【目的】具有1-氨基环丙烷-1-羧酸(ACC)脱氨酶活性的盐生植物碱蓬内生细菌的分离及生物学特性的研究有助于探索内生细菌与宿主植物耐盐性的关系。【方法】采用研磨法从健康碱蓬植株的根、茎、叶中分离具有ACC脱氨酶活性的碱蓬内生细菌,根据形态特征、生理生化、API鉴定系统和16S rRNA对菌种进行鉴定,并分别测定了菌株产ACC脱氨酶、铁载体、吲哚乙酸、赤霉素、脱落酸、蛋白酶及溶磷、固氮和拮抗病原菌的特性。【结果】将分离得到的内生细菌LP11、SS12、TW1和TW2分别鉴定为栖稻假单胞菌(Pseudomonas oryzihabitans)、假单胞菌(Pseudomonas sp.)、成团泛菌(Pantoea agglomerans)和恶臭假单胞菌(Pseudomonas putida),4株菌除具有较高的ACC脱氨酶活力之外,还可不同程度地产生铁载体、吲哚乙酸、赤霉素和脱落酸,且均有溶磷作用,但无固氮能力及蛋白酶活力,唯有菌株SS12对萝卜枯萎病菌(Fusariumoxysporum f.sp.conglutinans)和黄瓜枯萎病菌(F.oxysporum f.sp.cucumerinum)具有拮抗作用。【结论】从盐生植物碱蓬中分离到的假单胞菌属和泛菌属内生细菌,具有丰富多样的生物学特性。  相似文献

【目的】从3种蓝莓根际土壤中分离细菌,探究蓝莓根际土壤细菌多样性,并筛选具有产酸、促生长、抑菌性能的菌株,为蓝莓专用微生物肥料的研究提供优质菌株资源和理论基础。【方法】选用5种培养基分离3种蓝莓根际土壤细菌,并进行16S rRNA基因测序和系统发育分析。筛选产酸、产吲哚-3-乙酸(indole-3-acetic acid,IAA)和铁载体、固氮、溶磷和抑制灰葡萄孢生长的菌株,挑选最适菌株制备菌剂进行蓝莓苗盆栽实验验证促生能力,并检测菌剂对蓝莓元素吸收和根际土壤肥力的影响。【结果】从3种蓝莓根际土壤分离得到124株细菌,挑选70株代表性菌株进行16S rRNA基因测序,分布于3个门21个属,其中芽孢杆菌属(Bacillus)、假单胞菌属(Pseudomonas)、链霉菌属(Streptomyces)和红球菌属(Rhodococcus)为优势分离菌群。代表性菌株中,21.4%的菌株能产酸,21.4%的菌株产吲哚-3-乙酸,47.1%的菌株具有固氮潜力,65.7%的菌株具有解磷能力,14.3%的菌株能产铁载体。少量菌株同时具有产酸、产IAA、固氮、解磷和抑菌等能力。选取具有产酸和多种促生特征的菌株绿针假单胞菌CSM-70和双鱼假单胞菌CSM-129进行盆栽蓝莓苗处理,发现2株菌均能显著促进蓝莓苗的生长发育并调控根际土壤pH,其中菌株CSM-70处理还显著促进了蓝莓叶片氮、磷元素的吸收,提升了土壤速效钾、碱解氮的含量。【结论】蓝莓根际细菌多样性高且蕴藏着丰富的促生长菌株,绿针假单胞菌CSM-70和双鱼假单胞菌CSM-129能够促进蓝莓苗生长、调控根际土壤pH和肥力,并促进植株养分吸收,具有蓝莓专用微生物菌剂研制与应用的潜力。  相似文献

