Actinomycin D caused the production of hypomethylated DNA in cultured Friend erythroleukemia cells at cell culture concentrations of 1-4 ng per ml. Inhibition of DNA methyltransferase in cell-free assays was kinetically complex, with mixed-type inhibition. Cornish-Bowden graphical analysis was used to derive a Ki of about 35 nmol Act D per mg DNA. Although nuclei from drug-treated cells were found to contain hypomethylated DNA and DNA methyltransferase could be extracted from the nuclei, the methyl-accepting ability of DNA in whole nuclei themselves was not elevated. We conclude that the low level of Act D bound to DNA in the nuclei is sufficient to prevent the remethylation of hypomethylated sites.  相似文献

The formation of R-prime plasmids was selected in crosses involving soybean microsymbionts with genomic Tn5 insertions and carrying plasmid pJB3JI (with one IS2) copy as donors and Escherichia coli HB101 as recipient. Whereas the parent plasmid was 60 kb, recombinant plasmids between 76 kb and 121 kb were obtained. Restriction and Southern analyses confirmed the mobilization of Tn5 on four R-primes from Bradyrhizobium japonicum I-110 and on an R-prime plasmid from Rhizobium fredii HH303. There is a need for safe medications that can effectively support recovery by treating symptoms of protracted abstinence that may precipitate relapse in alcoholics, e.g. craving and disturbances in sleep and mood. This proof-of-concept study reports on the effectiveness of gabapentin 1200 mg for attenuating these symptoms in a non-treatment-seeking sample of cue-reactive, alcohol-dependent individuals. Subjects were 33 paid volunteers with current Diagnostic and Statistical Manual of Mental Disorders-IV alcohol dependence and a strength of craving rating 1 SD or greater for alcohol than water cues. Subjects were randomly assigned to gabapentin or placebo for 1 week and then participated in a within-subjects trial where each was exposed to standardized sets of pleasant, neutral and unpleasant visual stimuli followed by alcohol or water cues. Gabapentin was associated with significantly greater reductions than placebo on several measures of subjective craving for alcohol as well as for affectively evoked craving. Gabapentin was also associated with significant improvement on several measures of sleep quality. Side effects were minimal, and gabapentin effects were not found to resemble any major classes of abused drugs. Results suggest that gabapentin may be effective for treating the protracted abstinence phase in alcohol dependence and that a randomized clinical trial would be an appropriate next step. The study also suggests the value of cue-reactivity studies as proof-of-concept screens for potential antirelapse drugs.  相似文献

The Raymond A. Dart Collection of Human Skeletons (Dart Collection) is housed in the School of Anatomical Sciences at the University of the Witwatersrand, Johannesburg, South Africa, and comprises one of the largest documented cadaver‐derived human skeletal assemblages in the world. This collection originated in the early 1920s as a result of the efforts of Raymond Dart and continues to grow. The skeletons included represent varied indigenous and immigrant populations from southern Africa, Europe and Asia. This contribution documents the history of the collection and provides an updated inventory and demographic assessment of this valuable research collection. According to a recent inventory the Dart Collection currently comprises 2,605 skeletons representing individuals from regional SA African (76%), White (15%), Coloured (4%) and Indian (0.3%) populations. A large proportion of the skeletons (71%) represent males. The recorded ages at death range from the first year to over 100 years of age, but the majority of individuals died between the ages of 20 and 70. The Dart Collection has been affected by collection procedures based on availability. All of the cadavers collected before 1958, and large proportions subsequently, were derived from unclaimed bodies in regional South African hospitals. Some details of documentation (age at death, population group) are estimates and some aspects of the collection demographics (sex ratios) do not closely reflect any living South African population. At the end of Member 2 times, remnant mammillary-form stalactites in the roof and wall contacts show that an irregular wedge-shaped space existed from the Classic Section out to the Cone area. It was into this space that hyenas and other denning animals were able to gain access from an as yet unfilled Cone area. During the mining operations of the 1920’s, Eitzman (1958) recollects entering an impressive bone breccia resembling a charnel house. We suggest that it is most probable that the Grey Breccia corresponds to his main breccia as only the area on top of Member 2, between the dolomite walls, roof and stalagmite boss has the required volume. It also lies at the end of an incline first dug by the miners, (still in existence), and at the end of which he was able to note some of the deposits. The bone breccias at the same level at the back of the Cone suggest that other dens were created at about this time. As the roof continued to retreat upslope and more material was washed in or fell in from the surface so more sediment was winnowed into the back of the Cone. At some stage this allowed prey animals to enter the Cone area and gain access between the speleothem barrier to the area of the Classic Section. As most Australopithecine fragments came from this breccia, it seems likely that, as with Swartkrans, the hominid fragments are the result of predation. Then as sediments continued to wash into the Cone area, the Classic Section and the back of the Cone became inaccessible for denning purposes. Large speleothem remnants embedded in sediment starting part-way up the Cone shows that a roof continued to exist in this area for some time. In fact, even today, although bits of speleothems can be found in the eroded hardened surface sediments there are few, if any, dolomite blocks. We presume that they were removed along with the upper part of the infilled, cavern along with the country rock as the surface continued to erode. Although it may not be possible to reconstruct a complete strati-graphic sequence involving the whole of the site, we have presented summary of evidence here that shows it is possible to trace stratigraphic relationships from the base of the Ancient Entrance to the top of the Cone and which includes the Grey Breccia and the two other bone breccias. Present work is focussed on completion of a new chronology by comparing our site magnetostratigraphy with the global polarity timescale. The speleothem deposition today in any of the caves of the Makapansgat area, with its annual rainfall of about 700mm, is extremely low. It is commonly recognised by karst geomorphologists (eg, Ford and Williams, 1989) that massive deposits of speleothem are characteristic of humid tropical to semi-tropical karsts. Hence, at the time of their formation some 3 Ma or more ago, the massive speleothem deposits are evidence for a warmer, and certainly a much wetter, climate than that of today.  相似文献

