从牛羊粪堆肥中筛选出一株纤维素降解菌Aspergillus sp.YN1,主要研究了液体发酵培养基中碳源、氮源、培养温度、起始pH、通气量以及接种菌龄对菌株YN1的羧甲基纤维素酶活(CMC酶活)及滤纸酶活的影响。研究结果表明,在优化条件下,该菌的CMC酶活、滤纸酶活在培养第3天分别达到0.53U/mL和0.15U/mL。在酶学特性研究中,菌株YN1的CMC酶的最适反应温度为70°C,最适反应pH4.0(酶促反应为30min)。用不同温度处理1h或不同pH处理2h,YN1的CMC酶在30°C?50°C或pH3.0?4.0之间仍可保持80%以上的酶活性,对热及酸表现出较高的稳定性。  相似文献

一株产纤维素酶菌株的分离、鉴定及产酶特性   总被引:2,自引:0,他引:2  
【目的】筛选并鉴定一株产纤维素酶的菌株,初步探究该菌的产酶特性,为综合利用纤维素筛选菌源。【方法】在常温条件下,采用滤纸培养基对菌种富集,采用CMC-Na初筛纤维素降解菌,采用LB培养基分离纯化菌株,经形态学、生理生化特征试验、16S r RNA基因序列测定等分析筛选菌株的系统分类地位。单因素试验确定培养时间、培养温度、初始p H及Na Cl浓度对筛选菌株产酶活力的影响。【结果】从腐烂的玉米秸秆中分离出一株在常温下产纤维素酶细菌KZ-2,根据菌落形态特征、生理生化特征鉴定以及16S r RNA基因序列分析,初步鉴定KZ-2为肠杆菌(Enterobacter sp.),为潜在新种。产酶条件实验显示:该菌使用产酶发酵培养基120 h产酶量达到最大值,在25–35°C、初始p H 4.5–5.5、Na Cl浓度1.0%–2.0%范围内为最佳产酶条件,在最适条件下酶活可达80.93 U/m L。该菌株所产纤维素酶最适反应p H为7.0,最适反应温度为50°C。【结论】KZ-2是一株具有降解纤维素能力的细菌,在常温下即可分泌纤维素酶,并且该菌株为潜在新种,具有潜在的开发价值。  相似文献

通过对富含枯枝败叶的土壤样品进行富集培养,利用刚果红纤维素培养基初筛和酶活测定复筛得到产纤维素酶的一株真菌,将其命名为GC2-2,并对该菌株进行鉴定及酶学性质研究。结果表明该菌株是一株耐高温、碱性纤维素酶的真菌GC2-2。通过18S rDNA分子克隆测定,该菌为球孢枝孢菌,其滤纸酶的活力优于CMC酶的活力。该菌所产酶的最适反应条件为温度35°C,最适pH值7.5。  相似文献

【目的】从海洋样品中分离筛选出产葡萄糖氧化酶菌株。【方法】采用双层平板筛选法进行初筛、复筛确定一株酶活较好的菌株,命名为GOD2(Glucose oxidase)。通过形态学、生理生化特征及16S rRNA基因序列分析研究其分类地位,并对其产生的葡萄糖氧化酶进行分离纯化和部分酶学性质的研究。【结果】细菌GOD2为产葡萄糖氧化酶菌株且遗传稳定,初步鉴定该菌株为假单胞杆菌(Pseudomonas migulae),其所产酶最适反应温度为20°C,热稳定性较差,40°C剩余相对酶活80%;超过40°C酶活力迅速下降。【结论】GOD2是一株极具研究价值的产低温葡萄糖氧化酶菌株。目前没有关于利用该菌生产葡萄糖氧化酶的报道。  相似文献