青海可可西里嗜碱芽胞杆菌资源调查   总被引:3,自引:0,他引:3  
【目的】了解可可西里嗜碱芽胞杆菌资源多样性及产酶多样性,为芽胞杆菌功能资源挖掘和菌剂开发提供基础。【方法】采用Horikoshi I培养基,通过可培养法分离青海可可西里土壤中的嗜碱芽胞杆菌,利用16S r RNA基因序列初步鉴定分离获得的芽胞杆菌。采用透明圈法分析分离菌株的产蛋白酶、纤维素酶及木聚糖酶活性。【结果】从青海可可西里土壤中共分离获得66株嗜碱芽胞杆菌,根据16S r RNA基因序列相似性分析,发现它们隶属于6个属22个种,分别为芽胞杆菌属(Bacillus)、纤细芽胞杆菌属(Gracilibacillus)、喜盐芽胞杆菌属(Halobacillus)、咸海鲜芽胞杆菌属(Jeotgalibacillus)、类芽胞杆菌属(Paenibacillus)和嗜冷芽胞杆菌属(Psychrobacillus),其中以芽胞杆菌属(Bacillus)为优势属。2株嗜碱芽胞杆菌与它们最近匹配模式菌株的16S r RNA基因序列相似性为97.00%和98.65%,为潜在新种。三种酶活检测结果表明产酶菌株约占总分离菌株的95.00%,其中55株具有产蛋白酶活性,27株具有产纤维素酶活性,8株能够产木聚糖酶。【结论】青海可可西里蕴藏着较丰富的嗜碱芽胞杆菌资源及丰富的产酶资源,为后续嗜碱芽胞杆菌的挖掘提供理论基础。  相似文献

【背景】植物可通过改变根际微生物群落结构来减轻胁迫,但盐碱胁迫下小麦生长和根际细菌群落结构变化尚待明晰。【目的】明确盐碱胁迫下小麦幼苗生长及根际细菌群落结构变化。【方法】以添加NaCl和Na2CO3调节Hoagland营养液Na+浓度和pH来模拟盐碱胁迫,通过MiSeq高通量测序分析细菌群落结构。【结果】与无胁迫对照相比,单纯盐胁迫使小麦幼苗地上和地下部干重、叶绿素和根际土壤速效氮含量分别显著降低了25.0%、57.1%、21.2%和42.9%(P<0.05);小麦幼苗生长指标和根际土壤速效氮、速效磷、有机质含量及酶活性(除过氧化氢酶)随碱度升高而降低;所有处理的小麦幼苗根际优势菌门(丰度7.1%-71.5%)均为变形菌门(Proteobacteria)、放线菌门(Actinobacteria)和厚壁菌门(Firmicutes),优势属(丰度0.5%-5.1%)有肠杆菌属(Enterobacter)、嗜多甲基菌属(Methyloversatilis)、固氮菌属(Azotobacter)、假单胞菌属(Pseudomonas)和异根瘤菌属(Allorhizobium)。盐碱胁迫未显著...  相似文献

【背景】根际促生菌可以促进植物生长、提高植物抗性。茶树根际具有特殊的根土微生物生境,可以获得具促生作用的有益微生物。【目的】探究4株茶树根际促生菌菌株的分类地位及促生作用,筛选优良的根际促生菌菌株。【方法】通过形态、生理生化特征、16S rRNA基因序列同源性比对鉴定4株茶树根际促生菌,采用钼锑抗比色法测定溶磷量,通过比色法测定ACC脱氨酶活性、CAS法测定产铁载体能力、Salkowski法测定产IAA (Indoleacetic acid)的能力进行促生作用研究,通过盆栽实验测试白菜、空心菜、苋菜及水稻的株高及鲜重以分析促生效应。【结果】鉴定KKS-6-N1为放射型土壤杆菌(Agrobacteriumradiobacter), KKS-7-N7为铜绿假单胞菌(Pseudomonas aeruginosa),GD3为Pseudomonashunanensis,GD12为弯曲芽孢杆菌(Bacillusflexus)。固氮菌株KKS-6-N1可产铁载体;固氮菌株KKS-7-N7具有解磷及产铁载体能力,分泌的IAA含量高达101.29mg/L;解钾菌株GD3具溶磷能力,分泌的ACC脱氨酶酶活为8.09μmol/(mg·h),相对铁载体含量为0.31;具固氮解钾性能的菌株GD12分泌的ACC脱氨酶活性为14.46μmol/(mg·h)。盆栽试验表明,4个菌株对白菜、空心菜、苋菜的株高和鲜重均有明显促进作用,尤以GD3效果更甚。【结论】茶树根际促生菌菌株Pseudomonas hunanensis GD3促生作用显著,具有开发成微生物菌肥的潜力。  相似文献

