一株棘孢曲霉的鉴定及其柚苷酶合成规律   总被引:1,自引:0,他引:1  
【目的】对一株新分离的柚苷酶产生菌株JMUdb058进行鉴定,并研究该菌株柚苷酶合成的基本规律。【方法】利用形态观察、28S rDNA序列分析对JMUdb058进行鉴定;通过反相高效液相色谱测定JMUdb058粗酶液对柚皮苷的水解作用,鉴定其柚苷酶活性;分别用11种碳源和6种氮源进行摇瓶发酵,研究碳源、氮源对该菌株分泌柚苷酶的影响;在固态和液态条件下分别进行发酵,测定该菌株合成柚苷酶的能力。【结果】菌株JMUdb058的菌落形态和显微形态符合曲霉属黑色组的典型形态特征,其28S rDNA序列与棘孢曲霉(Aspergillus aculeatus)的相似度达100%;用液态和固态两种发酵方法制得的粗酶液均可将标准溶液中的柚皮苷水解产生普鲁宁和柚皮素,还可有效地水解琯溪蜜柚汁中的柚皮苷;用橘皮苷、柚皮苷、芸香苷和鼠李糖为碳源,胰蛋白胨和豆饼粉等有机物为氮源时可分泌柚苷酶;该菌种在固态发酵中表现出很强的柚苷酶发酵能力,用HPLC法测定的α-L-鼠李糖苷酶和柚苷酶活力分别达到5903和1939 U/gds,用Davis法测定的柚苷酶活力达到72232 U/gds。【结论】本研究首次发现棘孢曲霉能够分泌柚苷酶,含鼠李糖基团的物质可作为其产柚苷酶的诱导物。棘孢曲霉JMUdb058固态发酵产柚苷酶的能力突出,是一种高产柚苷酶的微生物新资源。  相似文献

【目的】以柚皮为原料,优化棘孢曲霉利用柑橘加工副产物固态发酵柚苷酶的条件。【方法】采用高效液相色谱法检测酶活力,通过单因素试验考察固水比、装样量、接种量、温度对柚苷酶发酵的影响,用正交试验优化发酵条件。【结果】单因素试验结果的显著性分析表明培养基的固水比、装样量和培养温度对柚苷酶产量有显著性影响,而接种量影响不显著;经正交试验确定的优化条件是:固水比1:1 (质量体积比),装样量5 g/250 mL三角瓶,温度为30 °C,接种1 mL孢子悬浮液,发酵8 d。在此优化条件下,柚苷酶酶活力为8.19 IU/g干物质,比初始培养基产柚苷酶活力提高7.38倍。【结论】通过对固水比、装样量和发酵温度进行优化,大幅度提高了棘孢曲霉固态发酵柑橘加工副产物的柚苷酶产量,为柚苷酶的生产提供了一种高产发酵工艺。  相似文献

【目的】从健康人肠道微生物菌群中分离能将柚皮苷高效转化为柚皮素的特定细菌菌株,对分离得到的菌株进行菌种鉴定并研究菌株对柚皮苷转化特性,目的是为柚皮素的高效生物合成提供新细菌资源。【方法】取健康人新鲜粪样,在厌氧工作站内培养24 h后进行梯度稀释并涂板,从板上挑取单菌落与底物柚皮苷进行厌氧培养,用高效液相色谱检测底物被转化情况。根据16S rDNA序列及相关生理生化特性分析,对所分离的柚皮苷转化菌进行菌种鉴定,并测定转化菌株对底物柚皮苷的转化动态和转化能力。【结果】从人肠道菌群中分离得到4株能将柚皮苷转化为柚皮素的细菌菌株,分别命名为AUH-JLD3、AUH-JLD7、AUHJLD104和AUH-JLD109。根据16S rDNA序列分析,结合形态学及相关生理生化特性等,将其分别鉴定为布劳特氏菌(Blautia sp.AUH-JLD3)、肠球菌(Enterococcus sp.AUH-JLD7)、拟杆菌(Bacteroides sp.AUHJLD104)和巴氏链球菌(Streptococcus pasteurianus subsp.AUH-JLD109)。转化动态研究结果表明,所分离的4株细菌菌株均能在12 h内将0.2 mmol/L底物柚皮苷转化为柚皮素;转化能力测定结果显示,菌株AUHJLD3、AUH-JLD7、AUH-JLD104及AUH-JLD109能够高效转化底物柚皮苷的最大浓度分别为0.2 mmol/L(平均转化率66.67%)、0.8 mmol/L(平均转化率86.49%)、0.2 mmol/L(平均转化率73.68%)以及1.6 mmol/L(平均转化率93.20%)。【结论】本研究首次从人肠道菌群中分离得到4株能将柚皮苷转化为柚皮素的细菌菌株,其中巴氏链球菌AUH-JLD109对底物柚皮苷转化能力最强。  相似文献

