杂合抗菌肽CecA-mil的改造及在毕赤酵母中的分泌表达   总被引:13,自引:0,他引:13  
参照毕赤氏巴斯德酵母(Pichia pastorts)偏好密码子,改造并化学合成杂合抗菌肽CecA-mil基因,改造后的CecA-mil基因克隆到pPICZα-A载体中,构建分泌型重组酵母表达载体pPICZα-A-CM,转化Pichia pastoris受体菌X-33。在醇氧化酶(AOX)启动子调控下,分子量约1.9kD的CecA-mil杂合抗菌肽获得表达,经表达条件优化,重组酵母菌的摇瓶发酵产率可达到245μg/mL。抗菌特性研究表明,该表达产物具有广谱抗菌活性,对多数G^-菌及G^ 菌均有较好的抑菌活性,特别是对氨苄青霉素抗性菌和卡那霉素抗性菌抑杀效果更好;具有热稳定性和酸稳定性。这些特点使得重组抗菌肽CecA-mil在食品防腐、疾病防治和动物饲料添加剂等方面显露出很好的应用前景。  相似文献

根据抗菌肽天蚕素A(cecropinA,CA)N端第1~7个氨基酸残基,马盖宁(magainin,M)N端第2~12个氨基酸残基,以毕赤酵母偏爱的密码子设计合成了杂合肽CA(1~7)-M(2~12)基因,同载体pPICZα-A连接后转化Pichia pastoris受体菌SMD1168,在醇氧化酶(AOX)启动子调控下,分子量约1.9kDa的CecA-Mag杂合抗菌肽获得表达,抗菌特性研究表明,该表达产物具有广谱抗菌活性,对多数G-菌及G 菌均有较好的抑菌活性。初步抑菌活性测定,显示该杂合肽对金黄色葡萄球菌、耐氨苄青霉素的大肠杆菌及枯草芽孢杆菌有良好的抑杀活性。酸稳定实验显示pH为3.2时仍具有相当高的活性。热稳定性实验显示该杂合肽100℃加热5min后仍具有抑菌活性。这些特点使得重组抗菌肽CecA-mag在疾病防治和动物饲料添加剂等方面显露出很好的应用前景。  相似文献

选用毕赤酵母偏爱密码子 ,设计合成了新型抗菌肽基因。所合成的magainin基因和cecA mil杂合肽基因全长分别为 1 0 1bp和 60bp ,并在其N端引入kex2裂解位点 ,以保证表达抗菌肽具有天然N端。其中 ,cecA mil杂合肽基因根据cecropinAN端第 1~ 7个氨基酸残基、melittinN端第 5~ 1 2个氨基酸残基所设计合成。基因分别克隆入pPICZα A质粒 ,构建分泌型重组酵母表达载体pPICZα A mag和pPICZα A CM。在AOX1 (醇氧化酶 )启动子调控下 ,类似天然抗菌肽大小的magainin及cecA mil蛋白获得分泌表达 ,其表达量分别为 1 0 5mg L和 1 1 8mg L。初步抑菌活性测定 ,显示两者对金黄色葡萄球菌及E .coliDH5α有较好的抑杀活性。  相似文献

为了在毕赤酵母中获得抗菌肽CM4表达,将抗菌肽CM4基因克隆入表达质粒pPIC9,SacⅠ线性化的重组表达质粒pPIC9-CM4电击转化P.pastorisGS115(his-),采用MM、MD板和PCR法筛选Mut 表型,甲醇诱导表达;抗菌活性检测、Tricine-SDS-PAGE及酸性活性电泳均表明抗菌肽CM4在毕赤酵母中成功地分泌表达;重组抗菌肽CM4对黑曲霉(Aspergillus niger)、绿色木霉(Trichoderma viride)都有很强的抑菌活性。  相似文献

目的:在毕赤酵母SMD1168中表达融合抗菌肽,并检测其体外抑菌活性。方法:本实验从实验室先前构建的重组质粒pVAX1-RHKJT中克隆出已构建好的融合抗菌肽RHKJT基因片段,将RHKJT基因片段插入至pGAPZaA真核表达质粒中,通过PCR和测序验证,构建pGAPZα-RHKJT重组真核表达质粒,将线性化的pGAPZα-RHKJT电转化至毕赤酵母SMD1168中获得重组酵母SMDp G-RHKJT,并通过PCR和RT-PCR验证,对重组毕赤酵母SMDpG-RHKJT进行发酵,并收集发酵上清液进行体外生物活性测定。结果:成功获得重组酵母SMDpG-RHKJT菌株,重组酵母发酵上清液对大肠杆菌标准菌、大肠杆菌耐药菌、沙门氏菌标准菌、金黄色葡萄球菌标准菌、金黄色葡萄球菌耐药菌、肺炎链球菌标准菌均具有显著的抑菌活性。结论:重组酵母表达的融合抗菌肽具有较广的抗菌谱和较高的抑菌活性,具有良好的潜在应用前景。  相似文献

