脊髓中血管紧张素Ⅱ的释放及其抗电针镇痛   总被引:9,自引:0,他引:9

本文采用放射免疫分析方法测定不同频率电针引起大鼠脊髓液中血管紧张素Ⅱ(AⅡ)免疫活性(ir)的变化,并观察阿片受体在其中起的作用。结果表明:(1)2Hz电针时大鼠脊髓灌流液中AⅡ-ir减少20%,但无统计学意义;15Hz电针使满足流液中AⅡ-ir降低62%(P〈0.01);100Hz时AⅡ-ir增加60%(P〈0.05)。(2)脊髓蛛网膜下腔(i.t.)注射阿片受体拮抗剂纳洛酮后,15HZ电针反使  相似文献

我室以往的研究表明,不同频率的电针可引起脊髓释放不同种类的阿片肽。本工作观察P物质(SP)释放的频率依赖性,电针频率选择2,4,8,15,30和100Hz,分别收集电针期间及电针前后各30min的脊髓灌流液,通过放射免疫方法测定大鼠电针有效组和电针无效组P物质免疫活性(SP-ir).结果如下:(1)电针有效组:2Hz引起SP-ir降低,与电针前相比,P<0.01;4Hz电针前后SP-ir比较,无统计学意义;8,15,30,100Hz时SP-ir均增加(P<0.01),其中15Hz时SP增加最多(P<0.001),表明刺激引起SP释放有频率依赖性。(2)电针无效组:不论应用何种频率,电针前后脊髓灌流液中SP-ir变化不大(均P>0.05)。提示,电针时脊髓液中SP含量变化与镇痛效果有密切关系。  相似文献

本研究发现,低频(2Hz)电针刺激时大鼠脊髓中P物质免疫活性(SP-ir)含量减少,中频(15Hz)、高频(100Hz)和变频(2/15Hz)刺激时SP-ir含量增多。脊髓蛛网膜下腔(i.t.)注射非肽类SP(NKI)受体拮抗剂CP96345和RP67580均能阻断中频、高频和变频的电针镇痛。i.t.注射阿片拮抗剂纳洛酮阻断低频和中频刺激时SP-ir含量的变化。结果提示,脊髓SP-ir在低频时释放  相似文献

目的和方法:研究肾上腺髓质素(Adm)、降钙素基因相关肽(CGRP)及C-型心房利太(CNP)对碱性成纤维细胞生长因子(bFGF)促血管平滑细胞(VSMC)增殖作用的影响及其机制。结果:孵育24h后,bFGF刺激VSMC增殖较对照组增加2.1倍(P〈0.01),细胞内蛋白磷酸化程度增加1.4倍(P〈0.01),PKC及MAPK活性分别增加1.5和2.5倍(P〈0.010;Adm.CGRPt CNP  相似文献

本实验观察了冠脉内注射降钙素基因相关肽(CGRP0.3μg/kg)对正常及不同程度冠脉狭窄犬的心功能的影响。结果表明正常犬冠脉内注射CGRP后,平均动脉压(MAP)下降1.2kPa(P<0.05),同时,心率(HR)、心输出量(CO)、左室收缩压峰值(LVSP)均不同程度增加;左室舒张末压(LVEDP)轻度降低。在中度狭窄30min后,冠脉内注射CGRP对HR、MAP无明显影响;而重度狭窄后注射CGRP,MAP由狭窄时降低逐渐增高,HR由增快而变慢。CO、LVSP均显著增高,LVEDP降低,此作用较冠脉狭窄前更为明显。提示CGRP扩张冠脉动脉,增加冠脉血流量和心排血量,增强心肌收缩力,对缺血心脏功能有保护作用。  相似文献

本文用免疫组化方法结合计算机图像处理技术观察大鼠后脚掌皮下注射福尔马林后脊髓背角P物质免疫阳性反应(SPLI)变化的节段性分布及中脑导水管周围灰质(PAG)内甲啡肽样免疫阳性反应(MELI)的变化。结果显示,注射福尔马林后,脊髓腰段(L1-2,L4-5)背角SPLI显著增强(P<.05),30min组与60min组相比较无显著变化(P>0.05);胸脊髓(T8)无显著变化(P>0.05);颈脊髓背角SPLI有增强趋势(0.05<P<0.1);PAG中MELI减弱,腹外侧部30min组比60min组变化更大(P<0.05)。PAG中MELI与脊髓背角SPLI变化的时相关系提示福尔马林致痛引起的脊髓背角P物质的增多可能与PAG中甲啡肽及阿片受体活动有关。  相似文献