从东祁连山高寒草甸土壤中分离得到一株纤维素酶高产菌株【3-2。根据菌体形态观察、革兰氏染色反应及16S rDNA序列测定以及序列同源性比较,确定鉴定该菌为芽孢杆菌属的成员(Bacillus sp.)。对该菌的产纤维素酶条件研究结果表明:该菌在5℃~45℃、初始pH 4.0~9.0的环境下均能产纤维素酶。在初始pH为8.0、盐浓度为2.0%~3.0%之间、培养温度为20℃培养条件下最适产酶。在5℃时,相对酶活仍能保持60%。  相似文献

从黄浦江淀山湖的水底沉积物中筛选分离得到一株产纤维素酶的菌株。经细菌形态观察,生理生化实验并结合16SrRNA序列分析,鉴定该菌为蜡状芽孢杆菌(Bacillus cereus),同时发现其在系统发育树中处于一个独立的分支,推测该菌为Bacillus属中一个新的亚种。对该菌株产酶及酶活特性进行了初步研究,发现纤维素酶的产生与细菌的生长密切相关。该菌株在37°C,pH7.0的条件下,发酵66h后纤维素酶活达到最高值4.58U/mL。  相似文献

产外切葡聚糖酶真菌的筛选鉴定及毕赤酵母中的表达   总被引:1,自引:0,他引:1  
【目的】外切葡聚糖酶是纤维素酶组分中一类对结晶纤维素有降解作用的酶类,如何提高外切葡聚糖酶活力是研究的关键问题。【方法】从筛选鉴定得到的一株产外切葡聚糖酶酶活较高的黑曲霉Asp-524菌株出发,通过PCR技术克隆得到外切葡聚糖酶基因序列,生物学信息分析后,构建了毕赤酵母诱导型表达载体,实现了该基因在毕赤酵母中的成功表达。【结果】抗性筛选得到的阳性转化子,用终浓度为1%甲醇诱导5 d后,酶活达到4.74 U/mL。酶学性质分析显示重组外切葡聚糖酶最适pH为5.0,pH稳定性分析显示在pH为4.0-6.0范围内相对稳定,酶活能保持在最高酶活力的80%以上,最适反应温度为50°C,经60°C保温1 h后,酶活仍能保持80%以上。【结论】结果说明该外切葡聚糖酶具有较好的热稳定性和pH稳定性,这一研究为纤维素酶的实际应用奠定了一定基础。  相似文献

从云南腾冲热泉水样中分离筛选得到一株产木聚糖酶的菌株。通过细菌形态观察、生理生化特性并结合16S rDNA序列分析,经鉴定,该菌为地芽孢杆菌(Geobacillus sp.),命名为Geobacillus sp.PZH1。对该菌株所产嗜热木聚糖酶及酶学特性进行了初步研究。SDS-PAGE和酶谱分析测得该酶分子量约为69 kD;酶的最适反应pH和最适反应温度分别为7.0和70°C,在pH 5.0-11.0和40°C-100°C范围内均有较高酶活;该酶在pH 5.0-12.0范围内和70°C以内具有较高的稳定性;40°C-100°C范围内,该木聚糖酶没有被检测到纤维素酶活性。  相似文献

本研究以褐色高温单孢菌(Thermomonospora fusca)为出发菌株,通过紫外线和60Co-γ射线联合诱变,获得了一株纤维素酶高产菌株AV8,CMC酶活力达到0.679 IU/mL,与出发菌株相比,其产酶能力提高3.53倍。通过对AV8产纤维素酶的培养条件进行测定,结果显示:产纤维素酶最适应温度为55℃;该菌的最适产CMC酶初始PH值为7.0,最适产FPA酶初始PH值为8.0;当培养到第7天时,产CMC酶达到高峰。当培养到第8天时产FPA酶达到高峰。  相似文献