【目的】建立采用3,5-二硝基水杨酸(DNS)法快速测定柚皮苷水解率的方法,并利用该方法对柚苷酶催化水解柚皮苷生成柚皮素的反应过程进行优化研究。【方法】利用棘孢曲霉JMUdb058发酵得到的柚苷酶催化水解柚皮苷,采用DNS法对柚皮苷酶解过程中还原糖的生成量进行分析,经过换算得到柚皮苷的水解率,并在此基础上通过单因素实验优化柚皮苷的酶解过程。【结果】在柚皮苷的水解过程中,还原糖的生成量与柚皮苷的水解量及柚皮素的生成量均呈现出良好的线性关系,因此可利用DNS法测定体系中还原糖的生成量,并通过换算得到柚皮素的生成量。利用该方法优化柚皮苷的酶解过程得到柚苷酶转化柚皮苷的最适温度为50°C、pH为5.0、酶用量为8 U/mL、底物浓度为0.2 g/100 mL。在此条件下,柚皮苷酶解150 min后可达到平衡,此时其水解率为85%。通过Lineweaver-Burk双倒数作图法测得Km为  相似文献

为了建立柚皮固态发酵产柚苷酶的工艺以及阐明其柚苷酶的应用价值,利用棘孢曲霉Aspergillus aculeatus JMUdb058对柚皮进行固态发酵,并研究其产酶特性。采用高效液相色谱法检测酶活力,通过单因素实验方法研究豆饼粉及培养基灭菌对柚苷酶发酵的影响;采用酶活力、还原糖以及生物量的变化解析发酵的动态规律;采用柚皮苷的水解活性表征柚苷酶对柑橘果汁的脱苦效果。结果表明,棘孢曲霉JMUdb058固态发酵柚皮可产生柚苷酶,额外添加豆饼粉可大幅度提高其柚苷酶活力;对培养基进行121℃20min的灭菌将导致柚苷酶活力急剧下降;当培养基组成为5g柚皮粉、0.75g豆饼粉、5.75mL蒸馏水,经30℃发酵8d时,柚苷酶和α-L-鼠李糖苷酶活力分别达到5.81IU/gds和6.08IU/gds;通过对产酶进行动力学拟合,发现柚苷酶和α-L-鼠李糖苷酶主要在棘孢曲霉的对数生长期内合成,它们的积累属于生长关联型;该柚苷酶可高效水解柚汁中的柚皮苷,柚皮苷去除率高达99.6%。因此,柚皮是固态发酵柚苷酶的良好原料,用棘孢曲霉对柚皮进行固态发酵是生产柚苷酶的有效方法。  相似文献

根据柚皮苷的物理及化学特性,设计和试验了一种新的柚苷酶产生菌筛选模型。在153株曲霉中筛选到一株产柚苷酶菌株AspergillusnigerZG84,摇瓶发酵酶活力为320u/ml。经自然分离和DES诱变处理,得到ZG86菌株,酶活力达1124u/ml。对其培养条件和产酶条件进行了研究。  相似文献

以α-鼠李糖苷酶活力和柚苷酶活力为考察指标, 研究了发酵条件对黑曲霉DB056产柚苷酶的影响。结果表明: 发酵温度、菌丝形态、培养基初始pH、接种量和装液量对黑曲霉DB056产柚苷酶都具有重要影响。初步优化得到黑曲霉DB056产柚苷酶的条件为: 培养基初始pH 8.0, 玻璃珠添加个数5个, 装液量45 mL, 接种量7%, 发酵温度34°C, 摇床转速190 r/min。采用此条件进行发酵, 高效液相色谱法检测α-鼠李糖苷酶活力最大可达1076.32 U/mL, 柚苷酶活力最大可达420.68 U/mL, 分别比初始条件提高了72.35%和78.03%。黑曲霉DB056不仅在对数生长期能快速合成柚苷酶, 在稳定期及衰亡期也会不断地分泌柚苷酶。阐明了发酵条件对黑曲霉DB056产柚苷酶的影响并获得了经过初步优化的发酵条件, 为进一步优化发酵条件, 提高黑曲霉DB056产柚苷酶的产量奠定了良好的基础。  相似文献

