采用在分离培养基中添加自然来源的抽提液,或加入一些特殊化合物,使其中处于Viable but non-culturable(VBNC)状态的微生物恢复其生长繁殖能力,从而得到分离。实验结果发现甜菜碱、丙酮酸钠、SOD以及过氧化氢酶可使分离到的微生物种类及菌落总数明显增加。还采用固液结合的方法来分离那些在普通平板培养基上不能形成肉眼可见菌落的那些微小菌落的微生物。采用这几种方法从4份土壤样品中共分离得到52株放线菌,103株细菌,17株真菌。对其中的放线菌和真菌进行了生物活性的测定,得到多株具有抗菌活性的微  相似文献

长木蜂属于野生蜂类,后代的生存和繁衍完全依靠雌蜂所提供的蜂粮,蜂粮保鲜时间的长短对蜂群的繁衍具有关键性作用。本文采用营养琼脂培养基、改良"LB"培养基、MRS(4.5)、MRS(6.5)培养基分别对芍药的手采花粉、长木蜂蜂花粉和不同酿制时间的蜂粮样品中的微生物进行菌落计数、分析和分离鉴定。酿制初期的蜂粮样品在营养琼脂培养基生长的细菌菌落数占优势,随酿制时间的延长而迅速下降;7d、8d蜂粮样品在改良"LB"培养基上生长的细菌菌落数占优势;10d之后的蜂粮样品在MRS(6.5)培养基上微生物菌落数占优势;分离的68株菌株,其中细菌43株,酵母17株,放线菌8株。从蜂粮中分离的酵母菌和放线菌菌株均来自长木蜂雌蜂的体表。分离的68株菌株中有12株有较强抑菌活性,尤其是放线菌NF-34菌株对真菌和细菌均有抑菌活性。  相似文献

从老鼠簕(Acanthus illicifolius L.)根际土壤和植物根、叶、茎中共分离得到97株微生物,其中包括27株放线菌,44株细菌以及26株真菌,并且土壤中的分离得到的微生物最多。对获得的菌株进行细胞毒活性、抗金黄色葡萄球菌和抗白色念珠菌的测定,结果发现细菌的活性百分比最高,而放线菌和真菌活性百分比相对较小。  相似文献

通过改良优化多种传统分离培养基,对凡口铅锌尾矿的酸性底泥可培养微生物资源进行分离,并结合多种分子标记基因对分离资源进行系统发育地位分析。结果表明,凡口铅锌尾底泥中分离获得细菌41株,分属于放线菌门(Actinomycetes)、变形菌门(Proteobacteria)和厚壁菌门(Firmicutes)的11目,18科,18属;真菌8株,分属于2门,5目,5科,5属;藻类1株,属于Galdieria属的红藻。改良ISP 2和改良9号培养基的分离效果较好,分离获得的微生物多样性较为良好。研究结果为矿山酸性废水微生物多样性研究提供参考,为酸性废水的修复及生态治理研究提供物质基础。  相似文献

为了探究从何种类型的自然生境中更易分离得到溶藻微生物,采用高氏1号培养基分别从水库底泥、湖泊底泥、农田土壤、林地土壤等四种来源共36份样品中分离了7 600株菌,并最终从中筛选得到了5株溶铜绿微囊藻(Microcystis aeruginosa)的溶藻菌,其中4株为假单胞菌(Pseudomonas sp.),1株为黄杆菌(Flavobacterium sp.),5株菌溶藻效率的变化范围为62%~95%。结果表明,当采用高氏1号培养基作为分离培养基时,湖泊底泥和水库底泥中的成功筛选概率最高,农田土壤次之,而林地土壤中则难以筛选得到,假单胞菌是较容易筛选得到的溶藻菌。  相似文献

【背景】红树林生境拥有丰富的微生物资源,分离获得更多的纯培养放线菌为新型抗生素的发现提供菌种资源。【目的】使用新型放线菌分离培养基,发掘广西北部湾茅尾海红树林生境放线菌资源,评估新型放线菌分离培养基分离效果。【方法】设计出新型放线菌分离培养基——椰子汁-海藻糖培养基,对广西北部湾茅尾海红树林根际土壤和红树林根皮放线菌进行分离。基于分子生物学鉴定方法,对获得的纯培养放线菌进行多样性分析。【结果】从红树林根际土壤样品中分离获得65株放线菌,分布在5个目10个科15个属,使用YIM171培养基分离到7个属,MA培养基分离到5个属,椰子汁-海藻糖培养基分离到11个属,其中包括4株潜在新种;从红树林根皮样品中分离获得19株放线菌,分布在4个目7个科10个属,使用YIM171和椰子汁-海藻糖培养基分别分离到7个属,包括4株潜在新种;使用椰子汁-海藻糖培养基在红树林生境两类样品中获得3株潜在新种。【结论】广西北部湾茅尾海红树林根际土壤和根皮中放线菌多样性丰富,使用椰子汁-海藻糖培养基分离放线菌效果较佳,可用作放线菌的分离培养基。  相似文献

