MicroRNAs (miRNAs) regulate gene expression and thereby influence cell development and function. Numerous studies have shown the significant roles of miRNAs in regulating immune cells including natural killer (NK) cells. However, little is known about the role of miRNAs in NK cells with aging. We previously demonstrated that the aged C57BL/6 mice have significantly decreased proportion of mature (CD27−CD11b+) NK cells compared with young mice, indicating impaired maturation of NK cells with aging. Here, we performed deep sequencing of CD27+ NK cells from young and aged mice. Profiling of the miRNome (global miRNA expression levels) revealed that 49 miRNAs displayed a twofold or greater difference in expression between young and aged NK cells. Among these, 30 miRNAs were upregulated and 19 miRNAs were downregulated in the aged NK cells. We found that the expression level of miR‐l8la‐5p was increased with the maturation of NK cells, and significantly decreased in NK cells from the aged mice. Knockdown of miR‐181a‐5p inhibited NK cell development in vitro and in vivo. Furthermore, miR‐181a‐5p is highly conserved in mice and human. MiR‐181a‐5p promoted the production of IFN‐γ and cytotoxicity in stimulated NK cells from both mice and human. Importantly, miR‐181a‐5p level markedly decreased in NK cells from PBMC of elderly people. Thus, our results demonstrated that the miRNAs profiles in NK cells change with aging, the decreased level of miR‐181a‐5p contributes to the defective NK cell development and function with aging. Manganese-enhanced MRI (MEMRI) has been described as a powerful tool to depict the architecture of neuronal circuits. In this study we investigated the potential use of in vivo MRI detection of manganese for tracing neuronal projections from the primary motor cortex (M1) in healthy marmosets (Callithrix Jacchus). We determined the optimal dose of manganese chloride (MnCl2) among 800, 400, 40 and 8nmol that led to manganese-induced hyperintensity furthest from the injection site, as specific to the corticospinal tract as possible, and that would not induce motor deficit. A commonly available 3T human clinical MRI scanner and human knee coil were used to follow hyperintensity in the corticospinal tract 24h after injection. A statistical parametric map of seven marmosets injected with the chosen dose, 8 nmol, showed the corticospinal tract and M1 connectivity with the basal ganglia, substantia nigra and thalamus. Safety was determined for the lowest dose that did not induce dexterity and grip strength deficit, and no behavioral effects could be seen in marmosets who received multiple injections of manganese one month apart. In conclusion, our study shows for the first time in marmosets, a reliable and reproducible way to perform longitudinal ME-MRI experiments to observe the integrity of the marmoset corticospinal tract on a clinical 3T MRI scanner. 相似文献

Background
Human papillomavirus (HPV) types 16 and 18 cause invasive cervical cancer and most invasive anal cancers (IACs). Overall, IAC rates are highest among men who have sex with men (MSM), especially MSM with HIV infection. Testosterone is prescribed for men showing hypogonadism and HIV-related wasting. While there are direct and indirect physiological effects of testosterone in males, its role in anal HPV16/18 infections in men is unknown.Methods
Free testosterone (FT) was measured in serum from 340 Multicenter AIDS Cohort Study (MACS) participants who were tested for anal HPV16/18-DNA approximately 36 months later. The effect of log10-transformed current FT level on anal HPV16/18 prevalence was modeled using Poisson regression with robust error variance. Multivariate models controlled for other HPV types, cumulative years of exogenous testosterone use, race, age, lifetime number of receptive anal intercourse partnerships, body mass index, tobacco smoking, HIV-infection and CD4+ T-cell counts among HIV-infected, and blood draw timing.Results
Participants were, on average, 60 (+5.4) years of age, White (86%), and HIV-uninfected (56%); Twenty-four percent tested positive for anal HPV16 and/or 18-DNA (HPV16 prevalence=17.1%, HPV18=9.1%). In adjusted analysis, each half-log10 increase of FT was associated with a 1.9-fold (95% Confidence Interval: 1.11, 3.24) higher HPV16/18 prevalence. Additionally, other Group 1 high-risk HPVs were associated with a 1.56-fold (1.03, 2.37) higher HPV16/18 prevalence. Traditional risk factors for HPV16/18 infection (age, tobacco smoking; lifetime number of sexual partners, including the number of receptive anal intercourse partnerships within 24 months preceding HPV testing) were poorly correlated with one another and not statistically significantly associated with higher prevalence of HPV16/18 infection in unadjusted and adjusted analyses.Conclusions
Higher free testosterone was associated with increased HPV16/18 prevalence measured approximately three years later, independent of sexual behavior and other potential confounders. The mechanisms underlying this association remain unclear and warrant further study. 相似文献6.