【目的】筛选并鉴定一株产纤维素酶的菌株,初步探究该菌的产酶特性,为综合利用纤维素筛选菌源。【方法】在常温条件下,采用滤纸培养基对菌种富集,采用CMC-Na初筛纤维素降解菌,采用LB培养基分离纯化菌株,经形态学、生理生化特征试验、16S r RNA基因序列测定等分析筛选菌株的系统分类地位。单因素试验确定培养时间、培养温度、初始p H及Na Cl浓度对筛选菌株产酶活力的影响。【结果】从腐烂的玉米秸秆中分离出一株在常温下产纤维素酶细菌KZ-2,根据菌落形态特征、生理生化特征鉴定以及16S r RNA基因序列分析,初步鉴定KZ-2为肠杆菌(Enterobacter sp.),为潜在新种。产酶条件实验显示:该菌使用产酶发酵培养基120 h产酶量达到最大值,在25–35°C、初始p H 4.5–5.5、Na Cl浓度1.0%–2.0%范围内为最佳产酶条件,在最适条件下酶活可达80.93 U/m L。该菌株所产纤维素酶最适反应p H为7.0,最适反应温度为50°C。【结论】KZ-2是一株具有降解纤维素能力的细菌,在常温下即可分泌纤维素酶,并且该菌株为潜在新种,具有潜在的开发价值。  相似文献

新生隐球菌的酚氧化酶及用于菌种鉴定的研究   总被引:4,自引:0,他引:4  
采用4%玉米浸汁咖啡酸琼脂(CACA)培养基。观察了具不同生物学特性的新生隐球菌的酚氧化酶活性,并对临床常见的多种酵母和酵母样真菌作了该酶的检测。结果,受试的3个变种、5种血清型和尿素酶阴性新生隐球菌均呈明确的阳性反应;150株常见酵母和酵母样真菌中43株新生隐球菌全部呈酚氧化酶阳性。107株其它酵母和酵母样真菌全部阴性。具各种不同生物学特性的新生隐球菌均特异性地产生酚氧化酶,用检测该酶的方法作该菌鉴定的特异性和敏感性均为100%,且可于72小时内得到结果。此外,结合尿素酶试验可以准确的鉴定出尿素酶阴性的新生隐球菌。  相似文献

【背景】低温β-半乳糖苷酶能在低温下仍保持较高的乳糖水解活性,筛选酶学特性适合在牛乳体系中高效水解乳糖的β-半乳糖苷酶生产菌株,是低乳糖牛乳加工产业关注的焦点。【目的】对天山中国一号冰川沉积物中分离的一株产低温β-半乳糖苷酶菌株的产酶条件和酶学特性进行研究。【方法】结合X-Gal平板法初筛和测定粗酶液酶活复筛,获得产低温β-半乳糖苷酶的菌株。通过形态学、生理生化试验及16S rRNA基因测序分析对筛选菌株进行鉴定,单因素摇瓶实验优化菌株的产酶条件,硫酸铵分级沉淀初步纯化β-半乳糖苷酶并对其酶学特性进行分析。【结果】通过形态学、生理生化特征和16S rRNA基因鉴定,确定菌株LW106为微杆菌属(Microbacterium)菌株;该菌株最适产酶温度为25°C,最佳产酶碳源为可溶性淀粉,培养基初始pH为7.0,接种量为3%;对初步纯化的低温β-半乳糖苷酶酶学性质的研究表明,LW106所产β-半乳糖苷酶的最适pH为6.0,最适反应温度为35°C,4°C时酶活为最大酶活的78%,4°C和pH 7.0时的稳定性最好,10 mmol/L的Na+对酶活性基本没有抑制作用,Ca~(2+)对酶活性具有一定的激活作用。【结论】菌株LW106所产低温β-半乳糖苷酶的酶学特性表明该酶在乳品低温加工领域具有进一步研究和应用的价值。  相似文献