[目的]以纤维素为唯一碳源,从四川省阿坝自治州黄龙沟的高山低温环境中分离筛选产纤维素酶的耐冷菌,并研究菌株的产酶特征.[方法]根据菌株的ITS序列分析及形态特征,对菌株进行鉴定.利用DNS法测定纤维素酶酶活性.[结果]从四川省阿坝自治州黄龙沟的高山腐殖土中筛选出一株产纤维素酶的耐冷菌HD1031,经鉴定该菌为玫红假裸囊菌(Pseudogymnoascus roseus).该菌可在4℃-25℃生长,最适生长温度为16℃-17℃.该菌在以微晶纤维素和玉米芯粉为碳源、硫酸铵和Tryptone为氮源的培养基中,17℃、160 r/min摇瓶发酵8d后产生纤维素酶,其中内切葡聚糖酶酶活为366.67 U/mL,滤纸酶酶活87.6 U/mL,β-葡萄糖苷酶酶活90.8 U/mL,酶最适反应pH为6.0,最适反应温度为50℃.[结论]筛选获得一株产纤维素酶的耐冷菌HD1031,此菌株所产纤维素酶在20℃-40℃下活性较高,对热敏感,具有低温纤维素酶的特点.  相似文献

Psychrotropic Bacillus sphaericus producing solvent stable cold-active lipase upon growth at low temperature was isolated from Gangotri glacier. Optimal parameters for lipase production were investigated and the strain was able to produce lipase even at 15 °C. An incubation period of 48 h and pH 8 was found to be conducive for cold-active lipase production. The addition of trybutyrin as substrate and lactose as additional carbon source increased lipase production. The enzyme was purified up to 17.74-fold by ammonium sulphate precipitation followed by DEAE cellulose column chromatography. The optimum temperature and pH for lipase activity were found to be 15 °C and 8.0, respectively. The lipase was found to be stable in the temperature range 20–30 °C and the pH range 6.0–9.0. The protein retained more than 83 % of its initial activity after exposure to organic solvents. The lipase exhibited significant stability in presence of acetone and DMSO retaining >90 % activity. The enzyme activity was inhibited by 10 mM CuSO4 and EDTA but showed no loss in activity after incubation with other metals or inhibitors examined in this study.  相似文献

[目的]筛选一株低温产纤维素酶菌株并进行鉴定,初步探索其酶学性质,为微生物肥料生产筛选菌种资源.[方法]常温条件下,采用CMC-刚果红染色法初筛纤维素降解菌株.采用低温条件诱导的方法,筛选耐低温且产纤维素酶能力最强的菌株,经形态学、生理生化特征试验、ITS序列等方面分析系统分类地位.单因素试验确定温度、pH及金属离子对纤维素酶活力的影响.[结果]从秸秆还田土壤中分离出一株在13℃低温环境下高效分解纤维素的真菌M11,鉴定M11为草酸青霉(Penicillium oxalicum).发酵试验表明:以玉米秸秆粉为唯一碳氮源,13℃、200 r/min摇床发酵培养9d时,纤维素酶活力最高为33.08 U/mL.对其酶学性质初步研究表明:该酶最适pH为5.0,最适反应温度为20℃,在5℃-20℃间酶活力仍能保持在90%以上.[结论]Penicillium oxalicum M11是一株高效的纤维素降解菌株,在低温条件下可分泌纤维素酶且活性显著,具有潜在的开发价值.  相似文献

