The purpose of this review is to discuss the impact of nanocarriers administered by pulmonary route to treat and to diagnose respiratory and non respiratory diseases. Indeed, during the past 10 years, the removal of chlorofluorocarbon propellants from industrial and household products intended for the pulmonary route has lead to the developments of new alternative products. Amongst these ones, on one hand, a lot of attention has been focused to improve the bioavailability of marketed drugs intended for respiratory diseases and to develop new concepts for pulmonary administration of drugs and, on the other hand, to use the pulmonary route to administer drugs for systemic diseases. This has led to some marketed products through the last decade. Although the introduction of nanotechnology permitted to step over numerous problems and to improve the bioavailability of drugs, there are, however, unresolved delivery problems to be still addressed. These scientific and industrial innovations and challenges are discussed along this review together with an analysis of the current situation concerning the industrial developments.  相似文献

在西双版纳热带雨林已鉴定蚊科昆虫9亚科76属267种。西双版纳地区的蚂蚁区系以热带至亚热带分布的东洋界成分最为丰富。在属级水平上,与马来西亚界关系最为密切。与澳洲界关系较密切;与非洲界和马拉加西界的关系知中。与新北界,新热带界和古北界的关系最为疏远。可见西双版纳的蚂蚁区系具有典型的热带亚洲起源特征,同时与澳洲和非洲的热带区系有一定的渊源关系。  相似文献

Disturbances (pulse, press and ramp) constitute a major force influencing, even determining, the structure and functions of ecological components – populations, communities and ecosystems. The capacity to weather a disturbance without loss is defined as resistance, whereas resilience is the capacity to recover from a disturbance after incurring losses, which may be considerable. This article seeks to resolve differences in the ecological definition of resistance and of resilience and to examine the importance of resilience as applied to ecological restoration. In restoration, interventions are designed and implemented with the aim of strengthening the resilience, that is, the capacity to recover, of degraded systems. In response to restorative measures, degraded systems may have both resistance and negative resilience to remain in the degraded state. The key aim of restoration is to overcome the resistance and negative resilience of the degraded state by strengthening positive resilience, the capacity to recover to the intact undegraded state. Restoration may be hindered by a lack of knowledge of acting disturbances (both past and present), of the previous intact condition and of appropriate interventions to implement. Resilience to a disturbance is determined after the disturbance has ceased. Thus, for current ongoing press and ramp disturbances, resilience can be hard to determine.  相似文献

It is clear that saltmarshes are a unique and important component of the coastal biosphere of Australia. Their contribution ranges from stabilisation of fine sediments and providing an excellent protective buffer between land and sea, to their diverse blend of terrestrial and marine fauna. Further, saltmarsh plants are highly specialised and adapted to fill a harsh niche allowing them to act in roles that other vegetation types cannot. Saltmarsh habitats are recognised for their importance to migratory waders under the Ramsar convention, but it is becoming increasingly evident that they are also important to a variety of commercially valuable fish and native mammal species. Activities that are detrimental to saltmarshes continue and need to be addressed in order to conserve remaining saltmarsh areas. In general, urbanisation of the catchment has lead to filling of saltmarshes, tidal restriction, use by recreational vehicles, grazing, trampling and increased sedimentation and nutrient runnoff allowing colonisation and invasion of mangroves. These disturbances have a number of ecological consequences ranging from weed infestation to complete changes in the species composition and ecology. Reversing the disturbance is not always simple and can require extensive groundwork to be successful. Rehabilitation of existing saltmarsh areas has been a successful means to enhance this habitat. In general, it requires relatively little effort to remove weeds and fence off areas to regenerate naturally. Saltmarsh areas have been shown to respond well to this type of manipulation. Restoration and creation require substantial effort and planning to ensure a successful outcome. However, given the right environmental combinations of elevation, tide and salinity, saltmarsh will establish and grow. To speed the process transplantation of saltmarsh plants can be considered either from donor sites or plants propagated in green houses.  相似文献