贻贝素(Mytilins)是一类主要在贝类血细胞中表达的阳离子小分子抗菌肽,包括多种同工型,它们在免疫系统中发挥了重要作用,Mytilin-1是其中的一种同工型。厚壳贻贝(Mytilus coruscus)Mytilin-1的一级结构由信号肽、成熟肽和C端的延伸肽组成;其成熟肽决定了Mytilin-1的生物学活性,它由34个氨基酸残基组成,其中8个保守的半胱氨酸残基在空间上能形成4对二硫键,对Mytilin-1的稳定性和活性起了关键作用。以厚壳贻贝的Mytilin-1成熟肽为重组DNA表达的目的蛋白,根据毕赤酵母(Pichia pastoris)的密码子偏爱性对编码该成熟肽的密码子进行优化,合成的目的基因"mMy1"与表达载体pPICZαA连接后电转至毕赤酵母X-33,通过高浓度博来霉素筛选高拷贝酵母转化子,获得的阳性转化子在29℃、250 r/min、pH6.0的条件下,使用1%甲醇诱导表达96 h;培养液上清经Tricine-SDS-PAGE和Western blot分析证明:重组mMy1在毕赤酵母X-33中得到成功表达,其分子量为4.8 kD。抑菌试验表明,重组mMy1对革兰氏阳性的金黄色葡萄球菌(Staphylococcus aureus)和枯草芽孢杆菌(Bacillus subtilis)以及革兰氏阴性的大肠杆菌(Escherchia coli)具有抑菌活性。  相似文献

杂合抗菌肽在毕赤酵母中的表达及其活性测定   总被引:3,自引:0,他引:3  
为获得溶血活性低、抗菌活性高的杂合抗菌肽,以家蝇抗菌肽Cec Md和中国林蛙抗菌肽Chensirin为母体肽,并结合毕赤酵母偏爱密码子的原则,设计出6条具有抗菌潜力的新型杂合抗菌肽,将其命名为CC22、CC28、CC29、CC30和CC34(1),CC34,利用SOE-PCR技术合成所需的目的基因,并将其克隆至毕赤酵母表达载体pGAPZαA,通过电击转化技术,将其转化至毕赤酵母SMD1168中,经含有Zeocin的抗性平板筛选阳性转化子,YPD液体培养72h后,经Tricine-SDS-PAGE检测出目的蛋白,然后采用高效液相色谱法对其进行纯化。检测结果显示,表达产物CC29对大肠杆菌、鸡沙门氏菌的最小抑菌浓度(MIC)均为25μg/ml;CC34(1)对大肠杆菌表现相对较弱的抑制作用,最小抑菌浓度为100μg/ml;CC34对鸡沙门氏菌和金黄色葡萄球菌的最小抑菌浓度为50μg/ml;且杂合抗菌肽对有益菌均没有表现出抑制作用。6条杂合肽的溶血活性均呈现较低水平,其中表现出抗菌活性的3条抗菌肽中,以CC29的溶血活性最低,CC34(1)和CC34相对次之。结合抑菌活性,CC29和CC34的抑菌效果较为明显,从而确定溶血活性低且抗菌活性较高的CC29和CC34为新型杂合抗菌肽。  相似文献

目的:用毕赤酵母真核系统表达有抑菌活性的家蚕抗菌肽(cecropin-XJ)。将pGEX-4T-1-cecropin-XJ上的抗菌肽基因cecropin—XJ克隆至穿梭质粒pSuperY上,用Bln Ⅰ酶切使之线性化后,采用电击法转化酵母SMD1168,转化子用小瓶发酵,经SDS—PAGE检测,表达产物可以在α信号因子的引导下,分泌到培养基中,且表达产物具有明显抑菌活性。  相似文献

