A spatio-temporal study of genetic variation in the Danish pine marten ( Martes martes ) populations from the Jutland peninsula and from the island of Sealand was performed using 11 microsatellite markers. Samples obtained from 1892 to 2003 were subdivided into historical (prior to 1970) and recent (from 1970) groups. As compared with the historical samples, there was a significant loss of genetic variation in the recent Jutland population, but not in Sealand. Effective population sizes were estimated using Bayesian-based software (TMVP). Historical effective population sizes were 5897 (90% highest probability density, HPD, limits: 1502–6849) in Jutland and 1300 (90% HPD limits: 224–5929) in Sealand, whereas recent effective population sizes were 14.7 (90% HPD limits: 10.9–23.5) in Jutland and 802 (90% HPD limits: 51.8–5510) in Sealand. Significant genetic differentiation ( F ST) was found between the two historical samples, between the two recent samples, and between the historical and the recent sample in Jutland; whereas the F ST value between the historical and the recent sample in Sealand was not significant. The significant genetic differentiation between the historical and the recent samples indicates changes in the genetic compositions over time, and the higher F ST values between the two recent samples, as compared with the two historical samples, indicates that the populations in Sealand and Jutland have drifted apart within a short time span. We developed polymerase chain reaction primers for nine dinucleotide microsatellite loci in the marine deep‐sea fish, the tusk (Brosme brosme). All markers were obtained from a partial genomic DNA library, and characterized in 100 unrelated individuals from one putative population. The number of alleles ranged from two to 60 with an average of 15/locus. The observed heterozygosity ranged from 0.020 to 0.826 (average 0.438). Several of the markers amplified multiple alleles from the two other gadoid species tested.  相似文献

Insect larval characteristics, including chaetotaxy, are used widely in systematics, including for classification and phylogenetic reconstruction. Despite their common use, basic aspects of larval morphology, including intraspecific variation, effects of relatedness between individuals, sex and asymmetry, are little investigated. In the larvae of the noctuid moth Orthosia gothica, properties of shape and size were separated to examine their effects separately. Siblings were found not to cover the entire variation of a population, and therefore specimens originating from a single female do not represent independent samples. This methodological bias may potentially lead to wrong conclusions regarding species characteristics. We observed slight differences between the left and right sides of the specimens studied, implying that one side should be examined consistently in studying larval chaetotaxy. We found no differences between sexes, but this may apply only to the species examined here; in general, sex should be determined and accounted for. We discovered considerable variation in seta numbers, which further emphasizes the importance of sufficient material, particularly in cladistic analyses in which setal counts are often used as characters.  相似文献

Ploidal level information is of particular importance in intricate polyploid complexes such as in arctic-alpine Draba . Relative DNA content is reported for the tetra- and hexaploid D. lactea and seven of its low-ploid relatives. Flow cytometry was used to study 200 plants from 93 populations, the screening based on relative fluorescence. Absolute DNA content was determined by Feulgen densitometry for 13 plants from seven species, and reference chromosome numbers were determined in 12 plants (1–3 per species) representing six species. The plants grouped into diploids (2 n  = 16), tetraploids (2 n  = 32), hexaploids (2 n  = 48), and two triploids. Each ploidal level showed a linear increase in relative DNA content, pointing to a relatively recent polyploid origin. The diploid level was confirmed in D. nivalis, D. subcapitata, D. fladnizensis , and D. lonchocarpa. Draba palanderiana , reported previously as di-, tetra- and octoploid, was diploid in all investigated accessions. Hexa- and tetraploids were observed in D. lactea , in approximately the same ratio (8 : 1) as reported previously. ABSTRACT. The protist Prorocentrum lima , a primary producer of the tumour promoter okadaic acid, is a member of the dinoflagellate class of marine microorganisms. Herein, we have identified and characterized a protein tyrosine kinase (designated PLIK 1A) in P. lima that autophosphorylates almost exclusively on tyrosine residues. PLIK 1A was shown to have an approximate molecular mass of 38 kDa by SDS-PAGE and a native molecular mass within the range of 47–55 kDa by Superdex-75 gel filtration. Phosphoamino acid analysis of autophosphorylated PLIK 1A revealed the presence of phosphotyrosine and autophosphorylated PLJK 1A reacted with monoclonal anti-phosphotyrosine antibodies in a Western immunoblot. In addition, two protein tyrosine phosphatases were identified in P. lima that had apparent molecular masses within the ranges of 150–168 kDa and 73–82 kDa as determined by Superdex-200 gel filtration. These P. lima phosphatases, termed PLPTP-I and PLPTP-II, efficiently dephosphorylated tyrosine phosphorylated myelin basic protein. owever, only PLPTP-I was capable of dephosphorylating the tyrosine phosphorylated substrate angiotensin. Both PLPTP-I and PLPTP-II were able to dephosphorylate tyrosine autophosphorylated PLIK 1A. These data provide the first evidence for reversible tyrosine protein phosphorylation in P. lima by protein tyrosine kinases and phosphatases  相似文献

Orchid seeds are minute and the first seedling stages of terrestrial species are underground, so there is little knowledge about the biology of the early life history and the size of seed/seedling populations relative to the number of plants that eventually emerge above ground. A recently developed field sowing technique makes it possible to explore a variety of topics such as the length of time that seeds spend as part of the soil seed bank, the phenology of seed germination, substrate requirements for germination, dependence of germination on fungal availability, substrate requirements and growth rate of seedlings, seedling phenology and development, and seedling-fungus specificity patterns in the soil. Several of these applications are relevant for conservation issues. Studies of Goodyera pubescens, Corallorhiza odontorhiza and Liparis lilifolia show considerable variation in germination strategies, patterns of seedling development and establishment, substrate requirements, and levels of specificity with the fungi.  相似文献

The- distribution of stegmata in Orchidales is given for 130 species representing 105 genera. The stegmata occur in leaves and stems as longitudinal files of silica cells. lining fibre bundles or vascular bundle sheaths. The silica bodies contained in orchid stegmata arc of two major kinds, spherical and conical. The species investigated are characterized either by a single kind of silica body or by lack of silica cells. Absence of stegmata and presence of stegmata with conical bodies are the two most common character states in the order. The stegmata containing spherical bodies characterize Vandeae and Eriinae, Podochilinae, and Dendrobiinae within Epidendreae
Comparison of character slates of the various groups within the family is used for a discussion of evolutionary polarity. The function of silica accumulation is reconsidered in the light of the epiphytic life-form of many of the investigated orchids.  相似文献