'Adapt to endure' has become a necessity in agriculture, but the means to do so remain largely undefined. The aim of this literature review is to analyse how the herd contributes to a livestock farming system's capacity to adapt to a changing world and evolve when the future is uncertain. We identify six categories of elements linked to the herd, called 'sources of flexibility', that are used to manage perturbation. The first three are: using the animal's adaptive capacities, using the diversity of species and breeds and combining the diversity of animal products. The last three are: organising the mobility of animals and livestock farmers, juggling the herd numbers and mastering the balance between productivity and herd survival. These sources of flexibility are described in the literature by studying the different ways in which they are used. For example, the 'juggle herd numbers' source is described by volume, categories of animals, type of transfer, such as births, purchases or gifts, and timing of use, especially linked to the timing of the perturbation. Identified studies also compare or rank sources and analyse the connections between them. The flexibility framework (management science) is used for this analysis according to the levels of organisation of a livestock farming system: a strategic level referring to long-term options and to the capacity to modify the system structure, and an operational level referring to adjustment decisions during the productive cycle, the presence or absence of intervention by the livestock farmer, and the time scales involved. We conclude that the decision to use one or another source (in terms of modalities, alternatives, scheduling and combinations) is made according to the production objectives, the structural means, the type/frequency/intensity of perturbations and the context/environment. Consequently, the flexibility of a livestock farming system cannot be assessed in absolute terms. Enhancing flexibility needs management of all elements and scales involved (and not only the herd), and requires diversity to be organised at different scales.  相似文献

Mammary tissue from rats that had been ovariectomized and adrenalectomized 4 weeks previously was compared to that from intact rats in terms of epithelial content and hormone-responsiveness in vitro. The endocrinectomy resulted in about a 30% enlargement of the gland, but led to a loss of only about 12% of the epithelium. This estrogen-depleted epithelium was able to acquire full responsiveness in vitro to insulin in terms of the accumulation of alpha-aminoisobutyric acid, and induction of glucose-6-phosphate and gluconate-6-phosphate dehydrogenases. It was also fully responsive to cortisol in relation to the induction of NADH-cytochrome C reductase, and to prolactin in terms of total RNA synthesis. However, estrogen-depletion led to an 82% loss in the ability of a unit amount of the epithelium to synthesize casein in response to these 3 hormones, and to a similar loss in relation to the accumulation of 25K casein mRNA. Estrogen administration in vivo could prevent and reverse the casein lesion. The disparity between constitutive and casein hormone-responsiveness in the absence of estrogen is discussed in relation to cell commitment.  相似文献

Abstract: Glial fibrillary acid protein (GFAP)-positive astrocytes isolated from the cerebral cortices of 3–10-day-old rats frequently showed increased intracellular Ca2+ concentration responses to l -glutamate and glutamate analogues. However, few of the acutely isolated cells responded to ATP, and no such cells responded to serotonin [5-hydroxytryptamine (5-HT)]. The same cell that failed to respond to ATP or 5-HT often responded to glutamate. Culturing acutely isolated cells in media containing horse serum decreased Ca2+ responses to glutamate but increased the responses to ATP and induced responses to 5-HT. In primary cultures prepared from the cerebral cortices of 1-day-old rats and cultured in horse serum, fewer of the cells responded to glutamate, but almost all cells responded to ATP and 5-HT. The lack of, or limited response to, 5-HT or ATP in the acutely isolated cells seems unlikely to be due to selective damage to the respective receptors because acutely isolated GFAP-negative cells showed responses to ATP, several different proteases and mechanical dissociation yielded cells that also responded to glutamate but not to ATP, and exposure of primary cultures to papain did not abolish Ca2+ responses to several transmitters. The responses of the acutely isolated cells to glutamate but limited or lack of responses to ATP and 5-HT also correspond to what has been seen so far for astrocytes in situ. Thus, the present studies provide direct evidence that some of the receptors seen in primary astrocyte cultures may reflect a response to culture conditions and that, in the context of the relevant information so far available, acutely isolated astrocytes seem to reflect better the in vivo state.  相似文献