次生盐渍化和土传病害是导致设施栽培连作障碍的主要因素,为了缓解设施栽培的连作障碍,从土壤中筛选多功能菌株并应用于连作障碍土壤中成为一种新的研究思路。文章通过对12株菌株的硝酸盐降解能力、产胞外聚合物(EPS)能力、降碱能力、抑菌能力进行验证,优选出硝酸盐降解能力最强的假单胞菌(Pseudomonas sp.)N11,EPS产量最高的琼氏不动杆菌(A c ine tobacte r junii)S27,抑菌效果最好的解淀粉芽孢杆菌(Bac illus amyloliquefacie ns)Y2,并将其制成混合菌剂运用于次生盐渍化土壤中。实验组对土壤中硝酸盐的降解率可达31.71%,而对照组仅为15.28%,说明混合菌剂降盐效果显著。筛选出的多功能菌株,可有效改善连作障碍土壤的次生盐渍化和土传病害问题,为提高设施蔬菜栽培产量提供可靠的技术手段。  相似文献

摘要:【目的】对南方红豆杉(Taxus chinensis var.mairei)根际溶无机磷细菌进行了分离、筛选与鉴定,并对获得的高效溶磷菌株进行了温室盆栽试验。本研究为通过生物途径改善南方红豆杉磷素供应,促进其生长提供了优良的菌株资源。【方法】利用选择培养基从南方红豆杉根际土壤中共分离出具溶磷能力的细菌;采用NBRI-BPB 培养基进行复筛获得溶磷能力较强的溶无机磷细菌;并采用钼锑抗比色法测量其在NBRIP 培养基中经4d 发酵后的可溶性磷含量;通过形态指标、生理生化测定、Biolog 系统和16S rDNA序列分析鉴定细菌种类;并进行了溶磷菌株的室内盆栽实生苗接种试验。【结论】从南方红豆杉根际共分离出4 株高效溶磷细菌,分别鉴定为荧光假单胞菌(Pseudomonas fluorescens)、蜡状芽胞杆菌(Bacillus cereus)、草木樨中华根瘤菌(Sinorhizobium meliloti)和地衣芽胞杆菌(Bacillus licheniformis);4株细菌对南方红豆杉苗期的生长有明显的促进作用。  相似文献

【目的】分析普通包装和气调包装(65%O2和35%CO2)生鲜冷却牛肉在贮藏(4°C)过程中的微生物多样性。【方法】通过16S rDNA V3区PCR-DGGE(变性梯度凝胶电泳)方法和16S rDNA克隆分析法研究生鲜冷却牛肉中微生物菌落结构及菌相变化规律。【结果】初始菌相主要有嗜冷杆菌属(Psychrobacter)和假单胞菌属(Pseudomonas)。贮藏过程中,普通包装和气调包装生鲜冷却牛肉中优势菌均为Brochothrix和Pseudomonas。气调包装冷却牛肉中细菌种类较少,两种包装生鲜牛肉在贮藏前期菌相变化明显。【结论】不同包装冷却牛肉中微生物菌落结构有较大差异,气调包装中CO2对Pseudomonas等细菌起到了一定的抑制作用。  相似文献

【目的】克隆产碱假单胞菌的脂肪酶基因,实现其在大肠杆菌中异源表达并进行酶学性质研究。【方法】通过基因文库构建和PCR,获得脂肪酶基因,并以pET30a(+)为表达载体、E.coli BL21(DE3)为宿主菌,在大肠杆菌中进行异源表达,表达产物经HisTrapTM亲和层析柱纯化后进行酶学性质研究。【结果】从产碱假单胞菌中克隆得到一个脂肪酶基因,大小为1 575 bp(GenBank登录号为JN674069)。该酶分子量为55 kD,最适底物为p-NPO,最适反应温度和pH分别为35°C、pH 9.0。重组酶经1 mmol/L的Cu2+处理30 min可使酶活提高至156%。在最适反应条件下重组酶的比活力为275 U/mg,Km和Vmax分别为80μmol/L和290 mmol/(min.g protein)。【结论】产碱假单胞菌脂肪酶基因的克隆与表达不仅积累了脂肪酶基因的资源,并为其在手性拆分中的应用奠定基础。  相似文献

