不同底物与固氮酶及其钼铁蛋白相互作用的荧光光谱分析

 本文研究了不同底物(N_2,H_2,N_2O,NaN_3,C_2H_2)对棕色固氮菌固氮酶及其钼铁蛋白荧光光谱的影响。结果表明,上述底物均能络合在钼铁蛋白及固氮酶上,但络合程度不同,从而为固氮酶系统有多个不同的底物络合中心,底物络合中心在钼铁蛋白分子上,铁蛋白对钼铁蛋白有变构作用,提供了光谱学证据。

Expression and characteristic of synthetic human epidermal growth factor (hEGF) in transgenic tobacco plants

To improve the accumulation of recombinant human epidermal growth factor (hEGF) in transgenic tobacco, a highly effective vector was constructed and transformed via Agrobacterium tumefaciens. The hEGF content in transgenic tobacco was up to 0.3% of the total soluble protein. Using the Vero E6 cell expansion assay and the MTT method for cell proliferation, hEGF produced by transgenic tobacco significantly stimulated Vero E6 cell expansion and proliferation, the same as commercial hEGF products.

节能型实验动物屏障设施的新型设计及应用

随着国内、外实验动物的发展,屏障设施的建立和应用越来越广泛。因此,许多单位投入大量资金搞硬件建设,但对设施运行后的节能考虑较少。目前,国内屏障设施的设计多采用分区控制、变频风机、换热器等来考虑节能,但在送、回风形式上并未改变传统的顶送风、四角回风形式,该送、回风形式由于受到平板饲养架的限制,设计上房间的换气次数多为15-20次/h,实际上动物饲养笼内的换气次数远远低于房间的换气次数,不能有效的保证笼内换气。本作者根据国内、外实验动物设施发展趋势,创新研制出定向流系统（已获得实用新型专利证书,专利号：ZL200520069682.8）饲养动物,同时采用动物饲养系统和房间送、回系统分开控制进行节能设计,有效的保证动物笼内换气和节能。该设计为国内首创,应用本设计夏季可节电47.4 kW.h,按工业用电0.8元/h计算,每小时节约运行成本近38元。     

玉米面水分含量对谷蠹生长发育和繁殖的影响

本文研究了玉米面的水分含量(21.33%、19.35%、18.67%、13.64%、10.98%)对谷蠹Rhyzopertha dominica Fabricius生长发育及产卵的影响。结果表明:玉米面的水分含量对谷蠹幼虫的发育历期影响显著,对卵和蛹的发育历期影响不显著;谷蠹卵的孵化率、幼虫和蛹的存活率在玉米面水分10.98%～19.35%的条件下增大,在玉米面水分21.33%条件下下降;谷蠹幼虫龄数也受到玉米面水分含量的影响,玉米面水分19.35%条件下幼虫龄数在3～5间变化,10.98%条件下幼虫龄数在3～8间变化;在试验的5种玉米面水分中,谷蠹只在19.35%和18.67%条件下才产卵,19.35%条件下日均产卵量和产卵期均大于18.67%条件下的相应值。     

Association between Dietary Fiber Intake and Physical Performance in Older Adults: A Nationwide Study in Taiwan

Background

Physical performance is a major determinant of health in older adults, and is related to lifestyle factors. Dietary fiber has multiple health benefits. It remains unclear whether fiber intake is independently linked to superior physical performance. We aimed to assess the association between dietary fiber and physical performance in older adults.

Methods

This was a cross-sectional study conducted with community-dwelling adults aged 55 years and older (n=2680) from the ongoing Healthy Aging Longitudinal Study (HALST) in Taiwan 2008-2010. Daily dietary fiber intake was assessed using a validated food frequency questionnaire. Physical performance was determined objectively by measuring gait speed, 6-minute walk distance, timed “up and go” (TUG), summary performance score, hand grip strength.

Results

Adjusting for all potential confounders, participants with higher fiber intake had significantly faster gait speed, longer 6-minute walk distance, faster TUG, higher summary performance score, and higher hand grip strength (all P <.05). Comparing with the highest quartile of fiber intake, the lowest quartile of fiber intake was significantly associated with the lowest sex-specific quartile of gait speed (adjusted OR, 2.18 in men [95% CI, 1.33-3.55] and 3.65 in women [95% CI, 2.20-6.05]), 6-minute walk distance (OR, 2.40 in men [95% CI, 1.38-4.17] and 4.32 in women [95% CI, 2.37-7.89]), TUG (OR, 2.42 in men [95% CI, 1.43-4.12] and 3.27 in women [95% CI, 1.94-5.52]), summary performance score (OR, 2.12 in men [95% CI, 1.19-3.78] and 5.47 in women [95% CI, 3.20-9.35]), and hand grip strength (OR, 2.64 in men [95% CI, 1.61-4.32] and 4.43 in women [95% CI, 2.62-7.50]).