目的探讨大鼠实验性肝癌发病中刺五加对肌体免疫功能和抗氧化酶活性的影响。方法46只SD雄性大鼠被随机分成对照组(喂普通饲料)、3-甲基4-双甲氨基偶氮苯(3-Me-DAB)组(喂含0.06%3Me-DAB饲料 10周)和刺五加组(饲喂同 3-Me-DAB外、另加入刺五加 4.5g/kg饲料,用常规方法检测全血谷光甘肽过氧化物酶(GSH-PX)、血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,用微量化学发光造检测吞噬细胞活性(PMN-CL)。结果1.PMN-CL检测峰值、积分值和吞噬细胞指数,3-MeDAB组较正常组和刺五加组均有显著升高(P<0.05和P<0.01)2.全血GSH-PX活性、SOD活性,刺五加组较3-MeDAB组均有显著升高(P<0.05)。MDA含量刺五加组和3-MeDAB组均较正常组升高(均P<0.05)。结论刺五加在大鼠实验性肝癌诱发过程中有提高抗氧化酶活性和对抗致癌剂引起的机体中性粒细胞吞噬功能代偿性增高的作用。  相似文献

本实验应用形态学、免疫组化染色,^3H-TdR掺入、流式细胞等技术探讨缺氧(〈1%O2和2.5%O2)对肺动脉平滑肌细胞(PASM)形态及增殖的影响。结果表明:缺氧24h使PASM表型从收缩型向合成型转换,胞浆内SM-a actin减少,线粒体和粗面内质网增多,缺氧48h以后线粒体肿胀,空泡化,内质网扩张,胞浆内出现髓鞘样结构。流式细胞分析显示缺氧PASM G2/M期细胞比例明显增多(P〈0.00  相似文献

Alleviating opiate withdrawal syndrome in addicts is a critical precondition to break away from drug and further to prevent reuse. Electroacupuncture (EA) was claimed to be effective for alleviating withdrawal syndrome, but the optimal protocol remained unclear. In the present study we found that (1) 100 Hz EA administered 12-24 h after the last morphine injection suppressed the withdrawal syndrome in rats, multiple sessions of EA were more effective than single session, with the after-effect lasting for at least 7 days. (2) A down-regulation of preprodynorphin (PPD) mRNA level was observed in spinal cord, PAG and hypothalamus 60 h after the last morphine injection, which could be reversed by multiple sessions, but not a single session of EA. (3) Accompanied with the decrease of PPD mRNA level, there was an up-regulation of p-CREB in the three CNS regions, which was abolished by 100 Hz EA treatment. The findings suggest that down-regulation of p-CREB and acceleration of dynorphin synthesis in spinal cord, PAG and hypothalamus may be implicated in the cumulative effect of multiple 100 Hz EA treatment for opioid detoxification.  相似文献

Previous studies have shown that rats subjected to low or high frequency electroacupuncture (EA) stimulation release enkephalins or dynorphins respectively to produce analgesia. This conclusion was tested in the present study by using cross tolerance technique for further analysing their receptor mechanisms. The main results were as follows: (1) In rats subjected to 2 Hz EA for 6 h, there was a gradual decrease in the analgesic effect, leading to a state of tolerance to 2 Hz EA analgesia. These rats, however, still responded to 100 Hz EA. Likewise, rats made tolerant to 100 Hz EA were still effective to 2 Hz EA stimulation, showing not significant cross tolerance between 2 Hz and 100 Hz EA analgesia. (2) Rats made-tolerant to 100 Hz EA analgesia showed a diminished response to intrathecal dynorphin A (1-13), a kappa agonist, whereas the analgesic effect of the delta agonist [D-Pen2, D-pen5] enkephalin (DPDPE) remained intact. (3) Rats made tolerant to 2 Hz EA analgesia showed a cross tolerance to DPDPE, but not to dynorphin A (1-13). Results obtained from aforementioned cross tolerance studies suggest that 2 Hz and 100 Hz EA analgesia are mediated by delta and kappa opioid receptors, respectively, at the spinal cord of the rat.  相似文献