An amidase (EC 3.5.1.4) in branch 2 of the nitrilase superfamily, from the thermophilic strain Geobacillus pallidus RAPc8, was produced at high expression levels (20 U/mg) in small-scale fermentations of Escherichia coli. The enzyme was purified to 90% homogeneity with specific activity of 1,800 U/mg in just two steps, namely, heat-treatment and gel permeation chromatography. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and electron microscopic (EM) analysis of the homogenous enzyme showed the native enzyme to be a homohexamer of 38 kDa subunits. Analysis of the biochemical properties of the amidase showed that the optimal temperature and pH for activity were 50 and 7.0°C, respectively. The amidase exhibited high thermal stability at 50 and 60°C, with half-lives greater than 5 h at both temperatures. At 70 and 80°C, the half-life values were 43 and 10 min, respectively. The amidase catalyzed the hydrolysis of low molecular weight aliphatic amides, with d-selectivity towards lactamide. Inhibition studies showed activation/inhibition data consistent with the presence of a catalytically active thiol group. Acyl transfer reactions were demonstrated with acetamide, propionamide, isobutyramide, and acrylamide as substrates and hydroxylamine as the acyl acceptor; the highest reaction rate being with isobutyramide. Immobilization by entrapment in polyacrylamide gels, covalent binding on Eupergit C beads at 4°C and on Amberlite-XAD57 resulted in low protein binding and low activity, but immobilization on Eupergit C beads at 25°C with cross-linking resulted in high protein binding yield and high immobilized specific activity (80% of non-immobilized activity). Characterization of Eupergit C-immobilized preparations showed that the optimum reaction temperature was unchanged, the pH range was somewhat broadened, and stability was enhanced giving half-lives of 52 min at 70°C and 30 min at 80°C. The amidase has potential for application under high temperature conditions as a biocatalyst for d-selective amide hydrolysis producing enantiomerically pure carboxylic acids and for production of novel amides by acyl transfer.  相似文献

A series of site-directed mutant glucose isomerase at tryptophan 139 from Thermoanaerobacterium saccharolyticum strain B6A were purified to gel electrophoretic homogeneity, and the biochemical properties were determined. W139F mutation is the most efficient mutant derivative with a tenfold increase in its catalytic efficiency toward glucose compared with the native GI. With a maximal activity at 80 °C of 59.58 U/mg on glucose, this mutant derivative is the most active type ever reported. The enzyme activity was maximal at 90 °C and like other glucose isomerase, this mutant enzyme required Co2+ or Mg2+ for enzyme activity and thermal stability (stable for 20 h at 80 °C in the absence of substrate). Its optimum pH was around 7.0, and it had 86 % of its maximum activity at pH 6.0 incubated for 12 h at 60 °C. This enzyme was determined as thermostable and weak-acid stable. These findings indicated that the mutant GI W139F from T. saccharolyticum strain B6A is appropriate for use as a potential candidate for high-fructose corn syrup producing enzyme.  相似文献

A Psychrotolerant alkaline protease producing bacterium IIIM-ST045 was isolated from a soil sample collected from the Thajiwas glacier of Kashmir, India and identified as Stenotrophomonas sp. on the basis of its biochemical properties and 16S ribosomal gene sequencing. The strain could grow well within a temperature range of 4–37°C however, showed optimum growth at 15°C. The strain was found to over-produce proteases when it was grown in media containing lactose as carbon source (157.50 U mg−1). The maximum specific enzyme activity (398 U mg−1) was obtained using soya oil as nitrogen source, however, the inorganic nitrogen sources urea, ammonium chloride and ammonium sulphate showed the lowest production of 38.9, 62.2 and 57.9 U mg−1. The enzyme was purified to 18.45 folds and the molecular weight of the partially purified protease was estimated to be ~55 kDa by SDS-PAGE analysis. The protease activity increased as the increase in enzyme concentration while as the optimum enzyme activity was found when casein (1% w/v) was used as substrate. The enzyme was highly active over a wide range of pH from 6.5 to 12.0 showing optimum activity at pH 10.0. The optimum temperature for the enzyme was 15°C. Proteolytic activity reduced gradually with higher temperatures with a decrease of 56% at 40°C. The purified enzyme was checked for the removal of protein containing tea stains using a silk cloth within a temperature range of 10–60°C. The best washing efficiency results obtained at low temperatures indicate that the enzyme may be used for cold washing purposes of delicate fabrics that otherwise are vulnerable to high temperatures.  相似文献

