Mobilization of different phosphate fractions in the rhizosphere   总被引:6,自引:1,他引:5

Availability of soil P fractions and mechanisms of acquisition by plants were studied. Plants mobilize soil P by desorption via depletion of P solution concentration around roots. In an oxisol, the process was enhanced by nitrate N nutrition of ryegrass, which increased soil pH, and by carboxylate release by white lupin. Ligand exchange and Fe/Al solubilization are assumed to be the mechanisms. Ammonium N nutrition of ryegrass decreased pH and allowed P mobilization in a luvisol but had no such effect in an oxisol, due to acid solubility of P in these soils. Organic P dissolved in soil solution contributed one third to the P uptake of field-grown barley on a luvisol. Laboratory experiments suggest that organic P is hydrolyzed by phosphatases at the root surface and replenished by micro-organisms.  相似文献

Organic phosphorus is often a major part of total phosphorus in soil solution. The role of this fraction as a P source for plants and the mechanism involved in its transfer from soil to plant is still unclear. We studied the utilization of organic phospharus in 0.01 M calcium chloride extracts by barley and its hydrolysis by isolated acid and alkaline phosphatases. Calcium chloride extracts were used as a nutrient solution in 24 hrs assays. Concentration of organic and inorganic P in equilibrium calcium chloride extracts was 7.8 and 1.8 µmol P L-1, respectively, which was similar to the soil solution P concentration. When soil microbial biomass was destroyed by autoclaving, organic P concentration increased to 64.8 µmol P L-1 whereas the inorganic P was hardly changed. Inoculation of the autoclaved soil with non-sterile soil and incubation for 5 days decreased the organic P concentration to 27.9 µmol P L-1 but did not change inorganic P. In this study barley plants utilized organic P from all extracts. The greatest reduction of organic P concentration occurred in fresh extracts of the autoclaved soil. Inorganic P was depleted to traces in all extracts. Organic P was hydrolyzed by isolated acid and alkaline phosphatases. We conclude that organic P in soil solution is a heterogeneous pool of organic P compounds originating from microbial biomass. Its initial availability to plants was nigh but its susceptibility to phosphatase hydrolysis was quickly reduced but not completely lost.  相似文献

The 2818 amino acids of neurofibromin, the product of the human NF1 gene, include a 230 amino acid Ras-GAP related domain (GRD). Functions which may be associated with the rest of the protein remain unknown. However, many NF1 mutations in neurofibromatosis 1 patients are found downstream of the GRD, suggesting that the C-terminal region of the protein is also functionally important. Since the C-terminal region of neurofibromin encompassing these mutations is homologous with the corresponding regions in the two Saccharomyces cerevisiae Ras-GAPs, Ira1p and Ira2p, we chose yeast as a model system for functional exploration of this region (Ira-C region). Three missense mutations that affect the Ira-C region of NF1 were used as a model for the mutagenesis of IRA1. The yeast phenotypes of heat shock sensitivity, iodine staining, sporulation efficiency, pseudohyphae formation, and GAP activity were scored. Even though none of the mutations directly affected the Ira1p-GRD, mutations at two of the three sites resulted in a decrease in the GAP activity present in ira1 cells. The third mutation appeared to disassociate the phenotypes of sporulation ability and GAP activity. This and other evidence suggest an effector function for Ira1p.  相似文献

Gene duplication leads to the formation of gene families, wherein purifying or neutral selection maintains the original gene function, while diversifying selection confers new functions onto duplicated genes. The B56 gene family is highly conserved; it is encoded by one gene in protists and fungi, and five genes in vertebrates. B56 regulates protein phosphatase 2A (PP2A), an abundant heterotrimeric serine/threonine phosphatase that functions as a tumor suppressor and consists of a scaffolding “A” and catalytic “C” subunit heterodimer bound to a regulatory “B” subunit. Individual regulatory B56 subunits confer disparate functions onto PP2A in various cell-cell signaling pathways. B56 proteins share a conserved central core domain, but have divergent N- and C-termini which play a role in isoform specificity. We carried out a nonsynonymous/synonymous substitution analysis to better understand the divergence of vertebrate B56 genes. When five B56 paralogs from ten vertebrate species were analyzed, the gene family displayed purifying selection; stronger purifying selection was revealed when individual B56 isoforms were analyzed separately. The B56 core experienced stronger purifying selection than the N- and C-termini, which correlates with the presence of several contacts between the core and the AC heterodimer. Indeed, the majority of the contact points that we analyzed between B56 and the AC heterodimer experienced strong purifying selection. B56 subfamilies showed distinct patterns of selection in their N- and C-termini. The C-terminus of the B56-1 subfamily and the N-terminus of the B56-2 subfamily exhibited strong purifying selection, suggesting that these termini carry out subfamily-specific functions, while the opposite termini exhibited diversifying selection and likely carry out isoform-specific functions. We also found reduced synonymous substitutions at the N- and C-termini when grouping B56 genes by species but not by isoform, suggesting species-specific codon bias may have a role in regulating B56 gene expression.  相似文献

Replenishment of soil solution organic and inorganic P in a sterile and nonsterile grassland soil amended with 0 and 235 kg P ha–1 for 13 consecutive years was investigated in a recirculating column system. In sterilized treatments, P liberated from soil biomass, initially increased solution organic and inorganic P concentrations to about 0.3 and 0.6 g P cm–3 in the 0 and 235 kg P treatment, respectively. Sterilization effects were larger than the residual fertilizer effect. Subsequently, in sterilized treatments were microbial activity was lacking, removal of solution P over the duration of the experiment reduced organic P concentration to the detection limit (0.001 g P cm–3). Organic P concentrations in the nonsterile treatment were maintained at about 0.015 g P cm–3 which was higher than inorganic P concentration. Inorganic P concentrations were about 0.002 and 0.008 g P cm–3 in the nonfertilized and the fertilized treatment, respectively. Inorganic P buffer power was greater in the nonsterile treatments, but abiotic buffering alone could not account for the measured inorganic P concentrations found during desorption. It was concluded that biomass P is a major factor controlling organic and inorganic P solution concentrations in this systems.  相似文献

The vasodilators glyceryl trinitrate (GTN) and pentaerythrityl tetranitrate (PETN) are supposed to be degraded in vivo to the lower nitrates PETriN, PEDN, PEMN, 1,2-GDN, 1,3-GDN, 1-GMN, and 2-GMN. We synthesized these bioactive metabolites as reference compounds for pharmacokinetic studies. The use of HPLC-methods for monitoring the stepwise reduction of PETN to lower nitrates and the syntheses of the glyceryl dinitrates proved advantageous. Furthermore, we measured the vasorelaxant properties of all metabolites by performing organ bath experiments with porcine pulmonary arteries. In general, the vasodilator potency increases with the number of nitrate moieties in the compound.  相似文献