Mytimacin是主要在无脊椎动物中表达的Macin抗菌肽家族中的一员,具有较强的抗病原微生物活性,是利用重组DNA技术开发天然抗菌剂的良好候选者。通过RT-PCR从青蛤(Cyclina sinensis)闭壳肌中克隆编码Mytimacin成熟肽的基因,经3次PCR在该基因的5’端添加Xho I限制性酶切位点和信号肽酶识别位点、3’端添加Xba I限制性酶切位点和6×His,获得目的基因"CsMm";以pPICZαA为表达载体、毕赤酵母(Pichia pastoris)X-33为工程菌,构建重组毕赤酵母X-33/pPICZαA-CsMm。通过高浓度博来霉素筛选高拷贝酵母转化子,在28℃、250 r/min条件下,使用1.5%的甲醇诱导表达72 h;使用固化金属离子亲和层析(IMAC)对表达产物进行纯化,并通过MALDI-TOF-TOF质谱分析对纯化产物进行鉴定。另外,通过涂布法和浊度法考察重组CsMm的抑菌活性。结果表明:基于X-33/pPICZαA-CsMm重组毕赤酵母的外源表达获得了表达量为25.6 mg/L的重组蛋白,经MALDI-TOF-TOF质谱鉴定其为分子量约7.8 kD的预期重组CsMm。抑菌试验证明重组CsMm对金黄色葡萄球菌(Staphylococcus aureus)、枯草芽孢杆菌(Bacillus subtilis)、大肠杆菌(Escherichia coli)和副溶血性弧菌(Vibrio Parahemolyticus)具有明显的抑菌活性。构建的重组毕赤酵母X-33/pPICZαA-CsMm能有效合成具有生物学活性的重组青蛤Mytimacin,旨为贝类来源天然小分子抗菌剂的开发提供可资参考的技术途径。  相似文献

利用毕赤酵母菌株表达家蝇抗菌肽domesticin基因并检测其抑菌活性。克隆家蝇domesticin基因与pPIC9k质粒相连,构建重组表达载体p PIC9K-domesticin,将其电击转化入毕赤酵母KM71中。甲醇诱导后利用Tricine-SDS-PAGE及Western Blot检测融合蛋白的表达,通过最小抑菌浓度测定表达产物的抑菌活性。结果显示家蝇抗菌肽Domesticin在毕赤酵母中成功表达,抑菌实验表明Domesticin对多种细菌具有抑制作用。Domesticin是一种对受试革兰氏阳性菌和革兰氏阴性菌均具有抑菌活性的广谱新型抗菌肽,有望成为新一代抗菌剂。  相似文献

Antibacterial peptides from various sources express different antibacterial activity. In order to obtain a high activity antibacterial peptide, the sequences of four antimicrobial peptides--Protegrin-1, 4 kDa Scorpion Defensin, Metalnikowin-2A and Sheep Myeloid Antibacterial Peptide SMAP-29--were exploited to generate a synthetic antimicrobial peptide cp gene, which was then cloned into the expression vector pPICZalpha-A. The constructed recombinant expression vector pPICZalpha-cp was transformed into Pichia pastoris X-33, in which the synthetic antimicrobial peptide (CP) could be expressed under the control of the inducible AOX1 promoter and secreted via the alpha mating factor leader of Saccharomyces cerevisiae. Results showed that recombinant plasmid is highly stable, and In vitro experiments showed that the recombinant antimicrobial peptide CP is heat and acid-stable, and it has high antibacterial activity against several Gram-positive and -negative bacteria. Only 1 microg of the recombinant antimicrobial peptide CP has an antibacterial activity equivalent to 64 U ampicillin. Thus, this recombinant antimicrobial peptide could serve as an attractive candidate for the development of therapeutic antimicrobial drugs.  相似文献

防御素是一类阳离子抗菌肽。研究从石斑鱼垂体SMART cDNA 文库中扩增出129 bp 石斑鱼-防御素成熟肽序列, 将其克隆到毕赤酵母表达载体pPCIZA 中, 构建了石斑鱼-防御素的真核表达载体, 电击转化毕赤酵母GS115。Western Blot 分析表明石斑鱼-防御素在酵母菌中获得了表达。体外抗菌实验表明纯化的重组蛋白具有抑制大肠杆菌以及嗜水气单胞菌的作用, 但是对革兰氏阳性菌, 如金黄色葡萄球菌和藤黄微球菌的生长没有抑制作用。实验结果表明酵母表达的石斑鱼-防御素能够特异地抑制革兰氏阴性菌的生长。    相似文献

In Drosophila, the response against various microorganisms involves different recognition and signaling pathways, as well as distinct antimicrobial effectors. On the one hand, the immune deficiency pathway regulates the expression of antimicrobial peptides that are active against Gram-negative bacteria. On the other hand, the Toll pathway is involved in the defense against filamentous fungi and controls the expression of antifungal peptide genes. The gene coding for the only known peptide with high activity against Gram-positive bacteria, Defensin, is regulated by both pathways. So far, survival experiments to Gram-positive bacteria have been performed with Micrococcus luteus and have failed to reveal the involvement of one or the other pathway in host defense against such infections. In this study, we report that the Toll pathway, but not that of immune deficiency, is required for resistance to other Gram-positive bacteria and that this response does not involve Defensin.  相似文献