Conclusions

Dietary fiber intake was independently associated with better physical performance.     

Effect of anions on cadmium(II) complexes with ligand 2-(pyrazin-2-yl)-1H-benzimidazole

Three new supramolecular complexes based on a 2-(pyrazin-2-yl)-1H-benzimidazole (Hpbi) and a series of Cd(II) salts have been solvothermally synthesized and structurally characterized by single-crystal X-ray diffraction analysis. Reaction of CdCl₂·2.5H₂O with Hpbi afforded a one-dimensional chain [Cd(Hpbi)Cl₂] (1), which exhibits a three-dimensional (3-D) supramolecular architecture through intermolecular X-H···Cl (X = N and C) hydrogen bonds and π-π stacking interactions. When using CdBr₂·4H₂O instead of CdCl₂·2.5H₂O under similar reaction conditions, a bisnuclear complex [Cd(Hpbi)₂Br₂] (2) is obtained, which obviously exhibits intermolecular X-H···Br (X = N and C) hydrogen bonds and π-π stacking interactions. When CdI₂ take place of CdCl₂·2.5H₂O, a mononuclear complex, [Cd(Hpbi)₂I₂] (3), is isolated, which shows a 3D supramolecule framework formed by intermolecule hydrogen bonds and π-π packing interactions. Interestingly, the Hpbi ligand exhibits the same coordination modes in complexes 1-3. It is noteworthy that the radius of anions plays an important role in affecting the structures and luminescent intensity of the final products. The TGA for 1-3 have been investigated and discussed in detail.     

Transcriptional activation of endoplasmic reticulum chaperone GRP78 by HCMV IE1-72 protein

Glucose-regulated protein 78 (GRP78), a key regulator of endoplasmic reticulum (ER) stress, facilitates cancer cell growth and viral replication. The mechanism leading to grp78 gene activation during viral infection is largely unknown. In this study, we show that the immediate-early 1 (IE1-72) protein of the human cytomegalovirus (HCMV) is essential for HCMV-mediated GRP78 activation. IE1-72 upregulated grp78 gene expression depending on the ATP-binding site, the zinc-finger domain and the putative leucine-zipper motif of IE1-72, as well as the ER stress response elements (ERSEs) on the grp78 promoter. The purified IE1-72 protein bound to the CCAAT box within ERSE in vitro, whereas deletion mutants of IE1-72 deficient in grp78 promoter stimulation failed to do so. Moreover, IE1-72 binding to the grp78 promoter in infected cells accompanied the recruitment of TATA box-binding protein-associated factor 1 (TAF1), a histone acetyltransferase, and the increased level of acetylated histone H4, an indicator of active-state chromatin. These results provide evidence that HCMV IE1-72 activates grp78 gene expression through direct promoter binding and modulation of the local chromatin structure, indicating an active viral mechanism of cellular chaperone induction for viral growth.     

双叉犀金龟成虫最佳食料筛选

在人工气候室条件下(26℃±1℃,RH 70%±5%,L∶D=14 h∶10 h),研究了6种水果对双叉犀金龟Allomyrina dichotoma成虫寿命以及子代数量的影响。结果显示,各处理雌虫寿命均显著长于雄虫,雌虫寿命46.67～55.08 d,雄虫寿命23.78～33.33 d,各处理间无显著差异;饲喂西瓜Citrullus lanatus(Thunb.) Matsum.etNakai子代数量最多,达39.44头/雌,与香蕉Musa nanaLour.、苹果Malus pumilaMill.和商品果冻处理无显著差异,饲喂葡萄Vitis viniferaL.和香瓜Cucumis meloL.处理均显著低于这4个处理;选择子代数量最多的西瓜、苹果、香蕉分别制作人工饲料果冻,用商品果冻作为对照,在15 d和35 d分别2次调查每对成虫的子代数,结果显示混合果冻、商品果冻、苹果果冻和西瓜果冻显著高于香蕉果冻,且各果冻处理在0～15 d和16～35 d的子代数量差异不同;综合比较两年水果饲喂和人工饲料果冻饲喂结果,饲喂混合果冻、商品果冻、西瓜果冻和苹果果冻4个处理的子代数量最高,平均达...