分离西沙群岛海域放线菌并研究其抗菌活性。采用干燥、辐射、冷冻及加热处理等11种样品预处理方式和10种培养基对海洋放线菌进行分离,对代表性菌株进行鉴定,并考察分离放线菌生长的海水依赖性。进一步以金黄色葡萄球菌、大肠埃希菌、啤酒酵母和扩展青霉为指示菌考察分离放线菌的抗菌活性。从西沙群岛海域样品中分离获得放线菌383株,其中专性海洋放线菌23株。选定93株代表菌株进行鉴定,93株菌隶属于9个科,11个属。不同培养基对分离放线菌菌株的数量及种类影响显著。6株放线菌对4种指示菌均有抑菌活性,其中4株为专性海洋放线菌,表明海洋环境具有丰富的放线菌资源,这些放线菌特别是专性海洋放线菌有望为新型抗菌物质的发现与开发提供菌种来源。  相似文献

【目的】探究含有抗肿瘤活性喜树碱的喜树(Camptotheca acuminata Decne.)种子中内生放线菌的多样性,以及从内生放线菌中寻找新型活性化合物产生菌。【方法】利用放线菌富集培养基分离喜树种子内生放线菌,并根据菌落形态及16S rRNA基因序列同源性分析进行菌种鉴定;以及利用与病原微生物共培养方法检测分离菌株的抗菌活性。【结果】从上海交通大学校园的喜树种子中共分离到33株内生疑似放线菌,经基于16S rRNA基因序列分析的分子鉴定,确定其中21株属于链霉菌,1株属于拟诺卡氏菌,另有3株由于序列相似性低于95%而无法分类;8株因未能克隆16S rRNA基因序列而未确定分类。这11株疑似内生放线菌是否是新放线菌(种)类群,有待后续深入研究。初步的抗细菌、真菌活性检测发现,分离到的内生放线菌中42.42%以上对立枯丝核菌(Rhizoctonia solani Kühn)有很强的抑制菌核形成的作用,54.54%内生菌有较强抑制芽孢杆菌生长的作用。并且挑选其中具有代表性的菌株Streptomyces sp.CXSZ1进行代谢产物的分析,初步发现其代谢产物中含有抗细菌活性成分。【结论】喜树种子内生放线菌的分离、鉴定及生物活性物质分离鉴定的研究工作,一方面可以为揭示喜树等高等植物种子内生菌的起源、演化等提供有价值的信息,同时也为新结构、新活性化合物发掘提供实验材料。  相似文献

Lipases are an important class of enzymes which catalyze the hydrolysis of long chain triglycerides and constitute the most prominent group of biocatalysts for biotechnological applications. There are a number of lipases, produced by some halophilic microorganisms. In this study, some lipase producing bacteria from the Maharla salt lake located in south of Iran were isolated. All isolates were screened for true lipase activity on plates containing olive oil. The lipase activity was measured using titrimetric methods. Among thirty three isolates, thirteen strains demonstrating orange zone around colonies under UV light, were selected for identification using the molecular methods and some morphological characteristics. The bacterium Bacillus vallismortis BCCS 007 with 3.41 ± 0.14 U/mL lipase activity was selected as the highest lipase producing isolate. This is the first report of isolation and molecular identification of lipase producing bacteria from the Maharla lake.  相似文献