Four derivatives of Rhizobium japonicum 110 were isolated on the basis of morphologically different colonies formed on yeast extract-mannitol-HM salts medium. All are able to nodulate Lee soybeans. The bacteria-plant associations formed by each clone have measurable acetylene-reducing activity, but those formed by two of these clones (designated L1-110 and L2-110) are 5- to 10-fold less efficient than those formed by the others (designated I-110 and S-110). These derivatives were not detectable with ordinary culture techniques since, because of cell adherence, genetically mixed colonies result. When a detergent (Tween 40 at 0.01%, vol/vol) was added to the dilution medium, separate clones resulted. The metabolic basis for the gross differences in colony morphology on yeast extract-mannitol-HM salts medium was found to be that L1-110 and L2-110 utilized p-mannitol for growth, whereas I-110 and S-110 did not. These clones differ analogously in ability to utilize D-arabitol. Clones I-110 and L1-110 were chosen for studies of growth rates on various carbohydrates. Although clone I-110 and clone L1-110 did not differ in growth rates on a number of sugars, such as gluconate, arabinose, glycerol, and mannose, they differed in growth rates on glucose and fructose. Although clone I-110 grew faster on glucose than did clone L1-110, clone L1-110 grew faster on fructose than did clone I-110. Clones I-110 and L1-110 showed identical responses to several antibiotics and deoxyribonucleic acid (DNA) synthesis inhibitors and identical susceptibility to some highly specific bacteriophages. Identical buoyant densities of their DNAs in isopycnic CsCl density gradients and identical thermal denaturation temperatures of their DNAs suggest that clones I-110 and L1-110 have the same DNA base composition. Preliminary DNA/DNA hybridization experiments show that strain I-110 DNA and strain L1-110 DNA have a high degree of common polynucleotide sequences.  相似文献

Reconstructing Plio-Pleistocene African paleoenvironments is important for models of early hominin evolution, but is often hampered by low-resolution or discontinuous climatic data. Here, we present high-resolution stable oxygen and carbon isotope time series data from two flowstones (secondary cave deposits) from the South African hominin-bearing Makapansgat Valley. The age of the older of the two flowstones (Collapsed Cone) is constrained by magnetostratigraphy to approximately 4-5 Ma; the younger flowstone (Buffalo Cave) grew between 2.0-1.5 Ma, as determined by magnetostratigraphy and orbital tuning of the isotopic data. The carbon isotope data is used as a proxy for the proportion of C(4) grasses in the local environment and the oxygen isotope data reflects monsoon rainfall intensity. The carbon isotope evidence indicates that in the late Miocene/early Pliocene, the local environment was dominated by C(3) vegetation, whereas, in the Plio-Pleistocene, it was composed of a mixture of C(3) and C(4) vegetation. This suggests that C(4) grasses became a significant part of the Makapansgat Valley ecosystem at approximately 4-5 Ma, towards the end of the late Neogene global expansion of C(4) grasses. After this initial expansion, South Africa experienced further fluctuations in the proportion of C(3) and C(4) vegetation during the Plio-Pleistocene, in response to regional and global climatic changes. Most notably, the Buffalo Cave flowstone provides evidence for C(4) grass expansion at ca. 1.7 Ma that we suggest was a response to African aridity caused by the onset of the Walker Circulation in the Pacific Ocean at this time.  相似文献

The study aimed to explore the suicidal process, suicidal communication and psychosocial situation of young suicide attempters in a rural community in Hanoi, Vietnam. Semi-structured interviews were conducted, in a community setting, with 19 suicide attempters aged 15-24 who had been consecutively hospitalized in an intensive care unit. In 12 of 19 cases, the first pressing, distinct and constant suicidal thoughts appeared less than one day before the suicide attempt in question. However, distress and mild, fleeting suicidal thoughts had been present up to six months before the suicide attempt in 16 cases. Five respondents had a suicide plan one to three days before attempting suicide. Altogether, 13 engaged in some form of suicidal communication before their attempt. This communication was, however, difficult for outsiders to interpret. Twelve of the respondents were victims of regular physical abuse and 16 had suffered psychological violence for at least one year before attempting suicide. Eighteen of the respondents used pesticides or raticides in their suicide attempts. None sought advice or consultation in the community despite long-standing psychosocial problems. The strategy of reducing the availability of suicide means (e.g., pesticides or raticides) in Asian countries should be complemented with a long-term suicide-preventive strategy that targets school dropouts and domestic violence, and promotes coping abilities and communication about psychological and social problems as well as recognition of signs of distress and suicidal communication.  相似文献
