plymouth Rock chickens, and their F1 and F2 reciprocal crosses. Intra- and intergenerational comparisons for growth related traits reflected considerable additive genetic variation. In contrast, those traits associated with reproduction exhibited heterosis. The role of sexual selection was seen in the evolution of prominent secondary sexual ornaments that lend to female choice and male-male competition. The large differences between parental lines in fearfulness to humans were only mitigated slightly in the intercross generations. Whereas, overall F1 generation heterosis was not transferred to the F2, there was developmental stability in the F2, as measured by relative asymmetry of bilateral traits. Through multigenerational analyses between the red junglefowl and the domestic White Plymouth Rocks, we observed plasticity and considerable residual genetic variation. These factors likely facilitated the adaptability of the chicken to a broad range of husbandry practices throughout the world.  相似文献