The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. 该研究采用田间小区试验,设计杏树(Prunus armeniaca)下清耕(CK)和杏树间作紫花苜蓿(T)2个处理,实地采集测定各样地不同土层紫花苜蓿的根系生物量以及杏树的侧根系生物量,并测定土壤pH、电导率及其土壤有机质和速效氮含量,分析果园间作模式下紫花苜蓿对果树侧根系垂直分布特征及其土壤理化性质的影响,为果园间作苜蓿模式的推广提供理论依据。结果表明:(1)CK与T处理下的杏树侧根生物量在土壤中的垂直分布都主要集中在20~60cm土层,其生物量分别为750.8g和737.6g,分别占总侧根生物量的64.4%和64.5%;紫花苜蓿根系生物量分布呈倒金字塔型,且主要分布在0~40cm土层(166.3g),其中0~20cm土层的根系生物量最高(97.4g),占根系总生物量的35.8%。(2)与CK处理相比,T处理可有效增加果园表层土壤的有机质含量、速效态氮含量、硝态氮含量和铵态氮含量,其中,在0~20cm土层分别显著增加17.1%、40.8%、28.5%和40.8%,在20~40cm土层分别显著增加36.1%、23.1%、60.2%和23.8%,并显著降低了表层土壤电导率,但对土壤pH无显著影响。研究认为,杏园间作牧草紫花苜蓿虽然杏树与苜蓿根系会发生较小资源的竞争,但有利于改善林下土壤的理化性质和养分状况,能够有效促进果树的生长发育。  相似文献

Douglas-fir, grown on strongly acid soil (pH-H2O 3.87), was fertilized with ammonium, nitrate or ammonium nitrate as N-source. Rhizosphere pH along the root axis was measured with microelectrodes. Pure ammonium supply resulted in acidification of the rhizosphere, almost along the entire root axis; only the extreme root tip was slightly alkaline compared with the bulk soil. With nitrate supply, the alkalization of the rhizosphere at the root tip was considerably stronger than with ammonium supply and the length of the alkalization zone greater, extending over the entire growth zone of the root. Acidification of the rhizosphere along the older parts of the root was less pronounced in the case of nitrate. It is concluded that nitrate nutrition enables the plant to protect its most essential root zone from the adverse effects of strong acidity by locally raising the rhizosphere pH.  相似文献

【背景】三七根际微生物群落特征与其土传根腐病害密切相关,而针对自然林下根腐三七的相关研究鲜见报道。【目的】比较分析自然林下与田间根腐三七根际土壤微生物群落的组成特征,结合土壤理化性质与酶活性分析,为三七根腐病害防治与仿野生栽培提供科学依据。【方法】采集自然林下与田间根腐三七根际土壤,利用高通量测序技术,分析土壤细菌与真菌群落的物种组成与多样性,并测定土壤理化性质和酶活性。【结果】自然林下与田间根腐三七根际土壤中细菌和真菌群落组成具有明显差异,自然林下根腐三七根际土壤中担子菌门(Basidiomycota)、酸杆菌门(Acidobacteria)和疣微菌门(Verrucomicrobia)的相对丰度较高,而田间根腐三七根际土壤中子囊菌门(Ascomycota)、变形菌门(Proteobacteria)和绿弯菌门(Chloroflexi)的相对丰度较高。在属分类水平,镰刀菌属(Fusarium)是自然林下根腐三七根际土壤中的优势菌群,相对丰度为17.30%,而癣囊腔菌属(Plectosphaerella)是田间根腐三七根际土壤中的优势菌群,相对丰度为22.55%;Candidatus Ba...  相似文献

