Trihelix转录因子家族是一类最近才被引发关注的基因家族, 其因在DNA结合结构域含有3个串联的螺旋结构(螺旋-环-螺旋-环-螺旋)而得名, 该结构域能特异地与DNA序列上的光应答元件GT元件结合, 所以该家族也被称为GT因子家族。在研究早期, 人们对该基因家族的认识只局限于它们对光依赖型靶基因的调控, 而最近10年的研究表明, 该家族基因在植物的花、气孔、表皮毛、胚胎和种子的发育等不同生长发育过程中, 以及在病害、盐胁迫、干旱胁迫和冷胁迫等生物胁迫和非生物胁迫应答过程中都扮演重要角色。文章主要从Trihelix转录因子家族的结构特点、分类以及最新的功能研究进展进行详细综述。  相似文献

植物同源结构域(plant homeodomain,PHD)是锌指结构域家族的一类转录调控因子,其最主要的功能是可以识别各种组蛋白修饰密码,包括组蛋白甲基化和乙酰化等;此外PHD结构域还可以与DNA结合。含有PHD结构域的蛋白,或者本身具有组蛋白修饰酶活性,或者可以与各类组蛋白修饰酶相互作用,还有部分与DNA甲基化相关,具有E3泛素连接酶活性,或者还可以作为染色质重塑因子,以各种不同的作用方式,在植物的生长发育过程中发挥了重要的作用。本文主要综述了结合各种类型组蛋白(包括H3K4me3/0、H3K9me3、H3R2和H3K14ac)以及DNA的PHD结构域的结构特点及其结合特异性、PHD结构域在植物中的进化保守性以及植物中已经发现的含有PHD结构域蛋白的功能及作用机制,为进一步了解该类蛋白在植物生长发育过程中如何发挥作用提供了参考。  相似文献

GntR家族转录调控因子是细菌中分布最为广泛的一类螺旋–转角–螺旋(helix-turn-helix,HTH)转录调控因子,此家族转录调控因子包含两个功能域,分别是N端的DNA结合结构域和C端的效应物结合结构域/寡聚化作用结构域。DNA结合结构域的氨基酸序列是非常保守的,但效应物结合结构域/寡聚化作用结构域的氨基酸序列却存在很大的差异性。目前许多GntR家族的转录调控因子已经被鉴定,这些转录因子调控细菌许多不同的细胞过程,如运动性、葡萄糖代谢、细菌的耐药性、病原细菌的致病力等。本文主要阐述了GntR家族转录调控因子的发现、二级结构、生物学功能、调控模式等方面的研究进展,旨在为研究者全面、深入地了解GntR家族转录调控因子的功能及作用机理提供帮助。  相似文献

脱水素(dehydrins,DHNs)是高等植物胚胎发育晚期产生的一类特异多肽,能够逆转细胞脱水所造成的伤害.为明确脱水素性质与功能的关系,本研究从3种棱型大麦分离到Dhn6基因,生物信息学分析其编码的蛋白质序列长度分别为523个(六棱)、502个(四棱)和486个(二棱)氨基酸残基;氨基酸突变位点分析发现该基因具有整体保守性和突变位点偏倚性.蛋白质性质与二级结构分析表明,DHN6是高度亲水的碱性蛋白质,线性结构和自由卷曲为二级结构的主要组分,K-片段参与α-螺旋结构的形成,由此推测DHN6的兼性α-螺旋结构域在稳定膜结构的水合保护体系过程中发挥着重要的生理功能.构建的21个物种DHN6蛋白质系统发生树,发现禾本科植物遗传距离较近,结合核苷酸序列的物种特异性,本研究认为Dhn6基因可作为物种鉴定依据.  相似文献

hnulp1是具有碱性螺旋-环-螺旋(bHLH)的新的一类转录因子.其C端含一个DUF654结构域,其序列在同源基因中相当保守,但该结构域功能未知.利用GAL4转录因子中的DNA结合结构域(DBD)和含有与DBD结合序列的荧光素酶报告基因(GAL4-Luc)质粒,构建了哺乳动物细胞转录因子活性分析系统,随后利用GAL4-Luc荧光素酶报告基因对5种含DUF654结构域的不同缺失片段转录抑制活性进行检测.检测结果表明,该基因DUF654结构域中从Δ228-407氨基酸区段具有强烈的转录抑制活性.该结果为进一步研究DUF654结构域的功能和hnulp1基因转录调控的机制奠定了基础.  相似文献