Left ventricular (LV) end-diastolic pressure (LVEDP) increase due to volume expansion (VExp) enhances mechanosensitive vagal cardiac afferent C-fiber activity (CNFA), thus decreasing renal sympathetic nerve activity (RSNA). Hypotensive hemorrhage (hHem) attenuates RSNA despite decreased LVEDP. We hypothesized that CNFA increases with any change in LVEDP. Coronary perfusion pressure (CPP), supposedly affected in both conditions, might also be a stimulus of CNFA. VExp and hHem were performed in anesthetized male Sprague-Dawley rats while blood pressure, heart rate, and RSNA were measured. Cervical vagotomy abolished RSNA response in both reflex responses. Single-unit CNFA was recorded while LVEDP was changed. Rapid changes (+/- 4, +/-6, +/-8 mmHg) were obtained by graded occlusion of the caval vein and descending aorta. Prolonged changes were obtained by VExp and hHem. Furthermore, CNFA was recorded in a modified Langendorff heart while CPP was changed (70, 100, 40 mmHg). Rapid LVEDP changes increased CNFA [caval vein occlusion: +16 +/- 3 Hz (approximately +602%); aortic occlusion: +15 +/- 3 Hz (approximately +553%); 70 units; P < 0.05]. VExp and hHem (n = 6) increased CNFA [VExp: +10 +/- 4 Hz (approximately +1,033%); hHem: +10 +/- 2 Hz (approximately +1,225%); P < 0.05]. An increase in CPP increased CNFA [+2 +/- 1 Hz (approximately +225%); P < 0.05], whereas a decrease in CPP decreased CNFA [-0.8 +/- 0.4 Hz (approximately -50%); P < 0.05]. All C fibers recorded originated from the LV. CNFA increased with any LVEDP change but changed equidirectionally with CPP. Thus neither LVEDP nor CPP fully accounts directly for afferent C-fiber and reflex sympathetic responses. The intrinsic afferent stimuli and receptive fields accounting for reflex sympathoinhibition still remain cryptic.  相似文献

目的:观察P物质(Substance P,SP)在慢性坐骨神经压迫损伤(chronic constriction injury,CCI)模型脊髓中表达的变化,探讨电针镇痛的机制是否与脊髓背角中SP表达的变化有关。方法:选择32只雄性、体重180-200 g的SD大鼠,并将其随机均分为4组(n=8)。空白组(Con组)为正常痛阈值大鼠;假电针组(CCI+A组)在损伤的坐骨神经旁置入电针,但无电流刺激;2 Hz组和100Hz组分别给予相应频率电流刺激30 min。在实验开始前和术后1、4、7、14、20、22天记录大鼠的热缩足反射潜伏期(Paw Withdrawal Latency,PWL)和机械刺激缩足反射阈值(Paw Withdrawal Threshold,PWT)。免疫组化方法检测脊髓背角SP的表达。结果:术后20天,电针治疗后,100 Hz组和2 Hz组PWT分别为(7.33±1.42)g和(7.80±1.42)g,均显著高于假电针组(2.60±1.46)g,差异有统计学意义(P0.05)。100 Hz组在术后20天后和2 Hz组在术后14天后PWL值均显著高于假电针组,差异有统计学意义(P0.05)。免疫组化显示:2 Hz组和100 Hz组大鼠脊髓背角中P物质阳性细胞显著低于假电针组(P0.05)。结论:坐骨神经旁电针刺激能够显著减轻CCI模型大鼠热痛觉及机械痛觉过敏,其机制可能与抑制脊髓背角SP的表达有关。  相似文献