【目的】黄顶菊、三叶鬼针草和豚草是我国危害较严重的3种菊科入侵植物。了解3种菊科植物生长过程中土壤养分和酶活性的变化,可以为研究其入侵机制提供依据。【方法】在中国农业科学院植物保护研究所廊坊中试基地开展同质园实验,比较分析了黄顶菊、三叶鬼针草和豚草3种菊科入侵植物在幼苗期、旺盛生长期和生殖生长期根际土壤养分和酶活性的变化。【结果】3种外来菊科植物的生长时期对土壤养分和酶活性存在显著影响。3种外来植物入侵域的土壤速效磷、硝态氮含量以及磷酸酶活性随生长时期变化都表现出先升高后降低的趋势,均在旺盛生长期达到最大值。入侵域土壤速效磷和硝态氮含量的变化趋势与狗尾草明显不同。3种外来植物入侵降低了土壤速效钾含量,提高了土壤脲酶、磷酸酶活性。【结论】3种外来菊科植物的入侵改变了土壤养分和酶活性,创造出更有利于自身生长和繁殖的条件,以利于其进一步扩张。  相似文献

【目的】筛选辣椒(Capsicum annuum L.)根腐病防病促生细菌并明确其防病促生效应。【方法】采集健康辣椒根围土壤样品,以辣椒根腐病病原真菌茄镰孢(Fusarium solani)和尖镰孢(Fusarium oxysporum)为指示菌,采用平板对峙法筛选生防细菌,采用选择性培养基筛选溶无机磷、溶有机磷、固氮菌和解钾菌等促生菌,钼锑抗比色法测定溶磷量,凯氏定氮法测定固氮量,火焰原子吸收光谱法测定解钾量。对特性良好组合的菌株进行16S rDNA序列分析鉴定并制作菌剂,最后采用盆栽法测定菌剂防病促生效果。【结果】共筛选得到323株特性良好的功能菌株,拮抗菌78株,溶有机磷菌87株,溶无机磷菌107株,固氮菌128株,解钾菌123株,部分菌株同时具有多个功能特性。互作组合得到6个特性良好的菌株组合,包括8株功能菌株,鉴定发现XP271和XP181为枯草芽胞杆菌(Bacillus subtilis),XP125为特基拉芽胞杆菌(Bacillus tequilensis),XP236为耐盐芽胞杆菌(Bacillus halotolerans),XP79为巨大芽胞杆菌(Bacillus ...  相似文献

The wide range of plant responses to ammonium nutrition can be used to study the way ammonium interferes with plant metabolism and to assess some characteristics related with ammonium tolerance by plants. In this work we investigated the hypothesis of plant tolerance to ammonium being related with the plants’ capacity to maintain high levels of inorganic nitrogen assimilation in the roots. Plants of several species (Spinacia oleracea L., Lycopersicon esculentum L., Lactuca sativa L., Pisum sativum L. and Lupinus albus L.) were grown in the presence of distinct concentrations (0.5, 1.5, 3 and 6 mM) of nitrate and ammonium. The relative contributions of the activity of the key enzymes glutamine synthetase (GS; under light and dark conditions) and glutamate dehydrogenase (GDH) were determined. The main plant organs of nitrogen assimilation (root or shoot) to plant tolerance to ammonium were assessed. The results show that only plants that are able to maintain high levels of GS activity in the dark (either in leaves or in roots) and high root GDH activities accumulate equal amounts of biomass independently of the nitrogen source available to the root medium and thus are ammonium tolerant. Plant species with high GS activities in the dark coincide with those displaying a high capacity for nitrogen metabolism in the roots. Therefore, the main location of nitrogen metabolism (shoots or roots) and the levels of GS activity in the dark are an important strategy for plant ammonium tolerance. The relative contribution of each of these parameters to species tolerance to ammonium is assessed. The efficient sequestration of ammonium in roots, presumably in the vacuoles, is considered as an additional mechanism contributing to plant tolerance to ammonium nutrition.  相似文献

Ammonium sulphate is a major component of the air pollutants deposited on forests in the Netherlands. Different amounts of NH4 + were added to Douglas-fir seedlings grown in tall containers of sand, to study the influence of high concentrations of NH4 + in the soil on the development of fine roots and the effects of nitrogen uptake on rhizosphere pH. At the end of this eight-month experiment part of the ammonium appeared to have nitrified into nitrate. High doses of ammonium negatively affected root length and root length per unit of dry matter (specific root length). Although Douglas fir shows a preferential ammonium uptake in nutrient solutions the increases in the pH of the rhizosphere in this experiment indicate that nitrogen was mostly taken up as nitrate. When the ammonium concentration in the soil is low, it cannot be taken up readily because of its low mobility in soil. Shoot growth was stimulated by high availability of nitrogen. The possible effects of high doses of ammonium on long-term forest vitality are discussed.  相似文献