微生物许多非核糖体肽类次生代谢产物主要是由非核糖体肽合成酶(NRPS)催化合成。参考Gontang发布的非核糖体肽合成酶(NRPS)通用引物设计扩增NRPS腺苷酰化结构域基因序列的特异引物,从海洋链霉菌L1的基因组DNA中扩增获得一个715 bp的NRPS基因序列。测序结果及比对分析表明该片段属于NRPS腺苷酰化结构域部分序列。对其拟翻译的氨基酸序列组成成分、理化性质进行分析,显示其包含AFD class I超基因家族核心结合区,为NRPS腺苷酰化结构域(A结构域)所在区域。对氨基酸序列的二级结构预测和三级结构模拟,发现与数据库中肠菌素合酶F组分的结构相似。为后续研究A结构域的特异性及完整NRPS基因簇克隆提供了参考。  相似文献

免疫球蛋白(Ig)的表达是B细胞特异性和发育阶段特异性的事件.Oct2、NF-kB和HLH蛋白与Ig基因细胞特异转录有关.Oct2含有POU结构域,属POU转录调控因子家族;NF-kB有rel-like结构域,属rel-like转录调控因子家族;HLH蛋白特征性结构为螺旋-环-螺旋(helix-loop-helix,HLH)结构域,已报道的HLH蛋白都与转录调节有关.  相似文献

SNF4基因编码的Snf4p具有调节Snf1复合体的蛋白激酶活性功能,根据已知的SNF4基因序列设计引物扩增获得S.cerevisiae YS2的SNF4基因完整序列。序列分析表明,SNF4基因的开放阅读框为969bp,编码322个氨基酸残基。应用生物信息方法预测其理化性质、疏水性、信号肽、亚细胞定位、活性位点及其高级结构。结果表明:Snf4p为具有一定亲水性的非跨膜胞内稳定酸性蛋白,功能结构域为CBS_pair superfamily结构域,二级结构主要由a-螺旋组成,空间结构是由4个CBS结构域构成两个CBS对围绕形成的二聚体。Snf4p的第一个CBS对区域的β片层结构是Snf1p、Sip2p的β发夹结构结合作用区。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Curcumin is the yellow pigment of turmeric that interacts irreversibly forming an adduct with thioredoxin reductase (TrxR), an enzyme responsible for redox control of cell and defence against oxidative stress. Docking at both the active sites of TrxR was performed to compare the potency of three naturally occurring curcuminoids, namely curcumin, demethoxy curcumin and bis-demethoxy curcumin. Results show that active sites of TrxR occur at the junction of E and F chains. Volume and area of both cavities is predicted. It has been concluded by distance mapping of the most active conformations that Se atom of catalytic residue SeCYS498, is at a distance of 3.56 from C13 of demethoxy curcumin at the E chain active site, whereas C13 carbon atom forms adduct with Se atom of SeCys 498. We report that at least one methoxy group in curcuminoids is necessary for interation with catalytic residues of thioredoxin. Pharmacophore of both active sites of the TrxR receptor for curcumin and demethoxy curcumin molecules has been drawn and proposed for design and synthesis of most probable potent antiproliferative synthetic drugs.  相似文献

Microbial resistance to antibiotics is an unresolved global concern, which needs urgent and coordinated action. One of the guidelines of the Centers for Disease Control and Preventions (CDC) to combat antibiotic resistance is the development of new antibiotics to treat drug-resistant bacteria. In our effort to find new antibiotics, we report the synthesis and antimicrobial studies of 30 new pyrazole derivatives. These novel molecules have been synthesized by using readily available starting materials and benign reaction conditions. Some of these molecules have shown activity with MIC values as low as 0.78?µg/mL against four bacterial strains; Staphylococcus aureus, methicillin-resistant S. aureus, Bacillus subtilis, and Acinetobacter baumannii. Furthermore, active molecules are non-toxic to mammalian cell line.

The young pistils in the melanthioid tribes, Hewardieae, Petrosavieae and Tricyrteae, are uniformly tricarpellate and syncarpous. They lack raphide idioblasts. All are multiovulate, with bitegmic ovules. The Petrosavieae are marked by the presence of septal glands and incomplete syncarpy. Tepals and stamens adhere to the ovary in the Hewardieae and the Petrosavieae but not in the Tricyrteae. Two vascular bundles occur in the stamens of the Hewartlieae and Tricyrtis latifolia. Ventral bundles in the upper part of the ovary of the Hewardieae are continuous with compound septal bundles and placental bundles in the lower part. Putative ventral bundles occur in the alternate position in the Tricyrteae and putative placental bundles in the opposite. position in the Petrosavieae. The dichtomously branched stigma in each carpel of the Tricyrteae is supplied by a bifurcated dorsal bundle.  相似文献

Cyclin-dependent kinases (CDKs) and Polo-like kinases (PLKs) play key role in the regulation of the cell cycle. The aim of our study was originally the further development of our recently discovered polo-like kinase 1 (PLK1) inhibitors. A series of new 2,4-disubstituted pyrimidine derivatives were synthesized around the original hit, but their PLK1 inhibitory activity was very poor. However the novel compounds showed nanomolar CDK9 inhibitory activity and very good antiproliferative effect on multiple myeloma cell lines (RPMI-8226).  相似文献