The impact of the presence of us humans as well as our activities to nature has led to over-exploitation of natural resources and to dramatic changes in land-use including the use of artificial fertilizers contributes to the deterioration of the natural environment. The population density, industrial processes and the use of fertilizers are the main causes for the eutrophication of river systems, estuaries and seas. There are several ways to determine the actual nutrient levels back to the 1950s, back to 1900 or even back to 1800. Available data indicate that the natural background concentrations of nutrients (pristine conditions or the period prior to the widespread use of artificial fertilizers and detergents) were dramatically lower than today. Available time series on chemical and biological data collected from different parts of the world show the (sometimes tremendous) increase in nutrient levels and the related productivity. The same time series, however, also show the decreases in values when measures were taken to reduce the nutrient emissions. Investigations of different systems all over the world show that nearly every system responses differently to eutrophication. Especially physical boundary conditions play an important role in the manifestation of the ultimate effect of local eutrophication. Apart from the physical boundary conditions also the transformation and retention of nutrients in estuarine and coastal systems contribute to system specific responses. Depending on all these different conditions, site specific responses with even site specific problems may occur. The challenge of this millennium is to really reach a balance between nature and mankind including its population size. A beneficial step in this discussion is assessing criteria to reduce eutrophication back to a level acceptable to both humans and nature.  相似文献

Chronic exposure to excessive levels of nutrients is postulated to affect the function of several organs and tissues and to contribute to the development of the many complications associated with obesity and the metabolic syndrome, including type 2 diabetes. To study the mechanisms by which excessive levels of glucose and fatty acids affect the pancreatic beta-cell and the secretion of insulin, we have established a chronic nutrient infusion model in the rat. The procedure consists of catheterizing the right jugular vein and left carotid artery under general anesthesia; allowing a 7-day recuperation period; connecting the catheters to the pumps using a swivel and counterweight system that enables the animal to move freely in the cage; and infusing glucose and/or Intralipid (a soybean oil emulsion which generates a mixture of approximately 80% unsaturated/20% saturated fatty acids when infused with heparin) for 72 hr. This model offers several advantages, including the possibility to finely modulate the target levels of circulating glucose and fatty acids; the option to co-infuse pharmacological compounds; and the relatively short time frame as opposed to dietary models. It can be used to examine the mechanisms of nutrient-induced dysfunction in a variety of organs and to test the effectiveness of drugs in this context.  相似文献

Farming is the basis of our civilization yet is more damaging to wild nature than any other sector of human activity. Here, we propose that in order to limit its impact into the future, conservation researchers and practitioners need to address several big topics--about the scale of future demand, about which crops and livestock to study, about whether low-yield or high-yield farming has the potential to be least harmful to nature, about the environmental performance of new and existing farming methods, and about the measures needed to enable promising approaches and techniques to deliver on their potential. Tackling these issues requires conservationists to explore the many consequences that decisions about agriculture have beyond the farm, to think broadly and imaginatively about the scale and scope of what is required to halt biodiversity loss, and to be brave enough to test and when necessary support counterintuitive measures.  相似文献

Guidance molecules present in both axonal and dendritic growth cones mediate neuronal responses to extracellular cues thereby ensuring correct neurite pathfinding and development of the nervous system. Little is known though about the mechanisms employed by neurons to deliver these receptors, specifically and efficiently, to the extending growth cone. A deeper understanding of this process is crucial if guidance receptors are to be manipulated to promote nervous system repair. Studies in other polarised cells, notably epithelial, have elucidated fundamental routes to the intracellular segregation of molecules mediated by endosomal pathways. Due to their extreme complexity and specialisation, neurons appear to have built upon these generic systems to evolve sophisticated trafficking networks. A striking feature is the axon initial segment which acts like a valve to tightly regulate the flux of molecules both entering and leaving the axon. Once in the growth cone, further controls operate to enhance the retention or rejection, as appropriate, of membrane receptors. We discuss the current state of knowledge regarding the intracellular trafficking of axon guidance receptors and how this relates to their developmental roles. We highlight the various facets still to be properly elucidated and by building on existing data regarding neuronal polarity and intracellular sorting mechanisms suggest ways to fill these gaps.  相似文献