旨在了解艾比湖湿地盐生植物盐角草根际与非根际中不同类型反硝化细菌的分布及其随季节变化情况,为温带干旱地区荒漠盐化生态系统的代表-艾比湖湿地在生态植被恢复过程中,由微生物推动的土壤氮素循环过程提供数据支撑。采集了艾比湖湿地夏、秋、春三个季节的盐角草根际和非根际土壤样本,通过高通量测序技术,比较分析了nirS-型和nirK-型两种类型的反硝化细菌的多样性和群落结构特点;利用RDA (redundancy analysis)探究了土壤理化因素对反硝化细菌多样性及群落结构的影响。艾比湖湿地盐角草根际与非根际中,nirS-型和nirK-型反硝化细菌多样性最高的为秋季根际土壤样本;各土壤样本中的反硝化细菌多样性均呈现根际>非根际。盐角草各土壤样本中的nirS-型反硝化细菌在门分类水平上隶属于变形菌门(Proteobacteria),厚壁菌门(Firmicutes)和放线菌门(Actinobacteria),而nirK-型反硝化细菌在门水平上分类仅包括了ProteobacteriaFirmicutesProteobacteria在各土壤样本中的占比均较高;其中Gamma-Proteobacteria的盐单胞菌属(Halomonas)和假单胞菌属(Pseudomonas)是各土壤样本所共有的nirS-型反硝化菌的优势菌属,但它们在每个土壤样本中的相对丰度各有差异。Alpha-Proteobacteria的根瘤菌属(Rhizobium)是盐角草各土壤样本中较为广泛存在的nirK-型反硝化细菌。艾比湖湿地盐角草各土壤样本中的反硝化细菌群落结构存在着一定的差异。RDA结果显示含水量、有机质、全氮和铵态氮等对各土壤样本中的nirS-型反硝化细菌的多样性影响较大,含水量、有机质、全氮、碱解氮等是nirK-型反硝化细菌多样性的主要影响因素。土壤电导率、全磷、全钾、全氮和碱解氮协同影响nirS-型反硝化细菌的群落结构,有机质、速效钾、速效磷、pH和硝态氮是nirK-型反硝化细菌群落结构组成的主要影响因素。艾比湖湿地反硝化细菌呈现季节性变化,nirS-型和nirK-型反硝化细菌以不同的主要菌属,共同推进湿地反硝化作用。而对于湿地生态系统的保护,则需要进行长期而广泛的土壤状态评估和土壤反硝化微生物菌群的动态监测。  相似文献

[背景]关于高原生境轮作制度对土壤固氮微生物群落组成及多样性的影响研究尚少。[目的]深入认识攀西高原不同轮作制度对农田土壤肥力及土壤固氮微生物nifH基因群落结构与多样性的影响,以期建立合理的轮作制度。[方法]以凉山州冕宁县不同作物轮作制度[包括光叶紫花苕-烤烟(分轮作15年和20年两种,分别为G1和G2)、苦荞-烤烟(KQ)、大麦-烤烟(DM)和撂荒(CK)]的土壤为研究对象,通过化学分析和Illumina MiSeq技术,对土壤理化性质、土壤固氮微生物nifH基因多样性及群落组成进行分析。[结果]撂荒土壤全氮、铵态氮、硝态氮、有机碳和含水量最显著(P<0.05)。KQ轮作下土壤有效磷和速效钾分别提高了43.0%和2.60%,而DM轮作下的土壤理化性质均下降。土壤固氮酶活以撂荒土壤最高,G2轮作最低。土壤固氮微生物nifH基因多样性以G1轮作最高、G2轮作最低,门水平上以变形菌门(Proteobacteria)是优势共有nifH基因类群,相对丰度占群落的63.0%-92.4%;属水平上,偶氮氢单胞菌属(Azohydromonas)是不同轮作制度下的优势物种,慢生根瘤属(Brad...  相似文献