Some microorganisms, including some bacteria isolated from soil, were found to secrete an extracellular soymilk-clotting enzyme. Among them, strain No. K-295G-7 showed the highest soymilk-clotting activity and stability of the production of the soymilk-clotting enzyme. The enzyme system (culture filtrate) coagulated protein in soymilk, a curd being formed at pH 5.8~6.7 and at 55~75°C. The optimum temperature for the soymilk-clotting activity was 75°C and the enzyme system was stable at temperatures below 50°C down to 35°C. About 80~100% of the original activity remained after 1 hr at pH 5~7 and 35°C.  相似文献

An extracellular detergent tolerant protease producing strain VSG-4 was isolated from tropical soil sample and identified as Bacillus subtilis based on morphological, biochemical characteristics as well as 16S-rRNA gene sequencing. The VSG-4 protease was purified to homogeneity using ammonium sulphate precipitation, dialysis and sephadex G-200 gel permeation chromatography with a 17.4 purification fold. The purified enzyme was active and stable over a broad range of pH (8.0–11.0, optimum at 9.0) and temperature (40°C to 60°C, optimum at 50°C). The thermostability of the enzyme was significantly increased by the addition CaCl2. This enzyme was strongly inhibited by PMSF and DFP, suggesting that it belongs to the serine protease superfamily. The purified VSG-4 alkaline protease showed remarkable stability in anionic (5 mM SDS) and ionic (1% Trion X-100 and 1% Tween 80) detergents. It retained 97±2% and 83.6±1.1% of its initial activity after 1 h preincubation in the presence of 1 % H2O2 and 1 % sodium perborate, respectively. Furthermore, the purified enzyme showed excellent stability and compatibility with some commercial laundry detergents besides its stain removal capacity. Considering these promising properties, VSG-4 protease may find tremendous application in laundry detergent formulations.  相似文献

Abstract

Extracellular lipase from an indigenous Bacillus aryabhattai SE3-PB was immobilized in alginate beads by entrapment method. After optimization of immobilization conditions, maximum immobilization efficiencies of 77%?±?1.53% and 75.99%?±?3.49% were recorded at optimum concentrations of 2% (w/v) sodium alginate and 0.2?M calcium chloride, respectively, for the entrapped enzyme. Biochemical properties of both free and immobilized lipase revealed no change in the optimum temperature and pH of both enzyme preparations, with maximum activity attained at 60?°C and 9.5, respectively. In comparison to free lipase, the immobilized enzyme exhibited improved stability over the studied pH range (8.5–9.5) and temperature (55–65?°C) when incubated for 3?h. Furthermore, the immobilized lipase showed enhanced enzyme-substrate affinity and higher catalytic efficiency when compared to soluble enzyme. The entrapped enzyme was also found to be more stable, retaining 61.51% and 49.44% of its original activity after being stored for 30 days at 4?°C and 25?°C, respectively. In addition, the insolubilized enzyme exhibited good reusability with 18.46% relative activity after being repeatedly used for six times. These findings suggest the efficient and sustainable use of the developed immobilized lipase for various biotechnological applications.  相似文献

温和气单孢菌YH311硫酸软骨素裂解酶的分离纯化与固定化   总被引:1,自引:0,他引:1  
通过硫酸铵沉淀、QAESephadex-A50柱层析及Sephadex-G150凝胶过滤等纯化步骤,对源自温和气单孢菌YH311的ChSase进行了分离纯化。结果表明,ChSase经上述纯化步骤后被纯化了55倍,其最终纯度可达95%以上,比活为31.86u/mg。经SDSPAGE及IFE测定可知该酶的分子量约为80kD,等电点为4.3~4.8。将纯化后的ChSase用海藻酸钠或纤维素固定化后,ChSase的热稳定性及贮存稳定性均可得到大幅度的提高:固定化酶用80℃水浴处理120min或于4℃冰箱放置30d后仍可保留50%以上的相对活力;但固定化酶的收率较低,仅为18.56%和18.86%。  相似文献