Naringinase produced by Penicillum decombens PTCC 5248 was immobilized by entrapping it in calcium alginate beads, hen egg white, gelatin open pore matrix and covalently attaching it to activated seeds of Ocimum basilicum. Optimum pH and temperature were found to be 4, 4.5, 4.5, 3.5 and 65 °C, 60 °C respectively for enzyme immobilized by each technique mentioned. Naringinase immobilized onto activated seeds of Ocimum basilicum was active for 7 days.  相似文献

Summary An extracellular naringinase (an enzyme complex consisting of α-L-rhamnosidase and β-D-glucosidase activity, EC 3.2.1.40) that hydrolyses naringin (a trihydroxy flavonoid) for the production of rhamnose and glucose was purified from the culture filtrate of Aspergillus niger 1344. The enzyme was purified 38-fold by ammonium sulphate precipitation, ion exchange and gel filtration chromatography with an overall recovery of 19% with a specific activity of 867 units per mg of protein. The molecular mass of the purified enzyme was estimated to be about 168 kDa by gel filtration chromatography on a Sephadex G-200 column and the molecular mass of the subunits was estimated to be 85 kDa by sodium dodecyl sulphate-Polyacrylamide gel electrophoresis (SDS-PAGE). The enzyme had an optimum pH of 4.0 and temperature of 50 °C, respectively. The naringinase was stable at 37 °C for 72 h, whereas at 40 °C the enzyme showed 50% inactivation after 96 h of incubation. Hg2+, SDS, p-chloromercuribenzoate, Cu2+ and Mn2+ completely inhibited the enzyme activity at a concentration of 2.5–10 mM, whereas, Ca2+, Co2+ and Mg2+ showed very little inactivation even at high concentrations (10–100 mM). The enzyme activity was strongly inhibited by rhamnose, the end product of naringin hydrolysis. The enzyme activity was accelerated by Mg2+ and remained stable for one year after storage at −20 °C. The purified enzyme preparation successfully hydrolysed naringin and rutin, but not hesperidin.  相似文献

Five strains of naringin-degrading bacteria were isolated and found to be positive for extracellular naringinase activity. The one that showed highest activity in the selective medium was identified by 16S rRNA analysis as Bacillus methylotrophicus. The best combination of carbon–nitrogen source was determined by employing two-level full factorial analyses, comprising 24 experiments in shake flasks. Sucrose–yeast extract showed significant increase in naringinase activity (7.46 U/L) compared to the basal medium. Naringinase production was found to be inducible and naringin was found to be the best inducer among naringin, naringenin, hesperidin, and L-rhamnose. Inoculum size of 2% (v/v) and age of 48 hr favored naringinase and biomass production. Highest naringinase activity of 8 U/L was observed at the initial medium pH of 6. Response surface modeling was applied based on central composite design to determine the effects of three independent variables (sucrose, yeast extract, and naringin) and their mutual interactions. In total, 20 experiments were conducted and a statistical model was developed, which predicted naringinase production of 10.61 U/L. Subsequently, verification experiments were conducted and validity of the model was verified. Bioreactor studies conducted with the optimized medium showed an enzyme production of 12.05 U/L within 34 hr of fermentation.  相似文献

A new Penicillium ulaiense strain showed carboxymethylcellulase, pectinase, protease on skim milk and naringinase activities, but no xylanase, cellulase, lipase, amylase, protease on gelatin, and ligninase activities. Affinity chromatography with immobilised triazine dyes was used to separate the main enzymes present in the naringinase complex produced by Aspergillus terreus CECT 2663. One alpha-L-rhamnosidase and two beta-D-glucosidases (beta G1 and beta G2) were separated by a simple two-step procedure involving chromatography with Red HE-3B immobilised on Sepharose 4B first at pH 5.5 and then at pH 4.7. Maximum activity of the beta-D-glucosidases was from pH 4 to 6 and at 65 degrees C. Both glucosidases were active on p -nitrophenol glucoside and prunin with respective Km values of 1.9 mm and 1.6 mm for beta G1 and 2.1 mm and 0.25 mm for beta G2. Only beta G1 hydrolysed cellobiose (Km = 5.7 mm).  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献