Adrenomedullin (AM) is a potent vasodilator peptide whose major source is the vascular wall. In the present study, the mechanism of release of AM was investigated in the rat mesenteric resistance artery. The isolated mesenteric vascular bed was perfused with Krebs solution at a constant flow rate (5 ml/min) and AM in the perfusate was measured by a highly sensitive enzyme immunoassay (Immunoenzymometric assay; IEMA) method. In preparations without endothelium, spontaneous release of AM was detected in the perfusate (68.7+/-5.8 fmol/ml, n=45). Periarterial nerve stimulation (PNS, 4 and 8 Hz) caused 11.4+/-3.9% (4 Hz) and 9.1+/-3.5% (8 Hz) decreases in the spontaneous release of AM. Removal of Ca2+ from the medium did not affect the spontaneous AM release, but abolished the PNS-induced inhibition of spontaneous AM release. Perfusion of 10nM calcitonin gene-related peptide (CGRP) or 0.1 microM capsaicin (inducer of CGRP release) inhibited significantly the spontaneous AM release. PNS (8 Hz)-induced inhibition of spontaneous AM release was antagonized by CGRP(8-37) (CGRP receptor antagonist). These results suggest that AM is mainly released from vascular smooth muscle cells of the rat mesenteric artery and endogenous or exogenous CGRP inhibits AM release.  相似文献

The neuropeptide calcitonin gene-related peptide (CGRP) may play a role in neurogenic inflammation, tissue remodeling of the uterine cervix, promoting vasodilation, parturition, and processing of sensory information in the spinal cord. CGRP-immunoreactive nerves of the cervix and spinal cord have been studied but cellular identification of the CGRP receptor has received little attention. CGRP-receptor component protein (CGRP-RCP) is a small protein associated with the CGRP receptor; thus, immunostaining for the CGRP-RCP can be used to identify sites of the CGRP receptor. We determined sites of CGRP-RCP immunoreactivity relative to the presence of CGRP-ir nerve fibers in the female rat uterine cervix, spinal cord, and dorsal root ganglia. CGRP-RCP immunoreactivity was expressed in the dorsal horn of the spinal cord, venules of the uterine cervix, and perikarya of sensory neurons in dorsal root ganglia. CGRP-immunoreactive fibers were adjacent to CGRP-RCP-immunoreactive vessels in the cervix and among CGRP-RCP-immunoreactive structures in the dorsal horn of the spinal cord. This suggests CGRP-RCP is associated with structures innervated by CGRP nerves and these interactions may be changed in tissues in response to an appropriate stimulus.  相似文献

目的:比较即刻电针天枢穴、足三里穴对肠易激综合征(ms)模型大鼠血浆降钙基因相关肽(CGRP)、内皮素(ET)水平及结肠组织中内皮素受体A(ETR-A)、CGRPmRNA表达的影响,旨在探讨电针即刻治疗IBS的部分机制。方法:采用WISTAR幼鼠制备肠易激综合征模型,随机分为空白对照组、模型组、天枢组、足三里组,每组8只。空白对照组不作任何处理,模型组只束缚不针刺,天枢组和足三里组在实验第8周电针治疗一次,留针20min。治疗结束后处死大鼠,取大鼠血浆及部分结肠组织进行生物活性物质检测。采用酶联免疫法检测血浆中CGRP、ET、结肠组织中ETR—A的含量,采用RT—PCR法检测结肠组织中CGRPmRNA表达。结果:(1)即刻电针对IBS模型大鼠血浆CGRP、ET水平的影响:与空白对照组比较,模型组CGRP水平明显降低(P〈0.01);与模型组比较,天枢组、足三里组CGRP水平明显升高(P〈0.01)。与空白对照组比较,模型组ET水平升高(P〈0.05);与模型组比较,天枢组ET水平明显降低(P〈0.01)。(2)即刻电针对IBS模型大鼠结肠组织ETR—A水平的影响:与空白对照组比较,模型组ETR—A水平明显升高(P〈0.01);与模型组比较,足三里组ETR-A水平明显降低(P〈0.01)。(3)即刻电针对IBS模型大鼠结肠组织CGRPmRNA表达的影响:与空白对照组比较,模型组、天枢组、足三里组CGRPmRNA表达明显增强(P〈0.01,P〈0.05);与模型组比较,足三里组CGRPmRNA表达减弱(P〈0.05)。结论:即刻电针介入后,能够调节机体的内环境紊乱和CGRP、ET的平衡失调。这种调节作用因穴位不同而具有不同的特点,天枢穴对血浆中CGRP、ET调节作用较强,足三里穴在受体和基因表达方面作用明显。  相似文献