【背景】碱蓬(Suaeda glauca Bunge)主要生长于含盐碱土壤,具有果实(种子)二型现象,而种子栖居有众多微生物,它们对种子萌发和幼苗建成具有重要影响。【目的】对碱蓬异型果实表生细菌的群落组成及相关细菌的促生属性进行观察研究,探讨不同类型果实表生细菌的群落组成及相关细菌的促生功能属性是否存在差异。【方法】利用纯培养方法对碱蓬二型果实的表生可培养细菌进行分离纯化,基于16S rRNA基因序列分析初步确定分离菌株的系统发育地位,并对获得菌株进行固氮、溶磷、解钾、产铁及产IAA能力的定性检测。【结果】碱蓬二型果实具有不同的表生细菌群落组成及分布比例,A型果实表生细菌分属于3纲15属,其优势属组成主要是短小杆菌属(34.33%)、芽孢杆菌属(13.43%)和泛菌属(10.45%);B型果实表生细菌分属于4纲20属,其优势属组成主要是短小杆菌属(12.77%)、芽孢杆菌属(17.02%)和冰冻小杆菌属(14.89%)。碱蓬二型果实的表生细菌多样性指数也存在差异,A型果实的Shannon多样性指数、Margalef丰富度指数和Shannon均匀度指数显著低于B型果实;而A型果实的Berger-Parker优势度指数显著高于B型果实。碱蓬二型果实表生细菌在固氮、溶磷、解钾、产铁及产IAA能力等促生属性上的菌株分布比例存在不同变化趋势,A型和B型果实均具有较高比例固氮菌株及较低比例溶磷菌株;但在解钾菌株和产IAA菌株分布比例上具有不同变化趋势,A型果实的解钾菌株所占比例显著高于B型果实,而产IAA能力菌株所占比例显著低于B型果实。【结论】盐生植物碱蓬具有明显不同的2种类型果实,其不同类型果实具有不同的表生细菌群落组成及分布比例,且在促生功能菌株的分布上也存在不同变化趋势。  相似文献

目的:探讨湖南某三甲医院重症监护病房(ICU)患者气管切开后肺部感染病原菌的类型及其耐药性,为临床经验性用药提供帮助。方法:回顾性分析ICU208例气管切开术后并发肺部感染患者的痰细菌培养及药物敏感性测定结果。结果:共分离出420株致病菌,革兰阴性菌293株,占69.76%,其中铜绿假单胞菌98株居首位;革兰阳性菌105株,占25.24%,其中金黄色葡萄球菌47株为最多;真菌占22株,占5.23%。分离出产超广谱B内酰胺酶(ESBLs)菌87株,耐甲氧西林金黄色葡萄球菌(MRSA)24株。所分离致病菌对常用抗菌药物均有不同程度的耐药,且为多重耐药。结论:ICU气管切开患者肺部感染病原菌以革兰阴性菌为主,耐药率高,临床应加强病原学监测,重视细菌的种类分布和耐药趋势,合理使用抗生素。  相似文献

Microorganisms assimilating methane at temperatures above 40 degrees C were isolated from various natural sources: ooze, mud, waste water of coal pits. The bacteria are obligate methylotrophs and are represented by two groups: (a) thermotolerant, growing at 37 to 45 degrees C; and (b) thermophilic, growing at 50 to 62 degrees C. The selective factor used to isolate various physiological forms of methylotrophs is corresponding temperatures of growth which allow to isolate from the same substrate meso-, thermotolerant, and thermophilic forms. Morphological and physiological properties of the strains are described. The thermotolerant cultures of methylotrophs are similar to Methylobacter vinelandii, though differ from it by some characteristics. The thermophilic microorganisms should be classed as a separate species Methylococcus thermophilus.  相似文献

Although some bacteria require an atmosphere with high CO(2) levels for their growth, CO(2) is not generally supplied to conventional screening cultures. Here, we isolated 84 bacterial strains exhibiting high-CO(2) dependence. Their phylogenetic affiliations imply that high-CO(2) culture has potential as an effective method to isolate unknown microorganisms.  相似文献

We used a two‐step enrichment approach to isolate root‐colonizing hexachlorocyclohexane (HCH)‐degrading microorganisms. The first step consists of the use of classical liquid enrichment to isolate γ‐HCH degraders. The γ‐HCH‐degrading microbes were attached in mass to corn seeds sown in soil with γ‐HCH, and after plant development we rescued bacteria growing on root tips. Bacteria were then subjected to a second enrichment round in which growth on liquid medium with γ‐HCH and inoculation of corn seeds were repeated. We then isolated bacteria on M9 minimal medium with γ‐HCH from root tips. We were able to isolate four Sphingomonas strains, all of which degraded α‐, β‐, γ‐ and δ‐HCH. Two of the strains were particularly good colonizers of corn roots, reaching high cell density in vegetated soil and partly removing γ‐HCH. In contrast, these bacteria performed poorly in unplanted soils. This study supports the hypothesis that the removal of persistent toxic chemicals can be accelerated by combinations of plants and bacteria, a process generally known as rhizoremediation.  相似文献