Human antimicrobial peptide CAP18/LL37 (hCAP18/LL37) was expressed in Pichia pastoris and its antibacterial activity was tested against pathogenic bacteria. The full length ORF of hCAP18/LL37 was cloned into the pPICZaA vector followed by integration into the genomic AOX1 gene of P. pastoris. Agar diffusion assay demonstrated that the different hCAP18/LL37 transformants showed various antibacterial activities against Staphylococcus aureus, Micrococcus luteus, and Salmonella gastroenteritis. The secreted form of hCAP18/LL37 exhibited its maximum activity after 72 h incubation with 2% methanol in MM media, not in BMM. This result suggests that the yeast secreted expression system can be used as a production tool of antimicrobial peptides for industrial or pharmaceutical application.  相似文献

抗菌肽是昆虫感染细菌后,由血细胞及脂肪细胞在瞬时分泌的抗菌物质。具有广谱抗菌活性和抑杀耐药菌株等优点。抗菌肽不仅抗菌谱广,而且对某些真菌、原虫、病毒及肿瘤有一定的杀灭和抑制作用,还能加速免疫和伤口愈合过程。防御素是一类在自然界中广泛存在的、具有微生物抗性的小分子抗菌肽。利用PCR技术,以果蝇总DNA为模板,扩增出两端加入了连接接头的防御素基因,将接头处理成粘性末端,将防御素基因与分泌型酵母表达载体pHBM905B重组,构建重组酵母表达载体pHBM905B/defensin,经"三明治"夹心平板筛选及菌落PCR鉴定证明目的基因已整合入酵母染色体中。挑选阳性克隆经甲醇诱导表达,以SDS-PAGE电泳对表达产物进行分析,证明表达蛋白的相对分子量约为10kD,与预期结果一致。用琼脂糖扩散法检测上清液的生物学活性,可以观察到明显的抑菌圈,显示其具有较强的抗菌活性。同时表达产物有极强的热稳定性,展示了诱人的应用前景,为进一步的开发研究奠定了基础。  相似文献

Thanatin, a 21-residue peptide, is an inducible insect peptide with a broad range of activity against bacteria and fungi. It has a C-terminal disulfide loop, like the frog skin secretion antimicrobial peptides of the brevinin family. In this study, we tried to find the effect of a number of amino acids between the disulfide bond. Thanatin showed stronger antibacterial activity to Gram negative bacteria than other mutants, except Th1; whereas, the mutant peptides with deletion had higher activity to Gram positive bacteria than thanatin. An increase in the number of amino acid(s) using the alanine residue decreased the antibacterial activity in all of the bacteria. Th1 with deletion of threonine at position 15 (Thr(1)(2)) showed similar antibacterial activity against Gram-negative bacteria, but had higher activity against the Gram positive bacteria. In order to study the structure-function relationship, we measured liposome disruption by the peptides and CD spectra of the peptides. Th1 also showed the highest liposome leaking activity and alpha-helical propensity in the sodium dodecyl sulfate solution, compared with other peptides. Liposome disruption activity was closely correlated with the anti-Gram positive bacterial activity. All of the peptides showed no hemolytic activity. A novel antimicrobial peptide, named Bicarinalin, has been isolated from the venom of the ant Tetramorium bicarinatum. Its amino acid sequence has been determined by de novo sequencing using mass spectrometry and by Edman degradation. Bicarinalin contained 20 amino acid residues and was C-terminally amidated as the majority of antimicrobial peptides isolated to date from insect venoms. Interestingly, this peptide had a linear structure and exhibited no meaningful similarity with any known peptides. Antibacterial activities against Staphylococcus aureus and S. xylosus strains were evaluated using a synthetic replicate. Bicarinalin had a potent and broad antibacterial activity of the same magnitude as Melittin and other hymenopteran antimicrobial peptides such as Pilosulin or Defensin. Moreover, this antimicrobial peptide has a weak hemolytic activity compared to Melittin on erythrocytes, suggesting potential for development into an anti-infective agent for use against emerging antibiotic-resistant pathogens.  相似文献

用热灭活的大肠杆菌DHSQ诱导粉纹夜蛾(Trichoplusia ni)离体细胞产生抗菌肽,用三氯乙酸沉淀法提取出该活性物质,采用琼脂糖孔穴扩散法和生长抑制测定法测定其抗菌谱,发现该抗菌物质具有较广的抗微生物活性,其中特别是对革兰氏阴性菌中的沙门氏茵和大肠杆茵,酵母菌中的白色念珠菌,植物病源真茵中的花生白绢病茵和小麦赤霉病茵具有较强的抑菌活性,从而表明该物质是一种既抗细菌,又抗真菌的抗微生物肽。  相似文献