We have previously reported that a single exposure to immobilization (IMO) in rats causes a long-term desensitization of the hypothalamic-pituitary-adrenal (HPA) response to the same (homotypic) stressor. Since there are reports showing that a single exposure to other stressors causes sensitization of the HPA response to heterotypic stressors and increases anxiety-like behavior, we studied in the present work the long-term effects of IMO on behavioral and HPA response to mild superimposed stressors. In Experiments 1 and 2, adult male Sprague–Dawley rats were subjected to 2 h of IMO and then exposed for 5 min to the elevated plus-maze (EPM) at 1, 3 or 7 days after IMO. Blood samples were taken at 15 min after initial exposure to the EPM. Increases in anxiety-like behavior and HPA responsiveness to the EPM were found at all times post-IMO. Changes in the resting levels of HPA hormones did not explain the enhanced HPA responsiveness to the EPM (Experiment 3). In Experiments 4 and 5, we studied the effects of a single exposure to a shorter session of IMO (1 h) on behavioral and HPA responses to a brief and mild session of foot-shocks done 10 days after IMO. Neither previous IMO nor exposure to shocks in control rats modified behavior in the EPM. However, a brief session of shocks in previously IMO-exposed rats dramatically increased anxiety in the EPM. HPA and freezing responses to shocks were similar in control and previous IMO groups. Therefore, a single exposure to IMO appears to induce long-lasting HPA and behavioral sensitization to mild superimposed stressors, although the two responses are likely to be at least partially independent. Long-term effects of IMO on the susceptibility to stress-induced endocrine and emotional disturbances may be relevant to the characterization of animal models of post-traumatic stress.  相似文献

The physiologically active lateral rootlets of all main trees in temperate forests are colonised by ectomycorrhizal fungi, forming so-called ectomycorrhizas. These symbiotic organs are the sites of exchange of nutrients, mainly P and N, provided from the fungal partner, and C from the host. Emerging from the ectomycorrhizas, fungal hyphae exploit the soil for the mobilisation and absorption of water and nutrient elements. By doing so, they connect the tree roots intimately with the soil and provide anchorage. The deposition of acidifying pollutants into forest ecosystems is a potential threat to the health and vitality of forest trees because it leads to the acidification and eutrophication of forest soils. Pollutants are also a threat to the functioning of ectomycorrhizas. Increased N concentrations in the soil lead to enhanced fungal N uptake and storage, and to enhanced N transfer to the host plants, and therefore to higher plant biomass of above ground parts. In consequence, there is a decrease of C allocation to the plant roots. This in turn leads to reduced ectomycorrhization, and to reduced production of external mycelia and fruiting bodies. Soil acidification leads to enhanced availability of Al, heavy metals, and radionuclides in the soil, all of which can be toxic to plants and fungi. Reduced growth of roots and hyphae are amongst the first symptoms. In ectomycorrhizas, the hyphae of the fungal tissues contain vacuolar polyphosphates which have the ability to bind Al, heavy metals, radionuclides and N. These electronegative polymers of phosphates represent an effective storage and detoxifying mechanism which otherwise is lacking in roots. Therefore, ectomycorrhizas have the potential to increase the tolerance of trees to acidifying pollutants and to the increased availability in the soil of toxic elements.  相似文献

Cold acclimation requires adjustment to a combination of light and low temperature, conditions which are potentially photoinhibitory. The photosynthetic response of plants to low temperature is dependent upon time of exposure and the developmental history of the leaves. Exposure of fully expanded leaves of winter cereals to short-term, low temperature shiftsinhibits whereas low temperature growthstimulates electron transport capacity and carbon assimilation. However, the photosynthetic response to low temperature is clearly species and cultivar dependent. Winter annuals and algae which actively grow and develop at low temperature and moderate irradiance acquire a resistance to irradiance 5- to 6-fold higher than their growth irradiance. Resistance to short-term photoinhibition (hours) in winter cereals is a reflection of the increased capacity to keep QA oxidized under high light conditions and low temperature. This is due to an increased capacity for photosynthesis. These characteristics reflect photosynthetic acclimation to low growth temperature and can be used to predict the freezing tolerance of cereals. It is proposed that the enhanced photosynthetic capacity reflects an increased flux of fixed carbon through to sucrose in source tissue as a consequence of the combined effects of increased storage of carbohydrate as fructans in the vacuole of leaf mesophyll cells and an enhanced export to the crown due to its increased sink activity. Long-term exposure (months) of cereals to low temperature photoinhibition indicates that this reduction of photochemical efficiency of PS II represents a stable, long-term down regulation of PS II to match the energy requirements for CO2 fixation. Thus, photoinhibition in vivo should be viewed as the capacity of plants to adjust photosynthetically to the prevailing environmental conditions rather than a process which necessarily results in damage or injury to plants. Not all cold tolerant, herbaceous annuals use the same mechanism to acquire resistance to photoinhibition. In contrast to annuals and algae, overwintering evergreens become dormant during the cold hardening period and generally remain susceptible to photoinhibition. It is concluded that the photosynthetic response to low temperatures and susceptibility to photoinhibition are consequences of the overwintering strategy of the plant species.  相似文献