从海南、广西与广东三省的红树林区采集了181个样品,不进行微生物分离而直接作发酵剂接种到发酵培养基进行发酵,取发酵上清液进行抗细菌、抗真菌与肿瘤细胞毒活性的测定。同时对样品进行可培养微生物的分离与生物活性测定。结果显示:不同样品类型的生物活性差异较大。在15个具有强抗活性的样品中,有5个样品分离到的单株菌均无任何生物活性,说明这5个样品的生物活性可能是由微生物的群体作用产生的,也可能是某种没有培养出的微生物产生的。初步表明了探索微生物混合培养获得生物活性代谢产物的可能性。  相似文献

An electron microscope study was conducted on samples of pine bark taken from stacks during consecutive stages of composting. It was found using scanning electron microscopy (SEM) that bacteria, actinomycetes and fungi were present in relatively low numbers on the bark surface before composting was initiated. After addition of urea and water to bark heaps, microbial numbers rose, particularly the bacterial fraction. A large number of actinomycetes were seen below the surface of the bark as composting progressed. Transmission electron microscopy (TEM) of bark in the late stages of composting demonstrated the presence of a variety of microbes within the bark cells. The microorganisms were seen, using SEM, to be degrading the surface of the bark chips, and, using TEM, to be attached to the lignified cell walls. Physiological studies on bacteria isolated at different stages of composting showed they had a number of enzymes such as carboxymethyl cellulase that could aid in the degradation of pine bark. The isolates consisted of Gram-negative and Gram-positive strains, some of which were spore formers. Most of the isolates, including some Gram-negative non-sporing bacteria, were able to grow over a wide range of temperatures from 30 to 60°C, and, in some cases, 70°C.  相似文献

One of the most important reasons of complications after organ transplantation may be the infections. The aim of the present work was to analyse of microorganisms isolated from patients, which were the recipients for kidney transplantation in 2001 year. The diagnostic material contained 140 samples from 53 patients, 40 (22.2%) samples from Euro-Collins fluid used for kidney storage before the transplantation and 3 end-pieces of catheter. The positive cultures were found in 125 (69.4%) samples. Gram-positive bacteria constituted 58.4%, Gram-negative bacteria--34.2%, fungi--7.4%. 140 strains of microorganisms were isolated from pharyngeal swabs and 55 strains of bacteria were isolated from palm swabs. Most of them were considered as a physiological flora. It was found 4-time significant bacteriuria among positive cultures from urine samples. In the cultures of fluid used for kidney storage 12 (30.0%) positive samples were obtained, out of which 16 strains of microorganisms were isolated. Among the strains of Staphylococcus 35.3% were MR. Among 18 strains of Gram-negative rods one strain was multiresistant to antibiotics. None of analyzed strains was ES beta L-producing. A high percentage of positive cultures from fluid used for kidney storage suggests the possibility of contamination of the organ with bacteria coming from kidney donor or during the storage, transport and actions connected with taking the organ to the transplantation.  相似文献

【目的】从新疆尉犁县黑湖中筛选分离获得嗜盐嗜碱微生物,并对筛选获得的微生物进行种属鉴定。【方法】采用传统分离鉴定技术,进行形态和生理生化特性研究和基于16S r RNA基因的序列分析。【结果】从样品中分离获得可培养嗜盐嗜碱菌25株,对其进行鉴定。根据生理生化特征、16S r RNA基因序列测定和系统发育分析表明,25株菌分布在古菌Halorubrum、Haloarcula、Natrialba、Halohasta和Halopiger等5个属。其中优势菌群为Halorubrum,次优势菌群为Natrialba。其中DH-66(KU663028)属于Halopiger属,16S r RNA基因序列同源性与该属的模式菌株Halopiger aswanensis 56T同源性最高,为95.75%,预示为潜在的新种(新种鉴定将另行报道)。25株嗜盐嗜碱菌生长条件实验表明,这些菌适应Na Cl的浓度范围为15%-30%、最适浓度为20%-25%,生长的p H范围为7.0-13.0、最适p H为9.0-10.0。各种水解酶类的分析表明,在分离的25株菌中产淀粉酶的菌有5株占20%、产蛋白酶的菌有4株占16%、产酯酶可水解吐温20的菌有15株占60%、可水解吐温40的有7株占28%、可水解吐温80的有4株占16%、产过氧化氢酶的菌有14株占56%。9株菌同时能产4种酶,2株菌同时能产3种酶。表明了嗜盐嗜碱菌产酶的多样性。19株菌硝酸盐还原为阳性。【结论】揭示了新疆尉犁县黑湖嗜盐嗜碱菌生理生化特性的多样性和系统发育多样性,而且蕴藏着较丰富的新的微生物类群,亟待系统研究和进一步开发利用。  相似文献