The effect of phosphate on the surface charge and cadmium (Cd) adsorption was examined in seven soils that varied in their variable-charge components. The effect of phosphate on immobilization and phytoavailability of Cd from one of the soils, treated with various levels of Cd (0–10 mg Cd kg–1 soil), was further evaluated using mustard (Brassica juncea L.) plants. Cadmium immobilization in soil was evaluated by a chemical fractionation scheme. Addition of phosphate, as KH2PO4, increased the pH, negative charge and Cd adsorption by the soils. Of the seven soils examined, the three allophanic soils (i.e., Egmont, Patua and Ramiha) exhibited greater increases in phosphate-induced pH, negative charge and Cd2+ adsorption over the other four non-allophanic soils (i.e., Ballantrae, Foxton, Manawatu ad Tokomaru). Increasing addition of Cd enhanced Cd concentration in plants, resulting in decreased plant growth (i.e., phytotoxicity). Addition of phosphate effectively reduced the phytotoxicity of Cd. There was a significant inverse relationship between dry matter yield and Cd concentration in soil solution. Addition of phosphate decreased the concentration of the soluble + exchangeable Cd fraction but increased the concentration of inorganic-bound Cd fraction in soil. The phosphate-induced alleviation of Cd phytotoxicity can be attributed primarily to Cd immobilization due to increases in pH and surface charge. 相似文献

We examined the effect of biosolid compost on the adsorption and complexation of cadmium (Cd) in two soils (Egmont and Manawatu) which varied in their organic matter content. The effect of biosolid compost on the uptake of Cd from the Manawatu soil, treated with various levels of Cd (0–10 mg Cd kg–1 soil), was also examined using mustard (Brassica juncea L.) plants. The transformation of Cd in soil was evaluated by a chemical fractionation scheme. Addition of biosolid compost increased negative charge in soil. The effect of biosolid compost on Cd adsorption varied between the soils, with a large portion of the sorbed Cd remaining in solution as an organic complex. Increasing addition of Cd increased Cd concentration in plants, resulting in decreased plant growth at high levels of Cd (i.e., phytotoxicity). Addition of biosolid compost was effective in reducing the phytotoxicity of Cd as indicated by the decrease in the concentration of NH4OAc extractable-Cd and soil solution-Cd. The solid-phase fractionation study indicated that the addition of biosolid compost decreased the concentration of the soluble and exchangeable Cd fraction but increased the concentration of organic-bound Cd fraction in soil. Alleviation of Cd phytotoxicity by biosolid compost can be attributed primarily to complexation of Cd by the organic matter in the biosolid compost. 相似文献

The new sECM biomaterials have been successfully used to perform 3D cell culture, drug and growth factor release, drug toxicity testing, and to develop a new anticancer drug evaluation model. The ready availability of these materials should facilitate progress in understanding regulation of cellular physiology as influenced by endogenous signals or exogenous pharmaceutical agents in 3-D tissue-like cell cultures. Importantly, the use of primary hepatocytes and soon, human liver stem cells, cultured in Extracel™ will expedite drug toxicity testing in vitro and in vivo. In addition, the application of the orthotopic engineered tumor xenograft model using Extracel™-containing tumor cells in nude mice should improve the selection of new anticancer agents that will show clinical efficacy in cancer patients. 相似文献

Microsporidia are believed to play little or no role in outbreaks of the jack pine budworm, Choristoneura pinuspinus Freeman (Lepidoptrera: Tortricidae), because the short duration (2–4 years) of those outbreaks may not permit significant build-up of the pathogen. We conducted the first survey of Nosema sp. (Microsporidia: Nosematidae) over the course of a recent jack pine budworm outbreak in Ontario. Between 2004 and 2010 the outbreak defoliated a cumulative total of 1.78 million ha. Microscopic examination of ∼15,000 overwintering larvae collected over 6 years in sites with densities of 3 larvae per branch or more revealed widespread occurrence of Nosema at generally high infection intensities. The pathogen was present in 69.5% of the 518 plots that were monitored. Prevalence of infection was generally low (below 40% in 84% of plots with infected larvae) but reached high levels (80–95%) locally and increased rapidly in most infestations within 1–2 years of onset. We hypothesize that the habit of early-instar larvae to feed on developing male flowers (pollen cones) after spring emergence is critical in allowing rapid build-up of Nosema by increasing efficiency of horizontal transmission (higher density of both infected larvae and egested spores). Nosema infection may contribute to the complexity of jack pine budworm outbreak patterns by affecting egg recruitment and early-instar survival at the stand level in concert with known effects of budworm-induced reductions in pollen cone production on those processes. 相似文献