AIMS: The aims of this study present were to assess and to evaluate in vitro the abilities of commercial probiotic strains derived from fermented milk products and related sources currently marketed in European countries, to inhibit, compete and displace the adhesion of selected potential pathogens to immobilized human mucus. METHODS AND RESULTS: The adhesion was assessed by measuring the radioactivity of bacteria adhered to the human mucus. We tested 12 probiotic strains against eight selected pathogens. All strains tested were able to adhere to mucus. All probiotic strains tested were able to inhibit and displace (P<0.05) the adhesion of Bacteroides, Clostridium, Staphylococcus and Enterobacter. In addition, the abilities to inhibit and to displace adhered pathogens depended on both the probiotic and the pathogen strains tested suggesting that several complementary mechanisms are implied in the processes. CONCLUSIONS: Our results indicate the need for a case-by-case assessment in order to select strains with the ability to inhibit or displace a specific pathogen. Probiotics could be useful to correct deviations observed in intestinal microbiota associated with specific diseases and also, to prevent pathogen infections. SIGNIFICANCE AND IMPACT OF THE STUDY: The competitive exclusion properties of probiotics as well as their ability to displace and inhibit pathogens are the most importance for therapeutic manipulation of the enteric microbiota. The application of such strategies could contribute to expand the beneficial properties on human health against pathogen infection.  相似文献

Computational methods based on continuum electrostatics are widely used in theoretical biochemistry to analyze the function of proteins. Continuum electrostatic methods in combination with quantum chemical and molecular mechanical methods can help to analyze even very complex biochemical systems. In this article, applications of these methods to proteins involved in photosynthesis are reviewed. After giving a short introduction to the basic concepts of the continuum electrostatic model based on the Poisson-Boltzmann equation, we describe the application of this approach to the docking of electron transfer proteins, to the comparison of isofunctional proteins, to the tuning of absorption spectra, to the analysis of the coupling of electron and proton transfer, to the analysis of the effect of membrane potentials on the energetics of membrane proteins, and to the kinetics of charge transfer reactions. Simulations as those reviewed in this article help to analyze molecular mechanisms on the basis of the structure of the protein, guide new experiments, and provide a better and deeper understanding of protein functions.  相似文献

Winter is a key driver of individual performance, community composition, and ecological interactions in terrestrial habitats. Although climate change research tends to focus on performance in the growing season, climate change is also modifying winter conditions rapidly. Changes to winter temperatures, the variability of winter conditions, and winter snow cover can interact to induce cold injury, alter energy and water balance, advance or retard phenology, and modify community interactions. Species vary in their susceptibility to these winter drivers, hampering efforts to predict biological responses to climate change. Existing frameworks for predicting the impacts of climate change do not incorporate the complexity of organismal responses to winter. Here, we synthesise organismal responses to winter climate change, and use this synthesis to build a framework to predict exposure and sensitivity to negative impacts. This framework can be used to estimate the vulnerability of species to winter climate change. We describe the importance of relationships between winter conditions and performance during the growing season in determining fitness, and demonstrate how summer and winter processes are linked. Incorporating winter into current models will require concerted effort from theoreticians and empiricists, and the expansion of current growing‐season studies to incorporate winter.  相似文献
